두릅 에탄올 추출물의 Chlorogenic acid 함량 분석 및 생리활성 (Analysis of Chlorogenic Acid Content and Biological Activities of Aralia elata Ethanol Extract)

  • 순창군에서 생산된 두릅을 40% EtOH을 이용하여 추출한 후 chlorogenic acid 함량, 항산화, 항균, 항염 및 소화효소 활성을 측정하였다. HPLC를 이용하여 chlorogenic acid 함량을 측정한 결과 7.06±0.01 mg/g 함유되어 있었다. DPPH 라디칼 소거활성(SC50)은 4.79±0.05 mg/mL, ABTS 라디칼 소거활성(SC50)은 5.79±0.05 mg/mL, 총 폴리페놀 함량은 170.0±1.8 mgGAE/g, 총 플라보노이드 함량은 105.5±4.1 mgQE/g으로 분석되었다. RAW 264.7 세포, Caco-2 세포에서 세포독성이 나타나지 않았으며, 농도 의존적으로 NO 생성이 억제되었다. RAW 264.7 세포에서 염증성 cytokine인 TNF-α생성은 8.9±0.1 ng/mL, IL-6 생성은 15.2±0.8 ng/mL, IL-1β생성은 30.9±0.9 pg/mL으로 억제되었으며, AEE의 처리 농도가 증가함에 따라 TNF-α, IL-1β, IL-6 생성이 농도 의존적으로 억제되었다. S. typhimurium, L. monocytogenes, H. pylori에 대한 항균활성이 우수하게 나타났으며, 두릅을 EtOH 추출물의 처리농도가 증가함에 따라 효소 활성인 α-amylase와 protease 효소활성도 증가하였다. 순창군에서 생산된 두릅은 chlorogenic acid를 다량 함유하고 있으며, 항산화, 항염, 항균, 소화효소 활성이 우수하게 나타나 향후 건강기능성 소재로 개발할 수 있을 것으로 판단된다.

  • This paper presents the active maximum power point tracking(MPPT) control of the photovoltaic(PV) module integrated converter(MIC) system considering the shadow influence. Conventional perturbation and observation(PO) and incremental conductance(IC) are the method finding MPP by the continued self-excitation vibration. The MPPT control is unable to be performed by rapid output change affected by the shadow. To solve this problem, the active MPPT in which the step value changes by output change is presented. In case there are the solar radiation, a temperature and shadow influence, the presented algorithm treats and compares the conventional control algorithm and output error. In addition, the validity of the algorithm is proved through the output error response characteristics.

Sensitivity of Pseudomonas syringae to Bovine Lactoferrin Hydrolysates and Identification of a Novel Inhibitory Peptide

  • The antimicrobial activity of bovine lactoferrin hydrolysates (bLFH) was measured against Pseudomonas strains (P. syringae and P. fluorescens) in vitro. To compare susceptibility to bLFH, minimal inhibitory concentration (MIC) values were determined using chemiluminescence assays and paper disc plate assays. Antimicrobial effect against P. fluorescens was not observed by either assay, suggesting that bLFH did not exhibit antimicrobial activity against P. fluorescens. However, a significant inhibition of P. syringae growth was observed in the presence of bLFH. The addition of bLFH in liquid or solid medium inhibited growth of P. syringae in a dose-dependent manner. Furthermore, a bLFH peptide with antimicrobial activity toward P. syringae was isolated and identified. The N-terminal amino acid sequences of thus obtained antimicrobial bLFH peptides were analyzed by a protein sequencer and were found to be Leu-Arg-Ile-Pro-Ser-Lys-Val-Asp-Ser-Ala and Phe-Lys-Cys-Arg-Arg-Trp-Gln-Trp-Arg-Met. The latter peptide sequence is known to be characteristic of lactoferricin. Therefore, in the present study, we identified a new antimicrobial peptide against P. syringae, present within the N-terminus and possessing the amino acid sequence of Leu-Arg-Ile-Pro-Ser-Lys-Val-Asp-Ser-Ala.

  • The objection of this research is to conform of practicable possibility and recycling of producing junk after citrus fruits is processed. In the middle of practicable possibility, with verification of antibiosis that tactiling sensibility in a microorganism. With extracting d-limonene oil that have 70${\sim}$90% a component of oil out of junk citrus peel, making certain the about 12000ppm(1.2%) concentration of it. By means of antibiosis property over the a pathogenic bacterium as well as a residence bacterium, considerating the limit of application against daily living supplies needs to antibiosis. Antibiosis effect of a stationing bacterium in the body permanently and by means of antibiosis verification of special bacteria Propionibacterium Acnes that causative skin trouble is selected, in based the antibiotic sencitivity test check up result of minimal inhibitory concentration(MIC).

Antimicrobial Activity of an Edible Wild Plant, Apiifolia Virgin's Bower (Clematis apiifolia DC)

  • An edible wild perennial plant with extremely potent antimicrobial activity was found and identified as apiifolia Virgin's Bower (Clematis apiifolia DC) which is easily found around wet wildernesses. Fresh fruit extract of C. apiifolia exhibited minimum inhibitory concentrations (MIC) in the vicinity of 0.1% against various yeasts and of less than or equal to 0.4% for non-lactic acid bacteria. MICs against lactic acid bacteria were about 2.0%. The antimicrobial activity of C. apiifolia fruit was even more potent than that of garlic which has been known for its potent antimicrobial activity. The principal antimicrobial compound of fruit extract of C. apiifolia was isolated and identified by high performance liquid chromatography and gas chromatography as protoanemonin (a gamma lactone of 4-hydroxy-2,4-pentadienoic acid). The antimicrobial activity of C. apiifolia was stable at high temperatures, and the activity was maintained after heating at $121^{\circ}C$ for 10 min. The antimicrobial compound of C. apiifolia was supposed to inhibit microorganisms by reacting with sulfhydryl groups of cellular proteins.

Thymol Rich Thymbra capitata Essential Oil Inhibits Quorum Sensing, Virulence and Biofilm Formation of Beta Lactamase Producing Pseudomonas aeruginosa

  • Infections with Pseudomonas aeruginosa are difficult to treat not only because it is often associated with multidrug-resistant infections but also it is able to form biofilm. The aim of this study was to evaluate the antibiofilm and anti-Quorum Sensing (QS) activities of Thymbra capitata essential oils (EOs) against Beta Lactamase (BL) producing P. aeruginosa and the reference strain P. aeruginosa 10145. GC/MS analysis showed that thymol (23.25%) is the most dominant compound in T. capitata EOs. The MICs of T. capitata EOs against P. aeruginosa (BL) and P. aeruginosa 10145 were 1.11%. At sub MIC (0.041, 0.014 and 0.0046%), the EOs of T. capitata remarkably inhibited the biofilm formation of both strains tested and complete inhibition of the biofilm formation was reported at 0.041%. The EOs of T. capitata were found to inhibit the swarming motility, aggregation ability and hydrophobic ability of P. aeruginosa (BL) and P. aeruginosa 10145. Interestingly, the EOs of T. capitata reduce the production of three secreted virulence factors that regulated by QS system including pyocyanin, rhamnolipids and LasA protease. The potent antibiofilm and anti-QS activities of T. capitata EOs can propose it as a new antibacterial agent to control pseudomonas infections.

Hydrogel Dressing with a Nano-Formula against Methicillin-Resistant Staphylococcus aureus and Pseudomonas aeruginosa Diabetic Foot Bacteria

  • This study proposes an alternative approach for the use of chitosan silver-based dressing for the control of foot infection with multidrug-resistant bacteria. Sixty-five bacterial isolates were isolated from 40 diabetic patients. Staphylococcus aureus (37%) and Pseudomonas aeruginosa (18.5%) were the predominant isolates in the ulcer samples. Ten antibiotics were in vitro tested against diabetic foot clinical bacterial isolates. The most resistant S. aureus and P. aeruginosa isolates were then selected for further study. Three chitosan sources were tested individually for chelating silver nanoparticles. Squilla chitosan silver nanoparticles (Sq. Cs-Ag0) showed the maximum activity against the resistant bacteria when mixed with amikacin that showed the maximum synergetic index. This, in turn, resulted in the reduction of the amikacin MIC value by 95%. For evaluation of the effectiveness of the prepared dressing using Artemia salina as the toxicity biomarker, the LC50 was found to be 549.5, 18,000, and 10,000 μg/ml for amikacin, Sq. Cs-Ag0, and dressing matrix, respectively. Loading the formula onto chitosan hydrogel dressing showed promising antibacterial activities, with responsive healing properties for the wounds in normal rats of those diabetic rats (polymicrobial infection). It is quite interesting to note that no emergence of any side effect on either kidney or liver biomedical functions was noticed.

Pharmacokinetic profiles of levofloxacin after intravenous, intramuscular and subcutaneous administration to rabbits (Oryctolagus cuniculus)

  • Levofloxacin pharmacokinetic profiles were evaluated in 6 healthy female rabbits after intravenous (I/V), intramuscular (I/M), or subcutaneous (S/C) administration routes at a single dose of 5 mg/kg in a 3 × 3 cross-over study. Plasma levofloxacin concentrations were detected using a validated Ultra Performance Liquid Chromatography method with a fluorescence detector. Levofloxacin was quantifiable up to 10 h post-drug administration. Mean AUC0-last values of 9.03 ± 2.66, 9.07 ± 1.80, and 9.28 ± 1.56 mg/h*L were obtained via I/V, I/M, and S/C, respectively. Plasma clearance was 0.6 mL/g*h after I/V administration. Peak plasma concentrations using the I/M and S/C routes were 3.33 ± 0.39 and 2.91 ± 0.56 ㎍/mL. Bioavailability values, after extravascular administration were complete, - 105% ± 27% (I/M) and 118% ± 40% (S/C). Average extraction ratio of levofloxacin after I/V administration was 7%. Additionally, levofloxacin administration effects on tear production and osmolarity were evaluated. Tear osmolarity decreased within 48 h post-drug administration. All 3 levofloxacin administration routes produced similar pharmacokinetic profiles. The studied dose is unlikely to be effective in rabbits; however, it was calculated that a daily dose of 29 mg/kg appears effective for I/V administration for pathogens with MIC < 0.5 ㎍/mL.

초석잠 추출물의 항산화, 항균 및 항염 활성 (Antioxidant, Antibacterial and Anti-inflammatory Effects of Stachys sieboldii Extract)

  • 순창군에서 재배한 초석잠을 건강식품을 개발하기 위한 기초자료를 확보하고자 초석잠 물 추출물과 60% 에탄올 추출물에 대한 항산화, 항균, 항염, 소화효소 활성을 측정하였다. DPPH 라디칼 소거능(IC50)은 SAW는 5.26 ± 0.05 mg/mL, SAE는 4.34 ± 0.04 mg/mL로 나타났으며, ABTS 라디칼 소거능(IC50)은 SAW는 6.44 ± 0.06 mg/mL, SAE는 5.05 ± 0.06 mg/mL로 나타났다. 총 폴리페놀 함량은 SAW는 106.25 ± 0.94 mgGAE/g, SAE는 124.61 ± 1.11 mgGAE/g, 총플라보노이드 함량은 SAW는 24.4 ± 0.24 mgQE/g, SAE는 45.2 ± 3.52 mgQE/g으로 분석되었다. CAA assay를 활용한 HepG2 세포내 항산화 활성은 400 ㎍/mL의 농도에서 SAW는 53.2 ± 1.8%, SAE는 54.1 ± 0.4%로 감소되었다. SAW의 MIC는 L. monocytogenes은 100 mg/mL, S. typhimurium와 H. pylori은 125 mg/mL로 측정되었으며, MBC는 L. monocytogenes와 S. typhimurium은 325 mg/mL, H. pylori은 400 mg/mL로 측정되었다. RAW 264.7 세포에서 각 추출물 모두100 ㎍/mL 이하의 농도에서 독성이 나타나지 않았으며, 추출물 100 ㎍/mL 농도에서 SAW는 44.3 ± 1.4%, SAE는 45.1 ± 1.0%로 NO 생성을 저하시켰다. 염증성 사이토카인인 TNF-𝛼, IL-1𝛽 및 IL-6 생성을 농도 의존적으로 억제시켰다. Caco-2 세포에서 SAW와 SAE 추출물 모두 독성이 나타나지 않았으며, 농도 의존적으로 NO 생성을 억제시켰다. 𝛼-Amylase와 protease 효소활성은 초석잠 추출물의 처리 농도가 증가함에 따라 효소의 활성도 증가하였다.

A Tuber Lectin from Arisaema jacquemontii Blume with Anti-insect and Anti-proliferative Properties

  • A tuber lectin from Arisaema jacquemontii Blume belonging to family Araceae was purified by employing a single step affinity chromatography using column of asialofetuin-linked amino activated silica beads and the bound lectin was eluted with 100 mM glycine-HCl buffer pH 2.5. The purified A. jacquemontii lectin (AJL) showed a single protein band with an apparent molecular mass of 13.4 kDa when submitted to SDS-polyacrylamide gel electrophoresis under reducing as well as non-reducing conditions. The native molecular mass of AJL determined by gel filtration on a Biogel P-200 column was 52 kDa and its carbohydrate content was estimated to be 3.40%. Thus AJL is a tetrameric glycoprotein. The purified lectin agglutinated erythrocytes from rabbit but not from human. Its activity was not inhibited by any of the mono- and disaccharides tested except N-acetyl-D-lactosamine having minimal inhibitory sugar concentration (MIC) 25 mM. Among the glycoproteins tested only asialofetuin was found to be inhibitory (MIC $125\;{\mu}g/mL$). A single band was obtained in native PAGE at pH 4.5 while PAGE at pH 8.3 showed two bands. Isoelectric focusing of AJL gave multiple bands in the pI range of 4.6-5.5. When incorporated in artificial diet AJL significantly affected the development of Bactrocera cucurbitae (Coquillett) larvae indicating the possibility of using this lectin in a biotechnological strategy for insect management of cucurbits. Larvae fed on artificial diet containing sub-lethal dose of AJL showed a significant decrease in acid phosphatase and alkaline phosphatase activity while esterase activity markedly increased as compared to larvae fed on diet without lectin. Out of various human cancer cell lines employed in sulphorhodamine B (SRB) assay, this lectin was found to have appreciable inhibitory effect on the in vitro proliferation of HCT-15, HOP-62, SW-620, HT-29, IMR-32, SKOV-3, Colo-205, PC-3, HEP-2 and A-549 cancer cell lines by 82, 77, 73, 70, 41, 41, 37, 29, 21 and 21% respectively.
