Antiserum Preparation of Recombinant Sweet Potato Latent Virus-Lotus (SPLV-Lotus) Coat Protein and Application for Virus-Infected Lotus Plant Detection

  • Lotus is one of the most important aquatic vegetables in China. Previously, we detected sweet potato latent virus from lotus (SPLV-lotus) and found that it has highly significant sequence diversity with SPLV-sweet potato isolates (SPLV-sp). Here, we developed serological methods for the detection of SPLV-lotus in Chinese lotus cultivation areas. Based on the high sensitivity of SPLV-lotus coat protein antiserum, rapid, sensitive and large-scale diagnosis methods of enzyme-linked immunosorbent assay (ELISA) and dot blot in lotus planting area were developed. The established ELISA and dot blot diagnostic methods can be used to detect SPLV-lotus from samples successfully. And our results also showed that the SPLV-lotus and sweet potato isolates appeared clearly distinction in serology. Our study provides a high-throughput, sensitive, and rapid diagnostic method based on serology that can detect SPLV on lotus, which is suggested to be included in viral disease management approach due to its good detection level.

자색고구마 Anthocyanin 색소의 추출조건 결정 (Determination of the Conditions for Anthocyanin Extraction from Purple-Flashed Sweet Potato)

  • To establish the optimum conditions for the extraction of anthocyanin pigment from purple-fleshed sweet potato, a suitable extraction solvent with the optimum citric acid concentration for acidification of the solvent, and the optimum extraction time and temperature were determined. Twenty percent ethanol solution acidified with citric acid was found to be a good solvent for the extraction of the pigment from purple-fleshed sweet potato. About 10 hour extraction at room temperature was appropriate for the extraction. pH of the extract was below 3 when more than 0.7% citric acid was added. The higher the concentration of citric acid added was, the higher the total optical density (TOD) of the extract was. However, the increase in TOD of the extract was insignificant when more than 1% of citric acid was added. Therefore, addition of 1% citric acid was determined for acidification of the extracting solvent. Though the initial rate of the pigment extraction increased as the extracting temperature increased, extraction at higher temperatures of 60 or 8$0^{\circ}C$ for an extended time caused a decrease in the extraction yield due to degradation of the pigment. The optimum extraction temperature for the anthocyanin pigment from purple-fleshed sweet potato with the solvent used was determined as 4$0^{\circ}C$.

Variation for Fruit Yield and Quality Characteristics in Sweet Pepper (Capsicum annuum L.) Germplasm Collection

  • Fifty-five sweet pepper (Capsicum annuum L.) germplasm from the Rural Development Administration (RDA) were analyzed for their fruit yield and fruit quality characters, including fruit weight, fruit size, pericarp thickness, total soluble content, fruit lobe, fruit shape, fruit surface and fruit color. Fruit yield per plot ranged from 200 g to 8150 g (average, 1884 g). Fruit fresh-weight per fruit ranged from 27.3 g to 200.0 g with an average of 97.2 g. Fruit length varied from 4.0 cm to 16.2 cm (average, 9.6 cm). Fruit length-to-width ratio ranged from 0.6 to 3.0 with an average 1.7. The fruit pericarp thickness ranged from 1.8 mm to 10.7 mm (average, 4.8 mm). Total soluble content was the highest ($8.5^{\circ}Brix$) in K156286 and the lowest ($5.3^{\circ}Brix$) in K156208 with the average value $6.7^{\circ}Brix$. Fruit lobes numbers varied from 2 to 4. Blocky, triangular, elongated, rectangular and round fruit shape identified among the sweet pepper accessions. Fruit surface varied from smooth to wrinkle and fruit color also varied. The presence of variability in fruit yield and quality traits within the sweet pepper genotypes can be utilized to develop high yielding sweet pepper variety with better fruit quality characters.

  • Suppressed Wave equation Estimation of Traveltime (SWEET, Shin et, al., 2000) 알고리즘을 이용하여 등방성 및 이방성 탄성 매질에 대한 ched 주시와 진폭을 계산하는 방법을 개발하였다. SWEET 알고리즘을 2차원 등방성 및 이방성 탄성 매질의 속도 구조에 적용하여 P파의 초동 주시와 진폭을 계산 할 수 있었다. 본 논문에서는 간단한 등방성 균질 탄성 모형, 복잡한 등방성 탄성 모형 및 간단한 이방성 균질 모형에 대한 수치 계산 결과를 보여 줄 것이다.

  • 본 논문에서는 4개의 스피커를 이용한 음향 재생 환경에서 청취자의 위치에 따라 최적의 sweet spot을 제공하도록 스피커를 구동하는 방법을 제안한다. 먼저 청취자 위치에 대하여 대칭 구조를 가지는 스피커 쌍(pair) 후보들을 선택한다. 다음, ringing 현상을 감소시키기 위하여 앞에서 선택한 스피커 쌍 후보 중에서 스피커 간 간격이 가장 먼 스피커 쌍과 이 스피커 쌍 사이에 있는 모든 스피커를 구동 스피커로 결정한다. 마지막으로, cross-talk cancellation 행렬의 크기를 제한하여 ringing 현상을 감소시킨다. 모의실험을 통해, 청취자 위치가 변할 때 제안하는 스피커 구동 방법을 사용하여 sweet spot 면적이 일정한 수준으로 유지되어 기존 방법에 비하여 성능이 향상된 것을 확인하였다.

  • $\beta$-Amylase was purified from sweet potato by acetone fractlonatlon, Sephadex A-50 ion exchange chromatography and Sepgadex G-200 gel chromatographyl The higher enzyme concentration was, the higher heat stability of enzyme became. After 1 hour 30 minute. At 6$0^{\circ}C$ in pH 5, enzyme under concentration of 30$\mu$l/ml lost its activity completely and over the concentration of 100$\mu$g/ml remained 25% of activity. The enzyme was stabilized at range of pH 4~10 and pH stability was increased by glycerol. Five moles of NaCl inhibited completely of the enzyme activity. SDS of 0.05% inhibited the enzyme completely after 12 hours at 37$^{\circ}C$ in pH5. One mole guanidine-HCl and 8M urea inhibited the entire enzyme after 13 hours at 37$^{\circ}C$ in pH 5.

Fruit Soft Rot of Sweet Persimmon Caused by Mucor piriformis in Korea

  • A fruit soft rot caused by Mucor piriformis occurred on sweet persimmon storages in Jinju, Changwon and Gimhae, Gyeongnam province, Korea, 2003. The disease infection usually started from wounding after cracking of fruits. At first, the lesions started with water soaked and rapidly softened and diseased lesion gradually expanded. Colonies on potato dextrose agar at $20^{\circ}C$ were whitish to olivaceous-buff Sporangia were globose, black and $96{\sim}153{\mu}m$ in size. Sporangiophores were $26{\sim}42{\mu}m$ in width. Sporangiospores were ellipsoid and $5.8{\sim}10.6{\times}4.3{\sim}7.6{\mu}m$ in size. Columella was obovoid, cylindrical-ellipsoidal, pyriform, subglobose and $80{\sim}125{\mu}m$ in size. Optimal temperature for mycelial growth was $20^{\circ}C$ on PDA. The causal organism was identified as M. piriformis. This is the first report of fruit soft rot on sweet persimmon caused by M. piriformis in Korea.

단감나무 둥근갈색무늬병의 발생 상태 (Occurrence of Leaf Blight of Sweet Persimmon Tree Caused by Pestalotiopsis theae in Korea)

  • Leaf blight outbreak was investigated in sweet persimmon tree orchards in Korea during a three-year period from 1995 to 1997. The man percentage of sweet perismmon leaves blighted by Pestalotiopsis theae was 15.9%. The highest disease incidence was surveyed to 20% in Milryang. The disease began from early June to late October, and peaked in September and October. The lesion size on leaf ranged mainly 1-3 cm. The typical symptoms were large grayish concentric lesions of oval patterns of either mesophyll or margin of the leaf, and olde trees were more susceptible than younger ones.

Mycoplasma성 고구마 위축병에 관한 연구 - 병증 및 병원체 (Studis on the Mycoplasmic Witches'-Broom of Sweet Totato in Korea (I))

  • The sweet potatoes infected with witches'-broom disease were collected in the growing fields in Jeon-bug area, Korea. As a possible control plant, Ipomoea batatas L.var. Suwon 147 was selected. The pathogen was identified by various methods ; such as mechanical transmission, antibiotic reactions and electron microscopy. In the results attained the author believed the pathogen of the sweet potato infected with witches' broom to be a mycoplasma-like organism. the results are as follows : 1. Mycoplasma-like bodies were occurred in the phloem region of the sweet potatoes infected with witches'-broom and its particles were sized in the range of about 200-2,500m.mu.. The membrane of the pathogen was observed to be made of an unit. 2. Responsibilities to the antibiotices were sensitive in case of tetracycline and terramycin, and root dipping method showed remarkable than foliage spray. 3. The infection was developed by the grafting transmission but by the insects, Myzus persicae and Cicadella viridis. 4. rosette, witches'-broom, stunt, yellowish, mosaic and necrosis were observed as the symptomps of the disease.

단감 자동정렬 포장장치 개발 (Development of an Automated Ordering and Packing Units for Sweet Persimmons)

  • The sweet persimmons are producted 227,394 tons per year in Korea. They are preserved at cold storage after wrapping them in packing vinyl to maintain quality. Using conventional handwork, a skillful worker could probably produce only 1,000 packings a day. Hiring many skilled-labors to wrap sweet persimmons in packing vinyl seems to be difficult because of the recent rural crisis. Therefore, it was necessary to be developed for the automated ordering and packing unit that is convenient and laborsaving. This study were summarized as follows : Success rates of 99%, 100%, and 88% were observed for ordering parts, sealing and cutting films, and sealing films in the inlet part, respectively. The time required per 1 cycle was 16 seconds and the developed unit performed 225 cycles an hour. The unit showed an ability of 1,125 pieces per hour. Therefore, the productivity was 10 times greater than that of skillful handwork.
