Studies on the Toxigenic Fungi Isolated from Local Grains and Foodstuffs (수종(數種) 곡류(穀類) 및 식품(食品)에서 분리(分離)한 진균(眞菌)의 유독성(有毒性)에 관(關)한 연구(硏究))

  • These studies were carried out to detect the presence of mycotoxin producing fungi in various kind of grains and foodstuffs in Korea. The experiments were divided into three parts: bacteriologic, toxicologic and electron microscopic studies. The results were summarized as follows: 1. From the 133 various samples, 426 colonies of fungi were isolated. In 405 of 426 colonies, it was possible to identify 17 genera. Among the identified strains, the predominant genera were Penicillium, Aspergillus and Alternaria. 2. In cytotoxicity test, 20 strains showed mild to severe toxic effects in mice and 24 strains showed toxic effects on HeLa cells among the 107 strains of experiments. 3. In electron microscopic studies of liver cells from animal which had been treated with toxin like substances, the liver cells showed the cytoplasmic changes: dilatation of endoplasmic reticulum, swelling of mitochondria, disappearance of mitochondrial cristae, increased number of lipid and glycogen particles. Nucleus and nuclear envelope alterations were also noted. 4. In fine structure of HeLa cells treated with culture filtrates of mycotoxin producing fungi and experimental strains had been noted a certain specific changes induced by culture filtrates. These alterations showed the disappearance of golgi complex, endoplasmic reticulum vacuolization and mitochondrial changes. 5. As a mass screening, the cytotoxicity tests of HeLa cells and histopathologic study of mice liver cells treated with toxin-like substances, employed are feasible to detect mycotoxin producing fungi.

Development of Cold Chain System Using Thermal Storage with Low-Energy Type (저 에너지형 축냉식 저온유통 시스템 개발)

  • The purpose of this study is to find the optimal conditions of PCM slurry manufacturing equipment for saving the marketing cost and keeping the original quality of products. In addition, the characteristics of the movable container for shipping or distributing products is analysed. The major results are as follows. 1. PCM thermal storage system is designed with the conditions of temperature($-5{\sim}10^{\circ}C$), cold chain time(30 minutes), and one time usage(50 liter). This system includes tank, freezer, circulating pump, cycle type heat exchanger, swelling tank, equipment of supplying PCM supplying unit includes cold tank, cycle type heat exchanger, suction unit and control equipments, etc. 2. After ability test of PCM thermal storage system, it shows that the required freezing time of PCM thermal storage system is less than one of the previous system. The reason is that churn (top and bottom) and compulsion circulation are occurred simultaneously and unit cooler type method is better than chiller type method. 3. By the experiment of transportation latent heat container, it is decided that the best container is $K_1$ with latent heat temperature($0{\sim}5^{\circ}C$) and density(0.15%). However, for $K_l\;and\;K_2$, it is necessary more studies on latent heat thermal conditions and conditions of making method.

Nuclear Remodeling and In Vitro Development Following Somatic Cell Nuclear Transfer in Swine

  • This study was conducted to investigate nuclear remodeling and developmental rate following nuclear transfer of fetal fibroblast cells, ear skin cells and oviduct epithelial cells into porcine recipient oocytes. To test par-thenogenetic activation, oocytes were treated with a 6-dimethylaminopurine (6-DMAP), a single DC-pulse (DC), calcium ionomycin (ionomycin), DC+6-DMAP and ionomycin + 6-DMAP after in vitro maturation. For nuclear transfer, in vitro matured oocytes were enucleated, and donor cells were transferred into oocytes. Cloned embryos were fused and stimulated with 6-DMAP for 4 h and cultured in vitro for 6 days. Among treatments for parthenogenesis, the activation rate of DC +6-DMAP treatment was significantly higher than that of single treatment roups (p<0.01), except for DC treatment group. However, the difference was not significant in activation rate compared to other complex treatment groups. Nuclear swelling of the cloned embryos was initiated at 60 min after stimulation and increased afterwards. Fusion rates were not different among different donor cells. Cleavage rates of DC treatment groups were significantly higher than those of DC+6-DMAP treatment groups (p<0.05) in case that fetal fibroblast and ear cells were used for nuclear donor. The cloned embryos from developed to blastocysts in oviduct epithelial cell nuclear transfer with DC+6-DMAP treatment was significantly higher compared to those with DC only treatment (p<0.05). However, no blastocyst was developed from nuclear transfer of fetal fibroblast and ear cells regardless of activation treatments. Based on these results, a proper activation stimulation may be necessary to increase the activation rate and the development to blastocyst in cloned porcine embryos.

Analysis of Sexed Sperm by Flow Cytometry in Hanwoo (Korean Native Cattle)

  • This study evaluated a sexed sperm ability to produce embryos by flow cytometer. Hanwoo bulls sperm were separated to X and Y sperm via Hoechst 33342 stained with near UV laser or performed the pre-sorted without near UV laser beam in flow cytometry. Pre-sorted sperm had significantly higher viability ($84{\pm}1.15%$, $p$<0.05) compared to other sorted groups in frozen-thawed semen. For fresh semen, pre-sorted sperm had the higher viability ($79{\pm}3%$, $p$<0.05) than those of the X and Y sperm ($44.7{\pm}1.67$ and $41.7{\pm}1.2%$) separated by differences of DNA content. On the other hand, pre-sorted and X sperm sorted according to differences in DNA content had significantly higher viabilities ($24.3{\pm}1.2$ and $25.7{\pm}0.9%$, $p$<0.05) compared to that of the sorted Y sperm ($13.7{\pm}1.2%$) in the hypoosmotic swelling test. The proportion acrosome reaction in the sorted X sperm was higher ($55.0{\pm}1.7$ and $45.0{\pm}1.5%$) than those of the sorted Y-sperm ($32.3{\pm}0.9%$, $p$<0.05). However, the sperm morphologies of the sorted groups were not significantly differences. In conclusion, the sex-sorting procedure by flow cytometry affected some characteristics of Hanwoo sperm. Further study is needed to determine the optimal procedures to enhance male and female embryos and sorting accuracy.

Pervaporation Dehydration of Acetic Acid Aqueous Solution using PVA/PAA Membrane with Na-Y Zeolite (Na-Y 제올라이트가 첨가된 PVA/PAA 분리막의 아세트산 수용액에서 투과 증발 연구)

  • Membranes were prepared by incorporating Na-Y zeolite particles into poly(vinyl alcohol) (PVA) cross-linked with poly(acrylic acid) (PAA). The membrane was characterized by FT-IR spectroscopy, contact angle measurement, swelling test, SEM analysis, and XRD analysis. The pervaporation separation of water/acetic acid mixtures was carried out using prepared membranes. From the results, it was shown that the hydrophilic property of prepared membrane increased with increase of zeolite contents and the PVA/PAA membrane with zeolite addition showed higher permeation flux than that of without zeolite membrane. The PVA/PAA membrane containing 8 wt% zeolite showed the highest permeation flux and separation in the feed solution containing 10 wt% acetic acid.

The Clinical Study of the Efficacy of Lizhongtang plus Baidusan(理中湯合敗毒散) on Allergic Rhinitis (理中湯合敗毒散의 알레르기성 비염 치료효과에 대한 임상적 연구)

  • Objectives : The purpose of this clinical report was to estimate the efficacy of Lizhongtang plus Baidusan(理中湯合敗毒散) on allergic rhinitis. Methods : We prescribed Lizhongtang plus Baidusan(理中湯合敗毒散) for 15 patients who visited Kabsan oriental clinic with allergic rhinitis. We inquired into distribution of age, case history period, improvement of symptoms, etc. We used the statistical methods of student t-test in order to analysis of the different of symptom before and after treatment(p 〈0.05). Results : 1. The distribution of sex & age was as follows; Total patient were women. 10-19 years 2 cases, , 30-39 years 5 cases, 40-49 years 6 cases and 50-59 years 2 cases. 2. The case of history period was as follows; under a year 1 case, 1-3 years 3 cases, 3-5 years 2 cases, 5-10 years 4 cases and over 10 years 5 cases. 3. The amount of herbal medicine they took as follows: 1 je(劑) 1 case, 2 je(劑) 7 cases, 3 je(劑) 5 cases and 4 je(劑) 2 cases. 4. The frequency of acupuncture and moxibustion was as follows; under 5 times 1 case, 6-10 times 10 cases, 11-15 times 3 cases, over 16 times 1 case. 5. The nasal symptoms except the nasal mucosa color(pale) were improved significantly after treatment(p 〈0.05). 6 The general symptoms except menstruation pain were improved significantly after treatment(p 〈0.05). 7. The curative influence of rhinitis by Lizhongtang plus Baidusan(理中湯合敗毒散) was as follows excellent 2 cases. good 11 cases, ineffectiveness 2 cases. Conclusion: If we administer Lizhongtang plus Baidusan(理中湯合敗毒散) to allergic rhinitis patients with cold in the Zhong Jiao of deficiency type (中焦虛寒), it improves not only allergic symptoms such as the watery rhinorrhea, turbinate swelling, sneeze, nasal obstruction, etc but also coldness of the limbs(手足冷症), dyspepsia, dysmenorrhea, etc. Therefore, we consider that administration of Lizhongtang plus Baidusan(理中湯合敗毒散) to allergic rhinitis patients with cold in the Zhong Jiao of deficiency type (中焦虛寒) improves in quality of life, as removes inconvenience of life and stress.

Preparation and Antimicrobial of Zinc Calcium Alginate Films according to Concentration (농도에 따른 알긴산 아연칼슘 필름의 제조 및 항균성)

  • Composite films of zinc calcium alginate were prepared by a film maker from 7 wt% sodium alginate solution and then they solidified into 3, 5 wt% content $ZnCl_2$, $CaCl_2$ solution followed by washing and drying at room temperature. The characteristics were measured with several methods (antimicrobial activity, water solubility, swelling ratio and viscosity, SEM, EDS) and the film properties were investigated. Composite films of zinc calcium alginate showed an increase in the water resistance by increasing $ZnCl_2$ and $CaCl_2$ content and the antimicrobial test showed that the calcium alginate as well as zinc alginate films result in excellent antimicrobial activity in the two strains, Staphylococcus and Escherichia coli. The results show the possible improvement of the physical properties of composite films.

In situ Gel Forming Stereocomplex Composed of Four-Arm PEG-PDLA and PEG-PLLA Block Copolymers

  • Injectable hydrogels are quite promising materials due to their potential to minimize invasive implantation and this provides versatile fitness irrespective of the damaged regions and facilitates the incorporation of bioactive agents or cells. In situ gel formation through stereocomplex formation is a promising candidate for injectable hydrogels. In this paper, a new series of enantiomeric, four-arm, PEG-PLA block copolymers and their stereocomplexed hydrogels were prepared by bulk ring-opening polymerization of D-lactide and L-lactide, respectively, with stannous octoate as a catalyst. The prepared polymers were characterized by $^1H$ nuclear magnetic resonance (NMR) spectroscopy, Fourier-transform infrared (FT IR) spectroscopy, gel permeation chromatography (GPC) and thermal gravitational analysis (TGA), confirming the tailored structure and chain lengths. The swelling and degradation behavior of the hydrogels formed from a selected copolymer series were observed in different concentrations. The degradation rate decreased with increasing polymer content in the solution. The rheological behavior indicated that the prepared hydrogel underwent in situ gelation and had favorable mechanical strength. In addition, its feasibility as an injectable scaffold was evaluated using a media dependence test for cell culture. A Tris solution was more favorable for in situ gel formation than PBS and DMEM solutions were. These results demonstrated the in situ formation of hydrogel through the construction of a stereocomplex with enantiomeric, 4-arm, PEG-PLA copolymers. Overall, enantiomeric, 4-arm, PEG-PLA copolymers are a new species of stereocomplexed hydrogels that are suitable for further research into injectable hydrogels.

An Investigation of the Characteristics of Sponge Cake with Ginseng Powder (인삼 분말을 첨가한 스폰지 케익의 품질 특성에 관한 연구)

  • The characteristics of sponge cake were investigated with various flour types and amounts of ginseng powder. Sponge cake made with medium flour in combination with 2 and 4% mixtures of ginseng powder was found to be optimal as a cake flour control, being unaffected by the addition of ginseng powder. The results of alveogram showed that increment of ginseng powder decreased over pressure(P), extensibility(L) and swelling index(G) value. Farinogram, consistency, water absorption, stability, time to breakdown and the farinograph quality number all decreased with increasing ginseng powder however, the longest development time was the medium flour with a 2% ginseng powder mixture. Water activities were not significantly affected by addition of ginseng powder, however the 2% ginseng powder mixture showed a slight decrease in water activity. Addition of ginseng powder caused an increase in darkness, as well as redness and yellowness. The results of the sensory test suggest that the 2% ginseng powder mixture was preferable for the cake flour control.

  • M$\acute{e}$n$\acute{e}$trier's disease is a rare form of acquired gastropathy characterized by giant rugal folds in the stomach and protein-losing gastropathy. Children with M$\acute{e}$n$\acute{e}$trier's disease tend to follow a benign self-limited course with symptoms typically completely resolving within 2 to 10 weeks in contrast to the chronic course in adults. A 9-year-old girl presented with a history of gradually worsening abdominal distension, increasing body weight, and abdominal pain for 2 weeks. Physical examination on admission indicated periorbital swelling, pitting edema in both the legs, and abdominal distension with mild diffuse tenderness and shifting dullness. Laboratory tests on admission showed hypoalbuminemia, hypoproteinemia, and peripheral eosinophilia. The test result for anti-cytomegalovirus immunoglobulin M was negative. Increased fecal alpha 1 anti-trypsin excretion was observed. Radiological findings showed massive ascites and pleural effusion in both the lungs. On gastroscopy, large gastric folds, erythema, erosion, and exudation were noted in the body and fundus of the stomach. Microscopic findings showed infiltration of eosinophils and neutrophils in the gastric mucosa. Her symptoms improved with conservative treatment from day 7 of hospitalization and resolved completely.
