Kinetic Study of Xylan Hydrolysis and Decomposition in Concentrated Sulfuric Acid Hydrolysis Process by $^1H$-NMR Spectroscopy ($^1H$-NMR에 의한 Xylan의 황산가수분해 과정에서 나타나는 반응 동력학 연구)

  • Proton-NMR spectroscopic method was applied to kinetic study of concentrated sulfuric acid hydrolysis reaction, especially focused on 2nd step of acid hydrolysis with deferent reaction time and temperature as main variables. Commercial xylan extracted from beech wood was used as model compound. In concentrated acid hydrolysis, xylan was converted to xylose, which is unstable in 2nd hydrolysis condition, which decomposed to furfural or other reaction products. Without neutralization steps, proton-NMR spectroscopic analysis method was valid for analysis of not only monosaccharide (xylose) but also other reaction products (furfural and formic acid) in acid hydrolyzates from concentrated acid hydrolysis of xylan, which was the main advantages of this analytical method. Higher temperature and longer reaction time at 2nd step acid hydrolysis led to less xylose concentration in xylan acid hydrolyzate, especially at $120^{\circ}C$ and 120 min, which meant hydrolyzed xylose was converted to furfural or other reaction products. Loss of xylose was not match with furfural formation, which meant part of furfural was degraded to other undetected compounds. Formation of formic acid was unexpected from acidic dehydration of pentose, which might come from the glucuronic acid at the side chain of xylan.

  • Linear alkyl sulfonates (LAS) constitute a large fraction of the surfactants used in cleaning processes in households, trade and industry Despite the industrial significance and the possible environmental impact of these compounds, the fast and inexpensive determination of LAS concentrations is still a difficult task. In this study, near infrared (NIR) spectroscopy which is a rapid spectroscopic analysis method compared with a traditional analytical method for the measurement of LAS concentration such as HPLC, GC and standard wet chemistry method. NIR spectra of LAS between 0.313 and 25.0% (w/v) in water were utilized to develop a calibration model. The best results (R = 0.998, SEP = 0.244% (w/v)) obtained by using partial least-squares regression with spectral data treatment and 2nd derivatization were comparable to the results (SEC = 0.186% (w/v), SEP = 0.206% (w/v)) obtained by using multiple linear least-squares regression (MLR). However, models based on derivative spectra have no significant advantage with MLR.

Assessment of Autoxidative Resistance for Organic Solvent by Pressure Monitoring Test

  • In the recycle process of organic solvent, the atmospheric oxygen can cause autoxidation and product peroxide. The time-saving method to evaluate the hazards has been required. In this study, oxygen pressure monitoring experiment was proposed as a new method to evaluate autoxidative resistances of solvents. Some of organic solvents were pressurized by oxygen and kept under isothermal condition. At the same time, the pressure in the vessel tracked. Iodometrical titration, thermal analysis and spectroscopic analysis were performed to measure peroxide concentration, the heat of reaction and chemical bonding change. From the results that THF has larger oxygen consumption rate than CPME, it is considered that autoxidative resistance of THF is lower than that of CPME. This method enables to obtain results in shorter time than other methods. These experimental results were consistent with the previous research with longer test durations [1-2].

Use of Nuclear Magnetic Resonance Spectroscopy in Analysis of Fennel Essential Oil

  • A simple and rapid method based on proton nuclear magnetic resonance spectroscopy was developed for determination of trans-anethole content in fennel essential oil. Spectra of pure trans-anethole, of the pure essential oil of fennel, and of the pure oil of fennel with thymol internal standard were recorded. The signal of $H-1^/$ was used for quantification of trans-anethole. This proton signal is well separated in the proton magnetic resonance spectrum of the compound. No reference compound is needed and cheap internal standard was used. The results obtained from spectroscopic analysis were compared with those obtained by gas chromatography. Additionally, the developed method was used for determination of the type of vegetable oil used as a carrier in commercial products, which cannot be quantified as such by gas chromatography. This study demonstrates the application of proton nuclear magnetic resonance spectroscopy as a quality control method for estimation of essential oil components.

  • Epimedii Herba has been used for tonic, cardiotonic, impotency, amnestic and diuretic. Icariin, a main component was isolated from the Epimedii Herba and identified by the spectroscopic methods. In order to evaluate the quality of Epimedii Herba purchased from various regions of Korea, quantitative determination of icariin in Epimedii Herba using HPLC method has been conducted. Quantitative analysis showed the average amount of icariin in commercial 42 samples was 0.34% on the dry weight basis.

  • Sanguisorbae Radix has hemostatic, analgesic and astrigent properties and it has been used for the treatment of burns, scalds and internal hemorrhage. Ziyuglycoside I was isolated from Sanguisorbae Radix and identified by the spectroscopic methods. In order to evaluate the quality of it, quantitative determination of Ziyuglycoside I in Sanguisorbae Radix using HPLC method has been conducted. Quantitative analysis of Ziyuglycoside I showed average 3.005% in 30 samples collected throughout the regions of Korea.

  • We developed a method to measure the concentration of highly concentrated colored samples using color coordinates. We present a color analysis to estimate the concentration of highly concentrated binary solutions with an empirical function and apply it to estimate the kinetics of a catalytic irreversible reaction of resazurin to resorufin. The developed method enabled one to measure the concentration of solutions whose range of concentrations is nonlinear and beyond the limit of UV-vis spectroscopic measurement directly without dilution.

  • Organosilicon compound containing silphenylene unit as an eletrolyte for the application of lithium-ion batteries was synthesized by hydrosilylation method between 1,4-bis(dimethylsilylhydro)benzene and 3-[2-(2-methoxyethoxy)ethoxy]-1-propene. As-prepared new organosilicon compounds containing spacer such as propyl group with ethylene glycol are synthesized to improve thermal stability and to promote conductivity. The products are characterized by spectroscopic analysis.

  • In space exploration, spectroscopic observation is useful for understanding objects' composition and physical properties. There are various methods for analyzing spectral data, and there are differences depending on the object and the wavelength. This paper introduces a method for analyzing visible & nearinfrared (VNIR) spectral data, which is mainly applied in lunar exploration. The main analysis methods include false color ratio image processing, reflectance pattern analysis, integrated band depth (IBD) processing, and continuum removal as preprocessing before analysis. These spectroscopic analysis methods help to understand the mineral properties of the lunar surface in the VNIR region and can be applied to other celestial bodies such as Mars.

  • GCSB-5 preparation is a purified extract from a mixture of 6 medicinal plants(Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used for the treatment of various bone disorders. The aim of this study was to investigate HPLC analysis method and screening of standard compound on Cibotii Rhizoma for quality standardization of a medicinal crude drug GCSB-5. Onitin-4-O-$\beta$-D-glucopyranoside was isolated from Cibotii Rhizoma as the standard compound and identified on the basis of spectroscopic data such as NMR. HPLC analysis method for the determination of onitin-4-O-$\beta$-D-glucopyranoside was established for the quality control of the medicinal plants of Cibotii Rhizoma species, GCSB-5 raw material and GCSB-5 preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline.
