Gynostemma pentaphyllum extract and Gypenoside L enhance skeletal muscle differentiation and mitochondrial metabolism by activating the PGC-1α pathway in C2C12 myotubes

  • BACKGROUND/OBJECTIVES: Peroxisome proliferator-activated receptor-gamma co-activator-1α (PGC-1α) has a central role in regulating muscle differentiation and mitochondrial metabolism. PGC-1α stimulates muscle growth and muscle fiber remodeling, concomitantly regulating lactate and lipid metabolism and promoting oxidative metabolism. Gynostemma pentaphyllum (Thumb.) has been widely employed as a traditional herbal medicine and possesses antioxidant, anti-obesity, anti-inflammatory, hypolipemic, hypoglycemic, and anticancer properties. We investigated whether G. pentaphyllum extract (GPE) and its active compound, gypenoside L (GL), affect muscle differentiation and mitochondrial metabolism via activation of the PGC-1α pathway in murine C2C12 myoblast cells. MATERIALS/METHODS: C2C12 cells were treated with GPE and GL, and quantitative reverse transcription polymerase chain reaction and western blot were used to analyze the mRNA and protein expression levels. Myh1 was determined using immunocytochemistry. Mitochondrial reactive oxygen species generation was measured using the 2'7'-dichlorofluorescein diacetate assay. RESULTS: GPE and GL promoted the differentiation of myoblasts into myotubes and elevated mRNA and protein expression levels of Myh1 (type IIx). GPE and GL also significantly increased the mRNA expression levels of the PGC-1α gene (Ppargc1a), lactate metabolism-regulatory genes (Esrra and Mct1), adipocyte-browning gene fibronectin type III domain-containing 5 gene (Fndc5), glycogen synthase gene (Gys), and lipid metabolism gene carnitine palmitoyltransferase 1b gene (Cpt1b). Moreover, GPE and GL induced the phosphorylation of AMP-activated protein kinase, p38, sirtuin1, and deacetylated PGC-1α. We also observed that treatment with GPE and GL significantly stimulated the expression of genes associated with the anti-oxidative stress response, such as Ucp2, Ucp3, Nrf2, and Sod2. CONCLUSIONS: The results indicated that GPE and GL enhance exercise performance by promoting myotube differentiation and mitochondrial metabolism through the upregulation of PGC-1α in C2C12 skeletal muscle.

미세먼지 PM10에 노출된 RAW264.7 세포에 대한 해당화 추출물의 항염증 활성 (Anti-inflammatory effects of Rosa rugosa extracts in RAW264.7 cells exposed to particulate matter (PM10))

  • 직경이 10 ㎛ 미만의 미세먼지 입자(particulate matter 10, PM10)는 한국을 비롯한 동아시아 지역에서 해로운 대기오염의 주요한 원인 중 하나로 최근 주목받고 있으며, 호흡기와 피부에 유해한 영향을 미치는 주범이 되고 있다. FD는 대식세포에서 활성산소 종(ROS)의 생성과 염증반응을 과도하게 촉진시켜 세포 손상과 더불어 세포 사멸을 유도한다. 장미속의 낙엽관목인 해당화(Rosa rugosa)는 다양한 질병을 치료하는 목적으로 예부터 동아시아 지역에서 민간요법 치료제로써 사용되었다. 본 연구에서는 PM10으로 유도된 RAW 264.7 대식세포에서의 염증반응을 억제하는 해당화 각 기관별 추출물의 항염증 효과를 탐색하고자 한다. 미세먼지 입자가 처리되지 않은 RAW 264.7 대식세포와 비교했을 때, PM10을 100 ㎍/ml의 농도로 처리했을 경우 lipopolysaccharide 처리와 유사하게 nitric oxide (NO) 생성을 눈에 띄게 증가시켰다. 또한 100 ㎍/ml의 해당화 줄기 추출물은 미세먼지 입자 단독 처리군에 비해 NO 생성을 45% 이상 감소시켰고, PM10에 의해 유도된 interleukin(IL)-1β, IL-6, inducible nitric oxide synthase, and cyclooxygenase-2의 발현을 유의하게 감소시켰다. 이러한 결과는 해당화 줄기 추출물의 항염증 효과가 PM10으로 자극된 RAW 264.7 세포에서 염증 유발 매개체의 억제에 의해 매개됨을 나타내며, 해당화 줄기가 유해한 공기 중 미세먼지 입자에 의해 유발되는 염증반응에 대한 보호 효과가 있는 천연 자원으로서 가치가 있음을 시사한다.

백출 용매추출 방법에 따른 항산화 활성 및 항염증 효과 (Antioxidant and Anti-inflammatory Activities of Atractylodes japonica According to Extract Methods)

  • 백출(Atractylodes japonica)은 국화과(Compositae)에 속하는 다년생 식물로 한국에서 이뇨작용, 수렴작용, 항알러지, 신경보호, 항암, 면역증강 및 위장관 보호효과 등으로 민간요법에서 널리 사용되고 있다. 백출의 열수 및 70% 에탄올 추출물에 대한 DPPH 및 ABTS 자유라디칼 소거능으로 항산화 활성을 측정하였다. 게다가 NO 생성, iNOS와 COX-2 단백질 발현을 웨스턴 블롯으로 측정하였으며, 염증성 사이토카인의 발현을 RT-PCR 방법으로 측정함으로 항염증 효과를 측정하였다. 열수 및 70% 에탄올 추출물들의 항산화 활성은 농도-의존적인 효과를 보였지만, 추출용매에 따른 차이는 나타나지 않았다. 70% 에탄올에 의한 백출 추출물에서 매우 강한 NO 활성 억제효과를 보였다. 백출 열수 및 에탄올 추출물은 NO 생성에 관여하는 iNOS 및 COX-2 단백질의 발현을 현저히 감소시켰다. 열수 및 70% 에탄올 추출물 모두 염증성 사이토카인인 IL-6 및 IL-1b는 현저히 억제시키지만, TNF-α의 발현은 약하게 억제시키는 결과를 얻었다. 본 연구를 통하여 백출 70% 에탄올 추출물이 NO 생성을 현저히 억제하며, 염증성 사이토카인의 발현을 현저히 감소시키는 효과를 보이므로 염증성 질환 치료 및 개선에 널리 사용될 수 있으리라 생각한다.

성장장애(成長障碍)를 주소(主訴)로 내원(來院)한 환아(患兒) 200례(例)에 대(對)한 임상적(臨床的) 관찰(觀察) (A Clinical Inquiry into 200 Cases of Children Coming to the Clinic Due to the Symptom of Growth Deficiency)

  • Over the period between January 1997 and December 1998, herbal medicine was more than three times administered to the patients coming to Na dong gyu's Oriental Medical Clinic on account of the symptom of growth deficiency. According to radiological opinions about the patients providing cooperation for measuring their height and weight as well as their bone age every three months, it was found that the growth plates were not closed. A research was conducted for 200 children randomly selected of patients in prepuberty (they grew by less than 5cm a year before treatment at a age of 12years for female children and 14years for male children). As a result, the following conclusion was drawn: 1. The randomly selected subjects were made up of 116 male and 84 female children in terms of gender. The age direstribution was most 10 to 12 years in 86 children(34.00%), followed by 8-10 years(27.50%) and 12 to 14 years(19.50%). 2. Considering the distribution of sick children's parental height, the fathers of 141 children(70.50%) measured less than 170cm high, the subaverage height, while the mothers of 172 children(86%) measured less than 160cm high, the subaverage height. It was shown that sick childen's height was genetically influenced by their parents. 3. Children patients's weight at a time of birth was most 3.1-3.5kg for 85 children(44%) and less than 2.5kg which came under the range of growth dificiency for 19 children(9.5%). 4. The highest proportian of the children patients with growth dificiency(56.33%) had the symptom of digestive disorders, of which 77 children patients(18.78%) had anorexia, 16.59% of children patients had the high level of respiratory disorders. Both the digestive disorder and the respiratory disorder put together, they had the high rate of 72.92%. Therefore, this indicates that both the digestive disorder and the respiratory disorder have a great effect on children's growth dificiency. 10.74% of chilren patients were shown to have allergic disorders, which indicates that they also exert an effect on growth deficiency. Specifically, 7.07% of the children patients had the high level of obesity, which shows that an excessive uptake of nutrition may rather induce children to have growth dificiency though an appropriate amount of nutritional uptake is necessary. 5. Comparing their bone age and their chronological age, 58 children patients(29.00%) showed that they were the same at the highest percent. 79 children patients(39.50%) showed that thier bone age was lower than their chronological age. And 63 children patients(31.50%) showed that their bone age was higher than their chronological age. 6. As regards the prescription administered to children patients for treating their growth dificiency, Growth tang A related to the kidney, the congenital factor, of the causes for growth dificiency in traditional Oriental Medicine was administered to 108 children patients(54%), whereas the Growth tang B related to the spleen, the acquired factor, was administered to 92 children patients(46%). 7. 116 male children patients with growth dificiency had the average value of growth for one year before treatment, 4.39cm, while 84 female children patients had the average value of growth for one year before treatment, 4.24cm. A total of 200 children had the average value of growth for one year before treatment, 4.33cm. The annual average value of growth in growth curve surveyed by the Korea Pediatrics Society was 5.79cm. Compared to this value, the one year average value of growth for 108 male and female children patients taking Growth tang A was shown be 8.44cm, which indicates a greater growth by 4.12cm(95.37%) in comparison with the average value of growth before treatment, 4.32cm, and a greater growth by 2.65cm(45.77%) compared to 5.79cm, the average value of growth in growth curve surveyed by the Korea Pediatrics Society. Also, the average value of growth before treatment for 92 male and female children patients taking Growth tang B was shown to be 8.47cm, which indicates a greater increase by 4.15cm(96.06%) compared to 4.32cm, the average value of growth before treatment and a greater increase by 2.67cm(46.29%) in comparison with 5.79cm, the average value of growth in growth curve surveyed by the Korea Pediatrics Society. Considering the average value of growth for male and female children patients taking Growth tang A and B, it was shown to be 8.46cm, which indicates a greater increase by 4.14cm(95.81%) compared to 4.32cm, the average value of growth before treatment, and a greater increase by 2.67cm(46.11%) compared to 5.79cm, the average value of growth in growth curve surveyed by the Korea Pediatrics Society.

동병하치 확산을 위한 전략적 방향과 이행방안 (Strategic Direction and Road Map of Expanding Prevention of Winter Disease in the Summer)

  • Objectives : The purpose of this study was to propose appropriate strategic directions and road maps for successful achievement of programs preventing winter disease in the summer. Methods : Details on programs preventing winter disease in the summer such as clear concept, theoretical basis, current status, intervention or available prescriptions and indication/contraindication/caution were prepared through the related journal review, upon which an observational study was devised and done for simulation to find out even a trivial problem and to guarantee the safety beforehand. The experimental group was divided into 5 groups by the size of pill and the way ginger is treated; 1cm pill with ginger group, 3cm pill group without ginger, 3cm pill group dipped into ginger, 3cm pill group applying ginger to acupoints and 3cm pill group with ginger Results 1. program preventing winter disease in the summer was defined as representative winter diseases such as common cold, influenza, chronic asthma, chronic bronchitis, allergic rhinitis, emphysema, chronic gastritis and rheumatoid arthritis, and preventive care in the summer, reinforcing deficient yang qi of five viscera by using exuberant yang qi from summer heat. 2. It was based upon historically established theories which is 'nourishing yang qi in the spring and summer', 'long summer, namely rainy spell in the summer overwhelms the winter, because of earth winning water according to the five phases theory' and 'To replenish yang qi is major principle to treat winter diseases, which can be most appropriately and timely applied to the patient with deficient yang qi of five viscera inherently, especially in the three dog days of the summer, because of exuberant exterior yang qi and deficient interior yang qi in the five viscera'. 3. In the adjacent China and Taiwan, acupoint applying method in the three dog days named 'San Fu Tie' have been stirring a boom throughout the nation, in which Xiaochuan Gao was used as a basic prescription and it mainly was applied at bilateral $BL_{13}$, $_{15}$ and $_{17}$ for about 4 hours. As far as domestic current status, the necessity of adopting the above method prior to Herbal formula was also recognised, because not a few koreans have apprehension for the safety of it including medicinal herbs and are reluctant to take it any more due to negative advertisement of narrow minded doctors' association. 4. Indication of acupoint applying method in the three dog days included most of winter diseases such as common cold, influenza, chronic asthma, chronic bronchitis, allergic rhinitis, emphysema, chronic gastritis. contraindication was pregnant woman and the weak such as infants and the old. More attention was paid to grasp firmly the normal reaction following the treatment for preventing side effect and teasing blister. recommendation was also given to abstain from food inducing phlegm and dampness such as meat, shrimp and crab as well as cold drinks and foods 5. In the simulation observational study based upon the above findings following review the related articles, no blister was shown on the acupoints icluding bilateral $BL_{13}$, $_{15}$ and $_{17}$ in every experimental group during 24hr observation following the acupoint applying treatment with pills made by modified and devised prescription. At 4 hr, the effectiveness of it reached a peak showing redness and mild tenderness and there is little difference between groups 3cm pills groups regardless of the way ginger was treated. abdominal distention and growling was found in all the volunteers during the treatment at CV 8. Strategic directions and road maps : Through successful fulfillment of the program preventing winter disease in the summer, Korean traditional medicine should be integrated into mainstream national health care services. Cultural access was thought to be as important as Scientific EBM approach. First of all, To evoke potential cultural homogeneity from campaigns and press advertisement was needed for promoting public awareness about preventing winter disease in the summer by enhancing immunity via acupoint applying treatment in the three dog days, and then indigenous name as Sambokcheop, protocol, Clinical Research Form for data collection of it should be developed and prepared. Once the first step was taken this summer, through a thorough data collection and scrutinized scientific evaluation, drawbacks should be compensted for and the efficacy and safety should be substantiated.

포도전정가지 추출물이 UVB에 유도된 HR-1 mice의 피부손상에 대한 광보호 효과 (Photoprotective Effect of Grape Pruning Stem Extracts on the UVB Induced HR-1 Mice Skin)

  • 본 연구는 포도수확 후 버려지는 가지를 이용한 포도전정가지 추출물(Grape Pruning Stem Extracts, GPSE)에 함유된 polyphenol류 중 항산화, 항염증, 항암 등의 효과가 있다고 알려진 rutin, procyanidin B3, quercetin, kaempferol의 함량을 분석하고, UVB로 유도된 HR-1 mice의 손상된 피부에 대한 피부 보습, 피부 증식 억제, 항염증 등의 효과를 측정하여 기능성식품, 기능성 화장품 소재로서의 응용 가능성을 확인하고자 하였다. 포도전정가지에서 polyphenol 성분을 80% EtOH로 추출하고 여과하여 농축한 후, 동결건조하여 $-20^{\circ}C$에 보관하면서 사용하였다. GPSE의 유효성분 함량은 HPLC를 사용하여 분석하였다. 피부손상을 유도하기 위해 UVB를 실험동물에 조사하였고, GPSE의 효능을 확인하기 위하여 TEWL assay, 피부조직의 H&E staining 및 COX-2 단백 발현 측정을 위한 면역조직화학적염색(immunohistochemical stain) 등을 실시하였다. 포도전정가지 시료 53 kg에서 EtOH 분획 추출물 2.34 kg을 추출하여 4.42%의 수율 결과를 얻었다. 유효성분 분석 결과, procyanidin B3, 0.28 mg/g, rutin 12.81 mg/g, quercetin 0.51 mg/g 및 kaempferol 8.24 mg/g로 나타났다. TEWL assay 결과 대조군과 비교, GPSE와 serum base의 혼합물이 도포된 그룹(농도 2,000 mg/kg~125 mg/kg)에서 모두 보습효과가 있는 것을 확인하였다(p<0.05). GPSE의 피부증식억제를 통한 광보호 효과를 확인한 결과 epidermis에는 GPSE 농도 2,000 mg/kg의 농도에서 suncream과 유사한 결과를 얻을 수 있었고(p<0.05), dermis의 두께를 측정한 결과 GPSE 농도 2,000 mg/kg~125 mg/kg 모든 농도에서 UVB 조사한 그룹 두께 $800{\mu}m$와 비교하여 suncream 도포군이 $580{\mu}m$, GPSE 2,000 mg/kg~125 mg/kg 모든 농도에서 $600{\mu}m$이하로 나타나 UVB에 의해 손상된 HR-1 mice의 피부증식 억제를 확인할 수 있었다(p<0.05). GPSE가 UVB에 의해 손상된 HR-1 mice 피부에 항염증 정도를 확인하기 위해 COX-2 단백발현을 보기위해 immunohistochemical stain한 결과 UVB를 조사한 대조군에 비해 GPSE를 도포한 그룹 모두에서 항염증 효과를 보였지만, GPSE의 농도 1,000 mg/kg에서는 suncream 22%보다 낮은 8%에서 COX-2의 단백발현이 낮은 것으로 나타나 우수한 항염증효과를 얻을 수 있었다(p<0.05). 본 연구 결과에서, GPSE 내 rutin, kaempferol, quercetin, procyanidin B3와 같은 유효성 페놀성 화합물이 확인되었으며, GPSE는 보습효과, 피부증식억제효과, 염증발현억제효과 및 자외선 손상에 의한 피부장벽 기능 개선효과 등이 있는 것으로 확인되었다. 결론적으로 GPSE는 피부보호 효과의 가능성이 있는 기능성 물질로서 기능성 식품 및 기능성 화장품 등의 원료로 활용성이 높을 것으로 판단된다.

일본 '고증파(考證派)' 의학에 관한 연구 (A Study on The 'Kao Zheng Pai'(考證派) of The Traditional Medicine of Japan)

  • 1.The 'Kao Zheng Pai'(考證派) comes from the 'Zhe Zhong Pai(折衷派)' and is a school that is influenced by the confucianism of the Qing dynasty. In Japan Inoue Kinga(井上金峨), Yoshida Koton(古田篁墩 $1745{\sim}1798$) became central members, and the rise of the methodology of historical research(考證學) influenced the members of the 'Zhe Zhong Pai', and the trend of historical research changed from confucianism to medicine, making a school of medicine based on the study of texts and proving that the classics were right. 2. Based on the function of 'Nei Qu Li'(內驅力) the 'Kao Zheng Pai', in the spirit of 'use confucianism as the base', researched letters, meanings and historical origins. Because they were influenced by the methodology of historical research(考證學) of the Qing era, they valued the evidential research of classic texts, and there was even one branch that did only historical research, the 'Rue Xue Kao Zheng Pai'(儒學考證派). Also, the 'Yi Xue Kao Zheng Pai'(醫學考證派) appeared by the influence of Yoshida Kouton and Kariya Ekisai(狩谷掖齋). 3. In the 'Kao Zheng Pai(考證派)'s theories and views the 'Yi Xue Kao Zheng Pai' did not look at medical scriptures like the "Huang Di Nei Jing"("黃帝內經") and did not do research on 'medical' related areas like acupuncture, the meridian and medicinal herbs. Since they were doctors that used medicine, they naturally were based on 'formulas'(方劑) and since their thoughts were based on the historical ideologies, they valued the "Shang Han Ja Bing Lun" which was revered as the 'ancestor of all formulas'(衆方之祖). 4. The lives of the important doctors of the 'Kao Zheng Pai' Meguro Dotaku(目黑道琢) Yamada Seichin(山田正珍), Yamada Kyoko(山田業廣), Mori Ritsi(森立之) Kitamura Naohara(喜多村直寬) are as follows. 1) Meguro Dotaku(目黑道琢 $1739{\sim}1798$) was born of lowly descent but, using his intelligence and knowledge, became a professor as a Shi Jing Yi(市井醫) and as a professor for 34 years at Ji Shou Guan(躋壽館) mastered the "Huang Di Nei Jing" after giving over 300 lectures. Since his pupil, Isawara Ken(伊澤蘭軒) taught the Lan Men Wu Zhe(蘭門五哲) and Shibue Chusai(澀江抽齋), Mori Ritsi(森立之), Okanishi Gentei(岡西玄亭), Kiyokawa Gendoh(淸川玄道) and Yamada Kyoko(山田業廣), Meguro Dotaku is considered the founder of the 'Yi Xue Kao Zheng Pai'. 2) The family of Yamada Seichin(山田正珍 $1749{\sim}1787$) had been medical officials in the Makufu(幕府) and the many books that his ancestors had left were the base of his art. Seichin learned from Shan Ben Bei Shan(山本北山), a 'Zhe Zhong Pai' scholar, and put his efforts into learning, teaching and researching the "Shang Han Lun"("傷寒論"). Living in a time between 'Gu Fang Pai'(古方派) member Nakanishi Goretada(中西惟忠) and 'Kao Zheng Pai' member Taki Motohiro(多紀元簡), he wrote 11 books, 2 of which express his thoughts and research clearly, the "Shang Han Lun Ji Cheng"("傷寒論集成") and "Shang Han Kao"("傷寒考"). His comparison of the 'six meridians'(3 yin, 3 yang) between the "Shang Han Lun" and the "Su Wen Re Lun"("素問 熱論") and his acknowledgement of the need and rationality of the concept of Yin-Yang and Deficient-Replete distinguishes him from the other 'Gu Fang Pai'. Also, his dissertation of the need for the concept doesn't use the theories of latter schools but uses the theory of the "Shang Han Lun" itself. He even researched the historical parts, such as terms like 'Shen Nong Chang Bai Cao'(神農嘗百草) and 'Cheng Qi Tang'(承氣湯). 3) The ancestor of Yamada Kyoko(山田業廣) was a court physician, and learned confucianism from Kao Zheng Pai's Ashikawa Genan(朝川善庵) and medicine from Isawa Ranken(伊澤蘭軒) and Taki Motokata(多紀元堅), and the secret to smallpox from Ikeda Keisui(池田京水). He later became a lecturer at the Edo Yi Xue Guan(醫學館) and was invited as the director to the Ji Zhong(濟衆) hospital. He also became the first owner of the Wen Zhi She(溫知社), whose main purpose was the revival of kampo, and launched the monthly magazine Wen Zi Yi Tan(溫知醫談). He also diagnosed and prescribed for the prince Ming Gong(明宮). His works include the "Jing Fang Bian"("經方辨"), "Shang Han Lun Si Ci"("傷寒論釋詞"), "Huang Zhao Zhu Jia Zhi Yan Ji Yao"("皇朝諸家治驗集要") and "Shang Han Ja Bing Lun Lei Juan"("傷寒雜病論類纂"). of these, the "Jing Fang Bian"("經方辨") states that the Shi Gao(石膏) used in the "Shang Han Lun" had three meanings-Fa Biao(發表), Qing Re(淸熱), Zi Yin(滋陰)-which were from 'symptoms', and first deducted the effects and then told of the reason. Another book, the "Jiu Zhe Tang Du Shu Ji"("九折堂讀書記") researched and translated the difficult parts of the "Shang Han Lun", "Jin Qui Yao Lue"("金匱要略"), "Qian Jin Fang"("千金方"), and "Wai Tai Mi Yao"("外臺秘要"). He usually analyzed the 'symptoms' of diseases but the composition, measurement, processing and application of medicine were all in the spectrum of 'analystic research' and 'researching analysis'. 4) The ancestors of Mori Ritsi(森立之 $1807{\sim}1885$) were warriors but he became a doctor by the will of his mother, and he learned from Shibue Chosai(澁江抽齋) and Isawaran Ken(伊澤蘭軒) and later became a pupil of Shou Gu Yi Zhai(狩谷掖齋), a historical research scholar. He then became a lecturer of medical herbs at the Yi Xue Guan, and later participated in the proofreading of "Yi Xin Fang"("醫心方") and with Chosai compiled the "Jing Ji Fang Gu Zhi"("經籍訪古志"). He visited the Chinese scholar Yang Shou Jing(楊守敬) in 1881 and exchanged books and ideas. Of his works, there are the collections(輯複本) of "Shen Nong Ben Cao Jing"("神農本草經") and "You Xiang Yi Hwa"("遊相醫話") and the records, notes, poems, and diaries such as "Zhi Yuan Man Lu"("枳園漫錄") and "Zhi Yuan Sui Bi"(枳園隨筆) that were not published. His thoughts were that in restoring the "Shen Nong Ben Cao Jing", "the herb to the doctor is like the "Shuo Wen Jie Zi"(說文解字) to the scholar", and he tried to restore the ancient herbal text using knowledge of medicine and investigation(考據), Also with Chosai he compiled the "Jing Ji Fang Gu Zhi"("經籍訪古志") using knowledge of ancient text. Ritzi left works on pure investigation, paid much attention to social problems, and through 12 years of poverty treated all people and animals in all branches of medicine, so he is called a 'half confucianist half doctor'(半儒半醫). 5) Kitamurana Ohira(喜多村直寬, $1804{\sim}1876$) learned scriptures and ancient texts from confucian scholar Asaka Gonsai(安積艮齋), and learned medicine from his father Huai Yaun(槐園), He became a teacher in the Yi Xue Guan in his middle ages, and to repay his country, he printed 266 volumes of "Yi Fang Lei Ju"("醫方類聚") and 1000 volumes of "Tai Ping Yu Lan"("太平禦覽") and devoted it to his country to be spread. His works are about 40 volumes including "Jin Qui Yao Lue Shu Yi"("金匱要略疏義") and "Lao Yi Zhi Yan"(老醫巵言) but most of them are researches on the "Shang Han Za Bing Lun". In his "Shang Han Lun Shu Yi"("傷寒論疏義") he shows the concept of the six meridians through the Yin-Yang, Superficial or internal, cold or hot, deficient or replete state of diseases, but did not match the names with the six meridians of the meridian theory, and this has something in common with the research based on the confucianism of Song(宋儒). In clinical treatment he was positive toward old and new methods and also the experience of civilians, but was negative toward western medicine. 6) The ancestor of the Taki family Tanbano Yasuyori(丹波康賴 $912{\sim}955$) became a Yi Bo Shi(醫博士) by his medical skills and compiled the "Yi Xin Fang"("醫心方"). His first son Tanbano Shigeaki(丹波重明) inherited the Shi Yao Yuan(施藥院) and the third son Tanbano Masatada(丹波雅忠) inherited the Dian You Tou(典藥頭). Masatada's descendents succeeded him for 25 generations until the family name was changed to Jin Bao(金保) and five generations later it was changed again to Duo Ji(多紀). The research scholar Taki Motohiro was in the third generation after the last name was changed to Taki, and his family kept an important part in the line of medical officers in Japan. Taki Motohiro(多紀元簡 $1755{\sim}1810$) was a teacher in the Yi Xue Guan where his father was residing, and became the physician for the general Jia Qi(家齊). He had a short temper and was not good at getting on in the world, and went against the will of the king and was banished from Ao Yi Shi(奧醫師). His most famous works, the "Shang Han Lun Ji Yi"("傷寒論輯義") and "Jin Qui Yao Lue Ji Yi"("金匱要略輯義") are the work of 20 years of collecting the theories of many schools and discussing, and is one of the most famous books on the "Shang Han Lun" in Japan. "Yi Sheng"("醫勝") is a collection of essays on research. Also there are the "Su Wen Shi"(素問識), "Ling Shu Shi"("靈樞識"), and the "Guan Ju Fang Yao Bu"("觀聚方要補"). Taki Motohiro(多紀元簡)'s position was succeeded by his third son Yuan Yin(元胤 $1789{\sim}1827$), and his works include works of research such as "Nan Jing Shu Jeng"(難經疏證), "Ti Ya"("體雅"), "Yao Ya"("藥雅"), "Ji Ya"(疾雅), "Ming Yi Gong An"(名醫公案), and "Yi Ji Kao"(醫籍考). The "Yi Ji Kao" is 80 volumes in length and lists about 3000 books on medicine in China before the Qing Dao Guang(道光), and under each title are the origin, number of volumes, state of existence, and, if possible, the preface, Ba Yu(跋語) and biography of the author. The younger sibling of Yuan Yin(元胤 $1789{\sim}1827$), Yuan Jian(元堅 $1795{\sim}1857$) expounded ancient writings at the Yi Xue Guan only after he reached middle age, was chosen for the Ao Yi Shi(奧醫師) and later became a Fa Yan(法眼), Fa Yin(法印) and Yu Chi(禦匙). He left about 15 texts, including "Su Wen Shao Shi"("素問紹識"), "Yi Xin Fang"("醫心方"), published in school, "Za Bing Guang Yao"("雜病廣要"), "Shang Han Guang Yao"("傷寒廣要"), and "Zhen Fu Yao Jue"("診腹要訣"). On the Taki family's founding and working of the Yi Xue Guan Yasuka Doumei(矢數道明) said they were "the people who took the initiative in Edo era kampo medicine" and evaluated their deeds in the fields of 'research of ancient text', the founding of Ji Shou Guan(躋壽館) and medical education', 'publication business', 'writing of medical text'. 5. The doctors of the 'Kao Zheng Pai' based their operations on the Edo Yi Xue Guan, and made groups with people with similar ideas to them, making a relationship 'net'. For example the three families of Duo Ji(多紀), Tang Chuan(湯川) and Xi Duo Cun(喜多村) married and adopted with and from each other and made prefaces and epitaphs for each other. Thus, the Taki family, the state science of the Makufu, the tendency of thinking, one's own interests and glory, one's own knowledge, the need of the society all played a role in the development of kampo medicine in the 18th and 19th century.

  • 1. The 'Kao Zheng Pai(考證派) comes from the 'Zhe Zhong Pai' and is a school that is influenced by the confucianism of the Qing dynasty. In Japan Inoue Kinga(井上金娥), Yoshida Koton(吉田篁墩) became central members, and the rise of the methodology of historical research(考證學) influenced the members of the 'Zhe Zhong Pai', and the trend of historical research changed from confucianism to medicine, making a school of medicine based on the study of texts and proving that the classics were right. 2. Based on the function of 'Nei Qu Li '(內驅力) the 'Kao Zheng Pai', in the spirit of 'use confucianism as the base', researched letters, meanings and historical origins. Because they were influenced by the methodology of historical research(考證學) of the Qing era, they valued the evidential research of classic texts, and there was even one branch that did only historical research, the 'Rue Xue Kao Zheng Pai'(儒學考證派). Also, the 'Yi Xue Kao Zheng Pai'(醫學考證派) appeared by the influence of Yoshida Kouton and Kariya Ekisai(狩谷掖齋). 3. In the 'Kao Zheng Pai(考證派)'s theories and views the 'Yi Xue Kao Zheng Pai' did not look at medical scriptures like the "Huang Di Nei Jing"("黃帝內經") and did not do research on 'medical' related areas like acupuncture, the meridian and medicinal herbs. Since they were doctors that used medicine, they naturally were based on 'formulas'(方劑) and since their thoughts were based on the historical ideologies, they valued the "Shang Han Ja Bing Lun" which was revered as the 'ancestor of all formulas'(衆方之祖). 4. The lives of the important doctors of the 'Kao Zheng Pai' Meguro Dotaku(目黑道琢) Yamada Seichin(山田正珍), Yamada Kyoko(山田業廣), Mori Ritsi(森立之) Kitamura Naohara(喜多村直寬) are as follows. 1) Meguro Dotaku(目黑道琢 1739${\sim}$1798) was born of lowly descent but, using his intelligence and knowledge, became a professor as a Shi Jing Yi(市井醫) and as a professor for 34 years at Ji Shou Guan mastered the "Huang Di Nei Jing" after giving over 300 lectures. Since his pupil, Isawara Ken taught the Lan Men Wu Zhe(蘭門五哲) and Shibue Chusai, Mori Ritsi(森立之), Okanishi Gentei(岡西玄亭), Kiyokawa Gendoh(淸川玄道) and Yamada Kyoko(山田業廣), Meguro Dotaku is considered the founder of the 'Yi Xue Kao Zheng Pai'. 2) The family of Yamada Seichin(山田正珍 1749${\sim}$1787) had been medical officials in the Makufu(幕府) and the many books that his ancestors had left were the base of his art. Seichin learned from Shan Ben Bei Shan(山本北山), a 'Zhe Zhong Pai' scholar, and put his efforts into learning, teaching and researching the "Shang Han Lun"("傷寒論"). Living in a time between 'Gu Fang Pai'(古方派) member Nakanishi Goretada(中西惟忠) and 'Kao Zheng Pai' member Taki Motohiro(多紀元簡), he wrote 11 books, 2 of which express his thoughts and research clearly, the "Shang Han Lun Ji Cheng"("傷寒論集成") and "Shang Han Kao"("傷寒考"). His comparison of the 'six meridians'(3 yin, 3 yang) between the "Shang Han Lun" and the "Su Wen Re Lun"("素問 熱論) and his acknowledgement of the need and rationality of the concept of Yin-Yang and Deficient-Replete distinguishes him from the other 'Gu Fang Pai'. Also, his dissertation of the need for the concept doesn't use the theories of latter schools but uses the theory of the "Shang Han Lun" itself. He even researched the historical parts, such as terms like 'Shen Nong Chang Bai Cao'(神農嘗百草) and 'Cheng Qi Tang'(承氣湯) 3) The ancestor of Yamada Kyoko(山田業廣) was a court physician, and learned confucianism from Kao Zheng Pai 's Ashikawa Genan(朝川善庵) and medicine from Isawa Ranken and Taki Motokata(多紀元堅), and the secret to smallpox from Ikeda Keisui(池田京水). He later became a lecturer at the Edo Yi Xue Guan(醫學館) and was invited as the director to the Ji Zhong(濟衆) hospital. He also became the first owner of the Wen Zhi She(溫知社), whose main purpose was the revival of kampo, and launched the monthly magazine Wen Zi Yi Tan(溫知醫談). He also diagnosed and prescribed for the prince Ming Gong(明宮). His works include the "Jing Fang Bian"("經方辨"), "Shang Han Lun Si Ci"("傷寒論釋司"), "Huang Zhao Zhu Jia Zhi Yan Ji Yao"("皇朝諸家治驗集要") and "Shang Han Ja Bing Lun Lei Juan"("傷寒雜病論類纂"). of these, the "Jing Fang Bian"("經方辨") states that the Shi Gao(石膏) used in the "Shang Han Lun" had three meanings-Fa Biao(發表), Qing Re(淸熱), Zi Yin(滋陰)-which were from 'symptoms', and first deducted the effects and then told of the reason. Another book, the "Jiu Zhe Tang Du Shu Ji"("九折堂讀書記") researched and translated the difficult parts of the "Shang Han Lun", "Jin Qui Yao Lue", "Qian Jin Fang"("千金方"), and "Wai Tai Mi Yao"("外臺秘要"). He usually analyzed the 'symptoms' of diseases but the composition, measurement, processing and application of medicine were all in the spectrum of 'analystic research' and 'researching analysis'. 4) The ancestors of Mori Rits(森立之 1807${\sim}$ 1885) were warriors but he became a doctor by the will of his mother, and he learned from Shibue Chosai(澁江抽齋) and Isawaran Ken and later became a pupil of Shou Gu Yi Zhai, a historical research scholar. 복합처방인 승마갈근탕(升摩葛根湯)의 항산화 및 항균효과 (Anti-oxidant and Anti-microbial Activities of Seungmakalgeuntang)

  • 복합처방인 승마갈근탕을 이용하여 화장품 및 식품에 응용하고자 항산화 및 항균효과를 검토하였다. 승마갈근탕의 전자공여능은 모든 추출물 100 ppm에서 50% 이상의 효과를 나타내었고, 특히 초임계 추출물의 경우 500 ppm에서 88%로 가장 높은 효과를 나타내었다. SOD 유사활성은 승마갈근탕 1,000 ppm에서 50% 이하의 다소 낮은 경향을 보였으나, 그 중 에탄올과 초임계 추출물의 경우 각각 46, 45의 효능을 나타내었으며, xanthine oxidase 저해활성의 경우 초임계 추출물 1,000 ppm에서 76%로 대조군인 vitamin C의 39%보다 높은 효과를 나타내었다. 승마갈근탕의 지방산패 억제능을 측정하기 위해 산화촉진제인 $Fe^{2+}$, $Cu^{2+}$ 이온을 첨가하여 측정한 결과 초임계 추출물의 $Cu^{2+}$ 첨가 시 500 ppm에서 68%의 효과를 나타내었고, $Fe^{2+}$를 첨가 시 100 ppm에서 60%의 지방산패 억제능을 나타내었다. 승마갈근탕의 clear zone 형성을 관찰한 결과, 추출물 모두 Escherichia coli를 제외한 모든 균주에 대해서 항균 효과가 나타내었다. Staphylococcus aureus와 Propionibacterium acnes의 경우 4 mg/disc에서 초임계 추출물에서는 1.45, 2.10 cm로 가장 높은 항균 효과를 나타내었고, Staphylococcus epidermidis의 경우, 에탄올 추출물이 1.82 cm의 항균효과를 나타내었다.

편백나무 잎 추출물의 성분분석과 면역효능에 관한 연구 (Analysis of the Component and Immunological Efficacy of Chamaecyparis obtusa Leaf Extract)

  • 본 연구는 전라도 장성지역의 편백나무 잎 추출물을 사용하여 in vitro에서 항염증 및 항알레르기 효과를 보고자 연구를 진행하였다. 편백나무 잎 추출물은 $50^{\circ}C$에서 감압 건조하여 실험에 사용하였으며, 총 폴리페놀 함량을 측정한 결과 $25.89{\pm}0.31mg\;GAE/g$로 나타났다. 설정된 GC-MS 분석법으로 편백나무잎 추출물의 6종 성분에 대한 함량 분석을 실시한 결과, ${\alpha}-Terpinene$ 3.03 mg/g, ${\alpha}-Terpineol$ 9.48 mg/g, limonene 5.96 mg/g, borneol 59.78 mg/g, myrcene 4.85 mg/g, sabinene 11.31 mg/g로 borneol이 가장 많은 것으로 나타났다. 편백나무 잎 추출물의 항산화 활성을 측정한 결과, $H_2O_2$$ABTS^+$ 라디칼에 대한 추출물의 $RC_{50}$이 각각 $5.47{\pm}0.13mg/mL$$4.00{\pm}0.01mg/mL$로 나타났다. 또한 마우스 유래의 대식세포주인 RAW 264.7 세포에서 LPS 100 ng/mL을 처리를 통한 염증유도 군에서 주요 인자인 NO 생성이 $28{\pm}0.38{\mu}M$까지 증가하였으나 편백나무 잎 추출물 $150{\mu}g/mL$ 처리 농도에서 $IC_{50}$ 으로 감소할 것으로 추정되므로, 편백나무 잎 추출물이 항염증 작용이 있음을 시사한다. 알레르기 주요 인자인 ${\beta}-hexosaminidase$의 경우 처리한 편백나무 잎 추출물의 농도 의존적으로 감소되어 항알레르기 효능이 있음을 알 수 있었으며, 인간 유래 섬유아세포인 CCD-986sk 세포에 대해 편백나무 잎 추출물의 농도 $100{\sim}800{\mu}g/mL$ 범위에서 세포독성을 전혀 보이지 않았다. 따라서 편백나무 잎 추출물이 향후 기능성 화장품, 연고 등의 산업화에 광범위하게 이용될 수 있을 것으로 사료된다.
