한국산 오수유성분에 관한 연구 (Studies on the Components of Evodia daniellii $H_{EMSLEY}$ in Korea)

  • Evodia daniellii $H_{EMSLEY}$, a plant belonging to Rutaceae, is cultivated in Korea and used as a folkmedicine for gastric inflammation, extermination of noxious insects, and headache. The seed oil of this plant also has been used in various diseases, for example, dermatitis, scabies and so forth. From the barks, fruit peels, and seed oil of Evodia daniellii $H_{EMSLEY}$, four crystalline compounds were isolated. Three compounds except one were characterized as methyl sinapate, bergapten, and evodiamine by m.p. determination, elemental analysis, UV, IR, NMR spectra and mass analysis. Even though methyl sinapate was a known synthetic compound, it was not yet reported as a natural product. From the seed oil, unsaponifiable fraction was separated and was analyzed. It was considered to be consisted with sterols, hydrocarbons and tocopherols.

  • Vicilin and legumin, the storage Proteins of seed, were Purified from ginseng (Panax ginseng C.A. Meyer) endosperm cells. They were immunized in rabbits, and antibodies were raised respectively. Using these two antibodies, double immunogold labeling of vicilin and legumin was carried out to determine the gap of synthetic time and the transport pattern of vicilin and legumin in the ginseng endosperm cells. Vicilin and legumin were synthesized at the same time at early embryo developmental stage. They were secreted from the Golgi bodies and accumulated into the small vacuoles. As the endosperm cells developed, vicilin and legumin localized in the small vacuoles were gradually transported toward the large central vacuole where they were stored. Protein bodies were derived from the vacuoles filled with proteins and distributed in the endosperm cells of mature red seed. Protein bodies were various in size from 1 to 8 ${\mu}{\textrm}{m}$ in which vicilin and legumin were mixed each other. The number of small particles labeled on the vicilin was greater than that of large particles labeled on the legumin in the protein bodies indicating that the amount of vicilin is higher than that of legumin in the protein bodies.

Cryopreservation of Forest Tree Seeds: A Mini-Review

  • Since forest trees form the basis of forest ecosystem, their prolong subsistence is crucial for various flora and fauna. The foremost challenges to sustain the forest ecosystem comprise of the declining forest tree population accompanied with structural changes due to afforestation and exploitation of forest area, environment changes, pests, pollution, and introgressive hybridization. For ex situ conservation approach, in vitro techniques encompass basic role for conserving tree genetic resources, predominantly where natural propagules like recalcitrant seed might not be appropriate for long-term conservation. The practice includes restricted growth techniques, conventional micropropagation, production and storage of synthetic seeds, and cryopreservation. Even though these practices have been applied chiefly to herbaceous species, but recently, woody species were also focused upon. Key conceptions, challenges and techniques for forest tree seed conservation are discussed briefly in this review with special emphasis on some successful cryopreservation approaches for long-term storage.

저결함 압전수정의 성장과 결함분석 (Growth of Low Defect Piezo-quartz and Defect Analysis)

  • 수열법을 이용하여 압전수정을 성장하고 각종 성장결함을 분석하였다. 4 중량퍼센트 NaOH, 성장온도 $340-360^{\circ}C$, 온도구배 $20-40^{\circ}C$의 성장조건에서 (0001)방향의 ZY 종자결정, 합성수정을 원료로 하였을 경우 성장속도는 0.25-0.65 mm/day 였다. 원료로써 합성수정을 사용하고 저전위 종자결정 위에서 수평 종자결정 배치법으로 성장된 압전수정의 전위밀도는 20.0개/$cm^2$, 에치채널 밀도는 5.0개/$cm^2,\;10\mu$ 이상의 함유물 농도는 2.4개/$cm^3$, 알파값은 0.019였으며, IEC 758규격에 의한 등급분류는 에치채널 밀도 1등급, 함유물 농도 Ia등급, 알파값 A등급이었다.

  • 블루 안료 15 : 6이라 불리는 ${\epsilon}$ 결정상의 동프탈로시아닌(${\epsilon}$-CuPc)는 LCD 패널에서 청색화소를 제공하는 중요한 소재로써, 본 연구에서는 ${\epsilon}$-CuPc 시료를 ${\epsilon}$-CuPc 나노 입자를 seed로 이용하는 방법으로 여러 조건에서 합성하여 적합한 용매 및 온도조건 등을 규명하였고, 이를 미리 합성한 ${\alpha}$${\beta}$ 결정상의 CuPc 시료들과 비교함으로써 ${\epsilon}$-CuPc 시료의 결정상, 결정 순도, 합성 수율 및 미세구조를 상호 비교 분석하였다. 시료들의 화학 구조 및 결정 구조는 푸레어 변환 적외선(FT-IR) 분광기 및 X선 회절 분석기(XRD)를 이용하여 각각 비교 분석하였다. 또한, 입자의 형상 및 크기는 주사 전자현미경(FE-SEM)으로 비교 분석하였고, 열적 특성은 열중량분석기(TGA)를 이용하여 비교 분석하였다.

산수유 종자 추출물의 화장품 방부효능에 관한 연구 (A Study on the Cosmetic Preservative Effects of Cornus officinalis seed Extracts)

  • 산수유 종자 추출물의 천연 방부제로서 가능성을 평가하기 위하여 항균활성 및 화장품 방부 효능 평가를 실시하였다. 산수유 종자는 각각 1,3-부틸렌글리콜과 에탄올을 이용하여 추출하였다. Paper disc method로 항균활성을 평가한 결과 Staphylococcus aureus, Staphylococcus epidermidis와 Propionibacterium acnes에서 항균활성이 나타났으며 산수유 종자 1,3-부틸렌글리콜 추출물(COS-A)이 여드름균에대해 $26.7{\pm}4.0mm$로 가장 높았다. 최소저해농도(MIC) 측정 결과 산수유 종자 에탄올 추출물(COS-B)이 가장 낮은 농도에서 S. epidermidis균의 생장을 억제하였다. 에멀션에서의 방부효능 평가를 위해 Challenge test를 실시한 결과 COS-A와 COS-B를 첨가한 에멀션에서 균 접종 7일 후 피부상재균이 100% 사멸되었다. 이러한 결과는 산수유 종자 추출물이 화장품에서 합성방부제를 대체할 천연 방부제로서 가능성이 있을 것으로 사료된다.

Algorithm for Predicting Functionally Equivalent Proteins from BLAST and HMMER Searches

  • In order to predict biologically significant attributes such as function from protein sequences, searching against large databases for homologous proteins is a common practice. In particular, BLAST and HMMER are widely used in a variety of biological fields. However, sequence-homologous proteins determined by BLAST and proteins having the same domains predicted by HMMER are not always functionally equivalent, even though their sequences are aligning with high similarity. Thus, accurate assignment of functionally equivalent proteins from aligned sequences remains a challenge in bioinformatics. We have developed the FEP-BH algorithm to predict functionally equivalent proteins from protein-protein pairs identified by BLAST and from protein-domain pairs predicted by HMMER. When examined against domain classes of the Pfam-A seed database, FEP-BH showed 71.53% accuracy, whereas BLAST and HMMER were 57.72% and 36.62%, respectively. We expect that the FEP-BH algorithm will be effective in predicting functionally equivalent proteins from BLAST and HMMER outputs and will also suit biologists who want to search out functionally equivalent proteins from among sequence-homologous proteins.

Synthesis of Large Bumpy Silver Nanostructures with Controlled Sizes and Shapes for Catalytic Applications

  • We have synthesized disc-like large silver nanomaterials that have nanostructured bumps on the surface using smaller nanoplate seeds. The size and shape of the bumpy nanostructures are rationally controlled by changing the concentrations of nanoplate seeds, silver ion, reductant, and citrate ion. Importantly, the synthetic mechanism of these bumpy nanostructures is remarkably similar to that of the conventional seed-mediated growth based on tiny seeds. We have further investigated the catalytic properties of the bumpy nanostructures for the reduction of 4-nitrophenol, which is associated with a concomitant color change from yellow to colorless.

  • The present study was undertaken to investigate the crystal growth onto enamel mineral and synthetic hydroxyapatite seeds in media resembling the enamel fluid composition. Effects of fluoride at low concentrations on the precipitation were also examined in a benchtop crystal growth model adopting a miniaturized reaction column. X-ray diffraction and Fourier transform infrared spectroscopy(FTIR), as well as chemical analyses, were employed for characterization of both seed materials before and after experimentation. Remarkable findings were that (1) both biological and synthetic seeds at the same total surface areas yielded rather similar precipitation rates at all levels of fluoride concentration in solution and (2) the precipitation rate was accelerated in a manner depending on fluoride concentrations in media. FTIR differential analysis disclosed that the precipitating phase was characterized as poorly crystallized apatite, which incorporated subtle carbonate. Most of the fluoride ions in soution were readily incorporated into crystals. The overall results support the view that the seeded crystal growth model is of value to gain insight into the mechanism of enamel crystal growth under fluoride regimens.

  • Pseudomonas cepacia로부터 유래한 quinolinone 대사물질인 2-(2-hepteny)-3-methyl-4-quinolinone (1), 2-heptyl-3-methyl-4-quinolinone, 2-nonyl-3-methyl-4-quinolinone 및 합성물질 ethyl 2-methyl-3-alkyl-4-quinolinone carboxylate의 식물생장촉진 효과를 보기 위해 종자발아검정 및 경엽처리 후의 생장량을 측정하였다. 또한 종자침지 및 관주 처리후의 초고, 생체중, 과실수도 조사하였다. 화합물 1은 종자발아 효과와 경엽처리 후에 일관된 생장촉진 효과를 보였다. 종자침지와 관주처리 후에, 화합물 1과 합성물질 ethyl 2-methyl-4-quinolinone-3-carboxylate (5)를 처리한 식물에서의 생체중과 과실수는 크게 증가하였다. 화합물1과 5는 대조관보다 각각 44%와 84%의 과실수 증가를 나타냈다.
