고환조직 정자채취술과 세포질내 정자주입술을 이용한 고환조직 정자의 수정률과 임신율 (Fertilization and Pregnancy Rate of Testicular Sperm after Testicular Sperm Extraction (TESE) with Intracytoplasmic Sperm Injection(ICSI))

  • This study was carried to determine the possibility of finding motile spermatozoa and fertilization, pregnancy rate after testicular sperm extraction(TESE) with ICSI in obstructive and non-obstructive azoospermic patients. In 154 cases(132 patients), obstructive azoospermia was 77 cases and non-obstructive azoospermia was 77 cases. In obstructive azoospermia, patients generally showed normal spermatogenesis and included vas agenesis(n=8), multiple vas obstruction(n=7), epididymal obstruction (n=54). Total of 982 retrieved oocytes were obtained and 84.4% were injected. The fertilization rates with 2 PN and cleavage rate were 72.5% and 62.3%, respectively. 30 pregnancies(38.9%) were achieved and the ongoing pregnancies were 22 cases (28.6%). In non-obstructive azoospermia, patients showed hypospermatogenesis(n=49), maturation arrest(n=4), Sertoli cell only syndrome (n=24). The various stages of spermatogenic cell could be retrieved by TESE and could be reached normal fertilization and embryo development with ICSI. Total of 1072 retrieved oocytes obtained and 80.2% were injected. The fertilization rates with 2 PN and cleavage rate were 52.8% and 68.9%, respectively. 22 pregnancies(30.1%) were achieved and the ongoing pregnancies were 19 cases(26.0%). Conclusively, the combination of TESE with ICSI using testicular spermatozoa can achieve normal fertilization and pregnancy rate and effective method in obstructive and non-obstructive azoospermic patients.

Cichorium intybus L. extract ameliorates testicular oxidative stress induced by lead acetate in male rats

  • Objective: Oxidative stress has been suggested as a possible mechanism for the adverse effects of heavy metal toxicity on male reproduction. Cichorium intybus L. is used in Iranian folk medicine as a hepatoprotective agent as well as for its supposed fertility-enhancing properties. The present study was performed to investigate whether the ethanolic extract of C. intybus leaves could protect male rats against lead-induced testicular oxidative stress. Methods: In this experimental study, adult Wistar rats were treated with 0.1% lead acetate in drinking water alone or with 50, 100, or 200 mg/kg body weight of C. intybus extract via gavage once daily for 70 days. The weight of their reproductive organs, levels of serum hormones, histometric parameters of the seminiferous tubules, epidydimal sperm quality, and oxidative stress status were evaluated. Results: The testis weight, seminiferous tubule diameter, epididymal sperm count, serum testosterone level, and testicular levels of superoxide dismutase and glutathione peroxidase were significantly reduced (p< 0.05) in the lead-treated rats. Moreover, significantly (p< 0.05) higher levels of malondialdehyde were observed in the lead-exposed group compared to the control. However, the co-administration of C. intybus ethanolic extract in lead-treated rats was associated with a significant improvement in reproductive parameters. Conclusion: We conclude that C. intybus leaf extract has the potential to prevent lead-induced testicular toxicity and to suppress the adverse effects of lead on male reproductive health.

Testicular Cell Indices and Peripheral Blood Testosterone Concentrations in Relation to Age and Semen Quality in Crossbred (Holstein Friesian×Tharparkar) Bulls

  • Present study analyzed the changes in peripheral blood testosterone concentrations and testicular cytogram in relation to age and semen quality in crossbred males. Three different age groups of crossbred males viz. bull calves (6 months, n = 5), young bulls (15 months, n = 5) and adult bulls (4 to 6 years, n = 8) were utilized for the study. Testicular fine needle aspiration cytology technique was used to quantify testicular cytology and their indices. Peripheral blood testosterone concentrations were measured using enzyme-linked immunosorbent assay method. Semen samples collected from adult bulls were microscopically evaluated for quality parameters. Mean peripheral blood testosterone concentrations in bull calves, young bulls and adult bulls were $2.28{\pm}0.09ng/mL$, $1.42{\pm}0.22ng/mL$ and $5.66{\pm}1.08ng/mL$ respectively, and that in adult bulls were significantly different (p<0.01) from young bulls and bull calves. There was no significant difference between the proportion of different testicular cells in bull calves and young bulls. Between young and adult bulls, significant differences (p<0.01) were observed in the proportion of spermatocytes, spermatozoa, and sperm: Sertoli cell ratio. The proportions of Sertoli cells showed a significant difference (p<0.01) between the three age groups. The number of primary spermatocytes had a positive correlation with peripheral blood testosterone concentrations in bull calves (r = 0.719, p<0.01). Number of Sertoli cells per 100 germ cells was negatively correlated with blood testosterone concentration in young bulls (r = -0.713, p<0.01). Among different semen parameters in adult bulls, ejaculate volume (r = 0.790, p<0.05) had positive relationship, and sperm motility had significant negative correlation (r = -0.711, p<0.05) with testosterone concentrations. The number of Sertoli cells and Sertoli cell index had a positive correlation with various semen quality parameters (p<0.001). Results of the present study conclude that number of Sertoli cells and Sertoli cell index are good indicators of semen quality, but peripheral blood testosterone concentrations may not have a direct relationship with various seminal attributes in crossbred bulls.

Effect of Green Tea on Prostate and Seminal Vesicle in Rats Exposed to 2,3,7,8-Tetrachlorodibenzo-p-Dioxin

  • 2,3,7,8-Tetrachlorodibenzo-p-Dioxin (TCDD), a ubiquitous environmental contaminant, causes a variety of adverse effects on the male reproductive system in rats. The effect of green tea extract (GTE) was investigated on the testicular function in Spragure-Dawley rats after a single exposure of 10$\mu\textrm{g}$ TCDD/kg body weight. The exposure of rat to TCDD significantly increased the weights of the epididymis and ventral prostate, yet significantly decresed the weight of the seminal vesicle when compared to the controls (p<0.05). In a combined treatment of TCDD with GTE, the organ weight changes caused by TCDD treatment disappeared. Significant decreases in sperm motility and sperm numbers were observed in the TCDD-treated rats, when compared to the control (p<0.05). GTE treatment reversed the decrease of sperm motility and sperm numbers caused by TCDD. There were no differences in sperm morphology, histological changes of the reproductive organs, and spermatogenesis between all the treated groups. In the ventral prostate and seminal vesicle, TCDD increased the CYP1A1 mRNA level, however, it did not affect the estrogen receptor $\beta$ (ER-$\beta$) mRNA level. GTE treatment did not influence the effect of TCDD on the levels of CYP1A1 and Er-$\beta$ mRNA. These results seem to indicate that green tea protects the testicular function against TCDD-induced reproductive toxicity, not because of a receptor-mediated mechanism but rather due to a secondary change of testes or accessory sex organs.

경골어류 잉어과 쉬리(Coreoleuciscus splendidus)의 정자형성과정 (The Spermatogenesis of Coreoleuciscus splendidus, Cyprinidae, Teleostei)

  • 경골어류 잉어과 모래무지아과에 속하는 쉬리(Coreoleuciscus splendidus)의 정자형성과정과 정자의 형태를 광학현미경과 전자현미경을 이용하여 관찰하였다. 쉬리의 정소는 부레와 창자 사이에 위치하고 있었고 장축 1.8 cm, 단축 3 mm 정도의 크기로 어두운 흰색을 띠고 있었다. 정자형성과정은 정소낭(testicular cyst)에서 이루어졌으며, 각 정소낭 내에서 다양한 분화시기의 생식세포가 분포하고 있었으나 번식기인 5월에는 정소의 상부는 다양한 분화시기의 생식세포와 정자가 하부에는 성숙한 정자만 관찰되었다. 정원세포는 전자밀도가 매우 낮고 미토콘드리아의 발달이 현저하였다. 제1정모세포는 원형으로 정원세포보다 크기가 작았고 제2정모세포는 제1정모세포보다 더 작아졌고 핵의 전자밀도는 더 높았다. 정세포의 초기발달시기에는 세포의 크기가 정모세포보다 작아졌고 두부의 전자밀도가 높아졌으며 편모가 형성되기 시작하였고 미토콘드리아는 핵 주변에 분포하였다. 정자완성과정 말기에는 핵의 염색질 응축이 뚜렷하였으며 핵은 세포질 한쪽에 치우쳐 있었고, 미토콘드리아는 편모 주변에 집중되었다. 완전히 성숙한 정자의 경우 두부형태는 구형이었고 두부에서 첨체는 관찰되지 않았으며 편모의 미세소관 배열은 9+2 구조를 이루고 있었다. 또한 편모 양쪽으로 lateral fin은 관찰되지 않았으며 7개의 조대원섬유가 발견되었다.

토끼 음경해면체평활근 이완 및 10주간 경구투여한 흰쥐의 생식기능에 미치는 KH204의 효과 (Effects of KH204 on the Relaxation Response of Rabbit Corpus Cavernosum and Reproductive Function in Male Rats)

  • KH204가 음경해면체 이완과 생식기능에 미치는 영향을 규명하기 위해 효능을 실험한 결과 토끼의 음경해면체를 $1{\times}10^{-6}M$ pheylephrine으로 수축시킨 후 KH204를 61, 183 및 549 mg/L의 축적용량으로 처리하여 각각 6.8, 10.1 및 16%의 용량의존적인 이완효과를 관찰하였다. 흰쥐를 이용한 생식기능 실험은 KH204를 61, 183 및 549 mg/kg으로 10주간 장기투여에서 생식장기인 고환조직의 중량 및 정자수 증가에 유의한 효과를 나타내었다. 정자 형태검사는 대조군과 비슷한 수치로 나타내어 고농도로 장기투여에도 정자군과 비슷한 수치로 나타내어 고농도로 장기투여에도 정자 기형에 문제가 없음을 확인하였다. 이상의 결과로 KH204가 특별한 부작용이 없이 음경해면체를 농도의존적으로 이완시키는 효과가 있어, 음경발기를 개선시키는 효과뿐만 아니라 고환성 원인 또는 정자생산 기능부전으로 보이는 남성불임에서 또한 효능이 있음을 시사하였다.

Heat stress and stallion fertility

  • The threat posed by increased surface temperatures worldwide has attracted the attention of researchers to the reaction of animals to heat stress. Spermatogenesis in animals such as stallions is a temperature-dependent process, ideally occurring at temperatures slightly below the core body temperature. Thus, proper thermoregulation is essential, especially because stallion spermatogenesis and the resulting spermatozoa are negatively affected by increased testicular temperature. Consequently, the failure of thermoregulation resulting in heat stress may diminish sperm quality and increase the likelihood of stallion infertility. In this review, we emphasize upon the impact of heat stress on spermatogenesis and the somatic and germ cells and describe the subsequent testicular alterations. In addition, we explore the functions and molecular responses of heat shock proteins, including HSP60, HSP70, HSP90, and HSP105, in heat-induced stress conditions. Finally, we discuss the use of various therapies to alleviate heat stress-induced reproductive harm by modulating distinct signaling pathways.

Bisphenol a induces reproductive dysfunction in male mice

  • It has been suggested that bisphenol A (BPA), a known endocrine disruptor, interferes with the endocrine system, causing reproductive dysfunction. Recently, BPA has been found in waste water due to incomplete sewage purification, possibly threatening health through its ingestion via tap water. In this study, young male mice (6 - 7 weeks old) were administered water containing BPA (50 mg·kg-1) for four weeks, while control mice consumed water without BPA. Serum, epididymal spermatozoa and testicular sections were assessed after sacrificing the mice on day 28. No significant differences were obtained between the groups in the body, testis and seminal vesicle weights. However, the epididymal sperm motility and count levels were significantly reduced in BPA-fed mice. Significantly higher hepatotoxicity levels were also observed in mice ingesting BPA as compared to the control mice. The level of serum testosterone was reduced, and testicular sections revealed incomplete and irregular spermatogenesis in BPA-ingested mice. The sperm proteasomal-proteolytic activity level has been implicated in sperm function and is measured in motile spermatozoa using fluorometric substrates. High ubiquitin C-terminal hydrolase activity levels were observed in the control mice without BPA. During a mating trial, a low pregnancy rate (71.4%) was observed in females mated with males who had consumed BPA (100% in the control mice). Overall, BPA adversely affected spermatogenesis and quality, as indicated by decreased sperm motility, concentration and serum testosterone levels, resulting in reduced fertility competence.

Maturational Changes in Binding Capacity of Fowl Sperm to the Epithelium of the Sperm Storage Tubules during Their Passage through the Male Reproductive Tract

  • The objective of this study was to examine the binding potential of sperm to the epithelium of the sperm storage tubules (SST) in vitro and in vivo to assess the functional maturation of fowl sperm. Sperm from the testis, epididymis, as well as the proximal, middle and distal vas deferens were incubated in vitro with either the uterovaginal junction (UVJ)- or infundibular tissue containing SST at $39^{\circ}C$ for 30 min. Aliquots of sperm were also artificially inseminated into the uteri of hens, and the UVJ and infundibulum were collected 24 h post artificial insemination (AI). After incubation and AI, tissues were washed to remove loosely adhered sperm and subjected to fluorescence staining with 4', 6-diamidino-2-phenylindole, dihydrochloride (DAPI) for counting the number of bound sperm per 0.25 mm2 of surface area. Sperm from the testis, epididymis, and the three segments of the vas deferens exhibited their differential (p<0.05) binding capacity, which increased gradually from the testicular to distal vas deferens sperm under both in vitro and in vivo conditions. Existing similar trend, sperm, regardless of their source had a lesser affinity to bind to the epithelium of the infundibular SST than to the UVJ-SST. These experimental results suggested that fowl sperm may undergo gradual changes in the process of functional maturation, whereby they gain the ability to bind to the epithelium of the SST during their passage through the male reproductive tract (MRT).

CORRELATION BETWEEN TESTICLE MEASUREMENTS AND LIBIDO AND SEMEN QUALITY IN RAMS

  • A study was conducted at Ijok, Malaysia, to determine the relationship of testicular measurements with libido and semen quality in tropical and imported temperate breeds of sheep. Ten rams each of Malin (M), Siamese Longtail (L), Cross of Merino with Border Leicester (C), Dorset (D) and Suffolk (S) were used for the study. Libido, semen volume and semen quality were recorded monthly for a year together with testicular length, width and circumference. The results showed that there were breed differences in volume and quality of semen where the tropical breeds had better semen compared to the temperate breeds. There was positive and significant correlation between testicle length and semen volume in all the breeds. Testicular length was found to be positively and significantly correlated with motility and sperm concentration in the tropical breeds (L and M). The relationship between libido and testicle measurements in the tropical breeds was not significant (p<0.05). There was variable relationship between the testicular measurements and libido in the temperate breeds where the relationship was significant and negative in breeds C and D and highly significant and positive in S. It was evident that the long testicles influenced the quality of the semen whereas testicles with greater circumference influenced the libido of the rams.
