Rhizopus nigricans로부터 원형질체의 분리 (Isolation of Protoplasts from Rhizopus nigricans)

  • Rhizopus nigricans의 포자와 균사체로부터 원형질체 생성을 위한 최적 조건을 조사하였다. 5% 2-deoxy-D-glucose가 첨가된 액체 배지에서 14시간 배양시킨 균사체보다 동일배지에서 4시간 배양하여 팽창시킨 포자로부터 더 많은 원형질체가 생성되었으며, Novozym 234(2%)를 단독으로 처리하였을 때보다는 5000 unit/ml 의 ${\beta}-glucuronidase$와 혼합하였을 때 원형질체의 생성율이 더 높았다. 삼투 안정제로 0.6 M $MgSO_4$를 사용한 복합 효소 용액(pH 6.0)에 2시간 처리하였을 때 팽창된 포자로부터 단위ml당 $8.0{\times}10^6$의 원형질체를 분리할 수 있었다.

  • Apexes of basidia in Rhodophyllus(synonyum of Entoloma) muraii var. albus are divided into four sections or depressed in the center. A spore is formed by inflation of the apex of the sterigma. The apex of the sterigma is swollen and changed from a papilla through a penisform and a club into a globeform. Six spots of globose spore are regularly or irregularly depressed with hilum axes. Finally the spores are cuboid. Basidia of Entoloma squamiferrum are developed from hymenium layer of crator-shaped parabasidium. Apex of basidium is flat or depressed in the center. Although four sterigmata are developed, only two sterigmata are symmetrically swollen to two spores in certain basidia. It means that two sterigmata among four sterigmata are infertile. The apex of the sterigma is swollen into a paillaform, and then turned into a penisform. It is swollen from a clubform into a globeform. Six spots of the surfaces of globose spore are deperssed with hilum axes. Finally the spore is cuboid, and then it is released from hilum. Four sterigmata of papillaform of E. violaceobrunneum are developed from cartor-shaped basidium. The apex of sterigma is swollen to a small globeform. And then it is swollen to a clubform. The clubform is again swollen to a ellipticalform, and then more than six spots of spore surfaces are ramomly depressed with hium axes. When the depression of surface of a elliptical spore is over, it is the multi-angular spore of the heterometrical-form.

Electron and Light Microscopic Studies on the Development of Oidia from Somatic Mycelium of Coprinus cinereus

  • Development of oidia, a type of thallic spores from monokaryotic mycelium of Coprinus cinereus was examined with electron microscope and light microscopes. Oidia formation in this fungus is unique in its mode of formation compared with other types of asexual sporogenesis. Oidiogenesis in C. cinereus is carried out in three steps: 1) Formation of oidiophore from the parent mycelium, 2) Formation of initials of oidial cells from swollen oidiophore, 3) Segmentation and detachment of mature oidial cell. Oidiophores appear to spring out singly as a swollen hyphal branches from the normal foot hyphae or sometimes coiled hypha. From the oidiophore, oidial branches sprout out forming a group of $2{\sim}6$, most often 4 oidial cells and each oidial cell undergoes a single mitosis resulting in 2 oidia. One of the sibling oidial cells in a group is frequently transformed into a new oidiophore, thus oidiogenic structures are tandemly produced at the several different levels.

Isolation and Identification of Fusicoccum Species from Quercus dentata

  • An imperfect fungus Fusiococcum species was isolated from Quercus dentata. A naturally infected Daimyo oak tree was collected and showed elongate wounds on the stem. The fungal cultures were initially white and cottony, and later turned dark gray. Numerous solitary pycnidia were developed on the medium surface, and typically spherical. Yellowish conidial masses were exuded from pycnidia on the culture plates. Conidial masses were swollen and measured as approximately 100 to $300{\mu}m$ in length. It appeared that conidia were usually held together in globose to oval drops. Conidia were hyaline, single-celled (nonseptate), ellipsoid to fusoid, and measured as approximately $8.0{\times}2.7{\mu}m$. Based on these cultural and morphological characteristics, the fungal isolate was identified as a species of Fusicoccum Corda. To preserve and examine fungal spores exuded from pycnidia on the medium surface, a vapor fixation procedure for scanning electron microscopy was employed in this study. The specimens were exposed to the vapor of 2% (v/v) glutaraldehyde and 2% (w/v) osmium tetroxide each for 2 h. With the vapor fixation we obtained excellent retention of conidial masses in this study. The simple and versatile procedure for demonstrating fungal spores and their exudation from fruiting bodies would facilitate characterization of diverse pathological and environmental isolates as they are in native environments.

Light and Electron Microscopic Study on the Development of Nosema Bombyics Naegeli in the Midgut of Silkworm Bombyx Mori L.

  • Infection effect of $Nosema$ $bombyics$ on the midgut of silkworm $Bombyx$ $mori$ and subsequent appearance of spores and the performance of larvae was studied. Autopsy of larvae showed white pustules on the surface of midgut at 5 days of post infection (pi). At later stage, important organs like midgut, silk gland and gonads reduced in size and all these organs showed white pustules. Light microscope observation of pustules revealed enormous spores. Spore multiplication was at a faster rate in young larvae. Infection of the adult larvae resulted in pebrinized pupa and moths. Larval weight, cocoon weight and cocoon shell ratio reduced as the post infection period increased. Transverse sections of midgut showed $N.$ $bombycis$ infection limited to a few columnar cells at 3-5 days of pi. At 7 days pi, cell volume increased, cells were swollen and elongated. Heavily infected cells looked like sacks filled with parasite and the apical region of certain cells were bulging into the gut lumen. Later at 8-9 days of pi, spores or its developing stages leaked into the lumen either freely or enclosed within the globules of host cytoplasm. Besides columnar cells, development of $N.$ $bombycis$ was observed in the regenerative cells and rarely in goblet cells. Development of $N.$ $bombycis$ was also observed in both longitudinal and circular muscles at the late pi period. The histopathological changes, deformities and spore production time in the host were all influenced by the spore dosage and age of the host.

Physio-Morphological Changes in a Riboflavin Producer Eremothecium ashbyii DT1 and UV Mutants in Submerged Fermentation

  • By UV-irradiation of Eremothecium ashbyii DTl, a higWy flavinogenic mutant (UV-18-57) and a nonflavinogenic mutant (UV -85) were obtained. The physio-morphological characteristics of these three strains were studied on glucose medium in submerged fermentation. Glucose utilization and mycelial growth occurred in 0 - 2 days of fermentation. By the third day, the biomass had declined. Extracellular riboflavin excretion was distinct from the second day, reaching a maximum rate by the fourth day. The hyphae of the highly flavinogenic mutant UV-18-57 were broader than DTl, while the nonflavinogenic UV-85 hyphae were very thin. Riboflavin accumulation was high in UV-18-57 (extracellular riboflavin,$825\mu\textrm{g}/ml$ , and intracellular, $490\mu\textrm{g}/ml$) and caused the mycelia to swell into bulbous forms. Riboflavin accumulation was less in DTl ($108\mu\textrm{g}/ml$ extracellular and $24\mu\textrm{g}/ml$ intracellular) and correspondingly its hyphae were thinner than those of UV-18-57 and swollen bulbous mycelia were not prominent. UV-85 was nonflavinogenic and, accordingly, its mOlphological characteristics included long thin filaments with no intracellular riboflavin accumulation. A large number of greenish fluorescence spores were seen in UV-18-57, whereas DTI had less spores and UV-85 was nonsporulating. Sporulation is correlated with riboflavin production. UV-18-57 had better mycelial integrity and lysis started only by the seventh day, whereas DTI and UV -85 started to lyze earlier by 4 -5 days. By the late stage of fermentation (eighth day), DTl had a few long, thin filaments indicating some secondary growth, whereas UV -85 showed a compact pellet form of mycelia. Most mycelia of UV-18-57 still appeared intact.

Penicillium verruculosum 으로부터 분생자 원형질체의 생성과 재생 (Formation and Regeneration of Conidial Protoplast from Penicilliun verruculosum)

  • 섬유소 분해능이 우수한 P. verruculosum 의 분생포자로부터 원형질체의 생성 및 재생에 대하여 조사하였다. 균의 원형질체 생성을 위한여 2-deoxy-D-glucose(2-DG) 가 첨가된 최소배지에서 분생포자를 10-12 시간 전배양 시킨후 시판 세포벽분해효소들을 작용시켜 본 결과 그중 Novozyme 234 (1%, w/v) 에서 50% 이상의 가장 좋은 원형질체 생성 수율을 보였다. 전배양 되지 않은 포자로부터는 원형질체 생성이 이루어지지 않았으며 원형질체 생성율을 향상시키기 위한 첨가테로서 2-DG 의 효과가 무첨가에 비해 2배 이상의 수율을 증가시켰다. 포자 원형질체의 재생율은 49.2% 로 동 균주의 균사체 원형질체의 재생율4.6-27.8%(삼투압 조절체로서 0.6 M magnesium sulfate) 보다 높았다. 원형질체 생성 및 재생을 위한 상투압 안정제로서는 0.6M ammonium sulfate 와0.6 M magnesium sulfate가 가장 적절하였다. 광학 형미경을 통하여 부풀린 포자로부터 원형질체의 생성과정과 두 가지 각기 다른 양상의 재생과정이 관찰되었다.
