This study was conducted to investigate the effect of U. atrum treatment on control of Botrytis leaf blight of lily and Botrytis gray mold of cucumber, and to evaluate the U. atrum as the biological control agent of Botrytis diseases. The antagonistic isolates CNU 9037 and CNU 9054 isolated from tomato leaves were identified as Ulocladium atrum Preuss based on morphological characteristics. This is the first record of U. atrum in Korea. In bioassays on dead leaves of tomato and cucumber, treatment of U. atrum colonized the dead leaves and suppressed sporulation of Botrytis as compared with the untreated control. The suppression of spoulation of Botrytis on dead leaf segments by U. atrum was higher when U. atrum was treated before Botrytis was treated. The effect of treatments with conidial suspension of U. atrum on leaf blight of lily and gray mold of cucumber caused by Botrytis elliptica and B. cinerea, respectively, was investigated under greenhouse conditions. Spraying U. atrum ($1{\times}10^6$ conidia per ml) at intervals of 1 week for three times resulted in a significant reduction of natural infections of lily leaves caused by B. elliptica. Protective value of U. atrum treatment was higher than that of the fungicide (procymidone) treatment. Spraying U. atrum also resulted in a significant reduction of cucumber gray mold caused by B. cinerea. Our results show that U. atrum has a potential for biological control against diseases caused by Botrytis spp, in lily and cucumber.
Effects of Zn, P and Fe on Cd uptake and accumulations by tomato (Lycopersicum esculentum Mill) and also their interactions on the uptake of Zn, Fe, Mn, P and Cd were investigated using batch type solution culture technique. Experiment 1 was a factorial scheme with 3 levels of Zn (0, 0.5, 2.5 ppm) and 3 levels of Cd (0, 0.2, 1.0 ppm). At 1.0 ppm Cd, significant yield reduction of dry matter and visual toxicity symptoms (yellowing and necrosis) of Cd was observed for all zinc levels. At this Cd level, increasing Zn treatment from 0 to 2.5 ppm increased Cd concentration from 199 to 235 ppm in leaves and from 124 to 145 ppm in stems. Similarly, Cd treatment did not suppress Zn uptake in leaves, and rather significantly increased in stems. Fe concentrations in leaves and stems were significantly reduced due to Cd treatment while Mn were increased by both Zn and Cd treatment. The results of experiment 2 with 3 levels of P (0.5, 2.0, 4.0m Mol) and 3 levels of Cd (0, 1.0, 2.0 ppm) in a factorial scheme also showed a growth reduction and visual toxic symptons from 1.0 ppm Cd level. Increasing P treatment tend to increase Cd concentrations in leaves and stems although it was not statistically significant. Increasing P concentration due to Cd treatment could be the 'concentration' effect as a result of reduced growth, while there was significant decrease in Fe concentration due to Cd treatment in spite of possible 'concentration' effect. Mn concentration was increased at 1.0 ppm Cd level and then dropped at 2.0 ppm Cd level. Zu concentration in leaves and stems showed significant increase as Cd treatment increased as observed in experiment 1. Experiment 3 had 3 levels of Fe (0.5, 1.0, 2.0 ppm) and 3 levels of Cd (0, 0.8, 1.6 ppm) treatments in a factorial design. Significant growth reduction and visual toxic symptoms as observed in experiment 1 and 2 were also observed from 0.8 ppm Cd level. Increasing Fe treatment obviously alleviated toxic symptoms, improved growth and significantly increased dry matter yield. At 0.8 ppm Cd treatment level, increasing Fe treatment from 0.5 to 2.0 ppm significantly decreased Cd concentration from 141 to 92 ppm in leaves and from 101 to 46 ppm in stems. At 1.6 ppm Cd treatment level the decrease was from 224 to 167 ppm in leaves and from 124 to 109 ppm in stems. As in the case of experiment 1 and 2, Fe concentration in leaves and stems were reduced as Cd treatment increased to 1.6 ppm at 0.5 and 1. 0 Fe treatment levels, whereas at 2.0 ppm Fe level, Cd treatment increased Fe concentration in leaves and stems showing significant interactions of Fe and Cd on Fe uptake. Cd effect on Zn and Mn showed similar results to experiment 1 and 2 and Fe treatments reduced Zn and Mn concentrations in plant tissue. The results of 3 experiments show that P and Zn did not manifest suppressive effect on Cd uptake, Fe significantly demonstrated it. Fe also alleviated Cd toxicity symptoms significantly in terms of visual symptoms and dry matter yield. Visual toxicity symptoms were definitely related to Fe status in plant tissue as well as possible physiological effect of Cd itself, and the results suggest that Fe requirement for normal growth increase as Cd element is present in plant tissue. Zn accumulated more in stems than in leaves whereas Cd, Fe and Mn showed the opposite trend in all experiments.
This experiment was conducted to investigate the effects of concentrated pig slurry using membrane filter on growth of tomato in nutriculture. Pig slurry was filtered by ultra filtration and concentrated by reverse osmosis process. Filtration of pig slurry was necessary to prevent the hose clogging in nutriculture. The concentrated pig slurry (CS) and nutrient solution (NS) were mixed by six different mixing ratios of 0:100, 20:80, 40:60, 60:40%, 80: 20 and 100%:0% based on nitrogen content. The chemical nutrient solution was the solution of National Horticulture Research Station for the growth of tomato. The concentration of nutrient solution was adjusted a range of $1.6{\sim}2.0mS/cm$ in EC. The plant height of tomato treated with CS 20+NS 80% was similar with NS 100% control plot. Plant height was highest in the plot of CS 20+NS 80%. The treatment of 100% concentrated pig slurry was lowest in the gowth characteristics of tomato. Number of cluster was very lower in 100% concentrated pig slurry compared with plot of chemical nutrient solution. In the beginning of growth stage, SPAD reading value was reduced in plot treated with CS 100%, but CS 20+NS 80% plot was higher compared to 100% concentrated pig slurry. SPAD value of tomato leaves was decreased as the amount of CS was increased. The SPAD value also in treatment of concentrated pig slurry was lower in the middle growth stage compared to control plot. The dry weight of stem and leaf were 107.4, 104.2g in plot of NS 100% and CS 20%+NS 80%, respectively. The fruit number and weight were decreased at high application plots of concentrated pig slurry, The fruit setting of tomato showed lowest in the plot treated with 100% concentrated pig slurry, and the growth of tomato severely decreased after application of 100% CS treatment. In conclusion, the growth characteristics such as plant height and fruit weight of tomato were not significantly different between the plots treated with mixture of 20% CS +80%NS and 100% nutrient solution treatment. In conclusion, the mixture solution of 20% of concentrated pig slurry and 80% of nutrient solution could be used as a nutrition solution of tomato nuticulture.

Five isolates of Tomato yellow leaf curl virus (TYLCV) collected from various regions of Korea were amplified using PCR and determined the sequences of full-length genome, respectively. The PCR-amplified DNA of each TYLCV isolate was introduced into a binary vector to construct infectious clone containing 1.9 copies of the corresponding viral genome. Various cultivars and breeding lines of tomato were inoculated with Agrobacterium tumefaciens harboring infectious clone of each TYLCV isolate to assess resistance against TYLCV. Susceptible cultivar 'Super-sunread' revealed typical yellowing and narrowing of the upper leaves. In contrast, breeding linesTY12, GC9, GC171, and GC173, which contained the TY-1 and/or TY-3 genes that confer resistance against TYLCV in nature, were completely symptomless, suggesting that the lines were resistant to challenging TYLCV isolates. Symptoms of TYLCV in susceptible tomato cultivars are significantly different from those of TYLCV in the resistant tomato cultivars at 30 days after agroinfiltration. Although genomic DNAs of TYLCV were detected from the breeding lines TY12, GC9, GC171, and GC173 using real-time PCR analysis with specific primers, levels of TYLCV DNA accumulation in the resistant breeding lines were much lower than those of TYLCV DNA accumulation in susceptible tomato cultivars. Similar symptom severity and levels of TYLCV DNA accumulation were observed from TYLCV infections mediated by Bemisia tabaci in the resistant and susceptible tomato cultivars. Concentration of agrobacterium did not affect the response of tomato cultivars against TYLCV inoculation. Taken together, these results suggest that TYLCV inoculation via agroinfiltration is as effective as inoculation through Bemisia tabaci and is useful for breeding programs of TYLCV-resistant tomato.

To develop an effective protection strategy against tomato bushy stunt virus (TBSV), tobacco plants expressing single-chain Fv antibodies (scFv), were established. A previous had shown that the replication activity of viral replicase was inhibited by the selected scFvs. Moreover, no systemic symptom was found after virus inoculation on leaves of wt N. benthamiana infiltrated with an Agrobacterium suspension resulting i3l expression of the scFvs. However, control plants showed systemic symptoms. In this study the localization of the scFvs within two transgenic plant lines, (CP28H3, CP-P55) was demonstrated using immunogold labelling. The gold particles, indicating the presence of scFv, were mostly found In the cytoplasm of the plant cells including chloroplasts and in the cell walls. However, they were hardly found in the vacuole, nucleoplasm and intercellular spaces. Gold particles often accumulated in either the cytosol or chloroplasts showing a specific labeling, There was no difference in type of gold labeling between both transgenic lines. The localization of the scFv in the cytoplasm further conforms the inhibition of the RNA-dependent RNA polymerase (RdRp) by the selected scFv because it is known that the RdRp is localized to membraneous cytosolic structures.
토마토 잎에 집적된 triflumizole 잔류량 동태에 미치는 온도 영향과 Cladosporium fulvum에 대한 triflumizole의 살균효과를 구명하였다. 8주간 키운 토마토 식물체에 triflumizole을 분무 살포한 후 식물생장용 항온실에서 키우면서 4가지 온도 조건을 처리하였다. 주기적으로 토마토 잎을 채취하여 HPLC 방법을 이용하여 triflumizole의 잔류량을 조사하였다. Triflumizole 잔류량의 경시적 동태는 약제살포후 적산온도를 독립변수로 사용하는 지수적 감소 몸형으로 나타낼수 있었으며, 이는 triflumizole의 잔류성이 온도 영향을 크게 받는다는 것을 뜻한다. 경기도 농촌진흥원에 위치한 토마토 비닐하우스에서 수행한 실험에서 얻은 자료를 이용하여 평가한 결과 이 모형의 타당성이 인정되었다. Triflumizole의 약효에 대한 in vitro 실험 결과, 이 살균제는 C. fulvum의 균사 생장과 포자형성을 뚜렷이 억제하였으며, 잎곰팡이병 방제를 위해서는 토마토 잎에 집적된 triflumizole 잔류량이 최소한 10 ppm 이상이 되어야 할 것으로 추측된다. 본 연구에서 연구된 지수적 감소 모형은 토마토 잎에 집적된 triflumizole 잔류량을 추정하는데 활용될 수 있으며, 실용적인 측면에서 볼 때, 토마토 잎곰팡이병을 효과적으로 방제하는데 필요한 triflumizole의 최소 잔류량을 식물체에 유지하기 위하여 triflumizole을 살포해야 할 시기를 결정하는데 활용될 수 있다.
Leaves of two-week old seedlings of tomato (Lycopersicon esculentum) were treated with various concentrations (0, 0.2 and 0.4 $\mu$g/1) of 2,4,5-trichlorobiphenyl (PCB-29) and subsequent growth of seedlings, symptoms of oxidative stress and activities of antioxidant enzymes were investigated. Compared with the non-treated control, foliar application of PCB-29 decreased both biomass and superoxide ($O_2$) radical production but increased hydrogen peroxide production and lipid peroxidation such as malondialdehyde (MDA) formation with increased activities of superoxide dismutase (SOD), ascorbate peroxidase (APX) and guaiacol peroxidase (GPX). Further studies on the isozymes of SOD, peroxidase (POD) and APX showed that all three isozymes of SOD such as Mn-SOD, Fe-SOD and Cu/Zn-SOD, two among four isozymes of POD and all three isozymes of APX were selectively increased in response to PCB. Therefore, we suggest that a possible cause for the reduction of seedling growth by PCB exposure is the oxidative stress including over production of hydrogen peroxide and the selective expression of specific isozymes of some antioxidant enzymes.
The American serpentine leafminer (ASL), Liriomyza trifolii (Burgess), was fist found from a vinyl house planting gerbera in Kwangju area on January 26, 1994. To learn its distribution and damage pattern on crop plants, a nationwide survey was carried out at 293 vinyl houses from 25 cities/counties in six provinces. Its damage was found at 22 vinyl houses in Kwangju and Chinju areas. The range of leaves damaged by the ASL was 5-100% on gerbera, 40-70% on chrysanthemum, 20% on cherry-tomato, and 5-20% on celery. Host crop plants included gerbera, chrysanthemum, celery, cherry-tomato, watermelon, and pumpkin.
Chrysanthemums showing leaf mottling were collected from three southern locations in Korea in 1998. Two kinds of viruses were isolated from the leaves and were identified as tomato aspermy virus ch-TAV) and chrysanthemum virus B (ch-CVB), according to their host range, morphology, intracellular location, agar gel double diffusion test, and double-stranded RNA (dsRNA) analysis. The purified ch-TAV was spherical particles of approximately 29 nm in diameter and ch-CVB was filamentous particles of 685 nm long. Inclusion bodies were not observed in ch-TAV and/or ch-CVB infected chrysanthemum. ch-TAV showed positive serological reaction with TAV antiserum (ATCC-127) but not with CMV-pepper antiserum. In dsRNA analysis, four kinds of viral dsRNA were observed on ch-TAV and one viral dsRNA was shown on ch-CVB. Rate of co-infection with TAV and CVB in commercial chrysanthemums was 20.9%. On the other hand, infection with CVB alone was 97.2%. However, chrysanthemums naturally infected with TAV alone were not found.

Tomato spotted wilt virus (TSWV) was occurred newly on the 4 ornamental plants of Impatiens balsamina (Balsam), Dahlia variabilis (Dahlia), Callistephus chinensis (Aster) and Zinnia elegans (Indian lilac) at Anyang area in Korea. TSWV produced the typical symptoms of single or double ring spots on the leaves and induced usually necrosis, wilt and/or severe mosaic.
