The present study examined the lipid lowering and antioxidant activity of Paecilomyces tenuipes. Eight week-old male Sprague-Dawley rats were fed one of the three diets, a reference diet without cholesterol addition (NC), a high fat (17 g/ 100 g)-high cholesterol (1 g/100 g) diet (HC) and a HC diet supplemented with 3% P. tenuipes (PT) for 30 days. Total lipid and total cholesterol were reduced significantly by 33 and 37%, respectively, in the PT diet group compared with controls. A similar reduction was found for low density lipoprotein (LDL) cholesterol levels, while plasma high density lipoprotein (HDL) cholesterol and triglyceride (TG) concentrations were not significantly different among groups. Hepatic total lipid and total cholesterol levels, but not hepatic TG levels, were significantly decreased in the PT group compared to the HC group. The administration of P. tenuipes increased the plasma total antioxidant potential and decreased the levels of lipid peroxidation. These results suggest that P. tenuipes exerts significant health benefits through the modulation of physiological functions including a variety of atherogenic lipid profiles and antioxidants in hypercholesterolemia.
Previously, we reported that a diet including sesame meal (SM) increased plasma total and high-density lipoprotein (HDL)-cholesterol concentrations in goats. In the present study, the components in the sesame meal that can increase plasma total and HDL-cholesterol concentrations have been examined. In experiment 1, we gave goats defatted sesame meal diet (DSM) to investigate the influence of ether extract fraction remained in sesame meal. Corn gluten meal diet (CGM) was also fed to goats as a high-protein diet to examine the influence of high dietary protein level caused by usage of sesame meal. Plasma total and HDL-cholesterol concentrations of goats fed DSM and CGM did not change during experimental periods though they were elevated by feeding SM. In experiment 2, the influence of sesame oil and corn oil added in diets on plasma total and HDL-cholesterol concentrations in goats was investigated. Plasma total and HDL-cholesterol concentrations were increased by feeding both corn oil diet and sesame oil diet. In conclusion, the increase in plasma HDL-cholesterol concentration by feeding sesame meal was resulted by the effect of ether extract fraction including sesame oil or some lipid-soluble components remained in sesame meal.
The objective of this study is to find the effect of improving hyperlipidemia through Platycodi radix saponin. A hypercholoesterol diet and Platycodi radix saponin methanol extract were supplemented to the 6 week old male Spargue Dawley rats for four weeks. Results showed that the serum total cholesterol, triglyceride and LDL-cholesterol level decreased in the group supplemented with Platycodi radix saponin methanol extract(p<0.05), however, the serum total lipid and HDL-cholesterol did not show any difference from the control group according to the supplement of Platycodi radix saponin methanol extract. In the group supplemented with Platycodi radix saponin methanol extract, the total cholesterol decreased by 20%, triglyceride by 36% and LDL-cholesterol by 25% respectively, and there was no difference according to the level of Platycodi radix saponin methanol extract. AI(atherogenic index) and CRF(cardiac risk factor) were low in the group supplemented with Platycodi radix saponin methanol extract. Total lipid, total cholesterol and triglyceride in liver and feces were significantly low in the group supplemented with the Platycodi radix saponin methanol extract compared with the control group(p<0.05). According to this result, it is expected that the Platycodi radix saponin methanol extract supplemented in the hypercholesterol diet can lower atherosclerosis, the cause of cardiovascular disease, by reducing not only serum cholesterol and triglyceride but also liver cholesterol and triglyceride.
The effect of fiber sources on lipid metabolism was investigated in relation to the level of dietary fat. After rats were fed each diet for 4 weeks, all animals were sacrificed to collect the liver, and blood samples by heart puncture. Total lipid, total cholesterol, phospholipid and HDL- cholesterol were determined in serum. Total lipid and total cholesterol were also determined in liver. These results were compared with the histochemical appearances by the method of Oil red-O staining. Higher level of fat in the diet appeared to increase the total lipid levels in serum and more fat in the liver cells were shown from the high fat diet judging from the E.M. pictures. Phospholipid concentration in serum was greater in the rats of high fat groups than those of the control or non-fat groups. High fat level caused to decrease the HDL- cholesterol concentration in serum. Pectin decreased serum and liver lipids and the deposit of fat in the liver cells. HDL-cholesterol levels in serum was increased by pectin. Whereas the cellulose supplementation had no effect to reduce serum and liver total lipid levels, and cellulose did not decreased the deposit of fat in the liver cells. From these results, it is concluded that dietary fat levels led to changes in the lipid metabolism of rats and pectin may exhibit a lowering total lipid contents in serum and liver. Cellulose may slightly elevate serum and liver levels, or have no lowering effect.
The study examines the effects of the Cnidium officinale Makino water extract on hyperlipidemia. For this, three levels (low, moderate, and high) of the Cnidium officinale Makino water extract were supplemented to six-week-old male sprague Dawley rats for five weeks. According to the results, serum total lipid, triglyceride, total cholesterol, and LDL cholesterol levels showed significant decreases (p<0.05) in the group with the Cnidium officinale Makino water extract. The serum total lipid level decreased by 8~22%; the triglyceride level, by 13~25%; the total cholesterol level, by 6~23%; and the LDL cholesterol level, by 14~25%. Total cholesterol and triglyceride levels in the liver were significantly lower in the group with the Cnidium officinale Makino water extract than in the control group (p<0.05), and fecal total cholesterol and triglyceride levels were significantly higher in the group with the Cnidium officinalle Makino water extract. These results suggest that the Cnidium officinale Makino water extract in the hypercholesterol diet may reduce hyperlipidemia, a major cause of cardiovascular disease, by reducing serum cholesterol and triglyceride level in the liver through the facilitation of their excretion by feces.

A simple, reproducible , and accurate enzymatic method using a cholesterol assay kit was developed to quantify total cholesterol content in egg yolk. Total egg yolk lipid was extracted with hexane : isopropanol(3 : 2, v/v) mixture. Samples containing various amount of the total lipid(0-3mg) in optically identifical culture tubes were reacted for 10 min in a water bath (37$^{\circ}C$) with the enzyme solution (5ml) from the cholesterol assay kit. Cholesterol content of the reaction mixturesin culture tubes was spectrophotometrically determined by two different ways : (1) using the culture tube as a curvette(designate culture tube method ; CTM) and (2) the quartz cvette containing the reaction mixture transferred from the culture tube (designate standard cvette method, SCM). CTM revealed lower cholesterol content in 0.1-1.0mg lipid sample range that SCM did, but not significant. For more than 2.0mg lipid sample, CTM gave significantly (p<0.01) lower cholesterol content relative to that by SCM, suggesting that SCM give a false positive result from the sample containing more than 2 mg lipid due to the interference of absorbance by lipid dispersed in the reaction solution . Cholesterol content of less than 1.0mg lipid sample by CTM was proportional to the amount of lipid used, but its linear relationship was not seen in more than 2mg lipid sample. Thus, to determine the appropriate lipid amounts (mg) analyzed . A constant level (41$\mu\textrm{g}$/mg) of cholesterol concentration was observed from the sample containing 0.1-1mg lipid. after which the cholesterol level was dropped to less than 41$\mu\textrm{g}$ /mg. Cholesterol concentration in egg yolk samples quantified by CTM was in accordance with that by GC method. These results suggest that CTM is an useful method for the quantification of cholesterol in egg yolk lipid and other lipids as well.
This study was performed to investigate the risk factors for coronary heart disease in elderly women. Seventy five elderly women over 65 years of age participated with 35 elderly men over 65 years of age, 40 middle-aged men and 31 middle-aged women as control subjects. The percentage of body fat(34.1 5.6%) in elderly women found to be significantly higher than in other groups and their waist/hip circumference ratio(WHR) was higher than in middle-aged women. The concentrations of plasma total cholesterol and LDL-cholesterol in elderly women were higher than in other groups, TG concentration higher than in middle aged women and HDL-cholesterol % lower than in other groups. Plasma cholesterol ester transfer protein(CETP) activities of elderly women were significantly higher than in middle-aged subjects, but were not different from those of elderly men. Plasma Apolipoprotein(Apo) A-I level in elderly women was higher than in middle-aged men but not different from the other groups. Differences among groups were not great in Lipoprotein (Lp)(a)levels. CETP activities were significantly correlated with age, body fat % total cholesterol, LDL-cholesterol and Lp(a). Therefore, it appears that the increased risk of coronary heart disease in elderly women is due to the increase of body fat, central adipose distribution, serum total cholesterol, LDL-cholesterol and CETP activities.
In order to independently examine the effects of long-chain saturated fatty acids and dietary cholesterol levels on plasma and hepatic cholesterol concentrations, six different diets were fed to male Mongolian gerbils (14 gerbils pes grcup) for an 8-week period. Purified diets contained 36% energy as fat (each saturated fatty acid tested comprised about 20% of the total fat energy) and 0.06% (w/w) cholesterol, corresponding to typical human consumption patterns in Western diets. Fat blends were formulated with natural fat sources. To determine the effects of different saturated fatty acids on plasma and liver cholesterol levels, four of the six diets contained constant levels of all nutrients except for the amounts of lauric acid (12 0), myristic acid (14 : 0), palmitic acid (16 0), and stearic acid (18 : 0). Dietary cholesterol effects were tested using 16 : 0-enriched diets containing 0, 0.006, and 0.06% (w/w) cholesterol. None of the plasma lipids were influenced by fatty acid treatment, including triglycerides, plasma total-, VLDL+LDL-, and HDL-cholesterol. However, hepatic esterified cholesterol concentrations were increased in the palmitic and stearic arid diet groups compared to the lauric and myristic acid diet group. The molar ratios of hepatic EC/FC were the highest in the palmitic acid diet (12.2 $\pm$0.6) and the lowest in the myristic acid diet (6.4$\pm$0.2). Dietary cholesterol significantly (p<0.001) increased the plasma total cholesterol which was due to the increase of both HDL-and VLDL+LDL-cholesterol. In the absence of dietary cholesterol and compared to other species, the gerbil exhibited a high molar ratio of hepatic EC/FC, which was funker elevated by dietary cholesterol feeding (0.06%). The results from this study indicate that hepatic cholesterol concentrations are sensitive to both low levels of dietary cholesterol and saturated fatty acid chain length and also, that plasma cholesterol concentrations are sensitive to low levels of dietary cholesterol .

This study was designed to investigate the hyperlipidemia-healing effect of liquid extract from Mageum-tang(麻芩湯), it was performed on the hyperlipidemia of rats induced by high cholesterol diet and Triton WR-1339. We prepared two types of hyperlipidemia model in rats induced by high cholesterol diet and Triton WR-1339. The amount of total cholesterol, HDL-cholesterol, LDL -cholesterol, triglyceride, phospholipid, free fatty acid, the activity of superoxide dismutase(SOD), and glutathione were measured. The liquid extract from Mageum-tang showed significant decreasing effects on total cholesterol, LDL- cholesterol, triglyceride, phospholipid, free fatty acid in the both model. And it showed significant increasing effects on HDL-cholesterol, the activity of SOD in both model and amount of glutathione in Triton WR-1339 model. These results suggest that liquid extract from Mageum-tang has healing efficacy on hyperlipidemia induced by high cholesterol diet and Triton WR-1339.
The benefits of milk on the rats supplemented with cholesterol and vitamin D2 in diet was investigated. A total of 150 male rats of Sprague-Dawley strain, 5 weeks of age, were divided into 6 groups with the diet. Cholesterol control group was fed the diet containing 1.0% choleterol and 50,000 IU/100 g of vitamin D2 for 24 months, and M0.5, M1, M2 and M5 of milk groups were fed the diet containing cholesterol, vitamin D2 and 0.5, 1, 2 and 5 of reference daily intakes of whole milk, respectively. No remarkable differences of growth, diet intake, and food effciency ratio among groups were observed though rats in cholesterol control group showed abrupt decrease of diet intake and body weight showed slightly higher relative organ weights than did the rats in control group. The rats in cholesterol control group showed the highest serum total cholesterol level. The rats in milk groups showed lower total cholesterol level and higher high density lipoprotein (HDL)-cholesterol fraction than did the rats in cholesterol control group though no tendency was observed among milk groups. In milk groups, aorta, heart, kidney and liver of rats showed milder calcification and necrosis or fat degeneration compared with those in cholesterol control group. The above results suggest that whole milk could have beneficial effect on the cholesterol-and -vitamin D2-induced atherosclerosis in rats.
