본 연구에서는 렌틸을 이용한 새로운 기능성 소재의 개발에 우선하여 세계적으로 소비량이 비교적 많은 벨루가, 레드, 그린, 프렌치 렌틸을 연구 소재로 선정하고, 0.2% HCl을 함유한 메탄올을 이용하여 추출물을 제조한 후 이들의 항산화 활성을 조사하였다. 총폴리페놀과 플라보노이드 함량은 각각 27.3~30.3 mg TAE/g과 13.14~16.29 mg QUE/g으로 다양하게 나타났으며, 그중 벨루가와 레드 렌틸이 상대적으로 높은 총폴리페놀 함량을 보였다. DPPH와 ABTS 라디칼 소거 활성 및 $H_2O_2$ 소거 활성은 총폴리페놀 함량이 상대적으로 높았던 벨루가와 레드 렌틸이 다른 품종에 비해 유의적으로 높은 활성을 보였으며, 항산화 활성 및 폴리페놀함량과의 비례적 상관관계가 있음을 확인할 수 있었다. ROS 소거 활성이 가장 높았던 벨루가 렌틸을 이용하여 linoleic acid에 대한 과산화 억제 효과를 FTC법으로 조사한 결과, $62.5{\sim}500{\mu}g/mL$에서 농도 의존적으로 유의적인 산화억제효과를 보였으며 $RC_{50}$은 $222.76{\mu}g/mL$로 확인되었다. 또한, 벨루가 및 레드 렌틸 추출물은 산화적 스트레스 발생을 통해 간세포 손상을 유발하는 것으로 알려진 알코올의 세포독성으로부터 우수한 간세포 보호 효과를 나타내었다. 따라서 향후 추가적인 세포실험과 동물실험을 통한 렌틸의 항산화 활성 검정 및 항산화 기전에 관한 연구가 필요할 것으로 생각되며, 이를 비롯한 다양한 생리활성에 대한 연구들이 이루어진다면 기능성 소재로서 렌틸의 산업적 응용이 활발해질 것으로 기대된다.
곤충은 넓은 범위의 온도영역에 사는 것으로 알려져 있으나, $40^{\circ}C$가 넘는 고온이나 빙결온도 이하의 저온에서는 생존이 어렵다. 본 연구는 사육온도 조건이 다른 환경에서 대사중심 조직인 지방체의 유전자 발현을 분석하기 위해, 온도조건을 달리하여 담배나방을 저온 사육충 ($3{\sim}10^{\circ}C$), 고온 사육충 ($35^{\circ}C$)로 나누고 상온 사육충 ($25^{\circ}C$)을 대조구로 사용하여 전사체 분석을 수행하였다. 저온에서 특이적으로 높은 발현을 보인 유전자는 표피단백질, ${\Delta}9$ 불포화효소, 글리세롤 3-인산 탈수소효소이며, 저온에서 발현이 낮아진 유전자는 키틴 합성효소, catalase, UDP-당전이 효소이다. 고온에서 특이적으로 높은 발현을 보인 유전자는 과산화물제거효소, metallothionein 2, phosphenolpyruvate carboxykinase, 트레할로스 운반단백질이었다. 고온에서 높고 저온에서 낮은 대조적 발현을 보인 유전자는 열충격단백질, glutathione peroxidase이었다. 이들 온도 특이적이거나 대조적 발현을 보이는 유전자는 기후변화에 관련한 특이마커로 활용이 가능할 것으로 사료된다.

We examined the ways in which fenobam could promote not only the transduction of PEP-1-FK506BP into cells and tissues but also the neuroprotective effect of PEP-1-FK506BP against ischemic damage. Fenobam strongly enhanced the protective effect of PEP-1-FK506BP against $H_2O_2$-induced toxicity and DNA fragmentation in C6 cells. In addition, combinational treatment of fenobam with PEP-1-FK506BP significantly inhibited the activation of Akt and MAPK induced by $H_2O_2$, compared to treatment with PEP-1-FK506BP alone. Interestingly, our results showed that fenobam significantly increased the transduction of PEP-1-FK506BP into both C6 cells and the hippocampus of gerbil brains. Subsequently, a transient ischemic gerbil model study demonstrated that fenobam pretreatment led to the increased neuroprotection of PEP-1-FK506BP in the CA1 region of the hippocampus. Therefore, these results suggest that fenobam can be a useful agent to enhance the transduction of therapeutic PEP-1-fusion proteins into cells and tissues, thereby promoting their neuroprotective effects.
현삼의 기내배양에서 TDZ처리가 비교적 재분화에 효율적이었고 형질전환 식물체를 선발하기 위하여 선발표지 유전자로 사용되는 NPTII gene이 항생제 kanamycin에 대한 저항성은 50 mg/L가 적당하였다. 선발배지에서 자란 현삼 식물체에서 DNA를 추출하여 PCR 분석을 통하여 특정 유전자 Gh-5 gene을 검정한 결과 형질전환되지 않은 식물체에서는 볼수가 없는 988 bp의 band가 형질전환된 식물체에서는 관찰되어 GST 유전자가 현삼의 염색체 안으로 삽입되었음을 확인하였다. 형질전환 효율 증진을 위하여 선발과정에서 암상태를 30일까지 유지할 경우 높은 형질전환 효율을 나타내었으나 그 이상의 처리는 오히려 형질전환 효율이 감소하는 결과를 나타내었다 (Table 5). 형질전환 식물체에서 GST의 활성이 형질전환 되지 않은 식물체의 2배로 나타났고 유도체의 적정 처리 농도는 50$\mu$M이며 유도체 처리 시간에 따라서는 12시간까지는 점차적으로 높아지는 경향이었으나 그 이상의 시간에서는 활성이 저하됨이 확인되었다. Fungus 피검균인 Asperigillus awamori에서 6, 12시간 처리 시 비교적 높은 활성을 보여주었으며 그 이상의 시간처리에서는 명확한 균사 억제를 나타내지 못하였으며, 특히 12시간에서 상대적으로 높은 활성을 나타내었다. Cladosporium herbarum에서 형질전환된 식물체의 활성이 훨씬 높게 나타났으며 6시간 처리에서 다른 처리에 비하여 높은 활성을 나타내었다. 피검균인 Saccharomyces cerevisiae에서는 형질전환 식물체에서 높은 활성을 보여주었으며 6, 12시간 처리에서 비슷하게 높은 활성을 보여 주었다. 박테리아 피검균 Bacillus subtillis에서는 50$\mu$M 이하의 유도체 처리에서 비교적 높은 활성을 나타내었다.
p53 유전자는 스트레스, DNA 손상, 저산소증 및 종양 발생에 대한 세포 반응의 전사 조절에서 중요한 역할을 담당한다. 최근에 발견된 다양한 종류의 p53의 생리 활성을 생각한다면 p53이 암 조절에 관여한다는 것은 놀랄만한 일이 아니다. 인간 암의 약 50%에는 p53 유전자의 돌연변이 또는 p53을 활성화시키는 기전의 결함을 통해 p53 단백질 기능의 불활성화가 나타난다. p53 기능의 이러한 장애는 p53 의존 반응으로부터 회피를 허용함으로써 종양의 진화에 결정적인 역할을 하게 된다. 최근의 많은 연구들은 p53의 돌연변이를 대폭 감소시키거나 p53의 종양 억제 기능을 복원하기 위하여 선택적인 저분자 화합물을 동정함으로써 p53의 돌연변이를 직접 표적하는 것에 초점을 두고 있다. 이들 저분자는 좋은 약물과 유사한 특성을 유지하면서 다양한 상호작용을 효과적으로 조절해야 한다. 이 중, p53의 음성조절인자 핵심인 MDM2의 발견은 p53과 MDM2 간의 상호작용을 차단하는 새로운 저분자 억제제의 설계를 제공하였다. 저분자 화합물 중 일부는 개념 증명 연구에서 임상 시험으로 옮겨졌으며 향후 맞춤형 항암제가 추가될 전망이다. 본 리뷰에서는 야생형 p53과 돌연변이 p53의 구조적 및 기능적 중요성과 p53을 직접 표적하는 치료제 개발, p53과 MDM2 간의 상호작용을 억제하는 화합물에 대하여 검토하였다.

Prostaglandin $E_2$ ($PGE_2$) promotes tumor-persistent inflammation, frequently resulting in cancer. Curcumin is a diphenolic turmeric that inhibits carcinogenesis and induces apoptosis. $PGE_2$ inhibits curcumin-induced apoptosis; however, the underlying inhibitory mechanisms in colon cancer cells remain unknown. The aim of the present study is to investigate the survival role of $PGE_2$ and whether addition of exogenous $PGE_2$ affects curcumininduced cell death. HCT-15 cells were treated with curcumin and $PGE_2$, and protein expression levels were investigated via Western blot. Reactive oxygen species (ROS) generation, lipid peroxidation, and intracellular glutathione (GSH) levels were confirmed using specific dyes. The nuclear factor-kappa B ($NF-{\kappa}B$) DNA-binding was measured by electrophoretic mobility shift assay (EMSA). $PGE_2$ inhibited curcumin-induced apoptosis by suppressing oxidative stress and degradation of PARP and lamin B. However, exposure of cells to the EP2 receptor antagonist, AH6809, and the PKA inhibitor, H89, before treatment with $PGE_2$ or curcumin abolished the protective effect of $PGE_2$ and enhanced curcumin-induced cell death. $PGE_2$ activates PKA, which is required for cAMP-mediated transcriptional activation of CREB. $PGE_2$ also activated the Ras/Raf/Erk pathway, and pretreatment with PD98059 abolished the protective effect of $PGE_2$. Furthermore, curcumin treatment greatly reduced phosphorylation of CREB, followed by a concomitant reduction of $NF-{\kappa}B$ (p50 and p65) subunit activation. $PGE_2$ markedly activated nuclear translocation of $NF-{\kappa}B$. EMSA confirmed the DNA-binding activities of $NF-{\kappa}B$ subunits. These results suggest that inhibition of curcumin-induced apoptosis by $PGE_2$ through activation of PKA, Ras, and $NF-{\kappa}B$ signaling pathways may provide a molecular basis for the reversal of curcumin-induced colon carcinoma cell death.
Fluidized bed opposed jet mill을 사용하여 초미세분쇄한 후 Turboplex classifier로 공기분급한 탈지미강분말의 입도는 air classifying wheel speed (ACWS)가 낮아짐에 따라 입자의 크기와 표준편차는 증가하고 단위부피당 표면적은 감소하였다. ACWS 21,000rpm에서 분급한 미강분말의 형태는 구형이었으며 입도 median이 $3.7{\mu}m$이었다. ACWS별로 회수된 시료의 단백질, 지방, 회분은 ACWS에 따라 전반적으로 증가하는 경향이었다. Mg, Zn, Fe 및 Mn 함량은 ACWS가 증가할수록 비교적 증가하는 경향을 보였으며, phytic acid함량은 중간 ACWS에서 약간 높은 값을 보였다. 식이섬유함량은 $25.2{\sim}31.5%$의 값을 보였고, ACWS 15,000rpm에서 최대값을 나타내었다. ACWS에 따른 시료의 색도는 ACWS가 높을수록 백색도(L)가 증가하였고, 적색도(a)와 황색도(B)는 감소하는 경향을 나타내었다. 보수력(WHC)은 12,000rpm에서 2.88g/g solid로 가장 높았고 ACWS가 높아짐에 따라 감소하는 추세를 보였으나, 보유력은 반대로 ACWS에 따라 약간 증가하는 경향을 보였다. 분급된 미강분말 현탁액의 유동특성은 Bingham body에 속하였으며 농도와 ACWS가 증가함에 따라 항복응력 및 점도가 상승하였다. 분급된 미강분말을 5% 수준으로 대체하여 케이크를 제조한 경우 대조구보다 케이크의 높이와 부피가 증가하였다.
Ethylene, known as a stress hormone regulate wide developmental processes including germination, root hair initiation, root and shoot primordial formation and elongation, leaf and flower senescence and abscission, fruit ripening. The acceleration of ethylene biosynthesis in plant associated with environmental and biological stresses. 1-Aminocycloprophane-1-carboxlyate deaminase(ACCD) is an enzyme that cleaves ACC into and ammonia, a precursor of the plant hormone ethylene. Plant growth-promoting rhizobacteria (PGPR) having ACCD can decrease endogenous ACC level of tissue, resulting in reduced production of ethylene in plants. ACC deaminse was a key enzyme for protect stressed plants from injurious effects of ethylene. ACCD gene was encoded from Pseudomonas flourescens, PGPR and was cloned in Escherichia coli. We expressed the recombinant ACCD(rACCD) containing 357 amino acids with molecular weight 39 kDa that revealed by SDS-PAGE and western blot. The rACCD was purified by Ni-NTA purification system. The active form of rACCD having enzyme activity converted ACC to a-ketobutyrate. The optimal pH for ACC deaminase activity was pH 8.5, but no activity below pH 7.0 and a less severe tapering activity at base condition resulting in loss of activity at over pH 11. The optimal temperature of the enzyme was $30^{\circ}$ and a slightly less severe tapering activity at 15 - 30$^{\circ}$, but no activity over $35^{\circ}$. P. flourescens ACC deaminase has a highly conserved residue that plays in allowing substrate accessibility to the active sites. The enzymatic properties of this rACCD will provide an important reference for analysis of newly isolated ACCD and identification of newly isolated PGPR containing ACCD.
We previously reported that the novel isoflavone-free peptide mixture (black soybean peptide, BSP) had several beneficial effects like antiobesity and hypotriglyceridemic effect. However, there are no reports for BSP on anti-hypertensive activity. BSP induced heme oxygenase-1 (HO-1) in HUVECs, thus investigated the HO-1-induced activity in HUVECs and the anti-hypertensive effects in SHR animal model. BSP significantly induced HO-1 expression both at transcriptional and protein levels in a time- and dose-dependent manner as measured by RT-PCR and Western blot analysis, respectively. These inductions were abolished by pretreatment of N-acetyl-cystein (NAC, 1~10 mM), but not by employing Tempol, a superoxide dismutase (SOD) mimetic (1~5 mM). As expected, enzymatic activity (~2 fold) determined by bilirubin formation assay and cGMP concentration (~6 fold) were significantly increased in BSP-treated cells. Based on the numerous evidences on the beneficial effects of HO-1 and our results, we investigated in vivo effects of BSP on the antihypertensive activity. The administration of BSP (1% in drinking water) significantly decreased mean blood pressure (BP) (from $218.6{\pm}6.99$ to $190.0{\pm}3.42$ mm Hg, p<0.01). This result indicates that BSP is strong inducer of HO-1 expression, which may be triggered by oxidative stress, and has anti-hypertensive activity.
본 연구는 점등 조절 방법이 육계의 생산성과 육질에 미치는 영향을 구명하기 위해 수행되었다. 공시계는 1일령 초생추 (Ross) 912수를 암수 구별하여 3처리 4반복 반복당 암컷 42수, 수컷 34수씩 공시하였다. 암수 모두 체중, 증체량, 사료섭취량 및 사료요구율은 유의차가 없었다 (P>0.05). 가슴육의 가열감량과 보수력은 처리구간에 유의적인 차이가 없었다. 전단력은 점등 처리구가 자연일조에 비해 높게 나타났다 (P<0.05). 계육의 일반성분 (수분, 조단백질, 조지방, 조회분)은 암수 모두 처리구간 차이가 없었다. 결론적으로, 점등시간을 조절해 주는 것이 육계에 스트레스를 줄이면서 생산성을 높여줄 수 있다고 사료된다.
