C57BL6 생쥐에서 NDM 헤어토닉의 모발성장 촉진 효과 (Effect of NDM Hair Tonic on Hair Growth Promotion in C57BL6 Mice)

  • NDM hair tonic is composed of several plant extracts which are known to be used in oriental medicine. This study was carried out to investigate the effect of NDM hair tonic on hair regrowth in an alopecia model of C57BL6 mice. Hair of six-weeks old mice were trimmed by electric clippers and electric shavers, so as not to damage the skin. The next day, mice without visible scraches were selected, randomized and separated in groups of 6 mice. There were four experimental groups including saline (negative control), 50% ethanol (vehicle control), 3% minoxidil (MXD), and NDM tonic. The test compounds were topically treated with 0.15 $m\ell$ per mouse per day for 2 weeks. The hair regrowth was determined photographically and histologically and the quantity of endocrine factors, IGF-1 and TGF-$\beta$, in the skin of mice was measured by PCR. No clinical signs were found in all animals. The topical treatment of NDM tonic for 2 weeks to dorsal skin accelerated hair regrowth faster than either the controls or MXD treatment. The NDM tonic treatment also promoted hair follicle development and elongation as compared with the controls or MXD treatment. Both NDM tonic and MXD treatment significantly increased the expression of IGF-1 and TGF-$\beta$ in the skin of C57BL6 mice as compared with the controls (p<0.05). These results suggest that NDM tonic has a hair growth activity and be useful for the treatment of baldness or alopecia.

Effects of Anticonvulsants on Acute and Tonic Pains in the Rat

  • Different neural substrates have been reported to be implicated in analgesic mechanisms in the acute phasic and the sustained tonic pains. To explore the differential antinociceptive action of diphenylhydantoin (DPH) and carbamazepine (CBZ) on the acute phasic and the tonic pains, changes in tail flick latency, hot plate latency and the formalin-induced nociceptive score were assessed prior to and after intraperitoneal administration of DPH (20 & 40 mg/Kg) and CBZ (20 mg/Kg). In 11 rats, CBZ was administered repeatedly for 6 days at the dose of 20 mg/Kg/day. Also studied were the effects of strychnine and picrotoxin (1 mg/Kg, i.p.) on the CBZ-produced changes in the formalin-induced pain behaviors. The tail flick and hot plate ltencies were not changes after administration of DPH and CBZ. However DPH strongly suppressed the formalin-induced tonic pain. A single and the repeated administration of CBZ inhibited both the early phasic and the late tonic pain responses to formalin in n similar manner. On the other hand, the antinociceptive actions of CBZ were not altered by strychnine or picrotoxin. These experimental findings lead to the conclusion that DPH and CBZ have differential antinociceptive action on the acute and the tonic pains and that their antinociceptive actions are independent of the GABA- and glycine-receptors.

Developmental changes in GABAA tonic inhibition are compromised by multiple mechanisms in preadolescent dentate gyrus granule cells

  • The sustained tonic currents ($I_{tonic}$) generated by ${\gamma}$-aminobutyric acid A receptors ($GABA_{A}Rs$) are implicated in diverse age-dependent brain functions. While various mechanisms regulating $I_{tonic}$ in the hippocampus are known, their combined role in $I_{tonic}$ regulation is not well understood in different age groups. In this study, we demonstrated that a developmental increase in GABA transporter (GAT) expression, combined with gradual decrease in $GABA_AR{\alpha}_5$ subunit, resulted in various $I_{tonic}$ in the dentate gyrus granule cells (DGGCs) of preadolescent rats. Both GAT-1 and GAT-3 expression gradually increased at infantile ($P_{6-8}$ and $P_{13-15}$) and juvenile ($P_{20-22}$ and $P_{27-29}$) stages, with stabilization observed thereafter in adolescents ($P_{34-36}$) and young adults ($P_{41-43}$). $I_{tonic}$ facilitation of a selective GAT-1 blocker (NO-711) was significantly less at $P_{6-8}$ than after $P_{13-15}$. The facilitation of $I_{tonic}$ by SNAP-5114, a GAT-3 inhibitor, was negligible in the absence of exogenous GABA at all tested ages. In contrast, $I_{tonic}$ in the presence of a nonselective GAT blocker (nipecotic acid, NPA) gradually decreased with age during the preadolescent period, which was mimicked by $I_{tonic}$ changes in the presence of exogenous GABA. $I_{tonic}$ sensitivity to L-655,708, a $GABA_AR{\alpha}_5$ subunit inverse agonist, gradually decreased during the preadolescent period in the presence of NPA or exogenous GABA. Finally, Western blot analysis showed that the expression of the $GABA_AR{\alpha}_5$ subunit in the dentate gyrus gradually decreased with age. Collectively, our results suggested that the $I_{tonic}$ regulation of altered GATs is under the final tune of $GABA_AR{\alpha}_5$ subunit activation in DGGCs at different ages.

Phasic and Tonic Inhibition are Maintained Respectively by CaMKII and PKA in the Rat Visual Cortex

  • Phasic and tonic ${\gamma}$-aminobutyric acidA ($GABA_A$) receptor-mediated inhibition critically regulate neuronal information processing. As these two inhibitory modalities have distinctive features in their receptor composition, subcellular localization of receptors, and the timing of receptor activation, it has been thought that they might exert distinct roles, if not completely separable, in the regulation of neuronal function. Inhibition should be maintained and regulated depending on changes in network activity, since maintenance of excitation-inhibition balance is essential for proper functioning of the nervous system. In the present study, we investigated how phasic and tonic inhibition are maintained and regulated by different signaling cascades. Inhibitory postsynaptic currents were measured as either electrically evoked events or spontaneous events to investigate regulation of phasic inhibition in layer 2/3 pyramidal neurons of the rat visual cortex. Tonic inhibition was assessed as changes in holding currents by the application of the $GABA_A$ receptor blocker bicuculline. Basal tone of phasic inhibition was maintained by intracellular $Ca^{2+}$ and $Ca^{2+}$/calmodulin-dependent protein kinase II (CaMKII). However, maintenance of tonic inhibition relied on protein kinase A activity. Depolarization of membrane potential (5 min of 0 mV holding) potentiated phasic inhibition via $Ca^{2+}$ and CaMKII but tonic inhibition was not affected. Thus, phasic and tonic inhibition seem to be independently maintained and regulated by different signaling cascades in the same cell. These results suggest that neuromodulatory signals might differentially regulate phasic and tonic inhibition in response to changes in brain states.

  • 본원 입원 치료받은 환자 1례의 경과는 다음과 같다. 1. 사물탕 가감방 복용 이후 tonic spasm의 발생빈도와 기타 임상증상의 변화가 없었다. 2. 육미지황탕 복용 이후 tonic spasm의 발생빈도가 1일 30회 이상에서 점차 감소하여 일주일간 복용 이후 10회 이하로 증상 발생되었으며 42일 복용 이후에는 0-1회로 발생빈도가 감소하였다. 3. 육미지황탕과 침구치료를 받는 동안 안혼(眼昏), 안삽(眼澁), 복근경직(腹筋硬直), 흉민(胸悶) 등의 증상이 소실되었다.

Charaterization of Ginsenosides-induce d Antinociception in Mice

  • We have investigated the antinociceptive efficacy of ginseng saponins in mice using l% formalin, which induce two phases of pain (acute and tonic pains) and is known to induce a clinically related pain. Ginseng total saponins (GTS) relieved both phases of pain with EDso of 162 mghg for acute and 92 mg/kg for tonic pain, respectively. Both protopanaxadiol (PD) and protopanaxatriol (PT) saponins did not attenuated acute phase of pain but relieved tonic phase of pain with EDso of 45 mg/kg for PD saponins and 105 mghg for PT saponins, respectively. Moreover, ginsenoside Rc, Rd, and Re among representative ginsenosides such as Rbl, Rc, Rd, Re and Rgl relieved slightly but significantly acute phase of pain and strongly attenuated tonic phase of pain but Rf relieved only tonic phase of pain. However, PD and PT saponins, and the individual ginsenosides tested except GTS did not greatly attenuate thermal noxious pain (tail-flick test). These results suggest that single ginsenoside or mixture of various ginsenosides mainly induce differential antinociception in mice.

사회.심리적 스트레스에 미치는 마인드 토닉(Mind Tonic) 음료의 영향 (Effects of Mind Tonic Drinks on Sociopsychological Stress)

  • The effects of anti-stress mind tonic (ASMT) drink (ASMT-0.1% and ASMT-0.5% concentration) were tested for the anti-stress action. ICR male mice (20$\pm$2 g) were fed with basic experimental diets free access to water containing these ingredients for 18 days. Psychological stress/sociopsychological stress were exposed by foot-shock for 1 hour for 3 days. Both ASMT-0.1 and ASMT-0.5 groups in the sociopsychological stress resulted in a significant decrease of 19.1% and 41.9% in plasma corticosterone (CS) secretion compared with psychological stress (control group). Noradrenaline (NA) secretions in the brain were significantly increased 23.4% and 35.9%, respectively, in ASMT-0.1 and ASMT-0.5 groups compared with control group. MHPG-SO4 (3-Methoxy-4-hydroxyphenylethyleneglycol sulfate) levels in the brain resulted in a marked decreases of 27.1% and 19.6%, respectively in ASMT-0.1 and ASMT-0.5 groups. NA/MHPG-SO4 ratios in the brain of ASMT-0.1 and ASMT-0.5 groups resulted in a significantly increase of about 70% compared with control group. These suggest that mind tonic anti-stress drink can effectively ridded the sociopsychological stress.

칼슘이 결장뉴 K-경축에 미치는 영향 (Effect of Calcium Ion on K-contracture in Guinea Pig's Taenia Coli)

  • The in vitro experiments for isometric contraction were done to investigate the different action mechanism of calcium ion on phasic and tonic components of K-contracture in guinea pig's taenia coli. The results were as follows: 1) The degrees of K-contracture were increased gradually from 4 mM to 15, 20, 40 & 100 mM of$[K^+]_0$. The maximal developed tension in tonic component was observed in 100 mM of $[K^+]_0$. 2) The phasic components were not so affected by 2, 4, 8 & 16 mM of $[Ca^{++}]_0$ in 100 mM K-contracture, but the tonic components were gradually increased in a dose-responsive manner. 3) The K-contracture was not influenced by norepinephrine, 1 mg/l, but was completely abolished by verapamil, 2 mg/l. 4) The phasic component was little affected by verapamil, 0.01 mg/l, 0.1 mg/l, and 1 mg/l, but the tonic component was completely disappeared at the concentration of verapamil 1 mg/l. The above results suggest that $[Ca^{++}]_0.$ primarily affected the tonic component than the phasic component of K-contracture in guinea pig's taenia coli.

  • Objectives : This study was carried out to examine the change of scalp and hair condition and to evaluate the efficacy of hair growth, subject's satisfaction and safety of the Narasoo healing shampoo and Hair tonic Methods : We offered 25 subjects Narasoo healing shampoo and Hair tonic and observed the changes among the starting point, 3rd week and 6th week. Study methods include the measurement of water and sebum content of the scalp, photo of scalp and hair, thermographic images and questionnaires. Results : It is estimated that Narasoo healing shampoo and Hair tonic have the positive effects on increase of water contents hair gloss, removal of excessive hair keratin, facilitation of hair growth, and decrease of sebum contents. Conclusions : Several meaningful effects of Narasoo healing shampoo and Hair tonic are observed in this study. Additional research is needed forward.

한방추출 천연혼합물의 탈모 유발 쥐에 대한 발모 효과 (Hair Growth Effect of Herbal Extract Mixture Ahn Tonic in Mice)

  • This paper aims to assess the effect of herbal extract mixture, Ahn Tonic, on hair growth and examine the stability of this percutaneous hair growth ointment. The hair on the back of the mice C57b1/6N was removed, and 1% of the TXN(testosterone) was then applied for a week to prevent the hair growth. The experimental group was then treated with Ahn Tonic, 0.2 mL per day. The degree of newly grown hair was observed with a vernier caliper. We also measured the proportion of the newly growing hair section to the entire shaved section in the 4th week and 8th week by distinguishing the section turning black from the shaved area. To observe the effect of the test chemical product on hair follicles and hair roots, the biopsy was executed between week 4 and week 8. Gene expressions, which operate as a factor for growing hair in the skin tissues extracted from each experimental animals, were also observed through a real-time PCR gene amplification method. The results showed that the Ahn tonic group had statistically significant hair restoring effect compared to the control group in terms of microscopy, biopsy, and gene expressions. Ahn Tonic is considered to have an impact on the hair growth.
