Green Tea Extract (CUMS6335) Inhibits Catecholamine Release in the Perfused Adrenal Medulla of Spontaneously Hypertensive Rats

  • The aim of the present study was to examine the effects of green tea extract (CUMS6335) on the release of CA evoked by cholinergic stimulation and direct membrane-depolarization in the perfused model of the adrenal gland isolated from the spontaneously hypertensive rats (SHRs), and to establish the mechanism of action. Furthermore, it was also to test whether there is species difference between animals, and between CUMS6335 and EGCG, one of biologically the most powerful catechin compounds found in green tea. CUMS6335 $(100\;{\mu}g/ml)$, when perfused into an adrenal vein for 60 min, time-dependently inhibited the CA secretory responses evoked by ACh (5.32mM), high $K^+$(56 mM), DMPP $(100\;{\mu}M)$, and McN-A-343 $(100\;{\mu}M)$ from the isolated perfused adrenal glands of SHRs. However, CUMS6335 itself did fail to affect basal catecholamine output. Also, in adrenal glands loaded with CUMS6335 $(100\;{\mu}g/ml)$, the CA secretory responses evoked by Bay-K-8644 $(10\;{\mu}M)$ and cyclopiazonic acid $(10\;{\mu}M)$ were also inhibited in a relatively time-dependent fashion. However, in the Presence of EGCG $(8.0\;{\mu}g/ml)$ for 60 min, the CA secretory response evoked by ACh, high $K^+$, DMPP, McN-A-343, Bay-K-8644 and cyclopiazonic acid were not affected except for last period. Collectively, these results indicate that CUMS6335 inhibits the CA secretion evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors as well as by direct membrane-depolarization from the perfused adrenal gland of the SHR. It seems that this inhibitory effect of CUMS6335 is exerted by blocking both the calcium influx into the rat adrenal medullary chromaffin cells and the uptake of $Ca^{2+}$ into the cytoplasmic calcium store, which are at least partly relevant to the direct interaction with the nicotinic receptor itself. It seems likely that there is much difference in mode of the CA-releasing action between CUMS6335 and EGCG.

Influence of Design Parameters of Grout Injection in Rock Mass using Numerical Analysis (암반 그라우팅 주입 설계변수가 주입성능에 미치는 영향의 수치해석적 평가)

  • In this paper, a numerical analysis of one-dimensional viscous fluid flow in a rock joint using UDEC code is performed to evaluate the effect of design parameters on injection performance. We consider injection pressure, fluid compressibility, time dependence of yield strength and viscosity of injected grout fluid, and mechanical deformation of joint as the design parameters, and penetration length and flow rate of injection are investigated as the injection performance. Numerical estimations of penetration length and flow rate were compared to analytical solution and were well comparable with each other. We showed that cumulative injection volume can be over-estimated by 1.2 times than the case that the time-dependent viscosity evolution is not considered. We also carried out a coupled fluid flow and mechanical deformation analysis and demonstrated that injection-induced joint opening may result in the increment of cumulative volume by 4.4 times of that from the flow only analysis in which joint aperture is kept constant.

Analysis of Groundwater Recharge Characteristics Using Relationship between Rainfall and Groundwater Level (강우량과 지하 수위를 이용한 지하수 함양특성 분석)

  • A dynamic model, which combined time series model with distributed-lag model, is applied to understand the relationship between rainfall and groundwater level. In the model, rainfall with distribution lags and past groundwater level as a dependent variables were used to estimate present groundwater level. The distribution of the lagged rainfall effects for groundwater levels was modeled by Almon polynomials. The model was applied to Banglim and Tanbu groundwater stations in Pyungchang river and Bocheong stream watershed which are representative basins for International Hydrological Program (IHP). The dynamic model represents observed groundwater levels very well and can be used to predict the levels. The model parameters reflect hydraulic characteristics of aquifer. In addition, from the parameters it appears that the increase in groundwater level due to rainfall takes place significantly within first two days of the rainfall event. The rainfall of the order of 18mm/day and 30mm/day at Banglim and Tanbu, respectively, had no significant effect on the groundwater levels.

Phenomenon of the absorptive optical bistabililty in amorphous $As_2S_3$ thin film (비정질 $As_2S_3$ 박막에서의 흡수형 광쌍안정 현상)

  • We observed the absorptive optical bistability in the single layer amorphous $As_2S_3$ thin film which has an isolated electron pairs at roomtemperature, for the frist time, $Ar^+$ laser beam (λ=514.5 nm) was used as a light source and was focused by thin lens. The experiment was performed in cavityless system and the occurrence of optical bistability in this material could be explained by the temperature-dependent absorption coefficient of the sample. Also, this effect was explained by the reversible shift of the absorption edge in this material. The condition of optical bistability is determined by detuning value ($a_0L$) and this value was 0.12 when thikness of that sample was about 0.8 ${\mu}{\textrm}{m}$. The switching was observed clealy when light intensity is 150~180 mW and the swiching time was about $10^{-4}$ s.

Histone Deacetylation Is Involved in Activation of CXCL10 Upon IFNγ Stimulation

  • Histone deacetylase (HDAC) activity is commonly associated with transcriptional repression. However, there is also evidence for a function in transcriptional activation. Previous studies have demonstrated a fundamental role of deacetylase activity in $IFN{\alpha}$-responsive gene transcription. In the case of type II IFN ($IFN{\gamma}$) results are controversial: some genes require HDAC activity, while transcription of others is repressed by HDAC. To investigate the effect of HDAC on transcription of an $IFN{\gamma}$-activated gene, real-time PCR was used to measure CXCL10 mRNA in Hela cells stimulated with $IFN{\gamma}$ in the presence or absence of the HDAC inhibitor TSA. Chromatin imunoprecipitation combined with real-time PCR was used to check acetylation of histone H4 and recruitment of the STAT1 complex to the ISRE locus of the CXCL10 gene. Activation of CXCL10 transcription in response to $IFN{\gamma}$ was paralleled by a decrease in histone H4 acetylation and an increase in recruitment of the STAT1 complex to the CXCL10 ISRE locus. The transcription of CXCL10 and histone H4 deacetylation were blocked by TSA, but the latter had no obvious affect on recruitment of the STAT1 complex. Our data indicate that $IFN{\gamma}$ and STAT-dependent gene transcription requires the participation of HDAC, as does the $IFN{\alpha}$-STAT pathway.

Effect of Sensory Integration Video Modeling on Self-initiation and Task Performance in Children with Intellectual Disability (감각통합활동 동영상이 지적장애아동의 자발성과 과제 수행율에 미치는 효과)

  • The aim of this research was to examine whether the sensory integration video modeling intervention would be beneficial to the children with intellectual disabilitiy. Four children with intellectual disabilitiy participated and the A-B-A design was utilized in this study. In the intervention phase, each individual watched his/her own 8 minutes long previously recorded video which was the independent variable in this study. The dependent variables were (1) the self-initiation and task performance of the four sensory integration activities, (2) performance time of Grooved Pegboard. During the intervention period, each individual participated in the occupational therapy session twice a week. The result showed that sensory integration video modeling increased self-initiation and task performance of the participants. Self-initiation and the task performance scores of the participants were maintained even after the intervention period. When Grooved Pegboard was administered, the performance time decreased. The findings indicated that sensory integration video modeling may be an effective intervention for improving self-initiation and task performance and reducing inattentiveness in children with intellectual disabilitiy. In the future research, it is suggested that the level of cognition and sensory processing capabilities of the participants be considered to validate the effectiveness of sensory integration video modeling.

Polymerization of Vinyl Monomers Initiated by Cobalt (II) Chloride (염화코발트(Ⅱ)에 의한 비닐단위체의 중합)

  • The polymerization of methyl methacrylate initiated by cobalt(II) chloride in tetrahydrofuran has been studied. The conversion was increased liearly with polymerization time at initial stage of 6${\sim}$7% conversion and then the rate of polymerization was slightly reduced, and after that the autoacceleration was observed. The rate of polymerization was increased with the concentration of cobalt(Ⅱ) chloride, but at the concentration of cobalt(II) chloride higher than $3.4{\times}10^{-4}$ mole/l the rate of polymerization was decreased. The rate of polymerization was dependent on the 1.38 power of the concentration of methyl methacrylate. The effect of the radical inhibitor DPPH was observed and the inhibition time was increased with the concentration of DPPH. The apparent overall activation energy was calculated to be 13.2 kcal/mole. In the copolymerization of methyl methacrylate ($M_1$) and styrene ($M_2$), the monomer reactivity ratio was found to be $r_1$ = 2.35, $r_2$ = 0.78. From the above results. the initiation mechanism of polymerization was discussed and it was considered that the polymerization proceeds mainly via diradical mechaism.

Determination of Indicators for Dry Aged Beef Quality

  • Previous studies on dry aged beef, which substantially increases the value of low-grade raw beef and non-preferred cuts, are currently limited to the observation of aged beef changes in laboratory settings or under particular aging conditions, whereas the factors influencing aging have so far been underexplored. Herein, we attempt to establish a technique for distinguishing between fresh and aged beef by observing changes in quality during beef aging. Specifically, we analyzed the effect of time on the quality of aged beef sourced from three Korean manufacturers and identified quality indicators that can be used to distinguish between fresh and aged beef, regardless of supplier. Storage/trimming/aging/cooking losses, moisture/fat/protein/collagen contents, and water holding capacity were tested as potential indicators, among other parameters. As a result, the quality of dry aged beef was shown to be supplier-dependent, which made the identification of factors for the above origin-independent discrimination difficult. Nevertheless, as storage loss, water holding capacity, and cooking loss significantly changed with dry aging time in all cases, these parameters were concluded to be potentially suited for discrimination purposes. The insights gained in this work may help promoting further research in this field and contribute to the development of a standard for consistent aged beef production.

Effect of Ferulic Acid Isolated from Cnidium Officinale on the Synthesis of Hyaluronic Acid (천궁으로부터 분리된 ferulic acid의 히알루론산 생성에 미치는 효과)

  • Hyaluronic acid (HA) is one of the major extracellular matrix components in skin. The HA content is reported to decline with age, which may contribute to decrease in skin moisture, wrinkle formation and the decrease in elasticity of the skin. Among the family of HA synthase genes (HAS-1, 2, 3) identified so far, HAS-2 plays crucial roles in the regulation of HA synthesis in human skin fibroblasts. In this study, we elucidated the effects of ferulic acid isolated from Cnidium officinale on HA production. Semi-quantitative RT-PCR and quantitative real-time PCR showed that ferulic acid increased mRNA level of HAS-2 gene and ELISA assay also revealed that ferulic acid increased HA production in human skin fibroblasts. Our study suggests that ferulic acid might prevent age-dependent skin deteriorations such as wrinkles, dryness and elasticity decrease, all of which could be ascribed to the reduction of the HA content in human skin.

Antiarrhythmic effects of ginsenoside Rg2 on calcium chloride-induced arrhythmias without oral toxicity

  • Background: Malignant arrhythmias require drug therapy. However, most of the currently available antiarrhythmic drugs have significant side effects. Ginsenoside Rg2 exhibits excellent cardioprotective effects and appears to be a promising candidate for cardiovascular drug development. So far, the oral toxicity and antiarrhythmic effects of Rg2 have not been evaluated. Methods: Acute oral toxicity of Rg2 was assessed by the Limit Test method in mice. Subchronic oral toxicity was determined by repeated dose 28-day toxicity study in rats. Antiarrhythmic activities of Rg2 were evaluated in calcium chloride-induced arrhythmic rats. Antiarrhythmic mechanism of Rg2 was investigated in arrhythmic rats and H9c2 cardiomyocytes. Results: The results of toxicity studies indicated that Rg2 exhibited no single-dose (10 g/kg) acute oral toxicity. And 28-day repeated dose treatment with Rg2 (1.75, 3.5 and 5 g/kg/d) demonstrated minimal, if any, subchronic toxicity. Serum biochemical examination showed that total cholesterol in the high-dose cohort was dramatically decreased, whereas prothrombin time was increased at Day 28, suggesting that Rg2 might regulate lipid metabolism and have a potential anticoagulant effect. Moreover, pretreatment with Rg2 showed antiarrhythmic effects on the rat model of calcium chloride induced arrhythmia, in terms of the reduced duration time, mortality, and incidence of malignant arrhythmias. The antiarrhythmic mechanism of Rg2 might be the inhibition of calcium influx through L-type calcium channels by suppressing the phosphorylation of Ca2+/calmodulin-dependent protein kinase II. Conclusion: Our findings support the development of Rg2 as a promising antiarrhythmic drug with fewer side effects for clinical use.
