  • It is very interest to use transparent milk for various determination because milk emulsion or colloidal state bring out opacity. The fat globule can disperse by butanone and casein complex can also disintergrate to monomer by action of Triton with aide of SDS and NaOH in same milk phase at time because it can be possible that butanone have amphiphilic characteristic solvent also miscible. Our formulution of solvent can treat almost all type of milk products except with proportion of Triton/Butanone such as 8:2 and 9:1 . In spite of growth inhibition to contact with solvent it call be use also for microbial counting because this formulution did not attactk wall. This is why present technique can be applied to diver determination for the reason of milk payment system, quality control, and national planning or control of national dairy.

Phytoecdysones from the Roots of Achyranthes japonica Nakai and their Anti-atopy Activity (우슬의 뿌리로부터 Phytoecdysones의 분리와 항 아토피 효과)

  • The roots of Achyranthes japonica Nakai were extracted with 100% aqueous and concentrated subfraction was separated with ultra-performance liquid chromatography-based activity profiling. Three compounds were isolated from the subfraction 5 through the repeated prep- high performance liquid chromatography column chromatography. According to the results of physico-chemical and spectroscopic data including NMR and MS, the chemical structures of the compounds were determined as ecdysterone (1), 25S-inokosterone (2), and 25R-inokosterone (3). Three phytoecdysones were showed weak inhibitory activity for thymus and activation-regulated chemokine expression levels in tumor necrosis factor (TNF)-${\alpha}$ plus IFN-${\gamma}$ induced HaCaT cells, respectively. However, those compounds 1-3 were exhibited the most potent inhibition (80-95% at $200{\mu}g/mL$) against TNF-${\alpha}$ expression levels in A23187 plus phorbol-myrisrate acetate-induced RBL-2H3 cells. As result, 100% aqueous extract of A. japonica has an excellent anti-atopy activity. It could be used to a large range of functional anti-atopy cosmetics.

The Study of DEP Degradation Properties by Combination US and UV Lamp of Different Wavelength (초음파 (US)와 다양한 파장범위의 자외선 (UV) 조사에 따른 DEP 분해특성에 관한 연구)

  • Diethyl phthalate (DEP) is widely spread in the natural environment as an endocrine disruption chemicals (EDs). Therefore, in this study, ultrasound (US) and ultraviolet (UVC), including various applied power density (10-40 W/L), UV wavelengths (365 nm, 254 nm and 185 nm) and frequencies (283 kHz, 935 kHz) were applied to a DEP contaminated solution. The pseudo-first order degradation rate constants were in the order of $10^{-1}$ to $10^{-4}\;min^{-1}$ depending on the processes. Photolytic and sonophotolytic DEP degradation rate also were high at shortest UV wavelength (VUV) due to the higher energy of photons, higher molar absorption coefficient of DEP and increased hydroxyl radical generation from homolysis of water. Sonolytic DEP degradation rate increased with increase of applied input power and the dominant reaction mechanism of DEP in sonolysis was estimated as hydroxyl radical reaction by the addition of t-BuOH, which is a common hydroxyl radical scavenger. Moreover, synergistic effect of were also observed for sonophotolytic degradation with various UV irradiation.

  • In this paper, we propose novel, small and integrated microwave chip filter using high dielectric substrates. A variety of dielectric substrates can be selected for the specifications of products according to dielectric, Q-factors, temperature stability ect. This paper describes an application of the very high dielectric constant (K=133) substrate for design of a band pass filter to a 5.8GHz Transmitter/receiver(T/R) module.

Structure Determination of the Extractives from the Taxus Cuspidata Fruits (주목열매 추출물 구조분석)

  • The fruits of Taxus cuspidata were collected, divided into seeds and fruits, and extracted with 95% EtOH. The extracts were evaporated under the reduced vacuum pressure, concentrated, then successively fractionated with a series of n-hexane, dichloromethane, ethyl acetate and water on a separatory funnel to get some freeze dried samples. A portion of the EtOAc (arils:1.65 g, seeds:1.04 g) and $H_2O$ (arils:7 g, seeds:10 g) soluble samples were chromatographed on a Sephadex column using MeOH-$H_2O$ (1:1, 1:3, 1:5, v/v), EtOH-hexane (3:1, v/v) mixture and 100% $H_2O$ as eluting solvents to isolate pure compounds from the fractions. The isolates were developed by cellulose TLC using t-BuOH-HOAc-$H_2O$ (TBA; 3:1:1, v/v/v) and 6% aqueous HOAc. Visualization was done under ultraviolet light and by spraying the vanillin-HCl-EtOH reagent (4.8:12:480, v/v/v). followed by heating. The structures of the isolates were characterized by $^1H$- and $^{13}C$-NMR, DEPT, 2D-NMR, LC/MS and EI-MS spectra. In addition to the NMR and MS spectra, acid hydrolysis and permethylation were used to determine the correct structure of the isolated sugar compound. Their structures were elucidated as (+)-catechin (1), (-)-epicatechin (2), (+)-gallocatechin (3), (-)-epigallocatechin (4) and ${\beta}$-D-fructofuranose-($2{\rightarrow}4$)-O-${\beta}$-D-glucopyranose($1{\rightarrow}4$)-O-${\alpha}$-D-glucopyranose ($1{\rightarrow}2$)-O-${\beta}$-D-fructofuranose (5) on the basis of the above experimental evidences.

Dynamic Behavior and Resonance Reduction of Two-Span Continuous Bridges for Korean Train eXpress (KTX용 2경간 연속교량의 동적거동 및 공진감소)

  • Dynamic behaviors of the two-span continuous bridge which is one of prototypes on Gyoung-Bu high-speed railway are analyzed and some methods for reducing the resonance of the bridge are proposed. The bridge is modeled as a two-span continuous beam and the load is a vehicle of TGV-K (2p+18T) with length of 380.15 meter traveling on the railway bridge at some constant velocity. The equations governing the dynamic behaviors of the bridge are partial differential equations produced by using a system with distributed mass and elasticity. The analysis of the governing equations is performed by the mode superposition method which has modal coordinates solved by Duhamel's integral. Without considering the train velocity the dynamic reponses can be greatly reduced at some special lengths of bridge. It is different from the results of simple bridges researched so far. When the dynamic responses increase rapidly to make a resonance phenomenon depending on the train velocities, the several methods are proposed to deduce the resonance.

Bioequivalence of Azela Tablet to Azeptin Tablet (Azelastine Hydrochloride 1 mg) (아젭틴 정(염산아젤라스틴 1 mg)에 대한 아젤라 정의 생물학적 동등성)

  • Azelastine, a phthalazinone derivative, is an antiallergic agent which demonstrates histamine $H_1-receptor$ antagonist activity and also inhibits histamine release from mast cells following antigen and non-antigen stimuli. Thus, azelastine may be useful in the management of both asthma and allergic disorders. The purpose of the present study was to evaluate the bioequivalence of two azelastine hydrochloride tablets, $Azeptin^{TM}$ (Bu Kwang Pharmaceutical Co., Ltd.) and $Azela^{TM}$ (Kyung Dong Pharmaceutical Co., Ltd.), according to the guidelines of Korea Food and Drug Administration (KFDA). Eighteen normal male volunteers, $22.44{\pm}2.01$ years in age and $61.99{\pm}6.18\;kg$ in body weight, were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After two tablets containing 1 mg of azelastine hydrochloride per tablet were orally administered, blood was taken at predetermined time intervals and the concentrations of azelastine in serum were determined using HPLC with fluorescence detector. Pharmacokinetic parameters such as $AUC_t$, $C_{max}\;and\;T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters. The results showed that the differences in $AUC_t$, $C_{max}\;and\;T_{max}$ between two tablets were -6.45%, -2.60% and -7.14%, respectively, when calculated against the $Azeptin^{TM}$ tablet. The powers $(1-{\beta})$ for $AUC_t\;and\;C_{max}$ were 96.65% and 88.47%, respectively. Minimum detectable differences $({\Delta})$ at ${\alpha}=0.05$ and $1-{\beta}=0.8$ were less than 20% (e.g., 14.40% and 17.65% for $AUC_t\;and\;C_{max}$, respectively). The 90% confidence intervals were within ${\pm}20%$ (e.g., $-14.87{\sim}1.97$ and $-12.92{\sim}7.72$ for $AUC_t\;and\;C_{max}$, respectively). Two parameters met the criteria of KFDA for bioequivalence, indicating that $Azela^{TM]$ tablet is bioequivalent to $Azeptin^{TM}$ tablet.

  • This study was carried out to investigate the effect of 70% ethanol extract and each fraction from Rhododendron brachycarpum D. Don leaves on cytotoxicity, anticancer, genotoxicity and immunological activity in vitro bioassay. Cytotoxicity for human normal cells (HEL299 and Chang) of the samples was shown below 35% in 0.5 mg/ml concentration of samples except aqueous fraction by SRB assay. DNA damage on the Chang cell of the samples alone in comet assay was observed very weak damage activity even in high concentration (1 mg/ml) of the samples. The anticancer effect of the samples on human cancer cell lines (A549, AGS, Hep3B, MCF7) was indicated that the cancer cells were inhibited gradually in proportion to the increase of the concentration of the samples by MTT assay. The growth of the Raji and Jurkat cells were hastened by adding butanol fraction among the samples. In the genotoxicity on $H_2O_2-induced$ DNA damage in Chang cells using alkaline comet assay, most of samples were shown a strong protective activity from DNA OTM values.

Preventive effects of Saponariae Vaccariae Semen and Melandryi Firmi Herba on hyperlipidemia and liver damage induced by alcohol (왕부류행(王不留行)과 경엽여루채(硬葉女婁菜)가 알콜 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향)

  • Objectives : This study was performed to compare the preventive effect of water extracts from Saponariae Vaccariae Semen(SVS) and Melandryi Firmi Herba(MFH) on hyperlipidemia and liver damage induced by alcohol. Methods : Except for the normal group, I fed rat on 25% alcohol for 55 days. And each extract was administrated for the same period. I measured the serum component in rat's blood, body weight and weight of liver. Results : At first, I observed effects of SVS and MFH on hyperlipidemia induced by alcohol. SVS and MFH group didn't show the significant decrease of total cholesterol in comparison with those of the control group. SVS and MFH group showed the significant increase of HDL-cholesterol in comparison with those of the control group. SVS group showed the significant decrease of triglyceride in comparison with those of the control group, but MFH group didn't show the significant decrease of triglyceride in comparison with those of the control group. SVS group showed the significant increase of body weight in comparison with those of the control group at 4weeks and 8weeks. But, MFH group didn't show the significant changes of body weight in comparison with those of the control group at 4weeks and 8weeks. By the way, SVS group showed the significant decrease of triglyceride in comparison with those of MFH group. SVS group showed the significant increase of body weight in comparison with those of MFH group at 8weeks. At second, I observed effects of SVS and MFH on liver damage induced by alcohol. SVS and MFH group showed the significant decrease of AST(aspartate aminotransferase), ALT(alanine aminotransferase), ALP(alkaline phosphatase) and LDH(lactate dehydrogenase) in comparison with those of the control group. SVS group showed the significant increase of liver weight in comparison with those of the control group, but MFH group didn't show the significant increase of liver weight in comparison with those of the control group. By the way, SVS group showed the significant increase of liver weight in comparison with those of MFH group. Conclusions : Reviewing these experimental results, it suggests that water extracts from SVS and MFH have pharmaceutical preventive efficacy on hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate the pharmaceutical efficacy of these in depth.

Effect of Death Education Program on Self-Esteem, Spiritual Well-Being, and Pain of Adults (죽음교육 프로그램이 성인의 자아존중감, 영적안녕, 통증에 미치는 효과)

  • This study was conducted to investigate the effects of a death education program on self-esteem, spiritual well-being, and pain in adults. This investigations employed a nonequivalent control group non-synchronized design, and was performed in C hospital located in Chungju, Chungcheongbukdo province, Korea from January 4-13, 2016. The study included 48 participants who agreed to enrolled. Participants were randomly divided into an experimental and a control group of 22 and 26 applicants each. Participants (n=22) in the experimental group participated in the death education program for 2 hours, three times a week for 2 weeks. The effects of treatment were measured using a structured questionnaire to evaluate self-esteem, spiritual well-being, and pain before and after 2 weeks of intervention. Data were analyzed using a t-test, chi-squared test, Fisher's Exact-test and a paired t-test. The experimental group showed significantly increased self-esteem (P<0.002) and spiritual well-being (P<0.015), and significantly reduced pain (P<0.014) compared to the control group after 2 weeks of intervention. Overall, the death education program was shown to be an effective nursing intervention for adults; therefore, of its use in various fields should be considered.
