Applications of Organic Fungicides Reduce Photosynthesis and Fruit Quality of Apple Trees

  • Two different pest control programs were applied on 8-year-old 'Ryoka'/M.26 apple trees (Malus domestica Borkh.). Lime sulfur or Bordeaux mixture with emulsified oil were applied 12 times from late March to mid-September as organic treatment, and synthetic chemicals were 7 times applied as control treatment. Over the entire apple-growing season, photosynthesis rates of apple trees were significantly lower in the organic treatment than in the control, and this photosynthetic differences were larger in July and August. Photosynthesis-related parameters such as stomatal conductance and transpiration behaved similarly to photosynthesis. The leaf area in the organic treatment was significantly smaller ($24.7cm^2$) than that in the control treatment ($30.7cm^2$). Organic leaves contained significantly less Chl. a ($15.5mg{\cdot}g^{-1}$) than did control leaves ($17.6mg{\cdot}g^{-1}$). Fruit yield per tree was significantly lower in the organic treatment (18.8 kg) than in the control (24.5 kg), because organic fruits experienced a higher rate of disease infection such as white rot (Botryosphaeria dothidae) and bitter rot (Glomerella cingulata) than did control fruits. Organic fruits had high flesh firmness but less color development (lower Hunter's a values). In this experiment, the pest control program with frequent applications of organic fungicides showed negative effects on photosynthesis and disease infection on leaves and fruits, and thus reduce the fruit quality and yield in 'Ryoka'/M.26 apple trees.

Growth Characteristics and Photosynthesis of Soybean Seedling to NaCl stress in Sand Culture (콩의 유묘기에 있어서 NaCl Stress에 의한 생육특성과 광합성 반응)

  • This atudy was conducted to determine the growth characteristics and photosynthesis of soybean (Glycine max L. cv. Keumjongkongl) 30 day old seedling to 100mM NaCl concentration containing 1/2 Hoagland`s nutrient solution in sand culture. The nodule formation of root is not found perfectly with NaCl stress. The leaf dry matter weight (g/plant) of stressed plant is more reduction in 77% to control than any other characters. The water content (%) is tend to increase but water potential (MPa) is tend to decrease at NaCl stress. The chlorophyll content (SPAD) is tend to increase at growing leaf age of control but decrease at NaCl stress. The photosynthesis, stomatal conductance and transpiration are tend to decrease sharply at NaCl stress.

Improvement of ex vitro acclimatization of mulberry plantlets by supplement of abscisic acid to the last subculture medium

  • Mulberry (Morus sp.) of the family Moraceae is very economically important in Asian countries including Korea, because its leaf and fruit have been commercially used in sericulture and horticultural industries. Therefore it is necessary to develop the optimal production system for rapid and cost-effective propagation of mulberry. Our studies focused on establishing an acclimatization method for the successful plantlet production of new cultivar 'Cheongsu' which was transferred ex vitro after in vitro culture. In particular, effect of abscisic acid (ABA) addition into the last subculture medium on plantlet response to subsequent ex vitro transfer and its growth was investigated. During acclimatization, stomatal conductance and transpiration rate of ABA-pretreated plantlets were significantly lower than those of non-treated plantlets. Net photosynthetic rate of ABA-pretreated plantlets decreased after ex vitro transfer but increased after 14 days, and it was mostly higher than that of non-treated plantlets. Moreover, relative water content as well as chlorophyll contents and its ratio were also higher in ABA-pretreated plantlets. On the other hand, proline was considerably higher than in control plantlets. After 1 month of ex vitro transfer, survival rate of ABA-pretreated plantlets was 85.6%, which increased by 29.1% in comparison with control (56.5%). More vigorous growth was also observed in ABA-pretreated plantlets. From these results, it was found that application of ABA to the last subculture medium could improve acclimatization and promote survival of mulberry plantlets after ex vitro transfer, inducing water stress tolerance and alleviating abiotic stresses.

  • The occurrence rate of russet in 'Whangkeumbae' pear showed significant difference by years because the russet occurrence is greatly affected by the amount of rainfall. This study was conducted to analyse the relationship between rainfall and russet occurrence by artificial treatment of high humidity. Under high relative humidity condition, stomatal resistance decreased and average fruit weight was higher since the increased net photosynthesis rate accumulation accelerated fruit growth. The russet occurrence began on July 25, when the growth speed of fruit weight and fruit surface is the most fast. Russet occurrence rate was higher in high relative humidity condition because the fruit growth was accelerated. Since the fruit calcium concentration change is extreme in late July, it is assumed that the deceased calcium content is related to the occurrence of russet in 'Whangkeumbae' pear, When the high relative humidity condition is maintained after rainfall, the amount of net photosynthesis rate increase and fruit growth is accelerated. Therefore, the unbalance in the amounts of transferred photosynthesis assimilation product, water and mineral elements would be one of the reasons for the russet occurrence in 'Whangkeumbae' pear.

Photosynthetic Characteristics and a Sensitive Indicator for $O_3$-exposed Platanus orientalis (오존에 노출된 버즘나무의 광합성 특성과 민감성 지표)

  • We investigated the effect of $O_3$ on the photosynthetic characteristics of oriental plane (Platanus orientalis L.) that is used as a side tree or ornamental tree in Korea. Two-year-old oriental plane seedlings were transplanted to pots and transferred into a closed $O_3$ chamber, Photosynthetic pigment content and photosynthetic characteristics of leaves were measured every three weeks during 100 ppb $O_3$ fumigation. There was no visible foliar injury by $O_3$ exposure and the content of photosynthetic pigments did not show significant differences between control and $O_3$-treated seedlings. But photosynthetic rate, stomatal conductance, and water use efficiency in leaves of $O_3$-treated seedlings were reduced after six weeks of ozone fumigation. In addition, reduction of carboxylation efficiency and photochemical efficiency was observed in leaves of $O_3$-treated seedlings after three weeks and six weeks. In accordance with our results, carboxylation efficiency, the most sensitive parameter to $O_3$ stress, was considered to be a suitable indicator of $O_3$ sensitivity.

Ecotypic Variation of Pulsatilla koreana Nakai Distributed in Korea (韓國에 分布하는 할미꽃 (Pulsatilla Koreana Nakai)의 生態型的 變異)

  • In order to investigate the ecotypic variations of pulsatilla koreana nakai distributed in korea, 8 local populations of the plant were selected in accordance with latitude and compared on the differences of leaf shape, elongstion, stomatal distribution and dhlorophyll content. The incised angle of the plant is 38。32' in cheju, showing a great difference as compared with 23。77'-30。 32' of the other sites. as to the incised rate, mt. togyu and cheju representsed relatively high values of 49.04% and 48.21% the parted leaved individuals were only observed along the warm current coastal area including cheju, sokcho and sunchon and were 54(62.8%), 2(2.5%) and 3(3.3%) in the sites, respectively. Especially, the individuals consisting of only parted learves were 5(5.8%) in cheju alone and none of the other sites showed such individuals at all. So p. cernua distributed in cheju and p. koreana in inland area can be considered to beecotypic variations of the same species from the upper investigations. According to the elongations of leaf and rachis, the local populations could be classified into 3 types, such as warm current coastal area type(cheju,sunchon and sokcho),southern inland area type(andong, chungju and mt. togyu)and central inland ara type(mt. komdan and hongchon). The dentate numbera of 39.80-43.73 in the warm current area were also different from those of 87% in mt.togyu and 15% in hongchon, while opposite leaflets account for 100% in the other sites. The content of chlorophyll ranged from 0.609 mg/g. d.w.(cheju)to 0.924 mg/g.d.w.(hongchon), showing an increasing trends as the latitudes grow higher. The number of stomata on the low epidermis is much larger than that on the upper epidermis in the whole area. But both aspects showed an increasing tendency, too as the latitude becomes higher.

Growth, Photosynthesis and Zinc Elimination Capacity of a Sorghum-Sudangrass Hybrid under Zinc Stress (고농도 아연 조건에서 수수-수단그라스 교잡종의 생장, 광합성 및 아연 제거능)

  • Plant biomass, photosystem II (PSII) photochemical activity, photosynthetic function, and zinc (Zn) accumulation were investigated in a sorghum-sudangrass hybrid (Sorghum bicolor ${\times}$ S. sudanense) exposed to various Zn concentrations to determine the elimination capacity of Zn from soils. Plant growth and biomass of the sorghum-sudangrass hybrid decreased with increasing Zn concentration. Symptoms of Zn toxicity, i.e., withering and discoloration of old leaves, were found at Zn concentrations over 800 ppm. PSII photochemical activity, as indicated by the values of $F_v/F_m$ and $F_v/F_o$, decreased significantly three days after exposure to Zn concentrations of 800 ppm or more. Photosynthetic $CO_2$ fixation rate (A) was high between Zn concentrations of 100-200 ppm ($22.5{\mu}mol$ $CO_2{\cdot}m^{-2}{\cdot}s^{-1}$), but it declined as Zn concentration increased. At Zn concentrations of 800 and 1600 ppm, A was 14.1 and $1.8{\mu}mol$ $CO_2{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. The patterns of stomatal conductance ($g_s$), transpiration rate (E), and water use efficiency (WUE) were all similar to that of photosynthetic $CO_2$ fixation rate, except for dark respiration ($R_d$), which showed an opposite pattern. Zn was accumulated in both above- and below-ground parts of plants, but was more in the below-ground parts. Magnesium (Mg) and iron (Fe) concentrations were significantly low in the leaves of plants, and symptoms of Mg or Fe deficiency, such as a decrease in the SPAD value, were found when plants were treated with Zn concentrations above 800 ppm. These results suggest that the sorghum-sudangrass hybrid is able to accumulate Zn to high level in plant body and eliminate it with its rapid growth and high biomass yield.

Effect of TDZ (Thidiazuron) on Shoot Proliferation of Peace Poplar (Peace 포플러(Populus koreana X P. trichocarpa)의 줄기형성에 미치는 Thidiazuron 효과)

  • Shoot formation was investigated from in vitro cultivation of exotic hybrid poplar (Populus koreana ${\times}$ P. trichocarpa) with a specific stomatal character occurring both upper and lower surface of leaves. Two different explants (stem and leaf segment) of Peace poplar were cultured on half strength Murashige and Skoog (MS) basal medium supplemented with the various concentrations of thidiazuron as a plant growth regulator. Most adventitious shoots were produced from excised ends of stem or mid-veins of leaf segments. The highest average numbers of shoots were 7.1 and 5.3 with the treatments of 0.02mg/L TDZ in both explants of stem and leaf segment. The highest shooting rates were achieved to 83.3% and 47.6% with the concentrations of 0.01mg/L and 0.02mg/L TDZ by axillary bud and leaf cultures, respectively.

Improvement of heat and drought photosynthetic tolerance in wheat by overaccumulation of glycinebetaine

  • Within their natural habitat, crops are often subjected to drought and heat stress, which suppress crop growth and decrease crop production. Causing overaccumulation of glycinebetaine (GB) has been used to enhance the crop yield under stress. Here, we investigated the response of wheat (Triticum aestivum L.) photosynthesis to drought, heat stress and their combination with a transgenic wheat line (T6) overaccumulating GB and its wild-type (WT) Shi4185. Drought stress (DS) was imposed by controlling irrigation until the relative water content (RWC) of the flag leaves decreased to between 78 and 82%. Heat stress (HS) was applied by exposing wheat plants to $40^{\circ}C$ for 4 h. A combination of drought and heat stress was applied by subjecting the drought-stressed plants to a heat stress as above. The results indicated that all stresses decreased photosynthesis, but the combination of drought and heat stress exacerbated the negative effects on photosynthesis more than exposure to drought or heat stress alone. Drought stress decreased the transpiration rate (Tr), stomatal conductance (Gs) and intercellular $CO_2$ concentration (Ci), while heat stress increased all of these; the deprivation of water was greater under drought stress than heat stress, but heat stress decreased the antioxidant enzyme activity to a greater extent. Overaccumulated GB could alleviate the decrease of photosynthesis caused by all stresses tested. These suggest that GB induces an increase of osmotic adjustments for drought tolerance, while its improvement of the antioxidative defense system including antioxidative enzymes and antioxidants may be more important for heat tolerance.

Plant let growth, leaf stomata, and photosynthesis of grape rootstock '5BB' as affected by inoculum density in bioreactor cultures (포도 왜성대목 '5BB'의 생물반응기 배양에서 접종밀도가 식물체 생장, 기공 및 광합성 특성에 미치는 영향)

  • In bioreactor cultures of plants, inoculum density is an important factor affecting growth and proliferation of the plantlets. To maximize shoot growth and proliferation of grape rootstock '5BB' in bioreactors, inoculum density varied at 15, 30, 45 and 60 single nodes in a 3-liter scale balloon type bioreactor, respectively and cultured for 40 days. Results suggested that the growth and the photosynthesis of the plantlet were greatly affected by inoculum density in the bioreactor. The inoculum density of 45 nodes resulted in the greatest growth (910.4 mg/shoot FW, 764.4 mg/root FW) followed by 30 nodes. $CO_2$ assimilation rate, stomatal conductance, transpiration rate of the plantlet were also highest at the inoculum density of 45 nodes. Significant reduces in shoot and root growth (426.5 mg/shoot FW, 248.4 mg/root FW) were observed at the inoculum density of 60 nodes. When the inoculum density decreased by 15 nodes, plantlets were malformed due to hyperhydricity, resulting in the highest transpiration rate and the lowest $CO_2$ assimilation rate. The plantlets stressed by the inoculum density at 15 nodes and 60 nodes showed larger number and irregular shape of stomata compared to the plantlets inoculated with 45 nodes.
