Blood Glucose Level, Insulin Content and Biochemical Variables of Complexcity Extract from Oriental Medicinal Plants on Diabetes Rats (한약자원 복합추출물이 당뇨쥐의 혈당, 인슐린 농도 및 생화학적 성상에 미치는 영향)

  • This study was conducted to examine the hypoglycemic effects of complexcity extract(DB 55) from five oriental medicinal plants on the intake of food, body weight, blood glucose level, insulin content, serum GOP, GPT, BUN and hematocrit value in streptozotocin(STZ)-induced diabetic rats. General nutritional composition was also assessed. Thirty- five male Sprague-Dawley rats were assigned to five groups : normal control group(NC), STZ-diabetic control group (STZ-control), STZ-diabetic DB-1 group(DB 55-1), STZ-diabetic DB-2 group(DB 55-2) and STZ-diabetic DB-3 group(DB 55-3). The animals were fed an experimental diet for 12 weeks. Body weight gain of the STZ-diabetic group was significantly lower but diet intake was significantly higher than the NC group. Blood glucose levels of the DB-1, DB-2 and DB-3 rats were significantly lower than the STZ-control animals. Insulin level of STZ-control rats was lower than the NC serum GOT, GPT and BUN levels were significantly higher in the NC group, but hematocrit value was not significant as compare to the STZ-diabetic group. Blood glucose level was lower observed for the DB 55-1, DB 55-2 and DB 55-3 group. The results will be useful in oriental diet therapy and in the developing functional food resources.

Immunohistochemical Study of Yanggyuksanhwa-tang on Focal Cerebral Ischemia of Diabetic Rats (당뇨흰쥐의 국소뇌허혈에 대한 양격산화탕(凉膈散火湯)의 면역조직화학적 연구)

  • This study evaluated neuroprotective effects of Yanggyuksanhwa-tang (YST), which have been known to be efficacy in the treatment of the stroke and diabetes. on focal cerebral ischemia of diabetic rats. On primary experiment, diabetic condition in rats was induced by streptozotocin injection, then, focal cerebral ischemia was induced by the middle cerebral artery occlusion (MCAO) under the diabetic condition. Then neuroprotective effect of YST was observed with changes of infarct size and volume, expressions of c-Fos, Bax, and hypoxia inducible factor (HIF)-1${\alpha}$ in the brain tissues by using 2% 2,3,5-triphenyltetrazolium chloride (TTC) staining and immunohistochemistry. YST treatment showed a significant decrease of infarct size and volume induced by MCAO in diabetic rats. YST treatment showed a significant decrease of c-Fos and Bax positive neurons in cortex penumbra. YST treatment showed a decrease of HIF-l${\alpha}$ positive neurons in cortex penumbra, but it was not significant statistically. These results suggest that YST has effects on neuroprotection against cerebral infarct under diabetic condition. And it is supposed that neuroprotective effect of YST reveals by anti-apoptosis mechanism.

Properties and Activitiy Screening of Chromone Derivatives (크로몬 유도체의 물성 및 활성검색)

  • We have synthesized 4-isonitroso-4H-1-benzopyran and 4-amino-2,3-dihydro -4H-1-benzopyran of chromone derivatives by using condensation method. Physico-chemical properties of these compounds were measured and analyzed by UV and HPLC method. The correlation coefficient of their methanol solutions by UV were 0.9992 and 0.9994, respectively. And oxime compound was resolved within 4 min and had a detection limit of 3 ng at S/N=3 by HPLC using a reversed phase column with three solvents(MeOH, $H_2O$, HAc). The amino compound was resolved within 4.5 min and had a detection limit of 10 ng at S/N=3 by HPLC under the same conditions. Anti-diabetic effect of chromone derivatives were investigated in the streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in male Sprague-Dawley rats by injections of STZ (45 mg/kg, i.v). The investigation of the hair growth effect of isonitrosobenzopyran and 4-aminobenzopyran on the hair of black mouse (C57BL/6) was also carried out. The administraion of their ethanol solution to the black mouse (C57BU/6) through skin them promoted the growth of hair.

The Effect of Some Antipolydipsia Oriental Prescriptions on Experimental Diabetic Rats -Cheongsimyeonja-eum, Kamijowiseunki-tang, Kikukjihwang-tang- (소갈증 치료제가 실험적 당뇨 흰쥐에 미치는 영향 -청심연자음, 가미조위승기탕, 기국지황탕-)

  • In order to study the effect of some oriental prescriptions for polydipsia on experimental diabetes mellitus and cataract, Cheongsimyeonja-eum, Kamijowiseungki-tang and Kikukjihwang-tang were administered to streptozotocin(STZ)-induced diabetic and galactose-induced cataractous rats. The increased level of serum glucose and urea nitrogen owing to 572 were significantly decreased by administration of Cheongsimyeonja-eum, Kamijowiseungki-tang and Kikukjihwang-tang. The increased level of serum triglyceride and total cholesterol owing to STZ were significantly decreased by administration of Kamijowiseungki-tang and Kikukjihwang-tang. Reduction of body weight owing to STZ was significantly recovered in 7th day since administration of Cheongsimyeonja-eum and Kamijowiseungki-tang. Weight of liver was significantly recovered by administration of Cheongsimyeonja-eum and Kamijowiseungki-tang and weight of kidney was significantly recovered only by administration of Kikukjihwang-tang. The polydipsia of water and food were significantly decreased by administration of Cheongsimyeonja-eum, Kamijowiseungki-tang and Kikukjihwang-tang. Onset of cataract in experimental group owing to galactose was not significantly different from control group.

Effect of Supplementation of Dietary Sea Tangle on the Renal Oxidative Stress in Diabetic Rats (식이 다시마의 섭취가 당뇨 쥐 신장의 산화적 스트레스 및 당뇨성 병변에 미치는 영향)

  • Diabetic nephropathy has been increasing, although blood glucose and blood pressure can be controlled by angiotensin converting enzyme(ACE) or advanced glycosylation end products(AGE) inhibitors in the diabetic patients. We investigated the effect of dietary supplementation of sea tangle on the blood glucose, and pathological scoring of diabetic kidneys in the streptozotocin(STZ) induced diabetic rats. Male Sprague-Dawley rats were divided into normal rats fed control diet and diabetic rats fed control diet or control diet supplemented with powder or oater extract of sea tangle. Diabetes was induced by a single injection of STZ(60mg/kg, ip) in citrate buffer. The animals were fed the experimental diet and water for 13 weeks. Dietary supplementation of sea tangle decreased blood glucose in the diabetic rats. However, dietary supplementation of sea tangle did not affect the antioxidant enzyme activities, MDA content and pathology of diabetic kidneys. These results indicate that decreased blood glucose by sea tangle could not delay the progression of diabetic kidney disease.

Effects of the C3G/D3G anthocyanins-rich black soybean testa extracts on improvement of lipid profiles in STZ-induced diabetic rats (Streptozotocin 유도 당뇨쥐에서 C3G 및 D3G 안토시안이 풍부한 검정콩 CJ-3호 종피 추출물의 혈중지질 개선효과)

  • Purpose: The purpose of this study was to examine the effect of black soybean (CJ-3) testa extracts on lipid profiles in streptozotocin (STZ)-induced diabetic rats. Methods: One control group and four STZ-induced diabetic groups with different doses of black soybean (CJ-3) testa extracts treatment [0 mg/kg (diabetic control, EX), 250 mg/kg (EX-250), 500 mg/kg (EX-500), 1,000 mg/kg (EX-1000)] were orally administered for 4 weeks. Results: All CJ-3 treatment groups had remarkably lower serum triglyceride (TG) levels than that of EX group (p < 0.05) whereas hepatic TG contents did not show any differences. Results from serum total cholesterol (TC) concentrations of EX-250 and EX-1000 groups were decreased compared to EX group (p < 0.05). Furthermore, protein levels of 3-hydroxy-3-methyl-glutaryl-Coenzyme A (HMG-CoA) reductase from the liver decreased in all treatment groups (p < 0.05). However, significant differences were not observed in serum glucose and insulin, and glucose transporter 4 (GLUT-4) protein expression in skeletal muscle tissue. Conclusion: These results suggest that black soybean testa extracts could be useful for improvement of hyperlipidemia and hypercholesteremia in diabetes.

Effects of YK-209 Mulberry Leaves on HMG-CoA Reductase and Lipid Composition of Liver in Streptozotocin-Induced Diabetic Rats (YK-209 뽕잎이 Streptozotocin 유발 당뇨쥐 간조직에서의 HMG-Coa Reductase 활성과 지질조성에 미치는 영향)

  • This study was conducted to investigate the effects of YK-209 mulberry leaves on HMG-CoA reductase activity and lipid composition of liver in streptozotocin-induced diabetic rats. Sprague-Dawley male rats weighing 100 $\pm$ 10 B were randomly assigned as a normal group and four STZ-induced diabetic groups according to the level of dietary mulberry leaves supplement. The experimental diets were fed ad libidum, so that diabetes was experimentally induced by intravenous injection of STZ 55 mg/kg of body weight after feeding for 3 weeks. Animals were sacrificed on the 9th day of diabetic states. The levels of serum triglyceride, total-cholesterol and LDL-cholesterol in DM group were higher than mulberry leaves supplemented groups and normal group, but those of the mulberry leaves supplemented groups were significantly decreased to normal level. In contrast, the leavels of serum HDL-cholesterol in DM group was significantly reduced than that of normal group, but mulberry leaves supplemented groups were increased to normal level. Atherogenic index in DM group was higher about 3 fold than the normal group but the DM-0.1Y and DM-0.2Y groups were maintained the normal level. Contents of total lipid and triglyceride of liver in DM group were significantly lower than that of normal group, but the mulberry Leaves supplemented groups increased than that of DM group. The contents of hepatic cholesterol in DM group was 160% higher than that of normal group, but the mulberry leaves supplementation groups maintained the normal level. The activity of hepatic 3-hydroxy -3-methylglutaryl Coenzyme A (HMG-CoA) reductase in DM group was 43% lower than that of normal group, but had no significant difference between DM-0.1Y, DM-0.2Y and normal groups. In conclusion, YK-209 mulberry loaves has improving effect of the lipid metabolism in STZ-induced diabetic rats through hepatic HMG-CoA reductase activity, and the change of lipids contents in serum and liver.

Effects of Cirsium japonicum Powder on Plasma Glucose and Lipid Level in Streptozotocin Induced Diabetic Rats (엉겅퀴 섭취가 Streptozotocin 유발 당뇨 흰쥐의 혈당과 지질수준에 미치는 영향)

  • This investigation was conducted to assess the influence of Cirsium japonicum consumption on the plasma glucose and lipid profiles in streptozotocin (STZ)-induced diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 230 g by injection of STZ into the tail vein at 45 mg/kg body weight. The rats were randomly assigned into four groups: a normal and STZ-control fed an AIN-93 diet group, and diabetic groups whose diets were supplemented with 10% Cirsium japonicum powder containing leaf or root for four weeks. To observe the effects of Cirsium japonicum in the animal model, the levels of glucose, cholesterol, HDL-cholesterol, triglyceride and free fatty acid in the plasma and the levels of glycogen in the tissue were determined after four weeks. Treatment of STZ-induced diabetic rats with leaf consumption significantly lowered the diet intake when compared with STZ-control rats. The relative weights of the liver and kidney differed significantly between the normal and diabetic groups. The weights of the kidneys in the leaf group were significantly lower than those in the STZ-control group. The level of hematocrit was also significantly lower in diabetic rats whose diets were supplemented with leaves when compared to those of STZ-control rats. The plasma glucose level was found to be significantly lower in the leaf group than the STZ-control group. The effect was significant after 2 weeks. The HDL-cholesterol levels increased in all of the diabetic experimental groups when compared to the STZ-control group. These results suggest that supplementation with Circicum japonicum leaves induced considerable hypoglycemic effect in STZ-induced diabetic rats and that these leaves may be useful for the management of diabetes mellitus.

Instant Gruel from Colored Barley and Oats for Improving Diabetic Conditions (유색보리와 귀리를 이용한 당뇨환자용 즉석죽의 당뇨 개선효과)

  • The abilities of instant gruel manufactured with colored barley and oats to improve diabetic conditions were investigated using diabetes-induced mice and rats. Mice or rats were divided into a diabetic control group and one experimental group (seven animals per group). The control groups were fed without instant gruel and experimental groups were fed basal diets supplemented with 10% instant gruel for 8 weeks. The streptozotocin (STZ)-induced diabetic rats experimental group showed a significant decrease in food intake compared to the control group. Both Type II diabetic mice and STZ-induced diabetic rats experimental groups showed higher increases in body weight than the control groups. The blood glucose levels of the experimental groups ($352{\pm}12.2$ mg/dL in Type II diabetic mice; $296.4{\pm}13.2$ mg/dL in STZ-induced diabetic rats) were lower than the untreated control groups ($426.0{\pm}15.4$ mg/dL in Type II diabetic mice; $514.0{\pm}17.6$ mg/dL in STZ-induced diabetic rats). The serum insulin levels of Type II diabetic mice increased by 38.3% in the experimental group ($12.8{\pm}1.1$ ng/mL) compared to the control group ($7.9{\pm}0.5$ ng/mL). The immunohistochemical density of insulin-secreting cells and glucagon-like peptide-1 (GLP-1)-secreting cells in the pancreas were significantly higher in the experimental groups than the control groups for Type II diabetic mice and STZ-induced diabetic rats. Therefore, we conclude that instant gruel manufactured with colored barley and oats stimulates the secretion of insulin and decreases blood sugar by activating insulin-secreting cells in the pancreatic islets of diabetic animals.

Effects of YK-209 Mulberry Leaves on Antioxidative Defense System of Liver in Streptozotocin-Induced Diabetic Rats (YK-209 뽕잎이 Streptozotocin 유발 당뇨쥐 간조직의 항산화계에 미치는 영향)

  • The purpose of this study was investigated the effects of YK-209 mulberry leaves on antioxidative defense system of liver in diabetic rats induced with streptozotocin (STZ). Male Sprague-Dawley rats weighing 100$\pm$10 g were randomly assigned to one normal and four STZ-induced diabetic groups; YK-209 mulberry leaves free diet (DM group),0.1% YK-209 mulberry leaves diet (DM-0.1Y group),0.2% YK-209 mulberry leaves diet (DM-0.2Y group) and 0.4% YK-209 mulberry leaves diet (DM-0.4Y group). Diabetes was induced by intravenous Injection of 55 mg/kg body weight of STZ in sodium citrate buffer (pH 4.3) via tail vein after 4 weeks feeding of experimental diets. Rats were sacrificed at the 9th day of diabetic states. Liver weight in all four diabetic groups were higher than normal group, but YK-209 mulberry supplementation groups were lower than DM group. Hepatic superoxide dismutase (SOD) activity was significantly decreased in all diabetic groups, compared with normal group. Hepatic glutathione peroxidase (GSHpx) activity was 7.3% decreased in DM group, compared with normal group, but those of DM-0.1Y and DM-0.2Y groups were maintained the normal level. The hepatic thiobarbituric acid reactive substances was markedly increased by 144% in DM group, compared with normal group, but those of DM-0. 1Y, DM-0.2Y groups were maintained the normal level. The contents of lipofuscin in liver were increased by 100% in DM group compared with normal group, but those of DM-0. 1Y, DM-0.2Y and DM-0.4Y groups were decreased to 42% 43% and 44%, respectively, compared with DM group. The hepatic superoxide radical (0$^2$-) contents in DM group were increased to 81%, compared with normal group, but those of DM-0.1Y and DM-0.4Y groups were similar to those of normal group. The present result indicate that YK-209 mulberry leaves regarded to suppress lipid peroxidation as an free radical scavenger system by the inhibition of oxidative stress.
