EDDS Effects on Heavy Metal Uptake by Bioenergy Plants (EDDS가 바이오에너지 작물의 중금속 흡수에 미치는 영향)

  • Plants grown in metal-contaminated sites have to be managed and disposed of safely even in phytoremediation because heavy metals can be transferred to other organisms through the food chain, which could result in bioaccumulation in organisms of a higher trophic level. However, if the harvested plants could be used for bioenergy, the ecological risk is reduced and phytoremediation improves economic feasibility. This study researched the effects of EDDS (Ethylenediamine disuccinate) on the heavy metal uptake performance of Brassica campetris and Sorghum biocolor, both of which have potential as bioenergy plants. The results showed that EDDS could increase Pb, Cu, Ni, Cd, and Zn concentrations in the roots and shoots of both of these plants. Furthermore, EDDS reduced the metal inhibition of the S. bicolor length growth. The translocation factors (TF) of S. bicolor and B. campestris are smaller than one for all five heavy metals tested and decreased by the following order: heavy metal + EDDS > heavy metals only > uncontaminated soil. The results suggest that with regard to plant growth and metal accumulation, S. bicolor treated with EDDS is more suitable than is B. campestris for the phytoremediation of soils contaminated with multiple metal species.

  • The effects of methanol. used as a solvent for the hydrophobic substrate progesterone. on the morphology of Rhizopus nigricans and 11$\alpha$-hydroxylation of progesterone was investigated. The methanol tolerance of the 11$\alpha$-hydroxylase system in polyacrylamide immobilized R. nigricans mycelia as well as in free mycelia has been induced by adding various unsaturated fatty acids. biotin and ions into the cultivation medium. Immobilization of the cell seemed to protect the cells from denaturation by methanol. It gave higher reaction rate of progesterone than the free mycelia in the presence of methanol.500 $\mu$g/l of biotin was found to be the most effective induction agent for the methanol tolerance among tested chemicals. R. nixricans cells sustained its enzymatic activity at higher methanol concentrations as a result of accumulation of unsaturated fatty acids. especially oleic acid. in the membrane phospholipid.

DNA Replication is not Required in Re-establishment of HMRE Silencer Function at the HSP82 Yeast Heat Shock Locus

  • We have exmained the re-establishment of HIMRE mediated silencing function on the transcriptional activity of yeast heast shock gene HSP82. To test whether the onset of SIR repression can occur in growing cells in the rpesence of a potent inhibitor of DNA replication, HMRa/HSP82 strains with SIR4- and SIR4S$^{+}$ genetic backgrounds were arrested in S phase by incubation of a culture in 200 mM hydroxyurea for 120 min. It was clear that following a 20 minute heat shock, silencing of the HMRa/HSP82 allele in cells pretreated with hydroxyurea does occur in a SIR4-dependen fashion, even though the kinetics of repression appears to be substantially delayed. We also have tested whether re- establishement of silencing at the HMR/hsp82 locus can occur in G1-arrested cells. Cell cycle arrest at G1 phase was achieved by treatment of early log a cell cultures with .alpha.-factor mating pheromone, which induces G1 arrest. The result suggests that passage through S phase (and therefore DNA replication) is nor required for re-establishing silencer-mediated repression at the HMNRa/HSP82 locus. Finally, to test whether de nono protein synthesis is required for re-establishment of silencer-mediated repression, cells were pretreated with cycloheximide (500 /.mu.g/ml) 120 min. It was apparent that inhibiting protein synthesis delays, but does not prevent, re-establishment of silencer-mediated repression. Altogether, these results indicate that re-establishment of silencer-mediated repression is not dependent on the DNA replication and has no requirement for protein synthesis.s.

Microbial Detection and Identification Using Biosensors

  • Various biosensors were evaluated for identifying and detecting foodborne pathogens in a rapid and effective manner. First, five strains of Escherichia coli and six strains of Salmonella were identified using Fourier transform infrared spectroscopy and a statistical program. For doing this, lipopolysaccharides (LPSs) and outer membrane proteins (OMPs) were extracted from a cell wall of each bacterial strain. As a result, each strain was identifed at the level of 97% for E. coli and 100% for Salmonella. Second, E. coli O157:H7, S. Enteritidis, and Listeria monocytogenes were identified by multiplex PCR products from four specific genes of each bacteria using a capillary electrophoresis (CE). Also, ground beef for E. coli O157:H7, lettuce for S. Enteritidis, and hot dog for L. monocytogenes were used to determine the possibility of detecting pathogens in foods. Foods inoculated with respective pathogen were cultivated for six hours and multiplex PCR products were obtained and assessed. The minimum detection levels of tested bacteria were <10 cells/g, <10 cells/g, and $10^4$ cells/g for E. coli O157:H7, S. Enteritidis, and L. monocytogenes, respectively. Third, it was possible to detect S. Typhimurium in a pure culture and lettuce by a bioluminescence-based detection assay using both recombinant bacteriophage P22::luxI and a bioluminescent bioreporter. In addition, bacteriophage T4 was quantitatively monitored using E. coli including luxCDABE genes.

  • To investigate the hemolytic toxicities of the Korean basidiomycetes, the cold-water extracts of 22 wild mushrooms were tested for their hemolytic activities on the erythrocytes of a mouse and a rat. As a result, three species including Collybia confluence, Laccaria vinaceoavellanea and Lenzites betulina showed strong hemolytic activities. Of these, L. vinaceoavellanea retained more than 80% of its hemolytic activity even after it was boiled for 30 minutes, while the other two were inactivited in five minutes.

  • A model test is carried out to investigate flow-induced vibration of a Tainter gate in estuary sulices. The gate model scaled with the ratio of 1:25 is made of acryl panel dimensioned 0.66 m in width, 0.5 m in height in the concrete test flume. Firstly, natural frequencies of the model gate are measured and the results are compared with the numerical results in order to verify the model. In the flow from the gate inside to the gate outside, the amplitudes of the vibration are measured under the different gate opening and downstream water level conditions. Also revised gate models with 20 mm bottom width are tested under the different gate openings and water levels. The results are analyzed to study the characteristics of the Tainter gate vibration in the sea ward flow. These test results are assessed in comparison with the results in the lake ward flow, as a result, presents the dynamic characteristics of the Tainter gate and a basic data for the guide manuals of gate management.

  • A new type of garlic peeling chamber has been developed which separates and peels garlic cloves using high-Pressure water Jets. The cylindrically-shaped chamber was fabricated with stainless steel plate and has a number of drilled holes, which function as solid-stream spray nozzles, on its interior wall. The chamber was divided into upper and lower section so that water can be applied at different pressures from each section. Three chambers of similar design were tested to evaluate their performances according to various pressure levels. The strongest correlation was observed between the pressure difference(lower-upper) and the proportion of unseparated and completely-peeled garlic cloves. Hanji-type garlic required higher or pressure difference to separate cloves from garlic bulbs than nanji-type garlic. On the other hand, protective leaves(skin) from the cloves were peeled more easily in case of hanji-type garlic. As a result of three consecutive peeling operations, the proportion of completely-peeled garlic cloves changed in the order of 80.1, 82.6, 84.1% in case of hanji-type, and 19.6, 29.1, 40.2% in case of nanji-type garlic.

  • The stabilizing effect of promazine hydrochloride injectin was tested by irradiation of sun lamp(5,000 Lux) on their acid solution sealed in ampules under various stabilizers. The result of experimentation on the color change, the remaining proportion and the half life of promazine hydrochloride solution is as follows. 1. Instability of promazine hydrochloride solution is mainly caused by the sunlight and oxygen. 2. The pH range of promazine hydrochloride injection is recommended to $pH\;4.4{\sim}5.2$ 3. The decomposition of promazine hydrochloride by the sunlight is composed of pseudo zero order reaction when It maintains colorless solution. 4. The half life of 0.5% promazine hydrochloride injection was 30 hrs. under sun lamp irradiation, but it was delayed to 84 hrs. by the simple stabilizer and to 175 hrs. by compound stabilizer. 5. The stabilizing effect of promazine .hydrochloride injection adding sod. metabisulfite$(Na_2S_2O_5)$ was most excellent in various simple stabilizers and adding $Na_2S_2O_5$ to nicotinic acid, $Na_2S_2O_5$ to nicotinamide were more excellent than other compound stabilizers.

A Study on Isokinetic Strength Ratios of Hip joints in Above-knee Amputees (대퇴절단환자의 고관절 등속성근력비율에 관한 연구)

  • The purpose of this study is to provide guideline of muscle strengthening exercise for preparing ambulation by presenting suitable ratio of muscle power of agonist & antagonist, and that of concentric & eccentric contraction on behalf of amputee's normal ambulation training and it's strenthening as well. 7 Subjects who have femur amputee for experimental group were able to ambulate naturally without inconvenience and 20 adult subjects of comparison group for comparison were considered to be free from disturbance of ambulation. The method of study was to measure the muscle power of hip pint, was to figure out the ratio of agonist & antagonist, concentric contraction & eccentric contraction, and was to find out mean and standard deviation of each measurement. Every numerical value of comparison was tested by Mann-whitney and comparison group's comparison between left & right value was done with t-test. Results are as followings : 1) Extension force was stronger than flexor force and had no remarkable difference(p<0.05) 2) For normal adults, adduction farce was stronger than abduction force and for amputees, abduction force was stronger while adduction force was the same as the normal without showing remarkable difference(p<0.05) According the result above, I make an assumption that maintaining a proper ratio of muscle power on strengthening exercise for amputee's ambulation training & rehabilitation and finally bring out an improvement of transfer and ambulation.

Effects of Delayed Pollination on Kernel Development in Corn

  • Receptivity of corn silk to pollen decreases with time. For effective pollination, it is important to study the receptivity of corn silks in relation to time elapsed after their emergence. Two commercial corn hybrids, 'Kwanganog' and 'Suwon 19', were tested for their silk growth and effects of delayed pollination on kernel development for 1 to 13 days after emergence of the first silk. Silks elongated rapidly for the first 3 days and then gradually decreased. Silks grew more than 30mm per day for the first 3 days and stopped growing on the 11 or 12th day after emergence of the first silk. Filled ear length decreased slowly for the first 8 days, and then decreased rapidly. Similar trends were observed for number of kernel rows and number of kernels per row or ear. The highest numbers of kernels per row or ear were observed when plants were pollinated 2-4 days after silking. These numbers decreased gradually up to 8 days, and then decreased sharply. This result indicates that 8 day-delayed pollination has no influence on kernel development. There were negative correlations between silk length and ear characteristics except kernel weight. Silk growth rate was positively correlated with filled ear length, row number, and kernel number. It might be assumed that delayed pollination by one week after the first silk emerged would not affect on kernel set.
