Occurrence of Gray Mold of Stachys sieboldii Caused by Botrytis cinerea (Botrytis cinerea에 의한 초석잠 잿빛곰팡이병 발생)

  • In March of 2005, gray mold disease caused by Botrytis cinereu on Stachys sieboldii Miq. was occurred in the mud cellar storage of Gyeongsangnam-do Agricultural Research and Extension Services, Korea. The symptoms started with water-soaked and rotten in the tubers. The conidia were one celled and mostly ellipsoid or ovoid in shape and light gray in color. The conidia were $5{\sim}16{\times}4{\sim}12{\mu}m$ in size and the conidiophores were $14{\sim}30{\mu}m$ in length. The pathogen formed conidia and sclerotia abundantly on PDA. The optimal temperature for mycelial growth and sclelotia formation was $20^{\circ}C$. On the basis of symptom, mycological characteristics and pathogenicity test on host plants, the fungus was identified as Botrytis cinerea Persoon: Fries. This is the first report on gray mold of S. sieboldii caused by B. cinerea in Korea.

The Desirable Development Direction of Farmer's Study Group in Agricultural Extension Work in Korea (품목별 농업인 연구모임의 발전방안)

  • The objective of this study is to suggest the developmental tasks for the successful fostering the Farmer's Study Group which have been guided by the agricultural extension work since 1994 in Korea. The 3,278 farmers study groups with 148,420 memberships have been organized in field of grains, cash crop, horticulture, livestock, improving living conditions, food processing and so on. Although the number of study groups have rapidly expanded, there also are tasks to be secured for their successful development in terms of (a) desirable phase of the study groups and goals of its fostering, (b) managements / administration of them, (c) long-term tasks to be reviewed. With regard to the desirable phase of the study groups and goals of its fostering, they should secure differential mission and the way of works compare to the other farmer's groups. Also, there should be consensus on the proper number of study groups to be fostered with a agricultural technical center at local level. With regard to the managements / administration of the study groups, there should be the different support, assistance and guidance according to the developmental level of the groups. For these, there should be evaluation works on the groups, and guidance manuals for the extension workers. Also extension works should facilitate the establishment of networks among groups over the counties. Finally, to establish the nationwide association in hierarchy and to make the cooperative works with similar farmer's group, doing the value-added activities rather than mere information exchange, and the introduction of capacity building programs for the extension workers are long-term tasks needed to review for the successful fostering study groups.

Detection of Cymbidium Mosaic Virus and Odontoglossum Ringspot Virus in Seed-Derived Plantlets of Phalaenopsis Imported by One-Step RT-PCR (One-Step RT-PCR 방법에 의한 수입 호접란묘의 심비디움 모자이크 바이러스와 오돈토글로섬 윤문 바이러스의 검정)

  • This experiment was carried out to detect the cymbidium mosaic virus (CymMV) and the odontoglossum ringspot virus (ORSV) in the seed-derived plantlets of Phalaenopsis imported from Taiwan by one-step reverse transcription-polymerase chain reaction (RT-PCR). Simple and rapid crude plant extracts for RT-PCR were prepared. The reverse transcription step was performed at $42^{\circ}C$ for 45 min and the following thermal cycling scheme was used for 36 reaction cycles: template predenaturation at $96^{\circ}C$ for 2 min, template denaturation at $96^{\circ}C$ for 30 s, primer annealing at $60^{\circ}C$ for 30 s, and DNA synthesis at $72^{\circ}C$ for 1 min. Of the 40 seed-derived plantlets of Phalaenopsis imported from Taiwan, all of them were infected with CymMV, but ORSV was not detected.

Gray Mold of Safflower Caused by Botrytis cinerea (Botrytis cinerea에 의한 잇꽃 잿빛곰팡이병)

  • The gray mold disease was severely occurred on safflower grown in experimental farm of Kyongsangnam-do Agricultural Research and Extension Services in 1999. At first, the infected leaves discolored gray or dark from the tip and gradually expanded to downword. The fungus also attacked stem, pod and calyx. The infected area became dark and dried, and frequently gray mold grown on the infected area. The causal organism was isolated and proved its pathogenecity according to Koch's postulate. Conidia of the fungus in mass were hyaline or gray, 1-celled, mostly ellipsoid or ovoid and sized $7.4{\sim}16.8{\times}5.3{\times}10.4\;{\mu}m$. Conidiophores were observed with their wide size of $2.9{\times}15.5\;{\mu}m$. The temperature range for mycelial growth was between $5^{\circ}C\;and\;30^{\circ}C$ with the optimum temperatures of $20^{\circ}C\;and\;25^{\circ}C$. The causal organism was identified as Botrytis cinerea and based on mycological characteristics examined. This is the first report on gray mold of safflower caused by Botrytis cinerea in Korea.

Stem Rot of Eupatorium lindleyanum Caused by Sclerotium rolfsii (Sclerotium rolfsii에 의한 골등골나물 흰비단병)

  • Stem rot symptoms of Eupatorium lindleyanum occurred sporadically in the herb exhibition field at Hamyang-gun Gyeongsangnam-do Agricultural Research and Extension Services in Korea. The typical symptom included water-soaking on the main stem, rotting, wilting, and blighting, which eventually leads to death of the plant. The sclerotia, white to brown, globoid with size of 1-3 mm, were formed over lesions and surface soil line. The optimum temperature for mycelial growth and sclerotia formation was $30^{\circ}C$ on PDA. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. Mycological characteristics and pathogenicity to host plants indicated that the infected fungus was Sclerotium rolfsii Saccardo. This is the first report of stem rot on Eupatorium lindleyanum caused by S. rolfsii in Korea.

Characteristics of mycelial growth and fruit body of Sparassis latifolia strains and selection of suitable incubation conditions in liquid spawn

  • Sparassis latifolia is called "Cauliflower Mushroom" and is known as an edible mushroom that has high content of ${\beta}$-glucan. Recently, artificial cultivation of S. latifolia has been done by bottle, plastic bag and wood cultivation in Korea. However it is not widely used because there are low incubation ratio and yield. For the high efficiency of production, we aim to find the superior strains and media for better mycelial and fruit body growth. First, we analyzed the genetic relationship among 31 strains and divided five groups with three kinds of URP primers. And then ten strains were selected from five groups based on the experiment of mycelial growth. The suitability of media for mycelial growth was different according to media type. The suitable solid and liquid media for mycelial growth of S. latifolia isolates were PDA and M2, respectively. In addition, with regard to C/N ratio, the mycelial growth increased even until C/N 160. Second, we investigated the production of fruitbody of the strains by plastic bag cultivation. The substrate was mixed with larch sawdust, corn flour, and wheat flour (8:1:1, v/v). Moisture content of substrate was controlled by about 60% with 10% molasses solution. Out of 31 strains, 19 strains formed primordia. The eight strains produced more than 140g/1kg in fresh weight. Third, molasses culture media was selected for the mycelial growth. And molasses suitable sugar content and input aeration were around 8Brix% and 0.3~0.6vvm, respectively. The longer the incubation period is, the more dried weight of mycelia increased, but medium volume decreased. Therefore, the best incubation period was 9 to 11 days depending on strains. In the future, research project entitled development of culture system and new variety for stable production of S. latifolia will be considered as a new item.

The Condition of Production of Artificial Fruiting Body of Cordyceps militaris (Cordyceps militaris 인공자실체 형성조건)

  • To compare the morphological and cultural characteristics of Cordyceps militaris, forming conditions of artificial fruiting body production were investigated in this experiment. The stroma shapes of artificial fruiting body were confirmed to club and/or stick. However, both shapes were same color as a orange. Perithecia of stroma was semi-egg shape and was ranged from $30{\sim}90{\times}90{\sim}130\;{\mu}m$ and it's asci was narrowly cylindric. Ascospore of perithecia was belong to filiform and multiseptate. The conidia of C. militaris was centurally grown and shaped with globose, long clavate type, floccose, centrally raised during anamorph stage. When it was cultured between glass and PP bottle, production of artificial fruiting body, pinheading ratio and total yield were higher in PP bottle. The optimum illumination was 1,000 lx for the those production. The culture medium of barley, wheat and hulled rice showed higher artificial fruiting body compared to that of silkworm. Pinheading and yield of it's isolates was decreased at more than three subsequent transculture.

Chilling Responses of Chilli Pepper Fruits at Different Ripening Stages to Low Temperature during Storage

  • This study was conducted to investigate effect of low temperature and ripeness stages on fruit quality of chilli pepper (cv. Nockgwang) during storage at 5, 7, and $10^{\circ}C$. Fruits at three ripeness stages were selected based on uniform fruit size and three maturities on peel color: S1 (fully developed fruit just before the onset of ripening), S2 (fruits in the onset of color change), and S3 (fruits in completely red in color). Lower temperature attributed to increase fruit weight loss (WL) and WL was higher in S2 than the other ripeness stages. The highest respiration rate and ethylene production was found at S2 fruit while those of fruit in green and red showed similar and remained lower level during storage. Electrolyte leakage (EL) was higher as storage temperature decreased. After 28 d storage, EL rates of fruits at $5^{\circ}C$ were 43, 36, and 17% in S1, S2, and S3 while those at $10^{\circ}C$ were 17, 19, and 14%, respectively. These results show that chilli pepper fruits are tended to lose more water at lower temperature partially associated with increased EL. Thus, threshold storage temperature must be considered for avoiding chilling during storage and for extending the storage life of fresh pepper fruits.
