Genetic Stability Studies in Micropropagated Date Palm (Phoenix dactylifera L.) Plants using Microsatellite Marker

  • Sixteen microsatellite markers (simple sequence repeat (SSR) markers) were employed to examine the genetic stability of 27 randomly chosen date palm (Phoenix dactylifera L.) plants produced through somatic embryogenesis with upto forty two in vitro subcultures. No microsatellite DNA variation was observed among all micropropagated plants. Our results indicate that the micropropagation protocol used for rapid in vitro multiplication is appropriate and suitable for clonal propagation of date palm and corroborated that somatic embryogenesis can also be used as one of the safe modes for production of true-to-type plants of date palm. This is the first report on the use of microsatellite DNA markers to establish the genetic stability in micropropagated date palm plants.

  • To enhance in vitro plantlet regeneration efficiency of soybean through embryogenesis, the culture conditions such as material part and size of immature seed, 2,4-D, pH and solidifying agents for somatic embryogenesis were investigated. Somatic embryogenesis was induced from the immature embryo, immature cotyledon and embryonic axis explants of the immature seed on MS medium supplemented with 2.0 mg/L 2,4-D. The highest rate (up to 22.9%) of somatic embryogenesis was obtained from the immature cotyledon, following embryonic axis and the immature embryo. The rate varied with the developmental stages of seed. The maximum rate (25.4%) of embryogenesis was obtained from 3-4 mm length of the seed (after 25 days of flowering). The optimum concentration of 2,4-D for embryogenesis was 10 mg/L. The optimum pH was at 5.8 and solidifying agent for medium was better with 0.4% gelrite than with agar. For rapid multiplication of shoot tips from the germinating somatic embryos, they were cultured on MS medium containing 2 mg/L indole-3-butyyic acid (IBA) and 1 mg/L 6-benzyladenine (BA). After then somatic embryos with one and three cotyledons were transferred to the growth regulator free medium. The medium exhibited the higher rate (ca. 50%) of development than the multiplication medium.

  • For the induction of somatic embryogenic callus, the penduncle explants of Corydalis remota for peatinata were cultured on MS basal media supplemented with 2,4-D, kinetin and zeatin. The highest embryogenic callus formation was observed on the media containing 2.0 mg/L of 2,4-D and 2.0 mg/L of zeatin. The somatic embryogenesis on the media with 0.5 mg/L of cytokinin (zeatin or kinetin) were excellent under light condition, however somatic embryos abnormally developed into plantlets. Normal dicotyledonary plantlets were found on MS medium supplemented with 1.0 mg/L of zeatin. When MS medium with 2,4-D plus cytokinin and with BAP were used, the secondary somatic embryogenesis took place in root explants of the regenerants derived from in vitro somatic embryogenic callus.

  • Carrot cotyledon explants cultured on MS medium with 1 mg/L 2,4-D were transferred to a hormone-free solid medium overlaid with filter paper in order to elucidate the effect of simple physical treatment on the development and germination of somatic embryos. Transfer of the explants cultured for one week on MS basal medium overlaid with 3 sheets of filter paper on to MS basal medium increased somatic embryo production 2-39 times over the one week culture on medium without filter paper overlay. Maturation and germination of somatic embryos was more prominent on medium overlaid with filter paper than on medium without filter paper. The explants cultured for one week on filter paper overlay added with liquid medium showed prominent decrease in somatic embryo formation compared to filter paper overlay only. It is suggested that the filter paper overlay affected the moisture environment of the somatic embryos developing on it.

  • The somatic chromosome numbers were counted to be 2n = 16 of Codonopsis lanceolata. Chromosomes 2 and 7 of Sokrisan and Koheung II has a hetero satellite on short arm. Chromosome 5 of Bakwoonsan showed homo satllite on short arms. Short arms of chromosomes 1 and 5 has homo satellite and short arms of chromosomes 2 and 7 has hetero satellite of Jilin.

  • Excised cotyledons and embryo axises of zygotic embryos of Rhus vemicifera were cultured on Murashige and Skoog(MS) medium with various concentrations of 2,4-D. About 3-5% of explants produced callus. Embryogenic callus was preferentially induced from basal parts of embryo axis of zygotic embryos seeds when they were cultured without removal of seed coats. Somatic embryos were developed from embryogenic callus in growth regulator-free medium after 2-3 subcultures on medium with 1.0mg/L 2,4-D and these embryos were matured to cotyledonary stage. Plantlets with well-developed shoots and roots from embryos were obtained on $\frac{1}{4}$MS medium with GA$_{3}$. After acclimatization of plantlets on artificial soil, they were exposed to soil pots.
