The Study of Trnascriptional Regulated Gene, $hrp^{2+}$, in Yeast

  • This study was designed to clone the SNF2/SW12 helicase-related genes from the fission yeast Schizosaccha-romyces pombe and thereafter to elucidate the common functions of the proteins in this family. The $hrp^{2+}$gene was cloned by polymerase chain reaction amplification using degenerative primers from conserved SNF2 motifs within the ERCC6 gene, which encodes a protein involved in DNA excision repair. Like other SNF2/SW12 family proteins, the deduced amino acid sequence of Hrp2 contains DNA-dependent ATPase/7 helicase domains as well as the chromodomain and the DNA binding domain. This configuration is similar to that of mCHD1 (mouse chromo-ATPase/helicase-DNA-dinding protein 1), suggesting that Hrp2 is a S. pombe homolog of mCHD1, which is thought to function in altering the chromatin structure to control the gene expression. To characterize the function of Hrp2, 4 Uracil-Hrp2 fusion protein, it was purified near homogeneity by affinity chromatography on $Ni^{2+}$-NTA agarose, DEAE-Sepharose ion exchange arid Sephacryl S-200 gel filtration chromatographies. The purified fusion protein exhibited DNA-dependent ATPase activity, which was stimulated by both double-stranded and single-stranded DNA. To determine the steady-state level of $hrp^{2+}$ transcripts during growth, cells were cultured in medium and collected at every 2hr to prepare total RNAs. The northern blot analysis showed that the level of $hrp^{2+}$ transcripts reached its maximum before the cells entered the exponential growth phase and then decreased gradually, This result implies that Hrp2 may be required at early stages of cell growth.h.

Thermal impacts on transcriptome of Pectoralis major muscle collected from commercial broilers, Thai native chickens and its crossbreeds

  • Objective: The main objective of this study was to define molecular mechanisms associated with thermal stress responses of chickens from commercial broilers (BR, Ross 308), Thai native chickens (NT) and crossbreeds between BR×NT (H75). Methods: Twenty days before reaching specific market age, chickens from each breed were divided into control and thermal-stressed groups. The stressed groups were exposed to a cyclic thermal challenge (35℃±1℃ for 6 h, followed by 26℃±1℃ for 18 h) for 20 days. Control group was raised under a constant temperature of 26℃±1℃. Pectoralis major (n = 4) from each group was collected for transcriptome analysis using HiSeq Illumina and analysis of glycogen and lactate. Gene expression patterns between control and thermal-stressed groups were compared within the same breeds. Results: Differentially expressed transcripts of 65, 59, and 246 transcripts for BR, NT, and H75, respectively, were revealed by RNA-Seq and recognized by Kyoto encyclopedia of genes and genomes database. Pathway analysis underlined altered glucose homeostasis and protein metabolisms in all breeds. The signals centered around phosphatidylinositol 3-kinase (PI3K)/Akt signaling, focal adhesion, and MAPK signaling in all breeds with slight differences in molecular signal transduction patterns among the breeds. An extensive apoptosis was underlined for BR. Roles of AMPK, MAPK signaling and regulation of actin cytoskeleton in adaptive response were suggested for H75 and NT chickens. Lower glycogen content was observed in the breast muscles of BR and NT (p<0.01) compared to their control counterparts. Only BR muscle exhibited increased lactate (p<0.01) upon exposure to the stress. Conclusion: The results provided a better comprehension regarding the associated biological pathways in response to the cyclic thermal stress in each breed and in chickens with different growth rates.

NA-Seq를 이용한 제주산 메밀의 발아초기 전사체 프로파일 분석 (Transcriptomic Profile Analysis of Jeju Buckwheat using RNA-Seq Data)

  • 본 연구에서는 메밀의 발아초기에 발현되는 전사체의 다양한 정보 수집을 위해 양절메밀과 대관 3-3호의 RNA를 추출하여 전사체 분석을 수행하였다. 제주산 양절메밀과 대관3-3호의 종자 및 발아 후 12, 24, 36시간별로 total RNA를 추출하고, llumina Hiseq 2000 플랫폼을 사용하여 시퀀싱 하였다. SolexaQA package의 DynamicTrim과 LengthsORT 프로그램으로 이용하여 raw 데이터 분석을 실시한 후, 어셈블리(assembly)와 annotation을 수행하였다. RNA-seq raw 데이터로부터 약 84.2%, 81.5%에 해당하는 16.5Gb, 16.2Gb의 transcriptome 데이터를 확보하였다. 47Mb에 해당하는 43,494개의 대표적인 전사체(representative transcripts)를 확보하였고, 그 중에서 annotation DB와 서열 유사도를 갖는 서열은 23,165개로 확인되었다. 메밀의 representative transcripts 유전자의 유전자 온톨로지(gene ontology) 분석결과, biological process는 metabolic process (49.49%)에서, cellular components는 cell (46.12%)에서, molecular function은 catalyltic activity (80.43%)에서 유전자가 많이 분포되어 있는 것을 확인하였다. 종자의 발아에 관련된 gibberellin receptor GID1C의 경우에는 양절메밀, 대관 3-3호의 발현양이 모두 시간이 지남에 따라 증가되는 것을 확인할 수 있었으며, gibberellin 20-oxidase1의 경우에는 양절메밀에서는 발아 후 12 시간이내에 증가되었으나, 대관 3-3호에서는 36시간까지 유전자 발현양 증가하는 것을 확인할 수 있었다. 이러한 제주산 메밀의 발아초기 단계별 전사체 분석 데이터는 종간의 기능적, 형태학적 차이를 일으키는 메커니즘 규명에 도움을 줄 것으로 사료된다.

흰쥐 부정소에서의 세포자연사에 미치는 Ethane 1,2-Dimethane Sulfonate(EDS)의 효과 (Effect of Ethane 1,2-Dimethane Sulfonate(EDS) on the Apoptosis in the Rat Epididymis)

  • Ethane 1,2-Dimethane sulfonate(EDS)은 Leydig cells(LC)만을 선별적 사멸을 유도하는 약물로서 가역적인 테스토스테론 결핍 흰쥐 모델을 만드는데 널리 사용된다. EDS 투여에 의해 유도된 'LC 녹아웃' 흰쥐의 경우 부정소와 저정낭과 같은 테스토스테론 의존성 부속 생식기관들의 급격한 무게 감소가 초래됨이 이전의 연구들에서 보고되었는데, 이러한 무게 감소의 상당 부분은 세포자연사에 의한 것으로 보인다. 본 연구는 흰쥐 부정소에서 세포자연사 관련 유전자들의 발현에 미치는 EDS 투여 효과를 조사한 것이다. 성숙한 수컷 흰쥐(SD strain, $300{\sim}350\;g\;B.W.$)에 EDS(75 mg/kg, i.p.)를 1회 복강주사하고 주사 후 0, 1, 2, 3, 4, 5, 6 그리고 7주가 경과한 날 희생시켰다. 희생 직후 조직 무게와 부정소 미부의 정자수를 측정하였다. 부정소로부터 total RNA를 추출한 후 세포자연사 관련 유전자들 가운데 bcl-2, bax, Fas 그리고 Fas ligand(Fas-L)의 발현 변화를 semi-quantitative RT-PCR로 측정하였다. 예상한 바처럼, 부정소 무게와 정자 수는 EDS 주사 후 $1{\sim}2$주 동안에 급격히 감소하였다. 이후 어느 정도 회복하였지만, 최종적으로 주사 후 7주경 부정소 무게(71%)와 정자 수(38%) 모두 최초 수준에는 미달하였다. 부정소에서의 bcl-2 전사 수준은 주사 직후부터 6주 후까지 지속되다가 주사 후 7주에 유의하게 상승하였다. 또한 bax 전사 수준은 주사 후 6주에 유의하게 감소하였으며 나머지 전 기간 동안에는 별다른 차이가 없었다. 한편 Fas 전사 수준은 EDS 주사 후 $1{\sim}2$ 주간 상승하였다가 주사 후 3주부터 정상 수준으로 감소하여 7주까지 지속되었다. 유사하게, Fas-L의 전사 수준도 주사 후 $1{\sim}3$주 동안 상승하였다가 주사 후 4주부터 정상 수준으로 복귀하였다. 본 연구의 결과는 EDS 주사가 흰쥐 부정소에서의 세포자연사 관련 유전자 발현을 유도할 가능성을 보여준 것이며, 특히 Fas와 Fas-L 유전자 활성이 초기 세포자연사 유도 과정에 중요하고, 그 결과로 부정소 무게 감소와 정자 수 감소가 초래되는 것으로 추정된다.

반성적 사고 중심의 모자 상호작용 증진 프로그램 효과 연구 : 발달지체유아 모자를 중심으로 (The Effects of an Intervention Program for Mother-child Interaction Behaviors Based on Reflective Thinking for Developmentally Delayed Young Children and their Mothers)

  • The study examined the effects of an intervention program based on reflective thinking on mother-child interaction behaviors. Three mothers with problematic mother-child interactions and their 4-year-old socially delayed boys participated in the program that consisted of four main activities like journal and video feedback, lectures on interactive strategies, mother-child interactive play and program evaluations and ran for twenty 120 minutes weekly sessions. The level of reflective thought was determined by analyzing interview transcripts, journal entries. The videotaped mother-child interaction behaviors were rated by the Maternal Behavior Rating Scale and Child Behavior Rating Scale. It was concluded that the program enhanced mother's reflection on mother-child interactions and changed interaction behaviors of 3 mother-child pairs positively.

Molecular cloning of cDNAs encoding antioxidant enzymes in Korean rock bream (Oplegnathus fasciatus)

  • Antioxidant enzyme genes play a key role in cell defense against the lethal effects of oxidative stresses in animals and have an essential function which has allowed the evolution of aerobic respiration starting from an ancient form of oxygen-insensitive life. Piscine antioxidant enzymes are also involved in the rapid response to various toxic chemicals as well as many biological stresses, indicating that they could be used as biomarkers for health and aquatic environment. With the purpose for developing fine molecular probing tool to assess the stresses in marine fish, we identified three major antioxidant enzyme genes (superoxide dismutase, catalase and glutathione-S-transferase) from Korean rock bream using expressed sequence tag analysis and/or high density filter screening. Here we report the molecular information on these gene transcripts including complete sequence data and expression profiles.

감각통합치료의 역사적 고찰 (Historical Review of Sensory Integration in Korea)

  • The significant historical events and developments in the area of approach for sensory integration in Korean Society of Sensory Integration Therapists(KSSIT) are reviewed to stimulate future planning and clinical application in professional and health promotion. The paper reviewed the purposes of establishment, the role of members, progress of regular meetings and education for members and seminars in KSSIT. This review is placed within the context of the progressive period of KSSIT history, 1997-2003. Historical data are used as primary sources as records of events in KSSIT's homepage, official transcripts, professional articles, and secondary sources such as news letters for occupational therapists and oral information from first members. This review examined current actions and thinking, and suggests further actions and thinking for progressive professional education programs, research, clinical applications and public relations in sensory integration approach in KSSIT.

Assembled and Annotated Genome of Plasmodiophora brassicae with Insights into Developmental Stage-Specific

  • Plasmodiophora brassicae is a major disease threat for Brassica oil and vegetable crop production worldwide. The causal agent is a Plasmodiophorid, which are obligate biotrophic plant-pathogenic protists in the Rhizarian kingdom. Although the Plasmodiophorids include other important agricultural pathogens such as Polymyxa betae, Spongospora subterranea, their biology remains poorly understood due to their intracellular biotrophic life style. I will present the assembled and annotated genome of P. brassicae, with insights into developmental stage-specific. We provide the first genomic data for pathogenic Rhizaria. The exploitation of the life stage specific transcripts will shed light in the understanding of the life cycle at a molecular basis, which will in the long run help to understand and control club root disease. Our data also fill an important gap for the understanding of the eukaryotic tree of life, since this is only the third genome of the eukaryotic kingdom of Rhizaria.

상호작용 읽기전략을 통해서 본 한국 대학생들의 독해과정에 관한 연구 (Investigating English reading processes of Korean college students through reciprocal reading strategy)

  • The purpose of the present study was to investigate the effectiveness of reciprocal teaching procedure for improving Korean college students' reading comprehension of English text. In particular, this study sought to explore the qualitative features, if any, in students' use of reading comprehension strategies presented in the process of Reciprocal Reading Procedure (RRP). In order to accomplish the goal of the study, transcripts of the students' dialogues, open-ended questionnaires, and researchers' observation notes were examined. The results of the study showed that the participants used different four kinds of reading strategies in the process of RRP (questioning, clarifying, predicting, summarizing). The findings also suggested that the readers with limited knowledge of vocabulary had difficulty in moving on to the next level. Additionally, future research direction and some pedagogical implications are presented for the practical EFL classroom.

Molecular Cloning and Characterization of a Lipocalin in the Bumblebee Bombus Ignitus

  • We have cloned and characterized a lipocalin from the bumblebee Bombus ignitus (Bi-lipocalin). The Bi-lipocalin gene spans 2284 bp and consists of four exons coding for 270 amino acid residues. Sequence analysis revealed that Bi-lipocalin possesses three structurally conserved regions (SCTs) that characterize lipocalins. Recombinant Bi-lipocalin, expressed as a 37 kDa protein in baculovirus-infected insect cells, was N-glycosylated, indicating that the carbohydrate moieties are necessary for secretion. Tissue distribution analysis revealed ubiquitous expression of Bi-lipocalin in all tissues examined. Bi-lipocalin transcripts were upregulated by stress, such as wounding, $H_2O_2$ exposure, and external temperature shock. These results indicate that Bi-lipocalin is a stress-inducible protein that acts on wounding, $H_2O_2$ overexposure and temperature stimulation.
