경옥고의 원방과 가미방 익수영진고가(加)꽃송이버섯이 항산화, 면역력 활성 및 기호도에 미치는 영향 (Effect of Original Kyungokgo & Iksuyongjingo plus Sparassis crispa on Antioxidant, Immunity Improvement and Sensory Evaluation)

  • Objectives : This study was conducted with objectives of comparatively investigating effects of original Kyungokgo and Iksuyongjingo plus Sparassis crispa which is gamibang of Kyungokgo on antioxidant improvement, immunity improvement, and sensory evaluation.Methods : Total phenol and DPPH scavenging activity were measured to determine antioxidant improvement of original Kyungokgo and Iksuyongjingo plus Sparassis crispa. While, blood and serum study were conducted and effect on the cytokines were observed from the immunosuppressed rat induced by methotrexate (MTX). Control group was administered with a drinking water, Kyungokgo group was administered with a original Kyungokgo 200 mg/kg, and Iksuyongjingo plus Sparassis crispa group was administered with a Iksuyongjingo plus Sparassis crispa 200 mg/kg for 20 days. Further, intensity and preference evaluation were performed as sensory evaluation.Results : It was observed that Iksuyongjingo plus Sparassis crispa significantly increased total phenol and DPPH scavenging activity, proving that these can improve antioxidant activities. In the immunosuppressed rat, Iksuyongjingo plus Sparassis crispa significantly decreased IL-2 level, significantly increased Il-10 level, and significantly increased neutrophils, RBC, HGB, and HCT levels, indicating that it is effective in increasing immunity. Further, Iksuyongjingo plus Sparassis crispa significantly improved intensity and preference in the sensory evaluation.Conclusions : From the above results, it is concluded that Iksuyongjingo plus Sparassis crispa which is gamibang of Kyungokgo can act effectively on improving antioxidant activity and immunity, and can attribute convenient intake by improving diet preference.

  • 꽃송이버섯(Sparassis crispa DSMZ 5201)의 균사를 사용하여 균사외 효소활성을 측정하였다. Yeast-malt extract-glucose 배지를 사용하여 $24^{\circ}C$에서 15일간 배양 후 배양여액을 조효소원으로 사용하였을 때, $\alpha$-amylase효소의 활성은 44.27 unit/$mg{\cdot}protein$이었다. 배양여액 중의 Protease, CMCase, $\beta$-glucosidase, chitinase 및 exo-$\beta$-1,4-glucanase의 세포외 효소활성은 상대적으로 높았으나, xylanase 효소활성은 낮게 나타났다.

꽃송이버섯의 구성성분과 약리효과에 대한 리뷰 (A Review on Phytochemistry and Pharmacology of Sparassis crispa)

  • Objective : Sparassis crispa has been recognized for its therapeutic value since the late 20th century because of its high β-glucan content. Since then, researches have been conducted on the pharmacological effects but most of these are individual studies on the effects of β-glucan from S. crispa and the comprehensive reviews are lacking. The purpose of this study was to review the compounds composition and pharmacological effects of S. crispa. Methods : This review analyzes the papers about chemical and nutritional composition and pharmacological effects of S. crispa. The data in this review is based on selected papers after reviewing all studies containing the keyword "sparassis crispa" for PubMed, NDSL, and J-Stage published before February 2019. Results : S. crispa is composed of protein, lipids, and carbohydrates. Most of the compounds are carbohydrates and the highest content is β-glucan. More than 40% of the dried fruiting body of S. crispa is composed of β-glucan. In addition, it contains polyphenols, flavonoids, terpenoids and phthalide-based compounds. Broad spectrum of its pharmacological actions have been established which include immunomodulatory, anticancer, antiinflammatory, antioxidant, hypoglycemic, antiobesic and neuroprotective effects. Conclusion : The most studied fields have been shown to have immunomodulatory and anticancer effects by inhibiting the proliferation of cancer cells and angiogenesis and increasing hematopoitic responses. Unique structure and characteristic of high molecular weight β-glucan are considered to have high immunomodulatory effects of S. crispa. And low molecular fractions or phthalides of S. crispa also have antioxidant, immunomodulatory and anticancer effects.

꽃송이버섯 추출물의 화장품소재로서의 가능성 평가 (A study on the possibility of extracts from Sparassis crispa for cosmetic ingredients)

  • 본 연구는 꽃송이버섯 추출물의 화장품소재로서 활용 가능성을 확인하는 것을 목적으로 한다. 꽃송이버섯 에탄올 추출물의 Straphylococcus epidermidis, Straphylococcus aureus, Escherichia coli, Candida albicans 균주로부터 항균효능을 확인하였으며 항산화효과 및 염증관련인자인 NO저해율을 확인하였다. 또한 꽃송이버섯 에탄올 추출물을 함유한 에멀션 제형의 물리적 안정성을 평가하기 위해 pH, 점도, 에멀션 입자를 관찰하였다. 그 결과 paper disc 실험의 경우 Straphylococcus aureus, Candida albicans 균주로부터 항균효능을 나타내었으며, 에멀션 제형 평가의 경우 안정한 점도와 pH를 나타내었다. 현미경으로 유화물 입자 관찰 시 추출물의 유무와 상관없이 제형 분리 현상은 확인되지 않았다. 이러한 결과는 꽃송이버섯 추출물의 화장품 소재로서의 가치를 제안할 수 있다.

  • Sparassis crispa (Wulf.) is an edible/medicinal mushroom and has been reported to biological activities such as antitumor, anti-angiogenesis, antioxidant and wound healing. However, there have not been many researches on osteoarthritis of S. crispa. The aim of this study was to investigate the effects of S. crispa extract on rats with osteoarthritis induced by MIA. Osteoarthritis is a gradually developmental disease that early stage, causes joint stiffness and complains of joint pain. In addition, it gives rise to edema and hypo-function. The results of this study, S. crispa extract effectively inhibited ROS production, increased the production of antioxidant protein SOD and catalase in knee joint cartilage tissue. In addition, S. crispa extract inhibited the expression of pro-inflammatory cytokines and enzymes such as NOX4 and $P47^{phox}$, which are involved in the expression of COX-2, iNOS and the production of ROS. Also, S. crispa extract inhibited the destruction of synovial tissue, cartilage tissue and proteoglycans in articular cartilage in rats.

Sawdust Media Affecting the Mycelial Growth and the Fruiting Body Formation of Sparassis crispa

  • Six strains of Sparassis crispa such as S. crispa DUM-01, DUM-02, DUM-03, DUM-04, DUM-05, and DUM-06 were tested for their mycelial growth on 6 different kinds of sawdust media and primordial formation on 10 different compositions of larch sawdust media. The highest mycelial growth was recorded on the larch sawdust. Of the 6 strains of S. crispa, S. crispa DUM-04 recorded the favorable formation of primordia. The primordial formation of S. crispa DUM-04 was more favorable on L-3 medium than 9 kinds of larch sawdust media. When S. crispa DUM-04 was cultured on the media of larch sawdust+pine sawdust, the formation of its fruiting bodies was more outstanding on the media of larch sawdust+pine sawdust than those of larch sawdust.

Generation and Evaluation of High ${\beta}$-Glucan Producing Mutant Strains of Sparassis crispa

  • A chemical mutagenesis technique was employed for development of mutant strains of Sparassis crispa targeting the shortened cultivation time and the high ${\beta}$-glucan content. The homogenized mycelial fragments of S. crispa IUM4010 strain were treated with 0.2 vol% methyl methanesulfonate, an alkylating agent, yielding 199 mutant strains. Subsequent screening in terms of growth and ${\beta}$-glucan content yielded two mutant strains, B4 and S7. Both mutants exhibited a significant increase in ${\beta}$-glucan productivity by producing 0.254 and 0.236 mg soluble ${\beta}$-glucan/mg dry cell weight for the B4 and S7 strains, respectively, whereas the wild type strain produced 0.102 mg soluble ${\beta}$-glucan/mg dry cell weight. The results demonstrate the usefulness of chemical mutagenesis for generation of mutant mushroom strains.

꽃송이버섯에서 추출한 β-glucan의 tyrosinase 활성과 멜라닌 합성 억제 효능 (Inhibitory Effect of β-Glucan Extracted from Cauliflower Mushroom Sparassis crispa on Tyrosinase Activity and Melanin Synthesis)

  • 천연물에서 피부 미백 효능을 가진 화합물을 개발하기 위한 노력이 많이 이루어지고 있다. 꽃송이 버섯(Sparassis crispa)은 β-glucan의 함량이 건조중량의 40% 이상으로서 항암과 면역증강 효과가 있는 것으로 입증되어 약용으로도 인정받고 있다. 이 연구에서는 S. crispa β-glucan의 멜라닌 생합성 및 tyrosinase 활성억제 효능을 확인함으로써 피부 미백제로서의 활용 가능성을 평가하고자 하였다. 외부 자극제인 α-melanocyte stimulating hormone (α-MSH)와 S. crispa β-glucan (10, 100, 1,000 ㎍/ml)을 B16F1 melanoma 세포에 투여하여 멜라닌 생성량과 tyrosinase 활성도를 측정하였다. Tyrosinase, tyrosinase related protein-1 (TRP-1), TRP-2, microphthalmia-associated transcription factor (MITF)의 발현정도에 대해서는 western blot으로 분석하였다. S. crispa β-glucan을 10, 100, 1,000 ㎍/ml의 농도로 투여하였을 때 α-MSH 만 투여군에 비하여 멜라닌 생성이 각각 13.9%, 18.7%, 39.5% 감소하였다. S. crispa β-glucan을 10, 100, 1,000 ㎍/ml의 농도로 투여하였을 때 β-glucan을 투여하지않은 α-MSH 유도군에 비하여 tyrosinase 활성도는 각각 15.6%, 26.9%, 43.2% 억제되었다. 또한 tyrosinase와 TRP-1, TRP-2, MITF 단백 발현도 효과적으로 감소시켰다. 본 연구 결과에 따르면 S. crispa β-glucan은 MITF 발현을 억제함으로써 tyrosinase 발현을 감소시켜 멜라닌 생성을 억제시킨 것으로 판단된다. 따라서 S. crispa β-glucan은 미백제로서 유용하게 활용할 수 있을 것으로 생각한다.

  • 꽃송이버섯(Sparassis crispa)은 항암 및 면역증강 효과가 뛰어나다고 알려져 있는 베타-글루칸의 함량이 건조중량의 40% 이상인 것으로 보고되고 있는 산림버섯이다. 일반적으로 버섯에서 추출한 베타-글루칸의 면역활성 및 항암 효과는 널리 알려져 있는 반면 꽃송이버섯 유래의 베타-글루칸에 의한 대식세포의 활성화 기작 및 자실체 추출물에 의한 항암 효과는 아직 보고되지 않았다. 본 연구에서는 꽃송이버섯 유래 베타-글루칸의 면역활성 효과를 확인하고자 마우스의 대식세포인 Raw 264.7 cell을 이용하였으며, 베타-글루칸 처리시 TNF-${\alpha}$와 interleukin-$1{\beta}$(IL-$1{\beta}$)와 같은 사이토카인의 발현 및 분비가 증가됨을 확인하였고, 이러한 과정에 필수적인 역할을 담당하는 전사 인자인 NF-${\kappa}B$가 활성화됨도 확인하였다. 뿐만 아니라 꽃송이버섯 추출물 처리 시 TNF-${\alpha}$의 분비가 증가됨과 함께 in vivo에서 암의 성장을 억제하는 기능이 있음을 확인하였다. 이러한 결과들은 꽃송이버섯이 면역활성을 증가시켜 암의 성장을 억제하는 건강 기능성 식품으로서의 개발 가능성이 높음을 의미하는 것이다.

꽃송이버섯(Sparassis crispa) 자실체로부터 ergosteol peroxide의 분리 및 동정 (Ergosterol peroxides from the fruit body of Sparassis crispa)

  • 꽃송이 버섯(S. crispa)을 80 % MeOH 수용액으로 추출하고, 추출물을 EtOAc, n-butyl alcohol 및 물 분획으로 나누었다. EtOAc 분획과 n-BuOH 분획에 대해 $SiO_2$ 및 ODS column chromatography를 반복적으로 실시하여 2종의 ergosterol peroxide를 분리, 정제하였다. infrared spectrometry, nuclear magnetic resonance 및 FAB-MS data를 해석, 화합물 1과 2를 ergosterol peroxide와 3-O-${\beta}$-D-glucopyranosyl ergosterol peroxide로 각각 구조동정 하였다. 두 화합물 모두 꽃송이 버섯으로부터는 이번 실험에서 처음으로 분리되었다.
