Antioxidative Activity and Nitrite Scavenging Ability of Methanol Extract from Salvia plebeia R. Br. (배암차즈기 메탄올 추출물의 항산화 활성 및 아질산염 소거능)

  • Efficacy of antioxidative activity and nitrite scavenging ability of methanol extract from Salvia plebeia was investigated. Electron-donating ability of extract at RC$_{50}$ was 51.10 ${\mu}$g/ml. After addition of 0.92 mg/ml extract, autooxidation of pyrogallol decreased to 62.11% by superoxide dismutase-like activity. In antioxidative activity of extract against linoleic acid during incubation times of 1,2, and 4 days at 40$^{\circ}$, lipid peroxidation values significantly decreased by 72.86%, 77.18%, and 78.32% with addition of 0.2 mg/ml, respectively. Total phenolic content was determined as gallic acid equivalents (GAE) and values revealed as 202.35${\pm}$ 1.26 GAE ${\mu}$g/ml of extract. Nitrite scavenging ability showed the most remark-able effect at pH 1.2, exhibited to 44.29% by addition of 0.2 mg/ml. These results suggest that methanol extract from Salvia plebeia can be used as bioactive and functional materials.

Antihypertensive Effects of Novel Isoflavone-Free Black Soy Peptide Mixture as HO-1 Inducer (Heme 산화효소 발현 유도체로서 Isoflavone-Free 검은콩 펩타이드의 항고혈압 활성)

  • We previously reported that the novel isoflavone-free peptide mixture (black soybean peptide, BSP) had several beneficial effects like antiobesity and hypotriglyceridemic effect. However, there are no reports for BSP on anti-hypertensive activity. BSP induced heme oxygenase-1 (HO-1) in HUVECs, thus investigated the HO-1-induced activity in HUVECs and the anti-hypertensive effects in SHR animal model. BSP significantly induced HO-1 expression both at transcriptional and protein levels in a time- and dose-dependent manner as measured by RT-PCR and Western blot analysis, respectively. These inductions were abolished by pretreatment of N-acetyl-cystein (NAC, 1~10 mM), but not by employing Tempol, a superoxide dismutase (SOD) mimetic (1~5 mM). As expected, enzymatic activity (~2 fold) determined by bilirubin formation assay and cGMP concentration (~6 fold) were significantly increased in BSP-treated cells. Based on the numerous evidences on the beneficial effects of HO-1 and our results, we investigated in vivo effects of BSP on the antihypertensive activity. The administration of BSP (1% in drinking water) significantly decreased mean blood pressure (BP) (from $218.6{\pm}6.99$ to $190.0{\pm}3.42$ mm Hg, p<0.01). This result indicates that BSP is strong inducer of HO-1 expression, which may be triggered by oxidative stress, and has anti-hypertensive activity.

Effects of Extraction Methods on the Antioxidative Activity of Artemisia sp. (추출 방법에 따른 쑥 추출물의 항산화 활성)

  • The effect of extraction methods, such as reflux extraction (RE), autoclave extraction (AE), low temperature high pressure extraction (LTPE) and ultrasonification extraction (USE) on antioxidant activity of various species of Artemisia (Artemisia capillaris T., Artemisia princeps P., Artemisia annua L.) was investigated. The extraction yield of RE and AE was higher than other methods tested for all Artemisia. The total polyphenol and flavonoid content of Artemisia sp. extracts from RE was highest of the extraction methods tested. The total polyphenol and flavonoid content of A. capillaris T. extracted by RE was 260.82 mg GAE/g and 11.52 mg RHE/g, respectively. The A. capillaris T. extract showed higher DPPH and ABTS radical scavenging activity than that of the other tested Artemisia sp. Nitrite scavenging activity and superoxide dismutase (SOD)-like activity of various extracts from RE was 45.48% and 68.29% (A. capillaris T.), 45.73% and 61.43% (A. princeps P.), and 44.25% and 58.19% (A. annua L.), respectively. The RE method was the most effective method for extracting antioxidant substances from various A. capillaris T. compared with AE, LTPE and USE. These results suggest that extracts of Artemisia sp. from RE can be used as bioactive and functional materials in the food industry.

Enhancement of Antioxidant Activities of Bark of Berberis koreana Palibin by Lactic Acid Fermentation (유산균 발효를 통한 매자나무 수피부의 항산화 활성 증진)

  • The bark of Berberis koreana Palibin was fermented by Bifidobacterium longum B6 and Lactobacillus paracasei at $37^{\circ}C$ for 72 hour, then extracted by water solvent at $100^{\circ}C$ for 180 min. Total pholyphenol and flavonoid contents were improved by fermentation process, compared to conventional water extraction. The barks of B. koreana Palibin extracts by B. longum B6 (FE-B.L.) and by L. paracasei (FE-L.P.) showed 17% and 16% cytotoxicity on human normal cell lines(HEK293) at 1.0 mg/$m{\ell}$ of the highest concentration, respectively, which was about 3~5% lower than 20% from normal extracts (NE). DPPH radical scavenging activity of the FE-B.L. and FE-L.P. were about 73% and 75.9% higher than 56.8% of NE. The highest inhibitory potency on xathine oxidase and superoxide dismutase (SOD)-like activities were also measured as 31.9% and 61.9% by adding FE-L.P. of 1.0 mg/$m{\ell}$. On tyrosinase inhibition test, the FE-L.P. showed highest activity as 75.9% at 1.0 mg/$m{\ell}$. Generally, FE-L.P. showed higher antioxidant activities as well as higher tyrosinase inhibition activity, possibly due to high contents of total pholyphenol and flavonoid. In general, fermentation of barks of B. koreana Palibin has relatively better biological activities than normal extracts. Specifically, the extracts fermented by L. paracasei showed higher activities than that from B. longum B6.

Physiological Function and Development of Beverage from Grifola frondosa (잎새버섯 추출물의 생리 기능 및 음료 제조)

  • Contents of polyphenol compounds and the physiological activity of extracts from Grifola frondosa by water and methanol extraction were investigated to determine their functional effects. A functional beverage was developed using the extracts. The yield and phenolic compounds content of the water extracts were highest (49.2% and 327 mg/100 g, respectively), while for the methanol extraction method they were 28.7% and 130 mg/100 g, respectively. DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity was 76.3% for the water extract and 65.4% for methanol extract, whereas the superoxide dismutase (SOD)-like activity was low ($26.3{\sim}36.8%\;at\;1,000{\mu}g/mL$ concentration) Angiotensin converting enzyme (ACE) inhibitory effect of water extract (75.1%) was higher compared to the methanol extracts (41.2%). Tyrosinase inhibition activity was 42.5% for the water extracts and 31.8% for the methanol extracts at $1,000{\mu}g/mL$ concentration. The most acceptable formulation for G. frondosa beverage developed was 0.5% G. frondosa water extract, 8.0% oligosaccharide, 2.0% green tea extract, 2.0% jujube extract, 1.0% Solomon's seal extract, 0.01% vitamin C, and 2.0% apple extract. The final product had 9.8 Brix and color values of L, 35.2+1.1; a, 3.2+0.2; b, 13.6+0.3.

Screening of Bioactive Compounds from Mushroom Pholiota sp. (비늘버섯으로부터 생리기능성 물질의 탐색)

  • Extracts from 63 kinds of Pholiota sp. fruiting bodies were prepared using water and methanol, and then their physiological functionalities were investigated. The methanol extracts from Pholiota adiposa PAD030 showed high fibrinolytic activity and those of P. adiposa ASI PAD-022 showed potential inhibitory activity of 76.8% against ${\beta}-hydroxy-{\beta}-methylglutaryl(HMG)-CoA$ reductase. The highest antioxidant and tyrosinase inhibitory activities were found in the water extracts of Pholiota sp. PSP-015 (72.7%) and methanol extracts of P. nameko PNA-024 (69.5%), respectively. However, superoxide dismutase(SOD)-like activity and elastase inhibitory activity were low in almost of the extracts. The HMG-CoA reductase inhibitor from the fruiting body of P. adiposa PAD-022 which showed the highest functionality was extracted maximally when powder of the fruiting body was shaked at $30^{\circ}C$ for 12 h by methanol and its HMG-CoA reductase inhibitory activity was 80.2%.

Antioxidative and Fibrinolytic Activity of Extracts from Soybean and Chungkukjang(Fermented Soybeans) Prepared from a Black Soybean Cultivar (검은콩의 품종에 따른 콩과 청국장 추출물의 항산화능 및 혈전용해능)

  • We sought to develop natural preservatives or functional health foods from black soybeans (Yakkong, Seoritae and Chungkukjang). These materials were extracted with distilled water or 70% (v/v) ethanol, and the extracts sanalyzed for polyphenol content and tested for antioxidative and fibrilytic activities. The polyphenol contents of water extracts from Yakkong and Yakkong Chungkukjang was 316.23 mg/100 g, 896.01 mg/100 g, respectively, whereas those from Seoritae and Seoritae Chungkukjang were 304.28 mg/100 g and 875.23 mg/100 g. The polyphenol contents of water extracts from Chungkukjang were 2.8-fold higher than those from black soybean. The electron-donating abilities (EDAs) and superoxide dismutase (SOD)-like activities of water extracts from black soybean Chungkukjang were higher than those of water extracts from black soybean, but both extracts had fibrinolytic activity, the highest activity was present in water extracts of Yakkong Chungkukjang. The polyphenol contents, and antioxidative and fibrinolytic activities of extracts from black soybean Chungkukjang were higher than those of other black soybean extracts from black soybeans. These results indicate that black soybean Chungkukjang can be used for development of a health food or as a natural antioxidant.

Antioxidant, Antimicrobial and Anticancer Properties of Seven Traditional Herb-combined Remedies (7가지 한약재 처방전에 대한 항산화·항균·항암활성에 대한 연구)

  • In this study, we investigated the antioxidant activities and compared other physiological properties including anti-cancer and antimicrobial effects of several traditional Korean herb-combined remedies such as Gilgyung-tang (GGT), Daihwangmokdan-tang (DHMDT), Sagan-tang (SGT), Socheonryongtang (SCRT), Sihocheonggan-tang (SHCGT), Sipyukmiyuki-eum (SYMYKE) and Hwangheuk-san (HHS), which were recorded in “Dong-eui-bo-gam” for “Ongjeo”. Total phenolic contents of the herb medicines were in a rich order of GGT < SYMYKE < SCRT < SHCGT < DHMDT < SGT < HHS. Among them, HHS appeared highest in superoxide dismutase-like activity, ferric reducing antioxidant power, scavenging of 2,2’-diphenyl-1-picrylhydrazyl, and 2,2’-azinobis(3-ethylbenzothiazoline-6-sulfonate) cationic radical activity. Interestingly, there was a positive relationship between their total phenolic contents and their antioxidant activities. Although all of them showed anti-proliferative activities in human colon cancer HCT-116 cells, HHS was seven times higher than GGT. Antimicrobial activities against Escherichia coli and Helicobacter pylori were revealed only on SGT, SCRT, SYMYKE and HHS. Taken together, these findings reveal the potential use of traditional Korean herbal formulas as functional ingredients in antioxidant and physiological materials.

Rutin alleviated lipopolysaccharide-induced damage in goat rumen epithelial cells

  • Objective: Rutin, also called vitamin P, is a flavonoids from plants. Previous studies have indicated that rutin can alleviate the injury of tissues and cells by inhibiting oxidative stress and ameliorating inflammation. There is no report on the protective effects of rutin on goat rumen epithelial cells (GRECs) at present. Hence, we investigated whether rutin can alleviate lipopolysaccharide (LPS)-induced damage in GRECs. Methods: GRECs were cultured in basal medium or basal medium containing 1 ㎍/mL LPS, or 1 ㎍/mL LPS and 20 ㎍/mL rutin. Six replicates were performed for each group. After 3-h culture, the GRECs were harvested to detect the relevant parameters. Results: Rutin significantly enhanced the cell activity (p<0.05) and transepithelial electrical resistance (TEER) (p<0.01) and significantly reduced the apoptosis rate (p<0.05) of LPS-induced GRECs. Rutin significantly increased superoxide dismutase, glutathione peroxidase, and catalase activity (p<0.01) and significantly decreased lactate dehydrogenase activity and reactive oxygen species and malondialdehyde (MDA) levels in LPS-induced GRECs (p<0.01). The mRNA and protein levels of interleukin 6 (IL-6), IL-1β, and C-X-C motif chemokine ligand 8 (CXCL8) and the mRNA level of tumor necrosis factor-α (TNF-α) and chemokine C-C motif ligand 5 (CCL5) were significantly increased in LPS-induced GRECs (p<0.05 or p<0.01), while rutin supplementation significantly decreased the mRNA and protein levels of IL-6, TNF-α, and CXCL8 in LPS-induced GRECs (p<0.05 or p<0.01). The mRNA level of toll-like receptor 2 (TLR2), and the mRNA and protein levels of TLR4 and nuclear factor κB (NF-κB) was significantly improved in LPS-induced GRECs (p<0.05 or p<0.01), whereas rutin supplementation could significantly reduce the mRNA and protein levels of TLR4 (p<0.05 or p<0.01). In Vitro Antioxidant Activity and Anticonvulsant Properties on Zebrafish PTZ-Induced Seizure Model of a Tilia viridis Aqueous Extract

  • Objectives: Tilia viridis (Bayer) Simonk. (Malvaceae) is widely distributed in Argentina and employed for its tranquilizing properties. Other species of the genus (Tilia europaea L., Tilia cordata Mill., Tilia platyphyllos Scop.) have been traditionally used for the treatment of epilepsy. Epilepsy affects approximately 65 million people worldwide and is characterized by an imbalance between excitatory and inhibitory processes in the brain, leading to unpredictable, unprovoked, recurrent seizures. Current pharmacological interventions often present mild to moderately severe side effects. Epilepsy has been associated with oxidative and nitrative stress as well as neuroinflammation. Herbal medicine therapies may offer new treatment options with multi-target antioxidant and anticonvulsant effects for patients whose seizures remain uncontrolled, potentially providing cost-effective solutions for individuals worldwide suffering from uncontrolled epilepsy.The aim of this study was to demonstrate the anticonvulsant activity of a standardized T. viridis aqueous extract (TE). Methods: Study of the constituents of TE, TE's antioxidant and anticonvulsant activities and toxicity, and analysis of the possible relation between the potential activities and the compounds present in the extract. In order to demonstrate TE's anticonvulsant activity a zebrafish model was used. The study also assessed TE's toxicity and antioxidant activity. To standardize the extract, total polyphenols and flavonoids were quantified and specific flavonoids were identified and quantified using HPLC-MS/MS and HPLC-UV. Results: TE exhibited anticonvulsant activity at low concentrations and demonstrated antioxidant effects by scavenging free radicals, exhibiting superoxide dismutase and peroxidase-like activities, as well as inhibiting lipoperoxidation. These actions can be attributed to the presence of polyphenols, particularly flavonoids. Conclusion: TE holds promise as a complementary herbal medicine in the treatment of epilepsy and may also offer benefits for other neuropathies associated with oxidative stress, such as Parkinson's disease and Alzheimer's disease.
