발열성 물질시험과 세균 내독소 시험의 비교 연구 (Comparison of LAL Test with Pyrogen Test)

  • To survey the possibility of replacing the pyrogen test with Limulus Amebocyte Lysate(LAL) test and to find out a standard methods suitable to our blood products made in Korea, 100 samples of 20% human serum albumin were tested by commercial LAL test kits and results of those were compared with rabbit pyrogen test. The LAL test is used both dinetic-chromogenically and kinetic-turbidimetrically. Both methods equally showed broad detection range (5.0~0.005 EU/ml), excellent sensitivity ($\geq$ 0.005 EU/ml) and predominant recovery rate within valid dilution range, but kinetic-turbidimetric method seemed to be more reproducible than kinetic-chromogenic method(kinetic-chromogenic method : S.D. = 15.88, kinetic-turbidimetric method : S.D. = 8.12). After heating the sample at 75$^{\circ}C$ for 15 min, the results showed a little elevated recovery rate with both methods. After performing the test on 100 albumin samples with both kits, the results were analysed using the USP standard (1.33 EU/ml). 7% of samples in kinetic-chromogenic methods and 1% of samples in kinetic-turbidimetric method exceeded the limit of endotoxin levels regulated for blood products in USA. Because this phenomenon was not observed in both methods at the same time and both methods have high sensitivity ($\geq$0.005 EU/ml), these results seemed to depend on nonspecific reaction. Considering its sensitivity and reproducibility, we could assure that LAL test is proper to detecting pyrogenic with good sensitivity.

해양생태독성시험 표준생물로서 미세조류의 특성 비교 연구(Skeletonema sp., Dunaliella tertiolecta) (Comparative Study on the Characteristics of Microalgae as Standard Species for Marine Ecotoxicity Tests (Skeletonema sp., Dunaliella tertiolecta))

  • 해양생태독성시험의 국제표준시험종 중 생산자에 속하는 대표적인 Skeletonema sp.와 Dunaliella tertiolecta의 생태독성학적 차이점을 알아보기 위해 각 표준시험법(규격)을 비교하였고 환경에 대한 종 적합성과 다양한 시험물질에 대한 민감도를 비교 분석하였다. 그 결과, 시험법의 경우 대부분 동일하였으나 시험 유효성의 기준에서 pH 변화 제한과 초기접종밀도에서 차이가 나타났으며 이는 D. tertiolecta의 낮은 성장률에 기인된 것으로 추정된다. 적합성에서는 두 종 모두 규격에서 요구하는 유효성의 기준을 연속 만족하여 시험수행의 일관성을 보였고 시험한계 염분범위는 Skeletonema sp.와 D. tertiolecta 각각 20 및 10 psu로 나타났다. 마지막으로 민감도의 경우, 시험규격에서 제시하는 참조물질, 실제 오염 배출수(선박평형수) 및 기타 다양한 화학물질에서 모두 Skeletonema sp.가 D. tertiolecta에 비해 독성 민감도가 상대적으로 높음을 확인하였으며 이는 해양생태독성시험 수행에 있어 생산자를 이용한 시험의 경우 최소 2종 이상의 다른 분류군의 미세조류를 이용하는 것이 시험결과의 신뢰성과 객관성을 높일 수 있는 방법임을 시사한다.

윤충류 Brachionus plicatilis 및 저서 요각류 Tigriopus japonicus의 초기 생활사를 이용한 해양생태독성시험 방법에 관한 연구 (Development of Ecotoxicological Standard Methods using Early Life Stage of Marine Rotifer Brachionus plicatilis and Benthic Copepod Tigriopus japonicus)

  • 해양생태독성평가를 위한 표준시험방법 개발을 위하여 해양생태계의 1차 소비자인 윤충류 및 저서 요각류를 이용한 시험방법을 정립하였다. 표준시험생물로 윤충류 Brachionus plicatilis와 저서 요각류 Tigriopus japonicus를 선정하였으며, endpoint는 윤충류 신생개체의 사망률(24 hr $LC_{50}$) 및 개체군성장률(48 hr $EC_{50}$)과 저서 요각류 유생($100{\sim}200\;{\mu}m$)의 사망률(48 hr $LC_{50}$)로 설정하였다. 표준시험방법은 미국재료시험협회 (ASTM)의 독성시험법을 참고하였으며, 표준시험생물은 국내 해양생태계의 특성 및 종의 유용성 등을 고려하여 재설정하였다. 윤충류 B. plicatilis와 저서 요각류 T. japonicus는 광염성으로 $5{\sim}35\;psu$ 구간에서 실험이 가능하고, 또한 배양이 용이한 점이 고려되었다. 상기 두 종을 이용한 독성시험은 시험생물 확보 및 배양이 쉽고 시험기간이 짧고 간단한 장점이 있는 반면 독성시험의 민감도가 무척추동물의 유생을 이용한 시험법보다 떨어지는 단점이 있다. 두 종 모두 해양유입 유해물질을 이용한 독성실험결과, 농도-반응의 선형관계가 뚜렷하였다. 카드뮴을 이용한 윤충류 개체군성장에 대한 실험실간 교차분석결과 $EC_{50}$이 각각 39.3 mg/L와 33.7 mg/L로 유사한 값을 보였다. 따라서 윤충류 및 저서 요각류는 독성시험생물로서 유용한 것으로 판단되며, 위의 두 종중 최소 1종은 해양생태 독성실험의 "battery test"에 포함할 것을 권장한다.

  • Research on the Development of EPUB SCP(Secure Content Protection) Technical Specification and International Standardization has been carried out as a R&D project granted by the Korea Copyright Commission since 2014. The research aims to build a standard framework for the EPUB DRM(Digital Rights Management) based on the Readium LCP(Licensed Content Protection) and KS EPUB DRM specifications, to develop element technologies for the framework, and to standardize it as an IDPF(International Digital Publishing Forum) industrial standard. One of the essential features of the EPUB DRM developed though this study is to support the lending model of the digital library. This paper introduces LSD(License Status Document) specification which is a DRM license management protocol for the digital library lending process, and also represents a model which can automatically test the implemented entities of the LSD specification, test scenarios and test materials.

  • This study is to investigate the bond characteristics of glass fiber reinforced polymer(GFRP) reinforcing bars in concrete by pullout test experimentally. Three different types of GFRP bars with different surface deformations were considered in this study. Also, standard deformed steel reinforcing bar with or without epoxy-coating were included for the comparisons of bond strength. All test procedures including specimens preparation, test apparatus and measuring devices were made according to the recommendation of CSA(Canadian Standards Association) Standard S806-02. From the test results, it was found that small surface indentations contributed to increase the bond strength of GFRP bar significantly. Based on the limited test results till now, the bond strength of GFRP bar with sand-coated deformation commercially available in foreign market is around $80\%$ of that of steel deformed bars.

  • This paper present adaptation method of rail potential test to verify DC railway system's performance and safety. For protection of person and utility caused by rail potential rise, international standard to limit rail potential rise is made and rail potential is tested at the time of plan and construction of DC railway system in foreign country. So, we need rail potential test at the opening time and prepare a continuous monitoring technique of rail potential and a limitation method of rail potential rise. For these, we describe cause of rail potential rise, international standard about rail potential, protection principle, an example of calculation and measurement, and adaptation method for performance test.

  • A Study of N-Insertion Preferences in KoreanKyung-A Kook, Ju-Won Kim, Ho-Young LeeSince n-insertion is not an obligatory process in Korean, it is necessary to investigate what factors influence n-insertion preferences and whether n-insertion preferences have been changed over time. To find answers to these questions, an n-insertion preference test using a questionnaire was conducted. 183 words were selected for this test and 167 subjects participated in the test. The results of this test show that the n-insertion preferences were influenced by the speakers' age, the number and structure of the syllable, word class, phonetic environments, and familiarity. It is suggested that the results of this test should be incorporated into the Principles of Standard Pronunciation and in the Grand Dictionary of Standard Korean.

경유차 적용 디젤산화촉매장치의 성능 변화 분석 (An Investigation of Performance Change of Diesel Oxidation Catalyst for Diesel Vehicle)

  • This paper is to investigated the analysis of performance characteristics of diesel oxidation catalyst (DOC) for diesel vehicle with 2.5L piston displacement. The performance evaluation test of DOC applied to test diesel vehicle was carried out for four kinds of DOCs manufactured from different company. The testing DOCs were randomly selected from the retrofit vehicle and then standard test vehicle that was representative for the application group was equipped with DOC for the test. In this verification test, the reduction rate of particulate matter (P.M.) and the deviation of the performance of the DOC were examined through CVS-75 mode of the standard vehicle and SOF reduction rate of specific DOC was investigated. It was found that some DOCs failed to pass the criteria of the P.M. reduction rate because of the reason seen catalyst aging even if they were same devices. In the result of SOF analysis, the specific DOC showed more PM reduction than SOF of PM. reduction exceptionally.

  • Using a three-dimensional (3-D) FE analyses, this paper provides the shape optimization of the standard test specimen for the torsion test, as well as a method for analyzing effects of misalignment under the angular and concentric misalignment. For verification, FE analysis is performed, which is designed for the perfectly full-model. To optimize the design shape of the torsion-controlled fatigue test specimen, we performed sensitivity analysis using shape parameters. Additionally, two kinds of misalignment (angular misalignment and concentric misalignment) are applied to the circular and tubular specimens to show effects of misalignments in the FE analysis. The present results will provide valuable information for designing shafts for every kind of mechanical system under torsional force.

자동차 배터리 지지 구조의 진동 피로 해석에 대한 연구 (Study on Vibration Fatigue Analysis of Automotive Battery Supporter)

  • In this paper, the vibration load and analysis results for automotive battery supporter were performed to provide efficient vibration tolerance performance prediction methods for single-product vibration tolerance testing, and the major influencing factors and considerations for setting up single-unit vibration tolerance tests were reviewed. A common applicable standard load was applied to efficiently predict the performance of single-unit vibrations through the frequency response analysis technique. The results similar to test results can be predicted by checking vulnerable parts of the vehicle components for vibration loads and applying scale factor to standard loads. In addition, it was confirmed that the test conditions with a frequency generating the same durability severity as the endurance test are needed for accurate prediction of the durability of the single-unit vibration tolerance test conditions, and the acceleration and frequency with the conditions that there is no significant nonlinear phenomena in the vibration system are established during the single-unit vibration tolerance test conditions.
