Nitrogen Compounds of Korea Ginseng and their Physiological Significance

  • Nitrogen compounds of Panax ginseng and their biological activities in plant and animal were reviewed. Major nitrogen compounds found in P. ginseng are free amino acids. Water solilble proteins, indouble proteins and peptides. Minor nitrogen compounds are dencichine. Glycolyroteins, amines, alkaloides, methoxy or alkyl pyrazine derivatives, free nucleosides and nucleic acid bases. 4-methyl-i-thiazoltethanol and pyroglutamic acid the contents of total nitrogen and protein in root Increased until 13 years old which was the highest age tinder investigation. Soluble protein content increased with the root weight and was higher in xylem pith than cortex-epidermis indicating the close relation with root growth. Arginine, which covered 58% of total free amino acids, may serve as storage nitrogen. Arginine seems to be changed into proline in rhizome. threonine in stem and again threonine and arginine in leaf. The greater the root weight the higher the polyamine stimulated Polyamine stimlllated the growth of root callus. Physiological roles of other minor nitrogen compounds are unknown although content is relatively high ((1.if) 6.w). Biochemical and pharmacological activities of some nitrogen compounds for animal were more investigated than physiological role there plant itself. Radiation and U.V protective function (heat stable protein). insulin-like activity in lipogenesis and livolysis (adenosine and pyroglutamic acid), depression of blood sugar content (glycopevtide). htmostatic and nellrotoxic activity (dencichine) and, sedative and hypnotic activity (4-methyl-i-thiazoleethanol) are reported. Heat stable protein increased with root age. The traditional quality criteria appear to be well in accordance with biological activities of nitrogen compounds. Chemical studies of nitrogen compounds seem relatively rare, probably due to difficulty of isolation, subsequently the investigations of biological activities are little.

  • It's crucial to control Alternaria blight and Anthracnose emerging mostly on ginseng leaves during the rainy season to increase the organic ginseng products. The purpose of this study is to investigate the efficay of lime-brodeaux spray on the ginseng leaves and evaluate the growth and yield of the ginseng, and the contents of ginsenoside and 70% ethanol extracts from 3-year-old ginseng variety, Cheonpoong. Lime-bordeaux sprayings were conducted in the ratio of 6-6 in June, 8-8 from July to September every 15 days. After June 10, the spraying have no effects on the growth leaf and stem, and there was no significant increase in chlorophyll contents. The ratio of intact leaf and root were distinctly increased because Alternaria blight and Anthracnose were decreased by spraying lime-bordeaux mixture. Root weight per plant and root yield were increased by 15%, and 62% in 3-year old ginseng, respectively, because the ratio of intact leaf and root were higher by using lime-bordeaux mixture. Furthermore, spraying of lime-bordeaux mixture is prone to increase the ratio of rusty root in ginseng. Spraying of lime-bordeaux mixture decreased both of the contents of ginsenoside and 70% ethanol extract by 13.7%, and 15.2% in 3-year-old ginseng, respectively.

Relationship between Vertical Root Distribution and Yield Traits in IRRI's New Plant Type Rice

  • This experiment was conducted to elucidate the relation-ship between vertical distribution of rice roots and yield traits under field conditions. Eight IRRI's new plant type rices (NPTRs) were tested in a volcanic ash soil paddy field under dense (IO 10 cm) and common (20 20 cm) planting densities. These lines were evaluated to have more spikelet numbers per panicle (SNP), lower filled grain rate (FGR), and lower rough grain weight per hill (RGWH). In dense planting, rough grain weight per stem (RGWS) was increased due to heavier culm and leaf dry weight (CLDW), and both RGWS and CLDW were related with the percentage of root distribution (%RWI) in the 10~30 cm soil layer, while in common planting, RGWS was not closely related with CLDW. SNP was highly related with root dry weight (RDW) in the 0~10cm soil layer. FGR was mainly affected by ROW in the 10~30 cm soil layer under both planting densities. RGWS was positively correlated with top dry weight (TDW) and harvest index (HI), and TDW was positively correlated with RWI under common planting or %RWI under dense planting, and HI was positively correlated with RWI in the 10~30 cm soil layer only under dense planting. RGWS was closely related with root weight index by dry weight (RWI) in the 10~30 cm soil layer and %RWI in the 0~30 cm or 10~30 cm soil layer under dense planting, and with only RWI in the 10~30 cm soil layer under common planting. But RGWH showed the close positive relationship with RDW and RWI in the 10~30 cm soil layer under dense planting, while under common planting, it showed the close positive relationship with RWI and %RWI in the 10~30 cm soil layer or %RWI in the 0~30 cm soil layer. The deeper root system in rice, especially under dense planting, is important for high yield of NPTRs focusing on the increment of top mass production and harvest index.

인삼의 연생별 지상부와 지하부의 주요 생장반응과 특성에 관한 비교분석 (Comparative Analysis on Major Growth Responses and Characteristics of Shoot and Root of Panax ginseng C. A. Meyer with Six Different Years Old)

  • This study was carried out to have the basic and applied information to develop the cultivation methods and models and to increase the productivity of high-quality ginseng. Plant height was dramatically increased from one year old to four years old, and then it was grown up very slowly. Stem length was shown similar tendency as the plant height did. Its range were from 9.4 cm in one year old to about 42.4 cm in four year-old ginseng. The leaf area was clearly increased until four years old, $10.1cm^2$ in one year old to $204.9cm^2$ in four years old, while it was slightly increased after four years old. Root length was continuously increased from one year old until four years old, and then it was grown up very slowly. Tap root length which was measured was appeared about 8 cm from three years old to six years old, showing not much different among the year-olds. The root diameter was continuously increased from one year old to six year-old ginseng, about 2.9 mm and about 19 mm, respectively. The moisture content was higher on the shoot than that on the root. It was slightly decreased as they were getting old. In shoot part, the rate of leaves and stems based on dry weight was about 1 : 1. In the root, the dry weight of tap root was heavier than that of lateral root. The dry weight of shoot was decreased after four years old, while it of the root was continuously increased until six years old, resulting the production of ginseng root. Based on the results of this study, growth characteristics of shoot and root of ginseng were dramatically increased from one year old to four years old and then their growths were appeared different between shoot and root, showing not much growing in shoot and keep growing in root.

Corky Root of Tomato Caused by Pyrenochaeta lycopersici in Korea

  • Corky root symptoms caused by Pyrenochaeta lycopersici were observed on the roots and stem base of tomato plants in Korea. Symptoms on infected plants typically appeared as stunting and generally lacking vigor, and infected plants die back from the foliage tips after fruits have set. Brown lesions appearing with bands around the roots were characteristic symptoms of the disease. The lesions become swollen and cracked along the length of the root with corky appearance. Based on cultural and morphological characteristics, the fungus from the diseased plants was identified as Pyrenochaeta lycopersici. Pycnidia were solitary, globose to subglobose, brown to black, darker around the neck region, and measured 173-215 $\mu\textrm{m}$ in diameter with septate setae up to 102-132$\times$6.5 $\mu\textrm{m}$. Conidia were hyaline, unicellular, and 4.2-4.7$\times$l.5-2.0 $\mu\textrm{m}$ long. Optimum temperature for mycelial growth of the p. lycopersici isolates ranged from $20^{\circ}C$ to $25^{\circ}C$. Fifteen isolates off lycopersici were tested for pathogenicity to susceptible and tolerant cultivars of tomato plants by artificial inoculation. Three isolates of P. lycopersici induced typical corky root discoloration on susceptible tomato cultivars but not on tolerant tomato. This is the Erst report in Korea of tomato corky root disease caused by P. lycopersici.

  • A new high performance liquid chromatographic method was applied for the determination of hyperin in Acanthopanax senticosus and A sessiliflorus. The stationary phase used was $\beta$-Bondapak $C_{18}$ reverse-phase column and a mobile phase program was a gradient of acetonitrile and distilled water at a flow rate of 1.0 ml/min. Hyperin was detected at 210 nm, and the analysis was successfully carried out within 20 min. Hyperin was detected in the one year-grown and two years-grown stem of A senticosus (0.47 and 0.13 mg/g, respectively) and A sessiliflorus (0.14 and 0.03 mg/g, respectively). Hyperin was detected in the main and branch root of A sessiliflorus (0.30 and 0.09 mg/g, respectively). But there is no detection of hyperin in the main and branch root of A senticosus.

  • Survey on potato parasitic nematodes has been undertaken in order to find distribution of the nemic fauna. 41 soil samples were taken from potato fields in Gyeongbug, Gyeongnam and Gangweon provinces. Twenty four species belonging to 16 different genera were identified. Potato cyst nematode, Globodera rostochiensis was not found in the areas. Potato-rot nematode, Ditylenchus destructor and stem nematode, Ditylenchus dipsaci were found from several potato fields and population density high and showed damage to the crops. Root-lesion nematode, Pratylenchus minyus, Pratylenchus penetrans, Pratytenchus thornei and Pratylenchus vulnus were found and their population of these four species were high depending on the fields. Root-knot nematode, Meloidogyne hapla and Meloidogyne incognita were found. M. hapla was found only in Gangweon province and the population density was high. M. incognita was found at Milyang in Gyeongnam province. Spiral nematode, Heticotylenchus digonichus, Helicotylenchus dihystera, Helicotylenchus pseudorobustus, Rotylenchus orientalis and Rotylenchus pini were found. Aphelenchoides saprophilus, Criconemoides informis, Ditylenchus destructor, D. dipsaci, Helicotylenchus digonichus, H. dihystera, Hemicriconemoides intermedius, Meloidogyne hapla, Psilenchus hilarulus, Pratylenchus minyus, and Xiphinema americanum were first found from potato fields in Korea.

Comparative Study on the Content and Cytotoxicity of Pseudolaric Acid B in the Five Plant Parts of Pseudolarix kaempferi

  • Pseudolaric acids of Pseudolarix kaempferi (Pinaceae) have been known as diterpenoids with potent anti-fungal-, anti-microbial, and cytotoxic activities. In the present study, the five MeOH extracts were prepared from the five plant part (root bark, stem bark, leaf, the inner part of root, and cone) to find the relation between the concentration of pseudolaric acids and cytotoxicity. Pseudolaric acids B and C were isolated from the root bark of P. kaempferi to use them as standard compounds. The five extracts were tested on cytotoxicity against six cancer cell lines, A549 (lung), HCT116 (colon), MDA-MB-231 (breast), SNU638 (stomach), and SK-hep-1 (liver) by SRB assay, but against K562 (leukemia) by SRB- or MTT assay. HPLC quantification were performed on a Shisheido Capcell PAK C18 column ($5{\mu}m$, $4.6mm{\times}250mm$) using 254 nm wavelength. The cytotoxicity ($IC_{50}$, $0.36{\mu}g/ml$ on K562 cell lines) of the root bark extract was potent and the content (101.1 mg/g extract) of pseudolaric acid B was very high in the root bark. These results suggest that the MeOH extract obtained from the root bark could be developed as the anti-cancer agent with a high quantity of pseudolaric acid B.

고려인삼의 식물부위 및 여러 조건과 관련한 에틸렌가스 방출 (Ethylene Release of Panax ginseng in Relation to Plant Part and Various Conditions)

  • Ethylene was released from leaf and fruit but root of Panax ginseng. Root callus showed higher ethylene release (ER) than fruit ER increased with leaf senesence. Fruit during ripening showed decreasing ER in the order of green stage, early stage of reddening and fully ripened stage. between leaves from the plant with fruits in different stages of ripening showed similar trend of fruit in ER but it was about 10 times higher in leaves than in fruits. Leaves of P. quinquefolius showed about 200 times higher ER than that of P ginseng on 22 July Fruits from the plant treated with ethephon showed higher ER after 109 days. Forty-five day-old seedlings grown with various growth regulators showed a significant decrease of stem length and significant increase of ER only in Uniconazole (0.1 ppm) and H-9 (0.0, 5 ppm) solution.

  • A cytosolic ascorbate peroxidase, hydrogen peroxide-scavenging enzyme, was characterized from Codonopsis lanceolata. The cytosolic ascorbate peroxidase cDNA (CAPX) was 983 nucleotides long and possess an open reading frame of 753 bp with 251 amino acids (MW 27.9 kDa) with pI 5.61. The deduced amino acid sequence of CAPX shows high homology to other known cytosolic APXs ($78{\sim}83%$), but the CAPX was clustered independently from compared ten plant APXs. The CAPX gene was highly expressed in leaf and stem tissues, but not in root. When Codonopsis leaves cut using scalpel were soaked in 1 mM hydorgen peroxide, the expression of CAPX gene was suppressed.
