Protective effects of extracts from six local strains of Pyropia yezoensis against oxidative damage in vitro and in zebrafish model

  • Pyropia yezoensis has been used as functional food in East Asia, especially in Korea and Japan, for more than five hundred years. This study aims to evaluate the antioxidant effect of polyphenols and proteins-rich extracts from P. yezoensis (PPPs) against 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH)-induced oxidative cell damage. Among six Korean local strains obtained from Jinhae (JiH), Haenam (HN), Jangheung (JaH), Jindo (JD), Wando (WD), and Sinan (SA) areas, the extracts of P. yezoensis from SA and JD are relatively higher in polyphenols and proteins contents. SA showed the lowest IC50 scavenging activities against 1,1-diphenyl-2-picryl-hydrazyl and alkyl radicals and displayed protective effects against reactive oxygen species (ROS) in AAPH-induced Vero cells. Especially, the PPPs extracts from SA and JD showed protective activities against AAPH-induced apoptosis, as observed by nuclear staining with Hoechst 33342. Furthermore, in vivo studies of the SA extract in zebrafish showed significantly reduced ROS generation, lipid peroxidation, and cell damage. This is the first study, to our knowledge, to evaluate the antioxidant bioactivity of PPP in the Korean Peninsula using a zebrafish model. Due to SA and JD both located in the west coast of Korea, we deduced that the chemical content of the different PPP extracts was mildly influenced by their geographic location, and this alga has potential of protective activity against AAPH-induced ROS both in vitro and in vivo.

  • Super-refraction of radar beams tends to occur primarily under a particular vertical structure of temperature and water vapor pressure profiles. A quality control process for the removal of anomalous propagation (AP) ehcoes are required because APs are easily misidentified as precipitation echoes. For this purpose, we collected X-band polarimetric radar parameters (differential reflectivity, cross-correlation coefficient, and differential phase) only including non-precipitation echoes (super-refraction and clear-sky ground echoes) and precipitation echoes, and compared the echo types regarding the relationships among radar reflectivities, polarimetric parameters, and the membership functions. We developed a removal algorithm for the non-precipitation echoes using the texture approach for the polarimetric parameters. The presented algorithm is qualitatively validated using the S-band Jindo radar in Jeollanam-do. Our algorithm shows the successful identification and removal of AP echoes.

  • This study was conducted to establish traditional manufacturing process of Hongju, a traditional spirit by traditional Kokja and Koji method based on different materials. In the fermentation process, the changes of temperature, pH, total acid, alcohol, total sugar and microflora in mash were analyzed. During the whole fermentation period, the temperature in mash made by Kokja method was reached to $30^{\circ}C$, and that of Koji method was reached $33^{\circ}C$. There was no significant difference in pH by different treatments and the initial pH in Koji method was pH 6.0 and the final pH was 4.0. The content of total acid was higher in mash processed by Kokja method than that of Koji method. Alcohol content was higher in mash processed by Koji method. The content of total sugar in all samples were reduced gradually according to progress of fermentation. Yeast populations in all samples were increased up to 6 days of fermentation and then decreased gradually. Lactic acid bacteria were increased until 3 days of fermentation and then decreased until the fermentation completed.

Development of Molecular Marker to Distinguish Octopus minor Sasaki Caught in Korea and that in China (한국산과 중국산 낙지구별을 위한 DNA 마커)

  • Octopus minor (O. minor) is widely distributed along the coastal regions of Korea, but most of them are caught in southern waters which are associated with one of the important fisheries stock. At present, O. minor from China has been introduced to the fishery markets in Korea. Here, we attempt to discriminate their origin for Korea or China using molecular techniques. Based on the O. minor mitochondrial DNA sequence, we developed a PCR-based origin discrimination system. The assay specificity was assessed by testing four individuals of O. minor from Sangdong, China, as well as 20 additional O. minor from Namhae, Muan, Yeosu and Jindo, Korea. Only four isolates of O. minor originated from China tested as positive in our distinction system. All PCR-positive products yielded identical sequences from Chinese O. minor, whereas Korean O. minor appeared to be PCR amplification. This result suggested that the primers used in this study are O. minor species specific, especially originated from China. The detection system appeared to be positive results in the use of 0.1 ng of Chinese O. minor DNA as template, however, the Korean O. minor even using $1{\mu}g$ of DNA showed no amplification. Consequently, the assay provides a simple, rapid and accurate method for the detection of Chinese O. minor.

Case Study of Diagnosis and Treatment of Thoracic Gunshot Trauma in a Dog by Computed Tomography (흉부 총상견에서 CT 촬영을 통한 흉부외상의 진단 및 치료 증례)

  • A 7-year-old female, Jindo dog was referred to the Veterinary Teaching Hospital with mild dyspnea and anorexia due to a gunshot trauma. The dog was wounded in the thoracic region 3 days ago. Plain radiographs showed the left 8th rib fracture, interstitial pattern in the left caudal lung field and pleural effusion. Abdominal radiographs showed the lead bullet. Computed tomographs(CT) showed the size of pulmonary contusion, laceration, lung parenchymal injuries, hemothorax and perforation of abdominal wall. The therapeutic plan was based on abnormalities seen on CT scans but not clearly seen in survey radiographs. Thoracic CT significantly provides even more informations compared with the corresponding radiographs in thoracic gunshot trauma. Although thoracic survey radiographs are useful as a screening tool, CT is highly sensitive in detecting thoracic injuries after thoracic trauma and is superior to routine thoracic survey radiographs in visualizing lung contusion, pneumothorax and hemothorax. Therefore, we recommend CT in the initial diagnostic work-up of patients with thoracic injuries and with suspected chest trauma because early and exact diagnosis of all thoracic injuries along with sufficient therapeutic consequences may reduce complications.

Identification of Harringtonine and Homoharringtonine and Their Contents in Korean Native Plumyew(Cephalotaxus koreana) (한국산 개비자(Cephalotaxus koreans)에서의 Harringtonine과 Homoharringtonine의 확인 및 함량 분석)

  • Harringtonine and homoharringtonine known as anti-cancer agents were isolated from Korean native plumyew(Cephalotaxus koreana) using column chromatography(CHCl3:MeOH=19:1, Rf=0.28). The structure of the mixture of two compounds was characterized by 1H-NMR. Comparison of our spectra of harringtonine and homoharringtonine with previously reported ones indicated that the two are identical. The contents of harringtonine and homoharringtonine in the needles, stems, and roots of Korean native plumyew were determined by high performance liquid chromatography(HPLC). The contents of both compounds varied with the site of location and the part of plant. The content of harringtonine was higher in needles and roots than in stems, whereas the content of homoharringtonlne was lower than harringtonine. Homoharringtonine contents in needles at Mt. Palgong, Mt. Dukyu, Mt. Baekyang, Mt. Jiri, and Namhae were higher than in stems and roots. But homoharringtonine contents in needles al Mt. Jokye and Jindo were lower than in stems and roots.

Physicochemical Properties of the Turmeric (Curcuma longa L.) in Jindo Korea (진도산 울금(Curcuma longa L.)의 이화학적 특성)

  • Studies on the physicochemical properties of physiological activity substance in turmeric (Curcuma longa L.) were analyzed for the use as an functional food materialization. The proximate compositions in the vacuum freeze dried turmeric were carbohydrate 72.90%, moisture 5.74%, crude protein 10.02%, crude fat 4.67%, and crude ash 6.69%, respectively. The mineral contents of turmeric were calcium (Ca) $2,294.77mg\;kg^{-1}$, potassium (K) $28,780.54mg\;kg^{-1}$, magnesium (Mg) $2,826.90mg\;kg^{-1}$, sodium (Na) $1,826.58mg\;kg^{-1}$, iron (Fe) $190.94mg\;kg^{-1}$, and manganese (Mn) $620.16mg\;kg^{-1}$. The vitamin contents of turmeric were pantothenic acid 1.040 mg/100 g, riboflavin 0.166 mg/100 g, thiamin 0.148 mg/100 g, pyridoxine 0.010 mg/100 g, and calciferol 0.008 mg/100 g, respectively. Total amino acid contents in protein of turmeric were 7.66 g%, and major amino acids were aspartic acid 1.45 g%, glutamic acid 1.07 g%, leucine 0.71 g%, phenylalanine 0.47 g%, and arginine 0.46 g%, respectively. The amount of free amino acids of turmeric were 225.81 mg%, and major free amino acids were asparagine, aspartic acid, glutamic acid, serine, and alanine. Especially, in the case of asparagine, it was highest. The compositions of fatty acid were saturated fatty acid 45.09%, monoenes 8.62%, and polyenes 46.30%.

Monthly Changes of Marine Benthic Algae and Community Structure at Gahakri, Southern Coast of Korea (한국 남해안 가학리의 해조류와 군집구조의 월별변화)

  • The marine algal flora and community structure of Gahakri in Jindo Gun, southern coast of Korea, was investigated monthly during September, 1993-August, 1994. As the results, a total of 87 species including 11 greens, 15 browns and 61 reds was identified. The marine algal flora was classified as the mixed flora based on the species composition. The dominant species of algal community throughout the year was Gloiopeltis furcata, and the subdominants were Sargassum thunbegii, Ulva pertusa and Hizikia fusiformis. In their vertical distribution, the species of algal community distributed yearly (or seasonally) from upper to lower intertidal zones were Gloiopeltis furcata-Corallina pilulifera, Myelophycus simplex, Gloiopeltis tenax (May$\~$October), Dumontia simplex, Sargassum thunbergii-Hizikia fusifomis, Ulva pertusa, Chondracanthus intemedius, Carpopeltis affinis.

Analysis of Essential oil and Variation of Estragol content in Different Growth Stages of Agastache rugosa Kuntze (배향초의 생육시기에 따른 정유성분 분석 및 Estragol 함량의 변이)

  • This study was carried out to compare the essential oil and its composition, and estragol content at different growth stages in the local collections of A. rugosa. Estragol was the major constituent in essential oil of all collections tested. Estragol content in essential oil was $82.3{\sim}89.1%$ in leaves, $81.1{\sim}89.2%$ in inflorescence, $72.0{\sim}45.2%$ in stem and there was no significant difference among nine local colletions. At full bloom stage, essential oil content was significantly higher than other growth stages and the highest content of estragol in essential oil was appeared at this stage. Especially, the content of essential oil in the inflorescence of Mokpo collection was 3.3%, which was the highest among the collections. Mokpo and Jindo collections produced much more essential oil and estragol than the other collections both at full bloom and ripening stages.

Summer Algal Flora of Dadohae National Park, Southwestern Coast of Korea (한국 남서해안 다도해 해상국립공원의 하계 해조상)

  • The summer algal flora and community of 9 islands in Dadohae National Park, southwestern coast of Korea, were investigated from June to September 2008. A total of 81 species (13 green, 22 brown and 46 red algae) of marine algae were identified. Among 9 islands, the number of species observed was the highest as 65 species at Jindo and the least as 37 species at Hongdo. The dominant species were Enteromorpha compressa, Ulva pertusa, Sargassum thunbergii, Gelidium amansii, Caulacanthus okamurae, Gloiopeltis furcata, and Symphyocladia latiuscula. The algal zonation of intertidal zone was figured out by Gloiopeltis furcata, Ishige okamurae, Caulacanthus okamurae-Enteromorpha compressa, Sargassum thungergii-Gelidium amansii, Symphyocladia latiuscula from upper to lower zone. The flora investigated could be classified into six functional groups such as coarsely branched form (38.1%), filamentous form (24.7%), sheet form (11.3%), thick leathery form (13.9%), jointed calcarious form (6.3%) and crustose form algae (5.7%). R/P and (R+C)/P values were 2.2 and 2.8, respectively. A cluster analysis of species occurrence was suggested that the number of marine algal species was different from greatly among the sampling sites.
