Supercritical $CO_2$ Extraction of Whole Berry Oil from Sea Buckthorn ($Hippopha\ddot{e}$ rhamnoides var. sp) Fruit

  • The whole berry, pulp, and seed of sea buckthorn fruit were extracted with supercritical $CO_2$ to produce edible oils. The effects of extraction pressure, temperature, and $CO_2$ flow rate on the oil yield and extraction rate were investigated, and the fatty acid composition, tocopherol, and carotenoid contents of the oils were compared. The results showed that the extraction rate was affected by pressure, temperature, and $CO_2$ flow rate and, in general, the yield increased with a rise in any of the 3 variables. Fatty acids in the whole berry and pulp oil were dominated by monounsaturated fatty acids (>64%), followed by saturated fatty acids (about 30%). In contrast, fatty acids in the seed oil consisted mainly of polyunsaturated (>60%) and monounsaturated fatty acids (>24%). The seed oil had a slightly higher content of tocopherols, but a much lower content of carotenoids, compared with the whole berry or pulp oil.

Formulation Design and Evaluation of Niosome Containing Itraconazole for Dermal Delivery System (니오좀 시스템을 이용한 이트라코나졸 외용제의 제제 설계 및 평가)

  • Itraconazole is a triazole antifungal agent to inhibit most fungal pathogens. However, it is difficult for itraconazloe to be delivered by topical system due to its poor aqueous solubility. First, niosomes containing drug were prepared with span 60, cholesterol. tocopherol and poloxamer 407 as vesicle forming agents in an effort to increase solubility of itraconazole. And then prepared niosomes were dispersed in O/W creams (containing xanthan gum, glycerin, vaseline, glyceryl monostearate and $Cerix^{\circledR}-5$) or gels (containing xanthan gum and poloxamer 407). Both creams and gels were evaluated with respect to their rheological properties, in vitro permeation through excised skin of hairless mouse. Creams or gels containing niosome showed pseudoplastic flow and hysteresis loop. For both creams and gels, viscosity was increased with increasing the content of glycerine or vaseline and the content of gel forming polymer, respectively. In creams, the permeability of drug to skin was decreased with increasing the viscosity of cream. The permeability of drug was affected by pH as well as viscosity of gel. In vitro permeation test results demonstrated that cream formulations showed better permeability than gels. In conclusion, these results suggest that creams formulation containing niosome can be useful for the topical delivery of intraconazole.

Antioxidant Characteristics in the Leaves of 14 Coniferous Trees under Field Conditions

  • We investigated antioxidant capacity in leaves of 14 coniferous trees under field conditions. We focused on understanding the species characteristics on antioxidant systems and screening the coniferous tree species with the best antioxidant systems using their characteristics. The antioxidant capacity of 14 coniferous trees was divided into three groups. First group was Thuja orientalis and Chamaecyparis obtusa and those species had the highest content of ${\beta}$-carotene and xanthophyll. Second group, C. obtusa and Juniperus chinensis, used antioxidant enzymes to mitigate stress. C. obtusa represented high activity at superoxide dismutase (SOD), glutathione reductase (GR), and peroxidase (POD), and J. chinensis exhibited high activity at SOD, POD, catalase (CAT). Third group employed antioxidant such as ascorbic acid and ${\alpha}$-tocopherol. The antioxidant content of T. orientalis was the highest while that of Pinus parviflora and C. obtusa were the lowest. Few species belonged in three groups simultaneously, and most species belonged in at least one or two groups. In summary, we proposed that C. obtusa and T. orientalis had the highest antioxidant capacity while P. parviflora and P. desiflora for. multicalus had the lowest antioxidant capacity.

Effect of Dietary Vitamin E supplement and Kinds of Lipid on the Serum vitamin E Concentration and Lipid Composition (지질의 종류를 달리하였을 때 식이내 Vitamin E 첨가가 흰쥐의 혈청 Vitamin E 농도 및 지질성분에 미치는 영향)

  • This study was performed to investigate the effect of dietary vitamin E supplement and kinds of lipid on the vitamin E concentration and lipid composition in serum of the rats fed fro seven weeks. Supplemented vitamin E was 300mg/kg diet as DL-α-tocopherol acetate, and 15% of corn oil or beef tallow was added to standard diet. The results were as follows ; 1) Body weight and feed efficiency ratio were significantly increased by supplement with vitamin E, corn oil and beef tallow. 2) Serum vitamin E concentration increased significantly by supplement of vitamin E and beef tallow on the diets, while one of corn oil fed group decreased significantly . 3) The contents of triglyceride, total cholesterol, phospholipid in serum were significantly increased in the beef tallow group, and the vitamin E only -supplemented group showed significantly lower serum cholesterol in serum increased significantly in the vitamin E only -fed group and vitamin E plus corn oil-fed group. 4) The content of HDL-cholesterol in serum increased significantly in the vitamin E only -fed group and vitmain e plus corn oil -fed group. 5) The ratio of VLDL, LDL-cholesterol content to HDL-cholesterol one in serum was significantly low in the vitamin E only-supplemented group and corn oil group, and was significantly higher in the beef tallow group than in the corn oil group.

Antioxidant Activity and Protection from DNA Damage by Water Extract from Pine (Pinus densiflora) Bark

  • Water extract from Pinus densiflora (WPD) was investigated for its antioxidant activity and its ability to provide protection from DNA damage. A series of antioxidant assays, including a 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assay, a reducing power assay, a metal-chelating assay, a superoxide radical scavenging assay, and a nitrite scavenging ability, as well as a DNA damage protection assay were performed. Total phenolic content was found to be 211.32 mg Tan/g WPD. The extract scavenged 50% DPPH free radical at a concentration of 21.35 ${\mu}g/mL$. At that same concentration, the reducing power ability of WPD was higher than that of ${\alpha}$-tocopherol. The extract chelated 68.9% ferrous ion at the concentration of 4 mg/mL. WPD showed better nitrite scavenging effect at the lower pH. Meanwhile, WPD exhibited a strong capability for DNA damage protection at 1 mg/mL concentration. Taken together, these data suggest water extract from Pinus densiflora could be used as a suitable natural antioxidant.

A Longitudinal Study of the Total Lipid, Total Cholesterol, and Vitamin E Contents and Fatty Acids Composition of Human Milk (수유 기간에 따른 모유의 총지질, 총콜레스테롤 및 비타민 E 함량과 총지방산 조성의 변화에 관한 연구)

  • This study was conducted longitudinally following the total lipid and cholesterol content, fatty acids composition and Vitamin E content in human milk from 2-5 days to 12 weeks postpartum of 45 lactating Korean mothers. All samples were from well-defined subjects and uniform collection procedures were used. Total lipid concentration increased from 1.98g/dl at 2-5 days to 3.09g/dl at 12 weeks postpartum. However, there was a large variation in the concentration of total lipids. Total cholesterol concentration tended to decrease from 20.68mg/dl at 2-5days to 17.19mg/dl at 12 weeks. In comparison with transitional and mature milk, human colostrum was characterized by a lower percentage of saturated fatty acids including medium chain length fatty acids and a higher percentage of monounsatureated fatty acids and polyunsaturated fatty acids. Therefore, the P/S and M/S ratios of human milk lipids decreased significanlty during lactation. P/M/S ratio of colostrum changed from 0.38 : 0.84 : 1.00 at 2-5 days to 0.29 : 0.61 : 1.00 at 12 weeks of mature milk. Total tocopherol concentration decreased significantly from 833 ug/dl at 2-5 days to 300 ug/dl at 12 weeks.

Properties of Chemical Compositions of Safflower Seeds (산지별 홍화씨의 화학적 성분 특성)

  • This study was carried out to investigate the chemical properties of safflower seeds cultivated at some areas in Korea. Safflower was used as one of the functional foods and medicinal plants for many centuries. Porximate compositions of safflower seeds were moisture, 4.8~8.1%, crude protein 16.5~19.5%, crude ash 2.7~3.3%, curde fiber 38.5~43.4%, and crude fat 16.6~24.4%, respectively. Glutamic acid, aspartic acid and arginine were the major amino acids and their contents were 2,587.4~3,143.5 mg%, 1,315.8~1,654.8 mg%, and 1,171.9~1,484.2 mg%. K, P, Ca, and Mg were major minerals and their contents were 611.6~886.3 mg%, 501.5~596.7 mg%, 208.5~641.2 mg%, and 530.6~639.5 mg%, respectively. The free sugars that were identified include raffinose, sucrose, glucose, fructose. ${\alpha}$-tocopherol content was 0.14~3.82 mg%. Contents of vitamin C was 0.43~3.39 mg%. The safflower seeds fatty acids were palmitic acid, stearic acid, oleic acid, linoleic acid and linoleic acid. The major fatty acid was linoleic acid it's content was 74.8~82.9%.

Comparative analysis of nutritional components in various parts of Hovenia dulcis Thunbergii

  • In this study, the nutritional compounds present in various parts of Hovenia dulcis Thunbergii (H. dulcis) were compared. Regarding the free sugar content, fruits exhibited the highest fructose concentration (9.42 g/100 g), whereas branches (2.46 g/100 g) and leaves (5.82 g/100 g) contained the highest glucose levels. The most common types of organic acids were citric and tartaric acids in the fruits, citric and malic acids in the branches, and malic and succinic acids in the leaves. The leaves exhibited the highest total amino acid content of 12,102.91 mg/100 g, whereas vitamin C is predominantly found in branches and leaves at 367.85 mg/100 g and 336.21 mg/100 g, respectively. In H. dulcis, β-carotene was present in high concentration in leaves (2.41 mg/100 g), whereas the branches (0.15 mg/100 g) and fruits (0.09 mg/100 g) contain smaller amounts of it. Vitamin E, α-tocopherol, was present in high concentrations in the fruit (11.01 mg/100 g), branches (6.61 mg/100 g), and leaves (11.01 mg/100 g).

Cultivar Comparison on Tocopherols, Tocotrienols, and Antioxidant Compounds in Rice Bran (미강의 토코페롤 및 토코트리에놀 함량과 항산화 물질의 품종간 비교)

  • The rice bran, a by-product of rice milling process, is well known for various functional components, such as tocopherol, tocotrienol, ${\gamma}$-oryzanol, carrying antioxidant activities. This study was conducted to investigate the antioxidant components and antioxidant activities in rice bran of different Korean rice cultivars. The 8 isomers of vitamin E, ${\gamma}$-oryzanol, flavonoids, and polyphenolics in rice bran from 16 Korean premium and high quality rice cultivars were quantified. DPPH and ABTS radical scavenging activities and reducing power of the ethanol extracts of rice bran were measured. 'Hopum' showed the highest total vitamin E content, $221.47{\mu}g/g$ among the cultivars, and 'Hanseol' showed the lowest content. The rice bran showed different compositions of ${\alpha}-$, ${\beta}-$, ${\gamma}-$, ${\delta}-$ tocopherol and tocotrienol among rice cultivars. The antioxidant contents were also different by cultivar; the ${\gamma}$-oryzanol contents ranged from 1.99 mg/g (Unkwang) to 4.30 mg/g (Chilbo), the polyphenol contents ranged from 427.22 mg gallic acid eq./100 g (Odaebyeo) to 775.80 mg gallic acid eq./100 g (Hopum). 'Hopum' also had the highest DPPH and ABTS free radical scavenging activities, 9.82% and 187.5 AEAC mg/100 g, respectively. In vitro, the rice bran extracts from 'Hopum' had significantly higher antioxidant activities than that of other cultivars.

Antioxidant Activity of Ulmus davidiana var. japonica N. and Hemipteleae davidii P. (느릅나무와 시무나무의 항산화 활성)

  • Cortex of Ulmus davidiana var. japonica N. and Hemipteleae davidii P. have been used without classification according to countries as oriental medicine for treatment of various disorders. This study was conducted to elucidate the difference in antioxidant activity and total phenol content of these medicinal materials. Root cortex and trunk cortex of two medicinal plants were extracted with 80% ethanol $(at\;85^{\circ}C)$ and water (at room temperature), respectively. All of the extracts at $200{\sim}5\;{\mu}g/ml$, showed more effective scavenging activity on superoxide radical than ascorbic acid, and 80% ethanol and water extracts of Ulmus root cortex (URC) was more effective. Scavenging activities on DPPH radical of 80% ethanol extracts from URC and Ulmus trunk cortex (UTC) at 10 ${\mu}g/ml$, (41.4, 35.6%) were higher than those of the other six extracts and ${\alpha}-tocopherol\;(2.3{\sim}24.0%)$. Inhibitory activities on human low density lipoprotein (LDL) oxidation of 80% ethanol extracts from UTC and URC (84.3, 81.7%) at $10\;{\mu}g/ml\;(31.3{\sim}78.2%)$ were higher than those of the other six extracts and ${\alpha}-tocopherol$. However, antioxidant activities on linoleic acid peroxidation of all of the extracts were relatively lower than that ${\alpha}-tocopherol$. Total phenol content of 80% ethanol extract from URC was the highest value as 50.8% and that of water extract from HRC was the lowest as 5.6% among the extracts. From these results, it is suggested that Ulmus root cortex was the most effective in antioxidant activity.
