Toxicity tests in our laboratory are conducted usually with mass-reared organisms. They are under the same environmentel conditions throughout seasons and are supplied at specific age. A total of 38 species of aquatic organisms are being reared. We have attempted to establish pruified strains or to select clones of various parthenogenic organisms. The merits or demerits of our culturing test organisms are discussed. The differences in the susceptibility among clones or strains of test organism are also discussed. For a single species test, algae, daphnia, fish are often used. However, we usually use early stages, but occasionally, adults fish are used for reproduction tests. As an another important aspect, the toxicity through food chains has been studied. In this study, we select a pair of species belonging different trophic levels. The differences between single species tests and multispecies tests will be discussed. Even a single species test intends to assess the effects of chemicals on ecosystem levels, however, this idea is not applicable to ecosystems. Single species tests with standard organisms and multispecies tests are contradictory in concept. One type of multispecles tests is indoor microcosms being composed of severel species artificially assembled, and another is composed of natural components (both indoor and outdoor). We have used three types of outdoor mesocosms using ponds and three types of artificial streams. The mesocosms is useful to not only to analyze the floral or faun61 changes but also to study the fate or behaviour of chemicals in naturd environments. Lastly, usefulness of the field observation or experiments or semi-field experiments will be discussed. This will enhance the exploitation of early warning systems utilizing indicator organisms or animal behaviour.aviour.
Helicobacter species have commonly been isolated from the gastric mucosa of humans and animals, however have not been known its association with clinical signs. This study was aimed to detect and identify Helicobacter species in the canine stomach by urease test and polymerase chain reaction(PCR). A total of 87 dogs in Kangwon and Kyunggi areas from August, 1998 to June, 1999 were examined. The detection rate of Helicobacter species by urease test for fundal biopsy samples was 83.9%, and positive rate was increased as incubation time was increased. Helicobacter species was detected in the seventy seven dogs(88.5%) of total 87 dogs by PCR. The fifty five strains of the 77 strains of Helicobacter species were identified as H heilmannii and the three strains were identified as H felis by PCR, but the nineteen strains were not identified.

Marine ecotoxicological standard method using fish larvae was established with the standard test species of Oryzias latipes(Japanese Medaka) and Paralichthys olivaceus(flounder) and with the 7 day $LC_{50}$ as endpoint. Test method referred to the USEPA(1994) with the replacement of test species found in the Korean water. Standard test species were selected in terms of the species supply and ecological importance in Korean waters. Japanese medaka can be reared with small tanks in the lab and has wide tolerance on salinity, and flounder eggs can be easily obtained from commercial fish hatcheries. General conditions for larval fish toxicity test are as follows. The possible salinity ranges for toxicity test were $0{\sim}35\;psu$ for medaka and >20 psu for flounder. Test type was designated as static non-renewal test if the dissolved oxygen in the test chamber does not fall below 4.0 mg/L. Ages of test species were selected as 7 days after hatched for medaka(about 5 mm TL) and 25 days for flounder(about 10 mm TL) because of the low natural mortality after these periods. Test can be accepted when the survival rates are over 80% in control. Also, species sensitivity on standard reference materials(copper, cadmium or zinc) must be provided with the toxicity test results.
A series of experiments was conducted to identify the potential of five phytoplankton species as standard test species for marine ecotoxicological tests. The candidate phytoplankton species are Skeletonema costatum, Heterosigma akashiwo, Prorocentrum micans, Isochrysis galbana, and Tetraselmis suecica. Salinity tolerance and sensitivity on potassium dichromate as a reference material were identified. Toxicity of eleven ocean dumped sewage sludges and four red tide expellent extracts were estimated by the inhibition of population growth rates (PGR) of marine diatom S. costatum, While most species revealed relatively weak tolerance on salinity, T. suecica demonstrated the highest salinity tolerance ranged from $5\~35$ psu and the others $15\~35$ psu. H. akashiwo revealed the highest sensitivity as 72h $IC_{50}$=0.76mg/L and T. suecica the lowest as 72h $IC_{50}$=8.89mg/L on potassium dichromate. Sludge extracts from industrial waste, domestic sewage and livestock farm waste sludge showed high toxicity as 72h $IC_{50}$<$2\%$ and lowest toxicity from filtration bed sludge as 72h $IC_{50}$=$30.50\%$ NOEC (No Observed Effective Concentration) of sludge extract ranged from <$0.4\%$ to $1.6\%$ and this indicated high phytotoxicity of ocean dumped sewage sludge. The test sensitivity of phytoplankton PGR inhibition was much higher than those of marine rotifer Brachionus plicatilis mortality test and bioluminescent inhibition test by marine bacteria Vibrio fischeri, and comparable with the sea urchin (Strongylocentrotus intermedius) fertilization test. As a result the phytotoxicity test using phytoplankton PGR inhibition ($IC_{50}$) must be a useful tool for marine phyto-toxicological evaluation of ocean dumped materials.
From phytochemical screening of the seven species of Caryophyllaceae, that is, one species of Melandrium, and Dianthus and Pseudostellaria, two species of Stellaria and Cerastium the following compounds were identified. 1) aldehyde, sugars, other reducing compounds and glycosides from the above mentioned seven species of Caryophyllaceae were identified by thin layer chromatography method and comparison of Rf values with the standard substances. 2) Saponins, steroids and terpenoids showed positive reaction with the Froth testing in all of seven plants. Five species of plant showed positive reaction with Liebermann-Buchard test. 3) Flavonoid and polyphenolic compounds were identified by the spots in TLC-test and chlorogenic acid showed the same Rf values with the standard substances. 4) The spots of alkaloid were shown at Rf=0.68 (Stellaria aquatica) and at Rf=0.71 (Pseudostellaria palibiniana) and at Rf=0.70, and 0.44 (Dianthus sinensis).
Araeosoma owstoni Mortensen, 1904, a newly recorded sea urchin, was collected from the Korea Strait by trawling during a research expedition in April 2017. The specimen was damaged and was severely peeled off on the aboral side during trawling. However, a test and the surface of the oral side of the sample were well preserved, allowing us to successfully identify it. The species was distinguished by the large and flexible test, the tiny apical section, and the interambulacra width which is twice of the ambulacra. Pedicellaria tridentate and triphyllous were presents, but tetradactyle pedicellaria was absent due to severe peeling on the aboral side. Moreover, a length of 1,212 bp sequence from mitochondrial COI gene was obtained and this sequence covered the general DNA barcoding region. The mean of interspecific divergence within A. owstoni from Korea and other eight species of Araeosoma from the GenBank was 6.8%. This value indicated that our species was clearly distinguishable from the others. Thus, the first Araeosoma species occurring in South Korea is presented in this study.
To evaluate the natural decay durability of valuable domestic softwoods which are used for preservative treatment in our country, we carried out decay test by European standard method. Of all test wood species, Japanese larch (Larix leptolepis) showed slightly high natural decay durability compared to other 4 wood species, Japanese red pine (Pinus densiflora), pitch pine (Pinus rigida), Japanese cedar (Cryptomeria japonica), and scots pine (Pinus sylvestris). However, all of evaluated domestic softwood species in this study caused high weight losses over about 30% in heartwood by test fungus, Poria placenta. We can hardly expect a good natural decay durability from these softwood species. According to the classification of the natural durability of European standard (EN 350-1), they are classified into "Not durable" or "Slightly durable". Therefore, if using these softwoods as exterior materials, we must do preservative treatment to ensure durability.
Indigenous species means a species that is likely, due to historical presence, to occur at a specified site for some portion of its life span. Therefore, indigenous species can be useful as an indicator to assess environmental risk caused by hazardous chemicals in a specific site. So far a few toxicity studies using freshwater species which are indigenous to Korea have been carried out. In this study, a freshwater shrimp (Neocardina denticulata) indigenous to Korea was used for acute toxicity test of heavy metals. Neocardina denticulata were exposed to cadmium chloride $(CdCl_2)$, copper chloride $(CuCl_2)$ and zinc chloride $(ZnCl_2)$ using automatic flow-through system for 96 hours. The 96h LC50s were calculated as 0.043 $(0.042{\sim}0.045)mg\;CdCl_2/L,\;0.104(0.098{\sim}0.113)mg\; CuCl_2/L\;and\;2.021\;(1.633{\sim}2.594)mg\;ZnCl_2/L$. When compaired with some international standard species such as medaka(Oryzias latipes), Neocardina denticulata had high sensitivity. Therefore, this study suggested that Neocardina denticulata have possibilities for a sensitive test species to test heavy metal toxicity in aqua-system.
This study was performed to investigate the difference in species of test fish on the determination of short-term bioconcentration factor in zebrafish(Brachydanio rerio), red sword tail(Xiphophorus hellieri) and goldfish(Carassius auratus). Experimental concentrations of carbamates were 0.05 and 0.10 ppm and chlorothalonil were 0.005 and 0.01 ppm for 3 and 5 days, respectively. This paper reports the measured BCF value on pesticides in various species of test fish, under steady state, and examined correlation between the BCF value and depuration rate constant or LC$_{50}$ or lipid content. Carbamates and chlorothalonil concentration in fish extract and BCF of carbamate and chlorothalonil were increased as incresing test concentration. Carbamates concentration in fish extract and BCF of carbamate were decreased as incresing test period, but chlorothalonil concentration in fish extract and BCF of chlorothalonil were increased as prolonging test period. Determined pesticide concentration in fish extract and BCF were highest in red sword tail, and followed by goldfish, and zebrafish. Determined depuration rate constant were highest in zebrafish, and followed by goldfish, and red sword tail. 96hr-LC$_{50}$ were highest in red sword tail, and followed by zebrafish, and goldfish. Lipid compositions were highest in red sword tail, and followed by goldfish, and zebrafish. Therefore, it is suggested that the difference of BCF between each pesticide due to those of lipid composition of fish and deputation rate constant, while LC$_{50}$ have no effect on BCF.
Seasonal variation in species composition of fish in the estuary of Saemangeum water in the western coast of Korea was determined using seasonal samples collected by an otter trawl from April 2001 to February 2002. Species compositions did not differ significantly among the 4 stations (p>0.05, Wilcoxon signed-rank test). The cumulative number of species and species diversity (H') approached asymptote after four 30-min trawl hauls. Therefore 30-min trawl samples from each of the four stations were pooled to represent each season. A total of 45 species 2758 individuals and 30,346.8 g of fish were collected during the study period. Of the fish collected Leiognathus nuchalis, Synechogobius hasta, Neosalanx jordani, Konosirus punctatus and Chaeturichthys stigmatias predominated in abundance accounting for $68\%$ of the total number of individuals. Seasonal variation of fish abundance showed a similar trend to those of the other western coastal waters of Korea. The resident species were collected only in spring and autumn. Number of species and biomass were highest in summer due to the catch of many migrating species. In winter catch amount was low, and only 7 species were collected.. Brackish water fishes were more abundant in the study area, and Johnius grypotus and Cynoglossus joyneri were not premominated compared to the other coastal waters. This difference in species compositions may be related to the lower salinity in this estuarine habitat than in the other western coastal waters of Korea.
