Glutathione S-transferase M1 Null Genotype and Hepatocellular Carcinoma Susceptibility in China and India: Evidence from an Updated Meta-analysis

  • Background: Glutathione S-transferase M1 (GSTM1) have been reported to be associated with hepatocellular carcinoma. However, the effect of the GSTMl null genotype was divergent in the literature and we therefore performed the present meta-analysis to explore the relationship in detail. Materials and Metbods: Reported studies were searched from 1990 to March 1, 2014 in PubMed and Wanfang Med Online. The total odds oatio (OR) and 95% CI were calculated and analyzed by Review Manager 5.1 and STATE 12. Results: Total OR was calculated from 26 articles with 3,769 cases and 5,517 controls and the association proved significant (OR [95%CI]=1.50 [1.25, 1.80], P<0.05) in the Chinese population. However, there was no significant association between hepatocellular carcinoma risk among subjects carrying the GSTM1 null genotype (OR [95%CI]=1.20 [0.88-1.64], P=0.24) in subgroups of publication in English and in Indian populations (OR [95%CI]=1.80 [0.80-4.20], P=0.15). Conclusions: The GSTM1 deletion polymorphism might not have a significant effect on the susceptibility of hepatocellular carcinoma overall.

식이 불포화지방산과 Vitamin E 함량이 흰쥐 간장내의 지질과산화에 미치는 영향 (Effect of Dietary Polyunsaturated Fatty Acid and $\omega$-Tocopherol on Lipid Peroxidation in Rat Liver)

  • High PUFA식이와 vitamin E 함량이 흰쥐 간장내의 지질과산화에 미치는 영향을 알아보기 위하여, 45Kcal% 고불포화 옥수수유의 정상적 vitamin E함량을 가한 식이군(PF군)과 45Kcal% 고불포화 옥수수유의 가한 PF군에 vitamin E를 5배 첨가한 식이군(PFE군), 45Kcal% 고불포화 옥수수유 식이에 vitamin E를 가하지 않은 식이군 (PFO군)을 실험군으로 하여 3주, 6주 및 9주간 사육한 후 간장에서의 vitamin E 함량과 과산화지질량 및 SOD와 GPX활성을 조사하였다. High PUFA식이에 vitamin E를 정상적으로 가한 PF군은 대조군과 차이가 나타나지 않았으나, vitamin E를 충분히 첨가한 식이 PFE군은 식이내 vitamin E 공급이 부족한 PFO군보다 vitamin E 함량과 GPX활성이 증가하였고, SOD활성과 과산화지질향은 현저하게 감소하였다. 식이 투여기간별로 PF군과 PFE군에 비해 PFO군이 실험기간이 경과할수록 vitamin E 함량과 GPX활성이 감소하였으나, SOD활성과 과산화지질향은 현저히 증가하였다.

Anti-oxidative Effect of a Protein from Cajanus indicus L against Acetaminophen-induced Hepato-nephro Toxicity

  • Overdoses of acetaminophen cause hepato-renal oxidative stress. The present study was undertaken to investigate the protective effect of a 43 kDa protein isolated from the herb Cajanus indicus, against acetaminophen-induced hepatic and renal toxicity. Male albino mice were treated with the protein for 4 days (intraperitoneally, 2 mg/kg body wt) prior or post to oral administration of acetaminophen (300 mg/kg body wt) for 2 days. Levels of different marker enzymes (namely, glutamate pyruvate transaminase and alkaline phosphatase), creatinine and blood urea nitrogen were measured in the experimental sera. Intracellular reactive oxygen species production and total antioxidant activity were also determined from acetaminophen and protein treated hepatocytes. Indices of different antioxidant enzymes (namely, superoxide dismutase, catalase, glutathione-S-transferase) as well as lipid peroxidation end-products and glutathione were determined in both liver and kidney homogenates. In addition, Cytochrome P450 activity was also measured from liver microsomes. Finally, histopathological studies were performed from liver sections of control, acetaminophen-treated and protein pre- and post-treated (along with acetaminophen) mice. Administration of acetaminophen increased all the serum markers and creatinine levels in mice sera along with the enhancement of hepatic and renal lipid peroxidation. Besides, application of acetaminophen to hepatocytes increased reactive oxygen species production and reduced the total antioxidant activity of the treated hepatocytes. It also reduced the levels of antioxidant enzymes and cellular reserves of glutathione in liver and kidney. In addition, acetaminophen enhanced the cytochrome P450 activity of liver microsomes. Treatment with the protein significantly reversed these changes to almost normal. Apart from these, histopathological changes also revealed the protective nature of the protein against acetaminophen induced necrotic damage of the liver tissues. Results suggest that the protein protects hepatic and renal tissues against oxidative damages and could be used as an effective protector against acetaminophen induced hepato-nephrotoxicity.

동충하초 추출물과 백삼추출물 투여가 방사선을 조사한 생쥐 간에서의 glutathione함량에 미치는 영향 (The Effect of glutathione contents of White Ginseng(WG) Extracts and Entomopathogenic Fugi Extracts on the liver in Mice that was irradiated by radiation)

  • 본 연구는 선형가속기의 고에너지 엑스선을 조사한 생쥐 간에 대한 백삼과 동충하초의 방사선방호효과를 연구하였다. ICR계 수컷생쥐 군에 7일 동안에 경구적으로 백삼(150 mg/kg/day)과 동충하초(200 mg/kg/day)를 각각 방사선조사 전에 투여했고 다른 생쥐 군에 5 Gy(1.01 Gy/min)의 방사선량으로 전신조사를 했고 대조군에 생리적 식염수 (0.1 ml)를 투여 한 후 간 조직에서 환원형 글루타치온(GSH)과 산화형 글루타치온(GSSG)의 함량을 각각 검사하였다. 그 결과 방사선조사군(Rad)보다 동충하초투여군(EF+Rad)과 백삼투여군(WG+Rad)에서 환원형 글루타치온 (GSH)함량이 유의성 있게 증가했으나 산화형 글루타치온(GSSG)의 함량은 유의성 있게 감소하였다. 총 환원형 글루타치온(total GSH)과 산화형 글루타치온(GSSG)의 함량 비율은 방사선조사군(Rad)보다 동충하초투여군(EF+Rad)과 백삼투여군(WG+Rad)에서 유의성 있게 감소하였다.

CD26: A Prognostic Marker of Acute Lymphoblastic Leukemia in Children in the Post Remission Induction Phase

  • Background: ALL is an irredeemable disease due to the resistance to treatment. There are several influences which are involved in such resistance to chemotherapy, including oxidative stress as a result of the generation of reactive oxygen species (ROS) and presence of hypodiploid cells. Cluster of differentiation 26 (CD26), also known as dipeptidyl peptidase-4, is a 110 kDa, multifunctional, membrane-bound glycoprotein. Aim and objectives: The aim of this study was to evaluate the clinical significance of serum CD26 in patients with acute lymphoblastic leukaemia patients in the post remission induction phase, as well as the relationship between CD26 activity and the oxidative stress status. Materials and Methods: CD26, total antioxidant status (TAS), total oxidant status (TOS), and oxidative stress index (OSI), in addition to activity of related enzymes myeloperoxidase, glutathione-s-transferase and xanthine oxidase, were analysed in sixty children with acute lymphoblastic leukaemia in the post remission induction phase. Results: The study showed significant elevation in CD26, TOS and OSI levels in patients with acute lymphoblastic leukaemia in the post remission induction phase in comparison to healthy control samples. In contrast, myeloperoxidase, glutathione-s-transferase and xanthine oxidase activities were decreased significantly. A significant correlation between CD26 concentration and some oxidative stress parameters was evident in ALL patients. Conclusions: Serum levels of CD26 appear to be useful as a new biomarker of oxidative stress in children with acute lymphoblastic leukaemia in the post remission induction phase, and levels of antioxidants must be regularly estimated during the treatment of children with ALL.

한라산 구상나무 건전개체와 쇠약개체의 항산화효소활성 및 토양특성 (Antioxidant Enzyme Activities and Soil Properties of Healthy and Declining Abies koreana (Wils.) in Mt. Halla)

  • 구상나무의 스트레스 적응능력을 보여주는 항산화효소활성, 건전개체와 쇠약한 개체의 토양의 물리화학적 특성을 알아보기 위하여 윗세오름, 영실 그리고 성판악(진달래밭)지역의 건전개체와 쇠퇴개체를 조사 분석하였다. 항산화효소활성으로는 Ascorbate peroxidase(APX)와 Glutathione Reductase(GR)를, 토양특성으로는 토성, pH, 유기물함량, 전 질소함량, 유효인산, 양이온치환용량, 양이온의 함량을 분석하였고, 구상나무 잎을 채취하여 식물체의 양분함량을 분석하였다. APX, GR 효소 활성에 있어서 건전개체와 쇠약한 개체 사이에는 큰 차이가 없었으나, 계절적으로 6월과 8월에 비해 9월의 항산화효소 활성도가 낮게 나타났다. 토양분석 결과 유기물함량, 전질소, 유효인산, 양이온치환용량, 양이온 함량이 건전개체가 있는 곳에 비해 쇠약한 개체가 있는 곳의 토양이 낮게 나타났다.

Galactosamine에 의해 유도된 녹각추출물이 간장해에 미치는 영향 (The Effect of old Antler on the Galactosamine-induced Hepatotoxicity in Rate)

  • 간장해에 미치는 녹각의 효과를 밝힐 목적으로 녹각을 성분별로 추출 분리하여 급여하고 galactosamine으로 간장해를 유도한 후 녹각추출물이 간기능 유지에 관여하는 효소활성 및 지질 대사에 미치는 영향을 생화학적 측면에서 고찰하였다. Cytochrome P-450은 녹각추출물 급여로 증가하였으며 갈락토사민 투여로 감소하였으나 neutral-ext.군에서 높은 증가를 보였다. Glutathione peroxidase 활성은 갈락토사민 투여로 감소하였으나, 녹각 추출물 급여로 활성이 증가하였다. 간조직의 글루타티올 함량은 녹각추출물 급여로 유의적 변동이 없었으며 갈락토사민 투여로 감소하였다. 과산화지질 함량은 대조군에 비해 각각의 녹각추출물 급여군에서 감소하여 특히 water-ext. 군에서 감소정도가 가장 현저하였다. 간조직중의 총 지방질, 총 콜레스테롤 및 트리글리세리드의 함량은 대조군에 비해 각각의 녹각추출물 급여군에서 감소하였으며, water-ext.군에서 가장 많이 하게 감소하였다. 이상의 결과로 미루어 볼 때 녹각의 water-ext.과 neutral-ext.에 함유되어 있는 성분이 간기능 유지에 관여하는 효소합성 증가를 통해 활성증가를 유도하고, 이로써 독성의 대사를 촉진시키므로 간독성을 예방할 수 있을 것으로 생각되며, 이와 같은 녹각추출물의 작용이 생체에 흡수되어진 xenobiotics의 해독작용과 연관될 것으로 생각된다.

대시호탕(大柴胡湯)이 alloxan으로 유발(誘發)된 고혈당(高血糖) 백서(白鼠)에 미치는 영향(影響) (Effects of Daesihotang and its component groups on diabetes, free radical and antioxidative defense system in alloxan induced diabetic rats)

  • The purpose of this study was to reseach the effect of Daesihotang and its component groups on diabetes, free radical and antioxidative defense system in alloxan-induced diabetic rats. The experimental group was divided into three groups: Daesihotang (DS), Soyangyak (DS1), Yangmyungyak (DS2). The results were obtained as follows: The level of glucose, triglyceride, total cholesterol, creatinine in serum were considerablely reduced by Daesihotang. And the level of HDL cholesterol, albumin, total protein in serum were increased by Daesihotang significantly. And the level of BUN has some decreased, but with no significancy. In the study of Daesihotang on free radical scavenging effect in vitro, it has shown that Daesihotang and its components group have significant suppressing effect on peroxidation of linoleic acid on concentration. And in other experiments as scavenging effect of DPPH radical, inhibitory effect of superoxide in xanthine-xanthine oxidase system and inhibitory effect on lipid peroxidation reaction by hydroxy radical in $H_2O_2-Fe^{2+}$ system, Daesihatang have some activity, but with no significancy. In the study of Daesihotang on antioxidative defense system in vivo, the activity of GOT and GPT in serum were significantly increased; and the amounts of lipid peroxide in serum was reduced significantly but in liver no significancy. In hepatic catalase activity, Daesihotang has showed significant effect. In the level of hepatic glutathione, Daesihotang increased the amount of glutathione significantly. And in the activity of glutathione-s-transferase, Daesihotang has decreasing effect but has no significancy. These result suggest that Daesihotang has strong effect on diabetes and it is useful to prevent diabetes, but has less effect on peroxidative damage by free radical.

Antioxidant Effects of Fermented Red Ginseng Extracts in Streptozotocin-Induced Diabetic Rats

  • The antioxidant activities of fermented red ginseng (FRG) were investigated in vitro and in vivo. The contents of total polyphenol and total flavonoid in FRG extracts were $17.01{\pm}2.00$ ${\mu}g/mg$ and $18.42{\pm}3.97$ ${\mu}g/mg$, respectively. These extracts were capable of directly scavenging ${\alpha}$, ${\alpha}$-diphenyl-picrylhydrazyl free radicals. The antioxidative effects of the FRG extracts in streptozotocin (STZ)-induced diabetic rats were also investigated. The activities of plasma alanine transaminase, aspartate transaminase, and ${\gamma}$-glutamyltransferase were significantly decreased by extract administration as compared to an STZ control group. Hepatic glutathione content depleted by STZ treatment was significantly increased by treatment of the FRG extracts, but the elevation of lipid peroxide content induced by STZ was significantly decreased by the extracts. Activities of superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase decreased after STZ-treatment were recovered by the treatment of the FRG extracts. These results indicate that FRG extracts have antioxidative effets in STZ-induced diabetic rats.

타우린보강이 건강한 성인영성의 혈중 항산화효소 활성과 지질과산화물 농도에 미치는 영향 (Effects of Oral Taurine Supplementation on Blood Antioxiant Enzyme Activities and Lipid Peroxidation in Healthy Female Adults)

  • Effects of oral taurine supplementation (6g/day)for 2-4 weeks on activities of red blood cell(RBC)total superoxide dismutase (SOD) and plasma glutathione peroxidase(GSH-Px) and the level of malondialdehyde(MDA) were evaluated in healthy female adults (23.6$\pm$0.3 years old). Compared to the value for 0 week plasma GSH-Px activity of the subjects was significantly lower after 2 weeks of taurine supplementation(p<0.05) and recovered to the value similar to 0 week after 4 weeks of taurine supplementation. RBC total SOD activity tended to be decreased after 2 weeks of taurine supplmentation compared to the values for 0 week although the difference between the means of the two group was not statistically significant. Plasma MDA level was not significantly decreased by taurine supplementation most probably due to the fact that the subjects participated in the present study were healthy and their antioxidant defense system had been in the 'normal' range. Plasma MDA concentration was negatively correlated with plasma taurine concentration(r=-0.2003m p<0.05) but tended to be positively correlated with plasma cholesterol concentration(r=0.2465, p=0.0645) as expected Plasma GSH-Px activity was positively associated with the percentage of 22:0 (r=0.2892, p<0.05) or 20:4w6(r=0.2939, p<0.05). On the other hand plasma MDA concentration was positively correlated with the percentage of 20:5w3 in plasma total lipids(r=0.2635 p<0.05) and negatively correlated with $\Delta$5 desaturation index of w6 fatty acids(20:3w6⇒20:4w6) in plamsa total lipids(r=-0.2714, p<0.05) as well as in phospholipids(r=-0.2864, p<0.05). From these results protective effect of taurine supplementation against lipid peroxidation and antioxidant defense system in humans appears to be minimal when the subjects are in a relatively healthy state. Further studies concerning the antioxidant efficacy of taurine should be conducted in human subjects under various disease states related to oxidative stress such as diabetes and artheroxclerosis.
