Characteristics of trace analysis of potential diesel oxygenates using the factorial design in solid-phase microextraction with GC/FID (고체상미량분석법(SPME-GC/FID)에서 요인배치법을 이용한 디젤첨가제의 미량분석의 특성 평가)

  • In this study, solid-phase microextraction (SPME) technique using GC/FID was studied as a possible alternative to liquid-liquid extraction for the analysis of EGBE, DGBE, DBM and TGME in water, and an optimization condition of trace analysis of EGBE, DGBE, DBM and TGME using the factorial design was described. Experiments used a fractional factorial design method followed by central composite design allowing optimization of a number of factors as well as statical analysis of results. The response surface analysis showed that the extraction efficiency can be described by a second-order polynomial equation in which the salts concentration, extraction temperature, extraction time and sonication time are the major influences. Using DOE, a new data-dependent method was developed that improved the quantity of confidently analyzed EGBE, DGBE, DBM and TGME in water samples.

Investigation of Tacrolimus Loaded Nanostructured Lipid Carriers for Topical Drug Delivery

  • The objective of this investigation was to develop nanostructured lipid carriers (NLCs) of tacrolimus by the hot homogenization technique by sonication. NLCs are commonly prepared by emulsification and lyophilization. The feasibility of fabricating tacrolimus-loaded NLCs was successfully demonstrated in this study. The developed NLCs were characterized in terms of their particle size, zeta potential, entrapment efficiency (EE) of tacrolimus, and morphology. Studies were conducted to evaluate the effectiveness of the NLCs in improving the penetration rate through hairless mouse skin. Tacrolimus-loaded NLCs were found to have an average size of $123.4{\pm}0.3\;nm$, a zeta potential of $-24.3{\pm}6.2\;mV$, and an EE of 50%. In vitro penetration tests revealed that the tacrolimus-loaded NLCs have a penetration rate that is 1.64 times that of the commercial tacrolimus ointment, Protopic$^{(R)}$.

  • Bromelain has been used as a meat-tenderizing agent in food processing. To increase the availability of bromelain, microencapsulation into liposome was carried out by the dehydration and rehydration method. Small unilamellar vesicles prepared by sonication treatment showed higher encapsulation efficiency (EE) than by the French press method. In the preparation of liposome, the effect of pH and centrifugal force on EE was also investigated and it showed a higher EE at acidic pH than at alkaline pH with centrifugation at 80, 000$\times$g. The actual EEs except unencapsulated bromelain which bound on the outside surface of liposome by electrostatic interaction also were investigated, and the optimal EE was at pH 4.6, at 0.6 of a ratio of bromelain to phosholipid(18.2%, w/w). Release of bromelain from liposomes was stimulated as the temperature increased at neutral pH.

Comparison of the Cell Surface Barrier and Enzymatic Modification System in Brevibacterium flavum and B. Lactofermentum

  • To investigate impediments to plasmid transformation in Brevibacterium flavum BF4 and B. lactofermentum BL1, cell surface barriers were determined by measuring growth inhibition whilst enzymatic barriers were determined by comparing DNA methylation properties. B. lactofermentum was more sensitive to growth inhibition by glycine than B. flavum. Release of cellular proteins during sonication was more rapid for B. lactofermentum than for B. flavum. Plasmid DNA (pCSL 17) isolated from B. flavum transformed recipient $McrBC^+$ strains of Escherichia coli with lower efficiency than $McrBC^-$. McrBC digestion of this DNA confirmed that B. flavum contain methylated cytidines in the target sequence of McrBc sequences but B. lactofermentum contained a different methylation pattern. DNA derived from the B. lactofermentum transformed recipient $EcoKR^+$ strains of E. coli with lower efficiency than $EcoKR^-$, indicating the presence of methylated adenosines in the target sequence of EcoK sequences. The present data describe the differences in the physical and enzymatic barriers between two species of corynebacteria and also provide some insight into the successful foreign gene expression in corynebacteria.

Preliminary Results of Extraction, Separation and Quantitation of Arsenic Species in Food and Dietary Supplements by HPLC-ICP-MS

  • Various extraction procedures were investigated using reference materials and samples to evaluate extraction efficiency and effectiveness. Inductively coupled plasma mass spectrometry (ICP-MS) was used to measure total arsenic and to quantitate arsenic species when coupled to an HPLC (high pressure liquid chromatography). Arsenic species were extracted from rice flour (NIST SRM 1568a) with water/methanol mixtures using accelerated solvent extraction (ASE). Total arsenic extraction efficiency ranged from 42 to 64%, for water and various methanol concentrations. From spinach (NIST SRM 1570), freeze-dried apple, and rice flour (NIST SRM 1568a), arsenic species were extracted with trifluoroacetic acid (TFA) at 100 ${^{\circ}C}$. Total arsenic extraction efficiency was 90% for spinach, 75% for freeze-dried apple, and 83% for rice flour. Enzymatic extraction with alpha-amylase and sonication resulted in extraction efficiency of 104% for rice flour, 98% for freeze-dried apple, and 7% for spinach. Chromatograms of arsenic species extracted by the optimum extraction methods were obtained, and the species were quantified. Arsenite (As(III)), arsenate (As(V)), dimethylarsinic acid (DMA), and monomethylarsonic acid (MMA) were found in the apple sample, and DMA and As(V) in the rice flour sample. As(V) and MMA were found in three herbal dietary supplement samples.

Change of the Protection Efficiency in Each Part of Developed Pesticide-Proof Clothes by Repeated Washings (개발 과수용 농약방제복의 반복세탁에 따른 부위별 농약 방호성능의 변화)

  • This study was conducted to evaluate of the protection efficiency in each part of developed pesticide-proof clothes by repeated washings. We investigated the effect of repeated laundering on mechanical properties of pesticide-proof clothes (not washed vs 5 times washed). We also examined pesticide infiltration rate into the pesticide-proof clothes by repeated laundering. The patches(TCL paper, surface area 50cm2)were attached to the inside of pesticide-proof clothes(head, chest, right upper-arm, right forearm, left thigh, left calf, back) which subjects had dressed in during pesticide spraying. The patches were detached from working clothes after work. For the extraction of pesticide in pesticide-proof clothes, sonication was applied for 30 min with methanol. The gas chromatography/mass spectrometry (GC/MS) was applied to identify the pesticide component. The results of this study are as follows: The force strength, water-vapour resistance and surface wetting resistance of pesticide-proof clothes decreased 5 times more in washed clothes. The concentration of pesticide was the highest in the head area of pesticide-proof clothes. In seven parts of TLC paper attached to the pesticide proof clothes, the concentration of pesticide was higher in the left thigh. The penetration part and concentration of pesticide increased as washing was repeated. Therefore the conclusion which can be drawn from this study is this: protection efficiency of pesticide-proof clothes decrease by repeated washings.

The Contribution of OH radical and the Effect of Adding Oxidants on the Sonolytic Degradation of 1,4-Dioxane (초음파를 이용한 1,4-Dioxane의 처리에서 OH 라디칼의 기여와 산화제 첨가의 효과)

  • This research investigates the effects of adding oxidants such as $Fe^0$, $Fe^{2+}$, and ${S_2O_8}^{2-}$ in the sonolysis of 1,4-D. Results indicate that the degradation could be divided into two steps (initiation and acceleration) kinetically. The initial portion agreed with zero-order expression, while the second portion could be fitted with pseudo first-order expression. In the presence of ${HCO_3}^-$, as a radical scavenger, the degradations of 1,4-D and TOC were suppressed, indicating that OH radical is an important factor in the sonolysis. The overall degradation efficiency of 79.0% in the sonolysis was achieved within 200 minutes. While $Fe^0$, $Fe^{2+}$, and ${S_2O_8}^{2-}$ were individually combined with sonication, the degradation efficiency of 1,4-D increased 18.6%, 19.1%, and 16.5% after 200 min, respectively. The addition of oxidants not only changed the kinetic model from zero to pseudo first order at initiation step, but also increased the rate constants in the acceleration step. The addition of oxidants in the sonolysis of 1,4-D also improved the mineralization of 1,4-D, however, the effect of adding oxidants on the rate increase was similar regardless of the oxidants.

Development of a Simple Cell Lysis Method for Recombinant DNA Using Bacteriophage Lambda Lysis Genes

  • In this study, we describe the development of a simple and efficient method for cell lysis via the insertion of a bacteriophage lambda lysis gene cluster into the pET22b expression vector in the following order; the T7 promoter, a gene for a target protein intended for production, Sam7 and R. This insertion of R and Sam7 into pET22b exerted no detrimental effects on cellular growth or the production of a target protein. The induction of the T7 promoter did not in itself result in the autolysis of cells in culture but the harvested cells were readily broken by freezing and thawing. We compared the efficiency of the cell lysis technique by freezing and thawing to that observed with sonication, and determined that both methods completely disintegrated the cells and released proteins into the solution. With our modification of pET22b, the lysis of cells became quite simple, efficient, and reliable. This strategy may prove useful for a broad variety of applications, particularly in experiments requiring extensive cell breakage, including library screening and culture condition exploration, in addition to protein purification.

Feasibility study on remediation for railway contaminated soil with waste-lubricant (윤활유 유래 철도오염 토양의 정화 타당성 연구)

  • Railway-contaminated soil is categorized by total petroleum hydrocarbon(TPH)-related contamination and heavy-metal contamination. The sources of TPH are diesel and lubricant. In this study, the feasibility of soil washing, chemical oxidation and ultra-sonication were investigated to treat lubricant-contaminated railway soil. tergitol, a non-ionic surfactant, was investigated as a washing agent. However, it is not effective to remove lubricant from soil even though tergitol is most effective washing agent for diesel-contaminated soil. Addition of alcohols with surfactant enhanced slightly washing efficiency of the lubricant-contaminated soil. To remediate railway-contaminated soil, source of pollution should be considered.

Dispersion of Highly Pure Single-Walled Carbon Nanotube in Aqueous Solution of Various Surfactants (다양한 계면활성제를 이용한 고순도 단일벽 탄소나노튜브의 수계 분산)

  • Practical application of single-walled carbon nanotubes (SWCNTs) qualified as a promising material has been limited by either poor dispersion or their insolubility in aqueous or organic media due to formation of bundling by relatively high surface energy. Thus, major attention to overcome this issue has been paid at surface modification of CNTs by functionalization, but this introduces defects to the sidewall of CNTs, consequently perturbing the inherent electronic and optical properties. Therefore, using surfactants is a general approach to disperse SWCNTs with lower damages by which bundled nanotubes could be dispersed up to the level of individuals or small bundles. Here, we have investigated various surfactants for their efficiency in dissolving purified SWCNTs produced by arc discharge in deionized water. To compare the surfactants respectively, we have determined the least amount of each surfactant to suspend the nanotubes under optimized experimental conditions(CNT amount, sonication power, and centrifugation speed, etc.) set on the basis of the most common surfactant (sodium dodecyl sulfate, SDS) and discussed the qualitative and quantitative characterization of SWCNT dispersions by UV-Vis absorption spectroscopy. Quantitative aspect about nanotube dispersion was that in particular N-methyl-2-pyrrolidone (NMP) and sodium dodecylbenzene sulfonate (NaDDBS) were found to be effective in dispersing individual tubes.
