A Successful Regeneration from Shoot Tips of Chrysanthemum morifolium (Ramat.) following Cryopreservation by Droplet-vitrification

  • This study describes an efficient and widely applicable droplet-vitrification following cryopreservation for shoot tips of Chrysanthemum morifolium (Ramat.) cvs. 'Borami' and 'Yes morning'. The shoot tips of Chrysanthemum were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.7 M). Precultured explants were treated with loading solution (LS, C6) containing glycerol 20% and sucrose 20% for 30 min and exposed to dehydration solution (B5) containing 40% of glycerol and 40% of sucrose for 60 min at $25^{\circ}C$, and then transferred onto droplets containing $2.5{\mu}l$ PVS3 on sterilized aluminum foils ($4cm{\times}0.5cm$) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regeneration rate (%) was obtained when shoot tips were precultured with treatment-2 (exposing of shoot tips to MS + 0.3M sucrose for 30 h and then treated with MS+0.5 M sucrose for 16 h) at $25^{\circ}C$ in both the cultivars. The viability of cooled samples, followed by culturing on $NH_4NO_3$-free MS medium for first 5 days was increased to two-fold (80.7%) regrowth rate over those cultured on normal MS medium or MS medium containing plant growth regulators. This result shows droplet-vitrification would be a promising method for cryobanking chrysanthemum germplasm.

Embryonic Development of Fertilized Eggs of Convict Grouper (Hyporthodus septemfasciatus) ♀×Giant Grouper (Epinephelus lanceolatus) ♂ (능성어 (Hyporthodus septemfasciatus) ♀×대왕바리 (Epinephelus lanceolatus) ♂ 수정란의 난발생)

  • The embryonic development and hatchability of the artificially fertilized eggs of convict grouper (Hyporthodus septemfasciatus) ♀${\times}$giant grouper (Epinephelus lanceolatus) ♂ (CGGG) were compared to those of the maternal pure species (convict grouper ♀${\times}$♂, CG) to establish a novel grouper hybrid for aquaculture industry. The fertilized eggs were divided into nine 5-L beakers (3,000~5,000 eggs/beaker) filled with UV sterilized seawater and incubated at a temperature range of $23.5{\sim}24.8^{\circ}C$ (32.1~32.8 psu). The percentages of fertilization and hatching of CGGG were $69.4{\pm}1.5%$ and $59.0{\pm}5.1%$, respectively and were significantly lower than those of CG (p<0.05). The CGGG proceeded normal embryonic development similar to that of CG, but showed an irregular cleavage, immature embryonic body and spinal deformity in hatched larvae. The incubation time from fertilization to hatching of CGGG was 31 hrs, which was approximately 2 hrs slower than that of CG. Our study provided the possibility of mass production of grouper hybrid CGGG larvae.

  • Corona 19 quarantine is often not implemented at degraded bus stops. We conducted this study to improve these problems. The inside and outside of the shelter can be sterilized with UV photocatalyst coating to maintain a hygienic and clean shelter, and passengers can breathe smoothly by reducing carbon dioxide in the shelter using CO2 sensors and fans. It is also expected that the body temperature can be measured using a body temperature sensor to prevent more than 20% of the spread of the coronavirus in the shelter by isolating suspected coronavirus patients in the first place. When the temperature measurement distance was less than 10cm, the accuracy of the temperature measurement was 98.9%. You can communicate your intention to ride safely and clearly without contacting the bus driver by using a ride bell that uses wireless communication inside the shelter.

Study on the Chlorine-Resistant Bacteria Isolated from Water Pipe Network (상수도관망에서 분리한 잔류염소 내성균에 관한 연구)

  • The free residual chlorine of tap water samples, collected from 266 faucets on the water pipe network in Daegu City, was between 0.1 and 0.79 mg/L. On microorganic tests, general bacteria and the coliform goup were not detected and thus the tap water was turned out to be fit to drink. In particular, samples of which free residual chlorine was 0.1 mg/L and over were cultured in R2A agar media at $25^{\circ}C$ for 7 days, and as a result heterotrophic bacteria were detected in 65.9% of samples; (1). The closer tap water got to the faucet from the stilling basin, the lower residual chlorine concentration became but the more the bacterial count became. And, more bacteria were detected in the R2A agar medium than in the PCA medium. (2). In the case of separated strains, most colonies were reddish or yellowish. 16S rRNA sequence was identified as Methylobacterium sp. and Williamsia sp., and yellow strain was identified as Sphingomonas sp., Sphingobium sp., Novosphingobium sp., Blastomonas sp., Rhodococcus sp. and Microbacterium sp. White strain was identified as Staphylococcus sp. (3). Sterilized tap water in polyethylene bottles was inoculated with separated strain and was left as it was for 2 months. As a result, bio-film was observed in tap water inoculated with Methylobacterium sp. and Sphingomonas sp. It was found that heterotrophic bacteria increased when free residual chlorine was removed from tap water in the water pipe network. Thus, there is a need to determine a base value for heterotrophic bacteria in order to check the cleanliness of tap water in the water pipe network.

Kinetic examination on synergistic effect of co-use of ultrasonic wave and ultraviolet radiation on bacterial disinfection in a household humidifier (초음파와 자외선(UV) 동시 적용이 가정용 가습기 내 살균에 미치는 시너지 효과에 대한 반응속도론적 연구)

  • This study examined the effect of ultraviolet (UV) application on bacterial disinfection in a commercialized humidifier using ultrasonic wave (UW). To accurately examine disinfection kinetics in tap-water condition, tap-water was sterilized using a filter, and then inoculated with pure cultures of E. coli and P. putida with known viable counts. The disinfection kinetic characteristics were experimentally compared when UV alone, UW alone, and UW+UV together were applied in disinfecting the added bacteria in the commercialized humidifier. When UV alone was applied, bacterial disinfection kinetics followed a first-order decay reaction, and showed an approximately 10-time weaker disinfection compared to the typical UV disinfection in water treatment or wastewater treatment. When UW alone was applied, bacterial disinfection kinetics followed a second-order decay reaction with a low disinfection rate constant of 0.0002 min-1(CFU/mL)-1. When UV and UW were applied together, however and interestingly, the disinfection rate constant (0.0211 min-1(CFU/mL)-1) was approximately 100 times increased than that for the UW alone case. These results revealed that the co-use of UV and UW can provide synergistic effect on bacterial disinfection in a tap-water condition in household humidifiers.

The contactless elevator button using the electrostatic capacity (정전 용량을 이용한 비접촉식 엘리베이터 버튼)

  • The elevator installed in the building consists of an elevator call button and an input button for selection to the target floor. The elevator button is input only when the elevator user directly presses it. Such passenger input can be infected with an infectious disease due to contamination of the button. A non-contact button is required as a means for solving this problem, which detects the proximity of an object by applying a capacitive method. It implements a function of measuring the body's body temperature by attaching an infrared heat sensor, and provides a sterilization function of a button by attaching a UV-LED. A button was selected, a body temperature was measured through an infrared temperature measurement sensor, and UV-LED was turned on and sterilized when there was no user. The contactless elevator button is expected to be effective in preventing infectious diseases as it can prevent infection of viruses carrying infectious diseases and can detect body temperature to select positive patients of CIVID 19.

  • Wild garlic (Allium victorialis var. platyphyllum, AVVP) is a nontimber forest product used as an edible and medicinal vegetable. AVVP is usually propagated form offspring bulbs but it takes a long time to harvest. Using tissue culture technology could overcome this problem. This study investigated the optimal conditions for shoot multiplication, root growth, and plant growth by scooping AVVP bulbs. AVVP bulbs harvested from Ulleung Island, Korea, the main producer of AVVP, were surface-sterilized and used for in vitro propagation. Shoot multiplication was performed by the scooping method. More than five multiple shoots were induced from scooped tissue in Quoirin and Lepoivre (QL) medium containing plant growth regulators (PGRs); the maximum number of multiple shoots were induced from scooped tissue in QL medium containing 0.45 μM thidiazuron (TDZ) after 16 weeks of culture. Roots were induced directly at the base of the shoots in all treatments. In vitro rooting depended on the type of PGRs, and the best root-inducing treatment was QL medium containing 9.84 μM indole-3-butyric acid (IBA). Plants with in vitro roots were transferred to pots containing artificial soil and successfully acclimatized for 4 weeks. The acclimatized plants showed a survival rate of 80% after 20 weeks and gradually promoted growth depending on the acclimatization period. The results of this study will be of great help to AVVP dissemination through sustainable mass propagation.

Natural Regeneration Potential of the Soil Seed Bank of Land Use Types in Ecosystems of Ogun River Watershed

  • Soil seed banks as natural storage of plant seeds play an important role in the maintenance and regeneration of watershed. Natural regeneration potential of the soil seed bank of Land use types (LUTs) in Ogun River watershed (ORW) was investigated. ORW was stratified using proportionate sampling technique into Guinea Savannah (GS), Rainforest (RF) and Swamp Forest (SF) Ecological Zones (EZs). Three LUTs: Natural Forest (NF), Disturbed Forest (DF) and Farmland (FL) were purposively selected in GS: GSNF, GSDF, GSFL; RF: RFNF, RFDF, RFFL and SF: SFNF, SFDF, SFFL, respectively. Systematic line transects was used in the laying of the sample plots. Sample plots of 25 m×25 m were established in alternate positions. Ten 1 m×1 m quadrats were randomly laid for soil core sampling from previously randomly selected ten plots. The core samples (10) were pooled per plot in each LUT and placed in individual trays. Ten trays with sterilized soil were used as control. The trays were watered regularly and checked for seedlings emergence fortnightly for 18 months. The experimental design used was 3×3 factorial experiments. ANOVA, Diversity index (H') and Similarity index (SI) were used to analyze the data. There was significant difference in seedling emergence among ecological zones and land use types (p<0.05). A total of 4,400 seedlings emerged from the soil samples. All species were distributed among 32 families. FL in the RF had the highest number of germinated seeds (705±37.33 seedlings) followed by DF in the RF (701±49.6 seedlings). The lowest emergence was in NF of the SF (199±28.41 seedlings). DF in the RF had highest number of species (34) distributed among 22 families. Emergence from soil seed bank of NF in ORW was generally with more of tree species than herbs that were predominant in FL and DF.

X-Ray Diffraction Analysis of Various Calcium Silicate-Based Materials

  • Background: The purpose of this study was to evaluate the composition of the crystal phases of various calcium silicate-based materials (CSMs): ProRoot white MTA (mineral trioxide aggregate) (WMTA), Ortho MTA (OM), Endocem MTA (EM), Retro MTA (RM), Endocem Zr (EN-Z), BiodentineTM (BD), EZ-sealTM (EZ), and OrthoMTA III (OM3). Methods: In a sample holder, 5 g of the powder sample was placed and the top surface of the material was packed flat using a sterilized glass slide. The prepared slides were mounted on an X-ray diffraction (XRD) instrument (D8 Advance; Bruker AXS GmbH, Germany). The X-ray beam 2θ angle range was set at 10~90° and scanned at 1.2° per minute. The Cu X-ray source set to operate at 40 kV and 40 mA in the continuous mode. The peaks in the diffraction pattern of each sample were analyzed using the software Diffrac (version 2.1). Then, the peaks were compared and matched with those of standard materials in the corresponding Powder Diffraction File (PDF-2, JCPDS International Center for Diffraction Data). A powder samples of the materials were analyzed using XRD and the peaks in diffraction pattern were compared to the Powder Diffraction File data. Results: Eight CSMs showed a similar diffraction pattern because their main component was calcium silicate. Eight CSMs showed similar diffraction peaks because calcium silicate was their main component. Two components were observed to have been added as radiopacifiers: bismuth oxide was detected in WMTA, OM, and EM while zirconium oxide was detected in RM, EN-Z, BD, EZ, and OM3. Unusual patterns were detected for the new material, OM3, which had strong peaks at low angles. Conclusion: It was caused by the presence of Brushite, which is believed to have resulted in crystal growth in a particular direction for a specific purpose.

Colonization of Pathogens in Earphones and Observation of Effective Sterilization Methods and Cycles

  • The use of earphones has recently been widely used around the world. In currently, students wear earphones a lot in a daily life. The types of earphones are open-earphones, Canalphones, and headphones. Many students don't periodically to sterilization their earphones. Therefore, it can be an incubator that can induced ear infections. The objective of this study was to detect the pathogenic bacteria from the earphones used by the students. A total of 3 type earphones swabs were collected by sterile cotton swabs. The swabs were inoculated onto BHI agar and incubated aerobically 48 hour at 37℃. 16s rRNA PCR, electrophoresis and sequencing were performed to confirm the identification of all the bacterial isolates. As a result, 24 pathogens were identified in sequencing. Three types of earphones were sterilized in three ways: ultraviolet (UV), 70% ethyl alcohol, and antibacterial wet tissue. If you use earphones for a long time without disinfecting them for a long time, it causes various diseases such as external ear infections. The findings of this study the users periodically to sterilization their respective earphones.
