The Ecological Character and Sequence Similarity Analysis of 18S rDNA from the Two Species of Grateloupia (Grateloupia, Rhodophyta) in Korea (두 종의 한국산 홍조 지누아리류(Grateloupia filicina, Grateloupia divaricata)의 생태특성과 18S rDNA 염기서열 상동성 분석)

  • The species of Grateloupia filicina, Grateloupia divaricata in East Sea were investigated taxonmically in order to clarify taxonomic position. The ecological character, external morphology, anatomy of vegetative structure. Blade length are $15{\sim}40\;cm$, erect from discoidal holdfast of $3{\sim}10\;mm$ in diameter. Stipe $1{\sim}2.5\;cm$ long, narrowly cylindrical below, compressed above Grateloupia filicina. Main axis are long and compressed, $3{\sim}7\;mm$ broad in broadest part. Colors are scarlet to light red. Blade length are $10{\sim}25\;cm$, erect from discoidal holdfast of $3{\sim}8\;cm$ in diameter Grateloupia divaricata. Stipe are single and simple $2{\sim}5\;mm$ broad. Thallus composed of cortex and medulla in section ; cortex composed of $9{\sim}10$ layers of anticlinally arranged cortical cell, divided into outer, middle and inner parts. Partial fragments of nuclear 18S rDNAs from the two species of Grateloupia (Rhodophyta) were amplified using the PCR reaction and sequenced to compare their similarity. The partial sequences showed 98.9% similarity each other. Grateloupia filicina has 371 bp sizes and Grateloupia divaricata has 372 bp size. The G+C contents of Grateloupia filicina is 54.3% and Grateloupia divaricata is 53.64%.

Evaluation of leaf morphology for distinguishing Prunus (Rosaceae) from Jeju, Korea (제주도산 벚나무속 잎 형질의 분류학적 검토)

  • Leaf morphology was examined for Prunus species from Jeju Island. Analyzed were the leaf blade length, petiole length, width of blade, number of veins and angle of the base to the mid-vein as quantitative characters and the distribution of trichome, position of glandsand leaf features qualitative characters. A total of 25 OTU were phenetically analyzed by UPGMA. The resuling phenograms slightly differ from the currently recognized taxonomic system in two points. Prunus mume was clustered with P. padus and P. buergeriana. Prunu spendula and P. yedoensis were separated from the cluster of P. jaamasakura, P. speciosa, P. sargentii and P. jamasakura var. quelpaertensis. Except for the members of subgenus Cerasus, subgenera Padus (P. buergeriana and P. padus), Microcerasus (P. japonica) and Amygdalus were well defined. Some morphological characters of leaves such as the ratio of blade length to width, the length of blade to petiole, number of veins, the distance between the gland and base, the angle of base to mid-vein, and the distribution of trichome were useful as diagnostic features for Prunus from Jeju Island.

Biochemical Variation and Systematic Status of the Genus Agkistrodon (Crotalidae) in Korea (韓國産 살모사 屬에 關한 遺傳的 變異 및 系流學的 硏究)

  • A total of 352 specimens of congeneric species of Agkistrodon was collected and morphometric analysis and starch-gel electrophoresis were carried out in order to investigate the taxonomic status of this genus. The results obtained in this study are as follows: Morphometric analysis 1. Three species are recognized based on Gloyd's criteria. There was no specimen that was doubtful to classify correctly. Therefore, it seems that Gloyd's morphological characters are good criteria to identify each species. 2. All three species are sympatric at two localities (Mt. Seolag, Gangwon-do, and Mt. Deogyu, Jeonra-bugdo) and A. caligino년 and A. b. brevicaudus are sympatric at Gwangneung and Mt. Yongmun, Gyonggi-do. No hybrids were found in these sympatric localities. 3. Notable sexual dimorphism was found in meristic characters. 4. A. saxatilis has signicantly more number of ventrals and shorter tail ratio than other two species. 5. There were no significant meristic character differences between A. caligino년 and A. b. brevicaudus. Genetic analysis 1. Among 26 loci investigated, 12 loci (46.1%) were identical in their mobility, 4 loci (15.4%) were nearly identical with minor frequency differences, and 10 loci (38.5%) showed interspecific mobility difference. 2. The average proportion of polymorphic loci was 9.03%. This is considerably less than that of other vertebrates. 3. The average S and D values between species are .695 and .342 respectively. These values indicate that three taxa are distinct species. 4. A. caliginosus is genetically more related to A. saxatilis than to Agkistrodon, namely A. b. brevicaudus, A. caligino년 and A. saxatilis, as proposed by Gloyd. Far from being mere morphological variants, as suggested by Kang and Yoon (1975), they are highly distinctive evolutionary units.

Chemosystematic and Phenotypic Characterization of Gram-positive Bacteria from Coastal Seawater, Korea (한국 근해에서 분리한 그람양성 세균의 화학 분류학적 및 표현형적 특성)

  • Twenty-five halotolerant gram-positlve bacteria were isolated from the coastal seawater 01 Cheju Island and Incheon J&yakdo Chemosystematic and phenotypic characteristics were used to iuvestigate the taxonomic position of these bacteria. According to their chemosystematic characteristics, the twenty-tive isolates were divided into 4 groups. Group 1 bacteria possesed 40.1 to 49.9 inol% m DNA G+C content, menaquinone-7 as a major quinone, and meso-Alpm as a diamino acid of peptidoglycan. Group 1 tam were identified as Bacilluspumilis, Bacillus lichenifbrrizis, Bacillus megaterium, Bncill~rsubtilis. Group 2 bacteria possessed 63.9 to 66.4 mol% and MK-8. They were all in the genus Arth~obaclm-. Group 3 bacteria possessed 31.0 to 37.6 mol% and MK-7. They were identified as Staphylococcus haeniol.viicvs, Siaph~~lococc~is sapropl~j~ticns, and Siaphylococcus intermediru.. Group 4 bacterium possessed 72.0mol% and MK-8 and was identified as ~Lficrococcus ltdtezm. Bacillus species accounted for 68% of h e total isolates.

Identification of Diachea leucopodia on Strawberry from Greenhouse in Korea

  • We have detected the slime mold, Diachea leucopodia (GNU06-10) in a strawberry greenhouse located in Sancheong-gun, Gyeongnam. Typical fruiting bodies had developed gregariously on the strawberry leaves, petioles, and plant debris on ground soil habitat, and also surprisingly on plastic pipes and a vinyl covering. Field samples were examined via stereomicroscopy, light microscopy, and SEM for the determination of morphological characteristics. Dark-brown to black spores formed gregariously within the stipitate cylindrical sporangium, and were covered by an iridescent peridium, which may be intact at maturity, or may have disintegrated. The upper portion of the peridium generally breaks up to expose the spores, whereas the lower portion was usually persistent. The results of energy dispersive X-ray spectrometer (EDS) analysis showed that lime was present in the stalk and columella but absent from the spores, capillitium, and peridium. The above characteristics confirm its taxonomic position in the genus Diachea. However, this genus is intermediate in character between the Physarales and Stemonitales of the Myxogastromycetidae. Hence, this genus had been classified as a member of the Stemonitales until the mid-1970's, on the basis of its iridescent peridium and noncalcareous capillitial system, similar to Comatricha of the Stemonitaceae. By way of contrast, emphasis on morphological characteristics, most notably the calcareous stalk and typical columella, places Diachea within the order Physarales. The presence of a phaneroplasmodium during the trophic stage and lime deposition in its sporophores, as was confirmed in this work, supported the inclusion of Diachea in the Physarales, and the noncalcareous capillitial system verified its identification as a member of the Didymiaceae. Further characteristics of the species D. leucopodia include the following: phaneroplasmodium, spore globose 7.5 ${\mu}m$ in diameter, very minutely roughened; sporangia $500{\mu}m\times1mm$, more or less cylindrical, gregarious, stalked 1.2mm; stalk and columella white.

Comparative Study on the Urohyal of the Subfamily Gobioninae of Korea (한국산(韓國産) 모래무지아과(亞科)(Gobioninae) 어류의 미설골(尾舌骨)에 대한 비교연구)

  • We reexamined the skulls of 14 gobionine genera of Korea to study taxonomic status of Gobioninae. The character transformation series found in the morphology of urohyal was very distinct among other skeletal characters. The gobioninae fishes of Korea were divided into 4 groups based on their morphology of urohyal as follows ; 1. Coreoleuciscus, Gnathopogon group ; The horizontal plate of urohyal has a ellipse shape with posterior edge wedged and the length is longer than that of vertical plate. 2. Hemibarbus, Squalidus group ; The horizontal plate has a elongated rhombeus shape with posterior edge pointed and the length is shorter than that of vertical plate. 3. Sarcocheilichthys, Pseudopungtungia, Pseudorasbora, Pungtungia group ; the horizontal plate has a expanded rhombeus shape, the length is longer than that of vertical plate, the hypohyal attatchment is not bifurcated, and the profile of the horizontal and vertical plate curved ventrally. 4. Ladislavia, Gobiobotia, Abbottina, Hicrophysogobio, Pseudogobio, Saurogobio group ; The horizontal plate has a pin or wedge shape expanded anteriorly, the length is shorter than that of vertical plate, and the hypohyal attatchment is thickened laterally. This results agreed with those by the adductor mandibulae complex, the geniohyoideus of cephalic muscles, and the cephalic-lateral line canals.

Phylogeny of Scopolia Jacq. s. str. based on ITS sequences (ITS 염기서열에 의한 미치광이풀속의 계통)

  • Internal transcribed spacer (ITS) sequences of nuclear ribosomal DNA were determined for 14 individuals representing eight taxa from Scopolia s. str. and related genera, Anisodus and Atropanthe. We found that the ITS sequences of Korean endemic species, S. parviflora, are significantly different from its allied species, S. japonica. This is contradictory to traditional taxonomic treatments in which those species are regarded as conspecific. S. parviflora exhibited closer relationship to S. carniolica, which is disjunctly distributed in Europe. In spite of substantially high sequence divergence between S. japonica and S. parvlflora/S. carniolica clade, morphological resemblance is evident among the species. Morphological stasis concept (retardation of morphological differentiation or evolution of similar characters among the disjuncts in a similar ecological habitat) was referred to understand this rather unusual evolutionary feature. S. lutescens, another Korean endemic species, shared almost identical ITS sequences with S. parviflora. Lack of diagnostic character distinguishing the taxa suggests that they are conspecific. Anisodus carniolicoides, which was originally described in Scopolia, was grouped with A. luridus and A. tanguticus. The monophyletic Anisodus formed a sister group relationship with a monotypic genus Atropanthe.

Cuticle micromorphology of leaves of Quercus L. (Fagaceae) and its taxonomic implications (참나무속(Quercus L.) 식물 잎의 큐티클 미세형태의 분류학적 의미)

  • Cuticle micromorphology of 22 species of Quercus and outgroup were examined by the SEM. Twenty-two species selected each two or three species in all section of the genus Quercus. The genus Trigonobalanus and Alnus are selected as outgroups. Ten characters of the inner surface and eight characters of the outer surface of the cuticle have been described. Some characters, such as the present of papillae, arrangement of subsidiary cell, shape of anticlinal cell wall are considered important character for infrageneric classification. A parsimony analysis of 18 characters resulted in 72 most parsimonious trees with its lengths of 66 steps. The topology obtained from the analysis showed two major lineages. Subgenus Cyclobalanopsis formed one clade by 75% and subgenus Quercus formed another clade by 57% bootstrap value. Based on the cuticle morphology, the two subgenus delimitation of Camus was supported. However, sect. Erythrobalanus and sect. Cerris formed one group, and sect. Lepidobalanus formed polytomy.

Taxonomy of Korean Asarum (Aristolochiaceae) by the Morphological Characters (한국산 족도리풀속(Asarum)의 외부형태학적 형질에 의한 분류)

  • In seven species of Korean Asarum, 44 qualitative and 28 quantitative characters were reviewed taxonomically. Among 44 qualitative characters observed in vegetative and reproductive organs, the states of 27 characters were common in all the seven species. Among 28 quantitative characters, five characters have the same values in all the seven species; the number of cataphyll (2-4), leaf (2), calyx lobe (5), stamen (12) and pistil (6). White spots on adaxial surface of leaf (effectively) distinguished A. maculatum and A. versicolor from the other species. Many long soft hairs on abaxial surface of leaf distinguished A. mandshuricum from other species with short hairs. The glandular hairs on inner surface of calyx tube were divided into 2 types, reflecting the taxonomic relationships between the species: Type 1 has non-stalked glandular hairs (A. maculatum, A. sieboldii and A. sp.), and Type 2 has stalked glandular hairs (A. patens, A. versicolor, A. mandshuricum and A. misandrum). The length, shape, colour of calyx lobe and the extent to which the calyx lobe recurves, as well as the diameter and colour of calyx tube throat, were confirmed to be taxonomically valuable. In the case of glandular hair density on the surface of calyx lobe, A. patens with low density and A. sp. with high density were clearly distinct from all the other species with intermediate hair density. Meanwhile, the stylar protuberance length of A. patens (2.9-3.3 mm) was strikingly different from other species (1.4-2.5 mm). Additionally, further examination of the identities of A. yeonbyeonense and A. yeonbyeonense var. viridiluteolum is required.

Cytotaxonomic study of Korean Euphorbia L. (Euphorbiaceae) (한국산 대극속(Euphorbia L., Euphorbiaceae)의 세포분류학적 연구)

  • Somatic chromosomes about 13 taxa of Korean Euphorbia L. was investigated to estimate its taxonomic significance. Somatic chromosome numbers of treated taxa were 2n= 12, 20, 22, 28, 40, 42, 56, therefore basic chromosome numbers of those were x=6, 7, 10, 11. The chromosome numbers of E. pallasii Turcz. (2n=20), E. hylonoma Hand.-Mazz (2n=20.), E. fauriei H. L$\acute{e}$v. & Vaniot ex H. L$\acute{e}$v (2n=28) and E. jolkini Boiss. (2n=28) were determined for the first time in this study. The chromosome numbers of four taxa were same as previous ones; E. sieboldiana Moor. & Decne. (2n=20), E. ebracteolata Hayata (2n=20), E. humifusa Willd. ex Schlecht. (2n=22). But those of six taxa were different; E. esula L (2n= 16, 20, 60, 64 vs 2n=20), E. helioscopia L. (2n=12, 42 vs 2n=42), E. lucorum Rupr. (2n=28, 40 vs 2n=56), E. pekinensis Rupr. in Maxim. (2n=24 vs 2n=28, 56), E. maculata L. (2n=28, 42 vs 2n=12), E. supina Raf. (n=7 vs 2n=40). E. ebracteolata, E. pallasii and E. hylonoma were distingushcd from the other taxa by the chromosome numbers, size and satellites, E. maculata, E. humifusa, E. supina had the different basic and somatic chromosome numbers in spite of the similar morphological. anatomical and palynological chracters. The chromosomal character of Korean Euphorbia was supported the Ma and Hu's systems, and as above results, it was found to be a good character in delimiting above sections and estimating relationships for some species.
