Sequencing of cDNA Clones Expressed in Adipose Tissues of Korean Cattle

  • To understand the molecular mechanisms that regulate intramuscular fat deposition and its release, cDNA clones expressed in adipose tissues of Korean cattle were identified by differential screening from adipose tissue cDNA library. By partial nucleotide sequencing of 486 clones and a search for sequence similarity in NCBI nucleotide databases, 245 clones revealed unique clones. By a functional grouping of the clones, 14% of the clones were categorized to metabolism and enzyme-related group (stearoyl CoA desaturase, lactate dehydrogenase, fatty acid synthase, ATP citrate lyase, lipoprotein lipase, acetyl CoA synthetase, etc), and 6% to signal transduction/cell cycle-related group (C/EBP, cAMP-regulated phosphoprotein, calmodulin, cyclin G1, cyclin H, etc), and 4% to cytoskeleton and extracellular matrix components (vimentin, ankyrin 2, gelosin, syntenin, talin, prefoldin 5). The obtained 245 clones will be useful to study lipid metabolism and signal transduction pathway in adipose tissues and to study obesity in human. Some clones were subjected to full-sequencing containing open reading frame. The cDNA clone of bovine homolog of human prefoldin 5 gene had a total length of 959 nucleotides coding for 139 amino acids. Comparison of the deduced amino acid sequences of bovine prefoldin 5 with those of human and mouse showed over 95% identity. The cDNA clone of bovine homolog of human ubiquitin-like/S30 ribosomal fusion protein gene had a total length of 484 nucleotides coding for 133 amino acids. Comparison of the deduced amino acid sequences of bovine ubiquitin-like/S30 ribosomal fusion protein gene with those of human, rat and mouse showed over 97% identity. The cDNA clone of bovine homolog of human proteolipid protein 2 mRNA had a total length of 928 nucleotides coding for 152 amino acids. Comparison of the deduced amino acid sequences of bovine proteolipid protein 2 with those of human and mouse showed 87.5% similarity. The cDNA clone of bovine homolog of rat thymosin beta 4 had a total length of 602 nucleotides coding for 44 amino acids. Comparison of the deduced amino acid sequences of bovine thymosin beta 4 gene with those of human, mouse and rat showed 93.1% similarity. The cDNA clone of bovine homolog of human myotrophin mRNA had a total length of 790 nucleotides coding for 118 amino acids. Comparison of the deduced amino acid sequences of bovine myotrophin gene with those of human, mouse and rat showed 83.9% similarity. The functional role of these clones in adipose tissues needs to be established.

Studies on the Contents of Free Amino Acids, Organic Acids and Isoflavones in Commercial Soybean Paste (시판된장의 유리아미노산, 유기산 및 Isoflavone의 함량에 관한 연구)

  • In order to establish the quality standard of commercial soybean paste, pH, color, moisture, crude protein, isoflavone, free amino acid and organic acid contents of the fermented soybean food in both traditional and commercial products were analyzed. The crude protein contents were from 5.0 to 11.9% in commercial soybean pasted of Samjang and traditional soybean paste of Chungugjang, and the latter showed higher value than those of the former. The contents of isoflavones in commercial soybean paste were less than those of traditional soybean paste. Total contents of free amino acids were distributed from 301 to 4,602mg%, respectively, in which Chungugjang showed the smallest value of 301mg% and the traditional soybean paste showed the most abundant one of 4,602mg%. The ratio of glutamic acid to total free amino acids were 15.9% and 30.1% in traditional soybean paste and commercial soybean paste, respectively. The major organic acid both commercial soybean paste and Samjang was citric acid containing 109.9~196.5mg%. On the other hand, that of traditional soybean paste and Chungugjang was oxalic acid containing 82.8~130.1mg%. This results might be caused by the differences of brewed periods, microflora, processing methods and used raw materials.

  • Methods developed for measuring digestibility and availability of amino acids in feedstuffs used in pig nutrition are reviewed. Digestibility is a proportion of an amino acid in a feed that is absorbed from the digestive tract and should be determined from the difference between the amount of amino acid consumed and passing the distal ileum. Techniques for ileal digesta sampling including various types of cannulas: a re-entrant, T-piece, IPV, IPVC and ileaorectal anastomosis are described and comparisons amongst these methods are presented. Other methodologies like mobile bag technique, in vitro assays and mathematical prediction method are also described. Significance and methodologies for measurement of endogenous nitrogen and amino acids losses at the distal ileum and their effect on the apparent and true nitrogen and amino acid digestibilities in feeds are discussed. Factors influencing the apparent and true amino acid digestibilities such as dry matter intake, protein, fibre and antinutritive compounds content in the diet are discussed. Amino acid bioavailability -the proportion of the total amino acid digested and absorbed in a form utilized in metabolism - measured by the growth assay may differ from its ileal digestibility. Chemical methods for determination of available lysine content in heat treated feeds are evaluated.

The Components of the Fermented Soy Sauce from Gorosoe and Bamboos SaP (고로쇠 및 대나무 수액간장의 성분조성)

  • Soy sauce was prepared with the addition of Gorosoe and bamboos sap instead of tap water to make ordinary soy sauce, respectively. The changes of such compounds during fermentation as minerals, free sugars, organic acids and free amino acids were analyzed. Total mineral contents in the 90 days fermented soy sauce from Gorosoe and bamboos sap were increased by 2.0 and 4.4 times as compared with those of control samples, respectively. Especially the increase of calcium, potassium and magnesium in soy sauce of sapes were derived from those of saps. The sugars were found to be only glucose and galactose in control samples, but they were detected not only above sugar but also fructose and sucrose in soy sauce of saps. Butyric acid in detected organic acid were dominant in all stray sauce samples. In the free amino acid composition of control samples, leucine, phenylalanine, isoleucine, lysine and glutamic acid were abundant amino acids. Amino acid such as isoleucine, leucine and phenylalanine were decreased, the rest 11 kinds of amino acids containing phosphoserine were increased during the fermentation of control samples. Increasing and decreasing patterns of free amino acids during fermentation of sap soy sauce were similar to control samples. Total nitrogen, amino acid nitrogen and ammonia nitrogen were increased during their fermentation.

Age-Related Changes in Sulfur Amino Acid Metabolism in Male C57BL/6 Mice

  • Alterations in sulfur amino acid metabolism are associated with an increased risk of a number of common late-life diseases, which raises the possibility that metabolism of sulfur amino acids may change with age. The present study was conducted to understand the age-related changes in hepatic metabolism of sulfur amino acids in 2-, 6-, 18- and 30-month-old male C57BL/6 mice. For this purpose, metabolite profiling of sulfur amino acids from methionine to taurine or glutathione (GSH) was performed. The levels of sulfur amino acids and their metabolites were not significantly different among 2-, 6- and 18-month-old mice, except for plasma GSH and hepatic homocysteine. Plasma total GSH and hepatic total homocysteine levels were significantly higher in 2-month-old mice than those in the other age groups. In contrast, 30-month-old mice exhibited increased hepatic methionine and cysteine, compared with all other groups, but decreased hepatic S-adenosylmethionine (SAM), S-adenosylhomocysteine and homocysteine, relative to 2-month-old mice. No differences in hepatic reduced GSH, GSH disulfide, or taurine were observed. The hepatic changes in homocysteine and cysteine may be attributed to upregulation of cystathionine ${\beta}-synthase$ and down-regulation of ${\gamma}-glutamylcysteine$ ligase in the aged mice. The elevation of hepatic cysteine levels may be involved in the maintenance of hepatic GSH levels. The opposite changes of methionine and SAM suggest that the regulatory role of SAM in hepatic sulfur amino acid metabolism may be impaired in 30-month-old mice.

Analysis of Minerals, Amino Acids,and Vitamin of Lespedeza cuneata (비수리의 미네랄, 이미노산, 비타민 분석)

  • For developing functional biomaterials, chemical analysis of Lespedeza cuneata including minerals, amino acids and vitamin were investigated. Minerals of L. cuneata were found to be calcium, potassium, magnesium, phosphorus, sodium, iron, manganese, zinc and copper. Among the free amino acid, proline was 33.77 mg% that were 67.2% of free amino acids and essential amino acids were 7.49 mg%. Total amino acids were analyzed as 2,817 mg% and the content of glutamic acid (496.00 mg%) was highest. In case of vitamin, the highest components was vitamin E with 33.03 mg%.

Chemical Composition and Bioactivity of Korean Green Tea (Camellia sinensis) Pollen collected by Honey Bee (한국 녹차 화분의 화학적 조성과 기능성 연구)

  • In this study, we investigated the nutritional composition including proximate, amino acid, vitamin, minerals, and the antioxidant activity of green tea (Camellia sinensis) pollen grains collected by Apis mellifera bees, for use as a health food. The crude protein and fat content was estimated at 26.14% and 3.49%, respectively. Eighteen amino acids were identified in green tea pollen, including 8 essential amino acids and 10 non-essential amino acids. The predominant amino acids were glutamic acid, proline and aspartic acid accounting for about 33.3% of total free amino acids. The concentration of vitamin C was the highest value of 35.7%, followed by $B_3$ and $B_2$ among the detected vitamins. The predominant minerals were potassium (790.32 mg/100g), followed by phosphorus (707.52 mg/100g) and sulfur (302.67 mg/100g), whereas copper, zinc and sodium were detected as minor elements. The antioxidant activity and phenolic content accounted for 33.8% at $500{\mu}g/mL$ extract and $2.55 {\mu}g/mg$, respectively.

Chemical Composition of Glycoprotein from Urechis unicinctus (개불(Urechis unicinctus) 당단백질의 성분조성과 특성)

  • To confirm the food quality of Urechis unicinctus which have been favored as a special raw seafood in southern area in Korea, the chemical composition of Urechis unicinctus and those glycoprotein were studied. Freeze dried Urechis unicinctus was composed of more than 70% of crude protein and 15% of total carbohydrate. The amino acids such as glycine(18.69%), glutamic acid(12.50%) and aspartic acid(9.37%) were noted as major components of total amino acids. The predominant free amino acids were alanine(32.98%), glycine(27.50%), asparagine(19.65%) and taurine(8.29%), and the sum of them were more than 88% to total free amino acids, so they may cause unique taste of Urechis unicinctus with sweetness. As the basis of sugar composition analysis, 56.35% of glucose and 30.49% of N acetylglucosamine were contained respectively, and they might also play an important role in a sweet taste. The leading carbohydrate moiety of glycoprotein from Urechis unicinctus was identified as glucose and N acetylglucosamine similarly to raw muscle, and they occupied more than 50% of total sugar content. Fucose(18.32%) and D glucuronic acid(12.31%) also detected in higher levels com pared to raw muscle. The total amino acid profile of glycoprotein showed a similar trend to raw muscle protein but glycine was detected a significantly lower than that in raw muscle. The glycoprotein from Urechis unicinctus was composed of 4 kinds of subunits, i.e. 25kDa, 20kDa, 18kDa and 12.5kDa of molecular weights through the SDS polyacrylamide gel electrophoresis. On the basis of the IR spectrum of absorptions appeared in 1,240cm-1 and 850cm-1, the glycoprotein had sulfate groups.

  • A series of experiments was conducted to determine the contents and distribution of amino acids in the hydrolysates of humic acids extracted from 3 plant materials. Wild grass hay, and leaves of forest trees were used in this study. Seventeen amino acids were analyzed and their amounts determined. Results obtained from the experiments are summarized in the following: 1. Contents and distribution of hydrolyzable amino acids in the humic acid fractions depend on the kind of plant materials and the allowed time for humification. 2. Neutral amino acids was the largest part of the total amino acids, followed by acidic amino acids, and basic amino acids. 3. The total amounts of amino acids in the hydrolyzable humic acid fractions of well humified residues were in the following order: wild grass hay > leaves of deciduous trees > leaves of coniferous trees 4. In general the relative amounts of lysine increased with humification progressing. S. Glycine and glutamic acid were the two major amino acids in common for the hydrolysate of humic acids extracted from well decomposed residues of plant materials. Alanine, glutamic acid, glycine, aspartic acid and leucine were the five major amino acid in common in raw materials without exception. 6. Arginine appeared to be absent in any of the hydrolysates of humic acids from well humified plant materials. 7. Phenylalanine and tyrosine were present in all hydrolysates and their relative contents increased with the humification of plant materials.

Analysis of the Change of Amino Acids by Abscisic acid and Polyamine Treatment in Radish Young Cotyledons (무우 유식물의 자엽에서 Abscisic acid와 Polyamine 처리에 의한 아미노산 변화의 분석)

  • The changes of amino acids in the cotyledons of spring - and winter radishes were analysed during the abscisic acid (ABA). ABA plus cycloheximide (CH) and ABA plus polyamine (PA) treatment. The total contents of amino acids were increased in spring radishes, and decreased in winter radishes by ABA. The contents of hydrophilic amino acids, proline, glycine, serine and cysteine in spring radishes (expecially proline), and of cysteine, leucine and phenylalanine in winter radishes (expecially cysteine and leucine)were increased, CH treatment resulted in the accumulation of amino acids by the inhibition of new synthesized protein, which synthesized against ABA induced dehydration both of radishes. On the contrary, the contents of amino acids were decreased in spring radishes, and increased in winter radishes during PA treatment. These results indicated that the accumulated hydrophilic amino acids and new synthesized proteins induced the adaptation of dehydration against stress and the role of ABA in accelerating of stress adaptation was mediated by polyamines.
