The biologically active compounds raphasatin and sulforaphene are formed during the hydrolysis of radishes by an endogenous myrosinase. Raphasatin is very unstable, and it is generated and simultaneously degraded to less active compounds during hydrolysis in aqueous media. This study determined the hydrolysis conditions to maximize the formation of raphasatin and sulforaphene by an endogenous myrosinase and minimize their degradation during the hydrolysis of radish roots. The reaction parameters, such as the reaction medium, reaction time, type of mixing, and reaction temperature were optimized. A stability test for raphasatin and sulforaphene was also performed during storage of the hydrolyzed products at $25^{\circ}C$ for 10 days. The formation and breakdown of raphasatin and sulforaphene in radish roots by endogenous enzymolysis was strongly influenced by the reaction medium, reaction time, and type of mixing. The production and stabilization of raphasatin in radishes was efficient in water and dichloromethane with shaking for 15 min at $25^{\circ}C$. For sulforaphene, the favorable condition was water as the reaction medium without shaking for 10 min at $25^{\circ}C$. The maximum yields of raphasatin and sulforaphene were achieved in a concurrent hydrolysis reaction without shaking in water for 10 min and then with shaking in dichloromethane for 15 min at $25^{\circ}C$. Under these conditions, the yields of raphasatin and sulforaphene were maximized at 12.89 and $1.93{\mu}mol/g$ of dry radish, respectively. The stabilities of raphasatin and sulforaphene in the hydrolyzed products were 56.4% and 86.5% after 10 days of storage in water and dichloromethane at $25^{\circ}C$.
We evaluated the potentials of 10 isothiocyanates (ITCs) from cruciferous vegetables and radish root hydrolysate for inhibiting the growth of oral pathogens, with an emphasis on assessing any structure-function relationship. Structural differences in ITCs impacted their antimicrobial activities against oral pathogens differently. The indolyl ITC (indol-3-carbinol) was the most potent inhibitor of the growth of oral pathogens, followed by aromatic ITCs (benzyl ITC (BITC) and phenylethyl ITC (PEITC)) and aliphatic ITCs (erucin, iberin, and sulforaphene). Sulforaphene, which is similar in structure, but has one double bond, showed higher antimicrobial activity than sulforaphane. Erucin, which has a thiol group, showed higher antimicrobial activity than sulforaphane, which has a sulfinyl group. BITC and iberin with a short chain exhibited higher antimicrobial potential than PEITC and sulforaphane with a longer chain, respectively. ITCs have strong antimicrobial activities and may be useful in the prevention and management of dental caries.
십자화과 채소의 주요 성분인 10종의 isothiocyanates(ITCs)와 무 가수분해물을 대상으로 6개의 넙치 어병세균에 대하여 항균활성을 측정하여 ITCs의 화학적 구조와 항균성과의 관계를 비교하였다. 항균활성은 sulforaphane, sulforaphene, PEITC, erucin, BITC, iberin, I3C가 높았으며, AITC, PITC, HITC는 낮았다. 어병 세균별로 ITCs에 대한 민감성은V. harveyi가 가장 높았으며, 그 다음으로E. tarda, P. damselae, S. parauberis, S. iniae, V. ichthyoenteri 순으로, 그람음성균이 그람양성균에 비하여 민감성이 높았다. 무가수분해물의 최소저해농도(MIC)는 S. iniae에 대해서 0.250 mg/mL(raphasatin의 농도)로 가장 높은 항균활성을 나타내었고, S. parauberis는 0.438 mg/mL, E. tarda와 V. harveyi는 0.500 mg/mL로 높은 항균활성을 나타내었다. ITCs의 화학구조에 따른 어병 세균에 대한 항균활성은 aliphatic ITCs 중에서는 sulforaphene, sulforaphane, erucin, iberin의 항균활성이 높았으며, benzene ring을 함유하고 있는 aromatic ITCs 중에서는 PEITC과 BITC이 가장 항균활성이 높았다. 이중결합이 없는 sulforaphane은 이중결합을 가지고 있는 sulforaphene에 비하여 항균활성이 대부분의 균주에 대해서 높았다. Thiol group을 가지고 있는 erucin은 sulfinyl group을 가지고 있는 sulforaphane에 비하여 일부균주에 대하여 높은 항균활성을 나타내었다. 탄소사슬의 길이가 긴 PEITC는 탄소사슬의 길이가 짧은 BITC에 비하여 4가지 균주에 대하여 항균활성이 높았다. 이상의 결과로 부터 십자화과 유래 ITCs는 넙치 어병 항균제로 활용할 수 있을 것으로 추정되었다.
Acne is a chronic inflammatory disease of the sebaceous gland attached to the hair follicles. Cutibacterium acnes is a major cause of inflammation caused by acne. It is well known that C. acnes secretes a lipolytic enzyme to break down lipids in sebum, and free fatty acids produced at this time accelerate the inflammatory reaction. There are several drugs used to treat acne; however, each one has various side effects. According to previous studies, sulforaphene (SFEN) has several functions associated with lipid metabolism, brain function, and antibacterial and anti-inflammatory activities. In this study, we examined the effects of SFEN on bacterial growth and inflammatory cytokine production induced by C. acnes. The results revealed that SFEN reduced the growth of C. acnes and inhibited proinflammatory cytokines in C. acnes-treated HaCaT keratinocytes through inhibiting NF-κB-related pathways. In addition, SFEN regulated the expression level of IL-1α, a representative pro-inflammatory cytokine expressed in co-cultured HaCaT keratinocytes and THP-1 monocytes induced by C. acnes. In conclusion, SFEN showed antibacterial activity against C. acnes and controlled the inflammatory response on keratinocytes and monocytes. This finding means that SFEN has potential as both a cosmetic material for acne prevention and a pharmaceutical material for acne treatment.
배무채의 형태적 특성은 양친인 배추와 무의 중간형태이다. 잎은 윗부분이 배추이고 아래 부분이 무를 닮았다. 잎의 중륵은 무처럼 둥글지만 직경이 3cm 이상으로 크고 배추처럼 흰 색이다. 뿌리는 처음에 중간부위가 부풀어져 무 모양이었는데 유전적 안정화 과정을 거치면서 재래종 배추(뿌리배추)처럼 큰 직근으로 바뀌었다. 꽃은 흰 색이며 종자 꼬투리는 선명하게 두 부분으로 나누어진다. 윗부분이 무로서 길이가 약 4cm 정도이며 그 속에 3-4개의 종자가 들고 아래 부분이 배추로서 길이가 약 3cm 정도이며 그 속에 7-8개의 종자가 들어있다. 종자는 배추와 비슷한 적갈색이며 천립중이 5.5g이고 mL당 약 120립 정도이다. 가을에 배추와 같이 재배하면 약 5kg 정도까지 자라며 외엽이 아주 무성하고 억세게 보인다. 속잎은 노란색을 띠며 엉성하지만 약 900g 정도의 구를 형성한다. 잎과 뿌리 모두 항암과 항균작용이 큰 기능성 물질 썰포라펜(sulforaphene)을 다량으로 함유하고 있다. 배무채는 복2배체 식물로서 자가불화합성이 없고 따라서 자가수정이 잘되지만 벌이 많이 오는 타가수분식물이다. 배무채는 그의 모계였던 배추와의 교잡에서 자방친일 때는 교잡이 아주 잘 되지만 부계일 때는 완전한 불화합이며 부계였던 무 및 양배추와 흑겨자 사이에는 상반교잡 모두 불화합성이다. 그러나 복2배체 식물인 유채 및 황겨자와의 사이에는 상반교잡 모두 화합성이며 갓과의 사이에는 상반교잡 모두 부분화합성이다.
