Secondary Structure for RNA Aptamers Binding to Guanine-Rich Sequence in the 5'-UTR RNA of N-Ras Oncogene

  • RNA molecules which bind to the G-rich sequence in the 5'-UTR RNA which plays an important role in expression of N-ras, were selected. The secondary structures of five selected RNA aptamers including primer sequence were found by the CLC RNA workbench ver. 4.2 program (www.clcbio.com) and investigated with RNA structural probes such as RNase T1 which has specificity for a G in single-stranded region, RNase V1 specific for double strand and nuclease S1 specific for single strand. The generalized secondary structure model was proposed and characterized. It was composed of a central long double strand region flanked by single strand region at both end sides. The double strand region had an internal single-strand region and bulges. The single strand loop in the right side was composed of four or five nucleotides.

Effect of Some Natural Products on the DNA Damaging Activity of 4NQO (4-nitroquinoline n-oxide) and Daunorubicin (Daunorubicin과 4NQO의 DNA damaging activity에 대한 천연물질의 영향)

  • The action mechanism of the inhibitory effect of some natural products on the DNA strand break and DNA damage was investigated in vitro and in vivo. In the E. coli chromosomal DNA strand break experiment in vitro, three mushroom water extracts were effective on the DNA strand breaking by daunorubicin. Phellinus linteus water extract inactivated daunorubicin, a DNA strand breaking agent, but did not protect DNA from daunorubicin-induced DNA strand breaking. Agaricus blazei water extract inhibited DNA strand breaking action of daunorubicin not only by daunorubicin inactivation, but also by DNA protection from daunorubicin. An inhibitory effect of Ganoderma lucidum water extract on the DNA strand break was based on the DNA protection rather than daunorubicin inactivation. In vivo mutagen assay system (SOS-chromotest), among three mushroom water extracts Phellinus linteus water extract was the most effective one on the inhibition of DNA damage by 4-NQO. The results suggest that all three mushroom water extracts inhibit daunorubicin-induced DNA damage and in vivo DNA damaging action of 4-NQO by the reaction of mutagen inactivation or DNA protection from the mutagen.

DNA Strand Breaks in Mitotic Germ Cells of Caenorhabditis elegans Evaluated by Comet Assay

  • DNA damage responses are important for the maintenance of genome stability and the survival of organisms. Such responses are activated in the presence of DNA damage and lead to cell cycle arrest, apoptosis, and DNA repair. In Caenorhabditis elegans, double-strand breaks induced by DNA damaging agents have been detected indirectly by antibodies against DSB recognizing proteins. In this study we used a comet assay to detect DNA strand breaks and to measure the elimination of DNA strand breaks in mitotic germline nuclei of C. elegans. We found that C. elegans brc-1 mutants were more sensitive to ionizing radiation and camptothecin than the N2 wild-type strain and repaired DNA strand breaks less efficiently than N2. This study is the first demonstration of direct measurement of DNA strand breaks in mitotic germline nuclei of C. elegans. This newly developed assay can be applied to detect DNA strand breaks in different C. elegans mutants that are sensitive to DNA damaging agents.

  • Bamboo strand board (BSB) was made with Phyllostachys banbusoides growing in Damyang district. Physical and mechanical properties of this BSB were summarized as follows; The specific gravity of BSB was 0.63∼0.79. Specific gravity decreased slightly with the thickness and length of BSB. Moisture content of BSB manufactured was 5.8∼6.9%. The absorption ate of BSB (42∼48%) did not show any relationship with the thickness and length of BSB. The thickness swelling rate of BSB was 13.9∼17.0%, relatively higher than any other panel products. Thickness swelling rate increased with the thickness of BSB, showing the strand thickness influenced much more on the rate of thickness swelling of BSB than the length of strand. The 3-point bending strength of BSB was 98∼126kgf/$\textrm{cm}^2$. Bending strength of showed the tendency of increase with the increased length of BSB, but with the decreased thickness. In particular, the length of BSB showed more effect on the increase of bending strength of BSB than the thickness of BSB. The compression strength perpendicular to BSB surface was 411 ∼ 465 kgf/$\textrm{cm}^2$, and the optimal length of strand for the 1mm- and 2mm-thickness of strand was 40mm and 60mm, respectively. Compression strength paralleled to BSD was 160∼221kgf/$\textrm{cm}^2$ and the optimal length of strand for the 2mm-thickness of strand appeared to be 60mm. The present work showed that appearance, physical and mechanical strength of BSB appeared quite positive in terms of board qualities, suggesting that bamboo would be appropriate for the production of board materials. In addition, our work showed that the crucial factor for determining the mechanical characteristics of BSB was the dimension of strand.

DNA Light-strand Preferential Recognition of Human Mitochondria Transcription Termination Factor mTERF

  • Transcription termination of the human mitochondrial genome requires specific binding to termination factor mTERF. In this study, mTERF was produced in E. coli and purified by two-step chromatography. mTERF-binding DNA sequences were isolated from a pool of randomized sequences by the repeated selection of bound sequences by gel-mobility shift assay and polymerase chain reaction. Sequencing and comparison of the 23 isolated clones revealed a 16-bp consensus sequence of 5'-GTG$\b{TGGC}$AGANCCNGG-3' in the light-strand (underlined residues were absolutely conserved), which nicely matched the genomic 13-bp terminator sequence 5'-$\b{TGGC}$AGAGCCCGG-3'. Moreover, mTERF binding assays of heteroduplex and single-stranded DNAs showed mTERF recognized the light strand in preference to the heavy strand. The preferential binding of mTERF with the light-strand may explain its distinct orientation-dependent termination activity.

An Experimental Study on the Fracture Characteristics of Wire Ropes (와이어 로프의 파단특성에 관한 실험적 연구)

  • This study examines fracture characteristics of wire rope of 6$\times$37+FC 14mm B degree, Which has been widely used for the strength support for machinery, cranes and shipping. The examination Is based on the fellowing experimental conditions. Firstly, this study analyzes the effect of the strength of wlre rope. When number of broken wires is occuring at the level of 5%, 10% and 15% degree respectively in the following two cases:1) Case of broken down to each strand equally. 2) Case of broken down to one strand only. Secondly, this study analyzes the effect of corrosion on the strength of wire rope. Corrosion times are 72hours, 144hours and 216hours corrosion respectively under following conditions. 1) Case of only a corrosion without wire breaking. 2) Case of corrosion after 5% degree wire breaking to each strand equally. Some outcomes of these experiment are as follows. 1) From the of tension test, We can find that the ratio of strength reduction case of wire broke down to one strand concentrically is greater than that ratio case of broke down to each strand equally. 2) From the corrosion test, We can also find that the ratio of strength reduction case of corroded after 5% degree wire breaking to each strand equally Is greater than that ratio case of corroded without wire breaking. 3) After comparing theoretical fracture load of wires with experimental fracture load of wires, we can find the fracture load of the case of broken down to each strand equally is most similar to theoretical fracture load. 4) It is proposed that safety rules on wire rope in the Industrial safety and healthy labor of law have to change “breaking over 10% of number of wires in one strand of wire rope(excluded filler wire )”has to be replaced by“breaking over 10% of number of all wires in all strand of wire rope(excluded filler wire)”

  • The nature of bond of untensioned prestressed strand in concrete differs from that of plain or deformed reinforcing bar as well as tensioned prestressed strand. There is a very limited amount of published research information regarding bonding of this type reinforcing. In order to use and design untensioned strand as reinforcing, relationships defining the load transfer characteristics of the strand are necessary. A program based upon pullout tests was designed to develop data relating the critical parameters for determining load transfer behavior of the untensioned strand. The purpose of this study is to investigate the characteristics of bond and development length between untensioned strand and concrete. The test variables include diameter of strands (9.3mm, 12.7mm) and development lengths. The maximum bond stress at the 9.3mm and 12.7mm strands decreases with the increase of the rate of development length. The untensioned prestressed strands displayed bond performance when secure development length more than 80$\%$ according to the development of deformed bars equation.

  • In this study the experimental results of fatigue specimen for the Strand Stay Cable Assembly of Extadosed bridges is investigated. The fatigue test and tensile experiment is conducted to 6 kinds of specimens. Test specimen OVM250-31 Strand Cable System manufactured by china OVM B-Machinery Co., Ltd, and OVM250-42 Parallel Strand Stay Cable Assembly manufactured by china OVM B-Machinery Co., Ltd, are passed for fatigue test and rupture tensile test. But Test specimen OVM250-42 Parallel Strand Stay Cable Assembly manufactured by korean A-Machinery Co., Ltd, is not passed for fatigue test conducted according to the "Recommendation for Stay Cable Design. The test result are compared to the fatigue criteria of PHI 2001 for cyclic load, and it is concluded that the current korean design code will be needed for representing the fatigue load in Hot Dip Galvanized Strand Stay Cable. It is verified that the new korean specification and quality criteria of Strand Stay Cable and exact experimental applied process will be needed.

  • The 4-probe method with a voltage tap on terminals has been used for the measurement of the critical current of multi-strand high-T$_{c}$ superconducting(HTS) cables. And the critical current of cables is obtained as the measured total current divided by the number of conductor when the terminal voltage exceeds the predetermined criterion of critical current. However, because of the non-uniform current distribution due to the different critical current, shapes, and other characteristics of each conductor, this is not applicable method to the multi-strand HTS cable. To determine the critical current of multi-strand HTS cable, the critical current of each conductor must be measured with different method. h this paper, the current distribution and the critical current of each conductors in multi-strand cable were measured with specially made Pick-up coils and voltage taps. It is presented that the real critical current of multi-strand is smaller than sum of each conductors. The main cause of non-uniform current distribution is the difference between the resistances appeared in each HTS wires.s.
