Genetic engineering of stone fruit species like apricot, plum, peach and cherry are hampered by the inefficient and low-level regeneration processes in tissue culture. The first transgenic stone fruit species have emerged from transformed hypocotyls. These great achievements were applauded by the scientific community contrary the fact that hypocotyl derived transgenic plants have no real brooding value. Tissue culture of different organs of valuable cultivars are recorded with an extremely low-level of regeneration in the literature. To improve the tissue culture basis of stone fruit plants an extensive tissue culture programme were launched and dozens of different media were compared including a series of hormone concentration in the tissue culture systems. Our continuous efforts were crowned by a very efficient method for achieving up to 30-40% regenerable petioles. Usually on a single petiole several well-separated meristems were induced. After 3-4 weeks of cultivation shoots were developed. The basic media $K_2$ were supplemented with 10g/L saccharose, 10g/L glucose and 10g/L maltose. The following plant hormones were used BAP 1mg/L, TDZ 1mg/L, 2-iP 1mg/L and IAA 0,1 mg/L concentrations. The Petri dishes were kept for 3 weeks in dark at a temperature 22$^{\circ}C$ for 8 hours and 22-24$^{\circ}C$ for 16 hours. The Petri dishes were sealed with Parafilm. The regeneration of the petioles were genotype independent and we were able to regenerate different plum cultivars with almost the same efficiency.
Interleukin-11 (IL-11) is a cytokine that plays a key regulatory role in the immune system. Recombinant human IL-11 (rhIL-11) exerts a preventative effect against apoptotic cell death and inhibits preadipocyte differentiation. IL-11 also is used to stimulate the bone marrow to produce platelets in order to prevent low platelets that may be caused by chemotherapy. Unfortunately, the high production cost of IL-11 associated. In this study, we investigated the feasibility of transgenic plants for the cost-effective production of rhIL-11. Production of rhIL-11 proteins in whole-plant expression system will be more economical when compared to the current E. coli based expression system. The human rhIL-11 gene was codon optimized to maximize plant host system expression. IL-11 expression vector under the control of a constitutive cauliflower mosaic virus 35S (CaMV 35S) promoter was introduced into tobacco by Agrobacterium-mediated transformation. The 5'-leader sequence (called ${\Omega}$) of tobacco mosaic virus (TMV) as a translational enhancer was added to construct. Transgenic tobacco plants expressing various levels of rhIL-11 protein were generated. Western blotting of the stably transformed lines demonstrated accumulation of the appropriately sized rhIL-11 protein in leaves. This research demonstrated the efficacy of using tobacco as an expression system for the production of rhIL-11.

TMV resistant lines (TRLs) originated from the Blo plant of Nicotiana tabacum cv. NC82 transformed with TMV coat protein cDNA which initially showed delayed disease symptom were selected for increased resistance in each subsequent generation. The result of field experiment of the transgenic tobacco lines in the fifth generation for TMV resistance and their response to other tobacco diseases (black shank, bacterial wilt, and powdery mildew) is described in this report. When fifteen TRLs of the fifth generation were tested for TMV resistance by mechanically inoculating the individual plants, over 95 percent of the plants of 6 lines showed complete resistance even 8 weeks after the inoculation. Average frequency of the resistant plants in TRLs of the fifth generation 8 weeks after the inoculation was 87%. Stable insertion and expression of TMV coat protein cDNA in the fifth generation of the transgenic tobacco plant were confirmed by PCR and immunoblot hybridization, respectively. All TRLs were resistant to the black shank but were susceptible to the bacterial wilt disease and the powdery mildew to the same degree as non-transgenic NC82 was. Therefore, it was indicated that the phenotypes related at least to disease resistance were not changed in the transgenic tobacco. The promoters of OsCaM1 and OsCaM3 were characterized after sequencing and fused to the reporter gene, GUS. The constructs were then transformed into the tobacco plant. Histochemical analysis of GUS showed different expression patterns in pOsCaM1::GUS and pOsCaM3:: GUS transgenic plants. The expression of pOsCaM1::GUS in 4- to 15-day-old seedlings in particular was observed only in the root, while the expression of pOsCaM3::GUS was detected in both the cotyledons and root. Also, pRCaM1::GUS was detected in all the tissues surrounding the root system, while the presence of pOsCaM3::GUS was observed in the root, except in the root meristem. However, in mature transgenic plants, the expression of pOsCaM1::GUS and OsRCaM3::GUS was scarcely detected. Under wounding stress, the GUS activity of pOsCaM1 and pOsCaM3 was strongly induced, and the activity of pOsCaM3 especially, was retained for long periods. In the phloem, pOsCaM3 activity induced by hormone treatments and abiotic stresses was also identified.
1990년 8월 충북 보은의 참깨 밭에서 전형적인 엽화병(葉化病; phyllody disease)이 발생하였다. 병징은 꽃이 잎처럼 녹색으로 변하고, 이병엽은 정상엽보다 작고 잔가지가 마치 빗자루 모양으로 총생하였고 개화 및 결실이 되지 않았다. 투과 전자현미경으로 병든 잎의 사관요소에서 phytoplasma가 존재하는 것을 확인하였다. 병든 잔가지에서 추출한 DNA를 중합효소 연쇄반응으로 분석한 결과 대추나무 빗자루병에 걸린 나무에서 증폭되는 DNA와 같은 크기의 band가 관찰되었다. 따라서 위의 시료는 phytoplasma 벙으로 확인 되었으며 참깨 엽화병으로 명명하였다. 또한 참깨 엽화병을 일으키는 phytoplasma와 대추 나무 빗자루병 phytoplasma를 PCR 증폭 및 제한효소로 절단하여 분석한 결과 이들은 서로 다른 그룹이라는 것을 알 수 있었다.
We previously identified and characterized a predominantly pollen-expressed gene of Petunia inflata that encodes a receptor-like kinase named PRK1. The extracellular domain of PRK1 contains leucine-rich repeats which have been implicated in protein-protein interactions, and the cytoplasmic domain was found to autophosphorylate on serine and tyrosine. To investigate the function PRK1 in pollen development, we transformed P. inflata plants with a construct containing the promoter of a predominantly pollen-expressed gene of tomato, LAT52, fused to an antisense PRK1 cDNA corresponding to part of the extracellular domain of PRK1, There transgenic plants were found to each produce approximately equal amounts of normal and aborted pollen. Analysis of the inheritance of the transgene inserts in two of the transgenic plants, ASRK-13 and ASRK-20, to their progeny revealed that certain transgene inserts cosegregated with the pollen abortion phenotype. Microscopic examination of the aborted pollen grains showed that their outer wall, the exine, was essentially normal, but that their cytoplasm contained only starch-like granules. Staining of the nuclei of the microspores at different stages of uninucleate stage. However, at subsequent stages half of the microspores completed mitosis and developed into normal binucleate pollen, but the other half initially remained uninucleate, then lost their nucleio. Analysis of the amounts of PRK1 mRNA and the antisense PRK1 transcript suggested that the pollen abortion phenotype most likely resulted from down-regulation of the PRK1 gene by the antisense PRK1 transgene. These results suggest that PRK1 plays an essential role in a signal transduction pathway that mediates post-meiotic development of microspores.

A cDNA encoding ${\gamma}-tocopherol$ methyltransferase (${\gamma}-TMT$) from Arabidopsis thaliana was overexpressed in lettuce (Latuca sativa L.) to improve the tocopherol composition. Seven lines of lettuce ($T_0$) containing the ${\gamma}-TMT$ transgene were produced by Agrobacterium-mediated transformation. The inheritance and expression of the transgene were confirmed by DNA and RNA gel blot analyses as well as quantification of tocopherols and ${\gamma}-TMT$ activities. The ratio of ${\alpha}-/{\gamma}-tocopherol$ content (TR) varied from 0.6 to 1.2 in non-transformed plants, while the $T_0$ plants had ratios of 0.8 to 320. The ratio ranged from 0.4 to 544 in 41 $T_1$ progenies of the $T_0$ transgenic line gTM3, and the phenotypic segregation indicated monogenic inheritance of the transgene (i.e., 3:1 = dominant:wild-type classes). There was a tight relationship between the TR phenotype and ${\gamma}-TMT$ activity, and enzyme activities were affected by the copy number and transcript levels of the transgene. The TR phenotype was stably expressed in $T_2$ progenies of $T_1$ plants. The results from this study indicated that a stable inheritance and expression of Arabidopsis ${\gamma}-TMT$ transgene in lettuce results in a higher enzyme activity and the conversion of the ${\gamma}-tocopherol$ pool to ${\alpha}-tocopherol$ in transgenic lettuce.

The practicability of transgenic tobacco engineered to express bacterial native mercuric reductase (MerA), responsible for the transport of $Hg^{2+}$ ions into the cell and their reduction to elemental mercury ($Hg^0$), without any codon modification, for phytoremediation of mercury pollution was evaluated. Transgenic tobacco plants reduce mercury ions to the metallic form; take up metallic mercury through their roots; and evolve the less toxic elemental mercury. Transformed tobacco produced a large amount of merA protein in leaves and showed a relatively higher resistance phenotype to $HgCl_2$ than wild type. Results suggest that the integrated merA gene, encoding mercuric reductase, a key enzyme of the bacterial mer operon, was stably integrated into the tobacco genome and translated to active MerA, which catalyzes the bioconversion of toxic $Hg^{2+}$ to the least toxic elemental $Hg^0$, and suggest that MerA is capable of reducing the $Hg^{2+}$, probably via NADPH as an electron donor. The transgenic tobacco expressing merA volatilized significantly more mercury than wild-type plants. This is first time we are reporting the expression of a bacterial native merA gene via the nuclear genome of Nicotiana tabacum, and enhanced mercury volatilization from tobacco transgenics. The study clearly indicates that transgenic tobacco plants are reasonable candidates for the remediation of mercurycontaminated areas.
OsDREB1D, a special DREB (dehydration responsive element binding protein) homologous gene, whose transcripts cannot be detected in rice (Oryza sativa L), either with or without stress treatments, was amplified from the rice genome DNA. The yeast one-hybrid assay revealed that OsDREB1D was able to form a complex with the dehydration responsive element/C-repeat motif. It can also bind with a sequence of LTRE (low temperature responsive element). To analyze the function of OsDREB1D, the gene was transformed and over-expressed in Arabidopsis thaliana cv. Columbia. Results indicated that the over-expression of OsDREB1D conferred cold and high-salt tolerance in transgenic plants, and that transgenic plants were also insensitive to ABA (abscisic acid). From these data, we deduced that this OsDREB1D gene functions similarly as other DREB transcription factors. The expression of OsDREB1D in rice may be controlled by a special mechanism for the redundancy of function.
벼에서 분리한 저분자량 HSP 유전자 (OsHSP17.9)를 오차드그래스 (orchardgrass)에 도입하기 위하여 Agrobacterium을 이용한 형질전환을 실시하여 다음의 결과를 얻었다. 오차드그래스의 종자유래의 캘러스를 OsHSP17.9 유전자가 도입된 Agrobacterium EHA101과 공동배양한 다음, hygromycin 선발된 캘러스로부터 hygromycin 저항성 식물체를 얻었다. PCR 및 Southern blot 분석 결과, 벼의 저분자량 HSP 유전자가 재분화된 식물체에 안정적으로 도입되었음을 확인하였으며, 품종 간의 형질전환 효율은 '포토맥'의 경우 16.5%, '프론티어' 의 경우 8.0%를 나타내었다. 또한 Northern blot 분석 결과, 도입된 유전자가 형질전환체에서 정상적으로 발현된다는 것을 확인하였으며, 형질전환체의 계통 간에 발현량의 차이를 나타내었다.
