Objective : In order to investigate anti-inflammatory effect of Alum objectively which is traditional remedy applicated by external preparation frequently, especially in Rhinologic field. Materials and Method : We studied the histopathologic and hematologic features, serum transaminase activities in the experimentally induced maxillary sinusitis in rabbits. 9 rabbits was divided into normal, control anti sample group(each 3 rabbits). We inoculated $10^{10}$ P. aeruginosa for experimental maxillary sinusitis to control group and sample group. Sample group was treated with aim solution(l0g/100cc) lcc via both nasal cavity (each 0.5cc) after 24hrs everyday for 14days. Results and Conclusion : 1. We confirmed that maxillary sinusitis was well induced by P. aeruginosa without occlusion of maxillary ostium. 2. There was no abnormal findings in serum transaminase(AST/ALT) activities even though application of Alum solution on nasal mucosa for 14days continuously. 3. Alum has evident anti-inflammatory effect of recovering mucosal surface injury, reduction of goblet cell, lymphocyte infiltration, edema and expansion of glandular tissue, dilatation and congestion of blood vessels and so on. 4. Alum has the effect that recover glandular tissue injury and decrease goblet cell increase by the result of dermal PAS staining increase and epidermal AB staining decrease in the qualitative analysis of epidermal and dermal mucopolysaccharide
A new local drug delivery device to treat otitis media (OM) has been developed. This device consists of a biodegradable poly(L-lactic acid) (PLLA) film containing antibiotic (ampicillin, AMP), which can be placed into the middle ear cavity and release the therapeutic concentration of AMP for prolonged period. Biodegradable films containing AMP (10 w/w%) were prepared by solution casting method using a suspension of the drug in a $PLLA/CH_{2}Cl_{2}$ solution (molecular weight of PLLA, 100,000 (100 K) and 300,000 (300 K), respectively). PLLA-AMP films were characterized by FTIR, DSC, and SEM. In vitro release of AMP from AMP-PLLA films were examined. The release pattern of AMP from AMP-PLLA films remained consistent from 1 day to 14 days, and the release rates of AMP from AMP-100K-PLLA film and AMP-300K-PLLA film were $0.7384\;{\mu}g/ml/day$, $0.4107\;{\mu}g/ml/day$, respectively.
A fluorine solution was used to measure the growth of S.mutans and the pH changes were also measured in order to find an effective and preventative dentistry lab within a limited time for the dental hygiene department curriculum and the following results were obtained. 1. In the growth comparison of S.mutans, the culture medium of the experiment group with fluorine weighed 0.3g and the culture medium with no fluorine weighed 0.5g, which shows that the growth rate of S.mutans is significantly decreased in the culture medium with the fluorine. 2. The pH7.8 of the culture medium was not nearly changed; it became 7.0 after culturing with fluoride, however the pH was significantly decreased to 3 in the culture medium that had no fluorine. 3. Since it has been proven that the fluorine can control the growth of germs, it is believed that the effect of fluorine as a cavity preventative should be emphasized to students and in addition, it will help students transmit the effect of fluoride to their patients since this knowledge has been acquired through practice. 4. It is considered that this is a simple test protocol providing effective results in the microorganism and preventive dentistry lab within a limited time and furthermore, it will furnish the students of dental hygiene with comprehensive accessibility to dental hygiene curriculum.
Porphyromonas endodontalis and Prevotella intermedia are black-pigmented anaerobic gram negative rods which have been isolated from infected root canals and submucous abscesses of endodontic origin. And they are associated with clinical symptoms such as pain, percussion, and foul odor. It has been reported that there are 3 serotypes according to capsule membrane in P. endodontalis and 2 DNA homology groups and 3 serotypes in P. intermedia, but there is no data available regarding genetic diversity for the species P. endodontalis and P. intermedia. The purpose of this study is to investigate genetic diversities between individual strains of P. endodontalis and P. intermedia which are indistinguishable by serotyping and biotyping using bacterial DNA restriction endonuclease analysis. 45 teeth with at least one clinical symptoms, with single canal, and with pulp necrosis were sampled. For sampling bacteria, access cavity was prepared after disinfecting tooth and its surroundings. Then the paper point was inserted to the apex of the canal, leave there for 15 seconds, and finally it was placed into PRAS Ringer's solution and PBS solution. P. endodontalis and P. intermedia were identified by biochemical test and IIF after subculturing black and brown colonies which were produced after 7 days of incubation on BAP in anaerobic chamber. P. endodontalis and P. intermedia strains were grown in BHI broth and whole genomic DNA was extracted by phenol-chloroform extraction technique and digested by restriction endonuclease, Eco RI and Pst I. The resulting DNA fragments were separated by agarose gel electrophoresis, stained with EtBr and photographed under UV light. The results were as follows : 1. In both P. endodontalis and P. intermedia, different serotypes could be found within a root canal of same patient. 2. There were obvious genetic heterogeneity within a patient and within a serotype in both P. endodontalis and P. intermedia. 3. P. endodontalis serotype c, isolated from different patients, exhibited limited genotypic diversity.
Objectives : Tongqiao-tang(TQT) has been commonly used for the treatment of common cold, rhinitis etc. Nowadays, TQT becomes one of the most frequently used medicines for allergic rhinitis, but the mechanism of TQT in vivo isn't investigated yet. This study was performed to investigate the effect of TQT on OVA-induced allergic rhinitis mouse model by calculating serum cytokines and IgE. Methods : 8 weeks aged male BALB/c mice were divided into three groups: the normal group, the control group and the medicated group (the TQT group). Each group was consisted of 15 mice. The TQT group was administered TQT extract orally one time a day (1g/kg) from the $1^{st}$ day of experiment till the $26^{th}$ day. The control group and the normal group were administered normal saline by the same method of the TQT group. To induce the allergic rhinitis in the control group and the TQT group, mice of each group were sensitized intraperitoneally with ovalbumin (OVA) solution at the $1^{st}$, the $7^{th}$ and the $14^{th}$ day. After then, intranasal sensitization was performed by dropping 0.1% OVA solution in nasal cavity at the $22^{th}$, the $24^{th}$ and the $26^{th}$ day. At the $27^{th}$ day, the mice were killed and the changes of interferon-${\gamma}$, interleukin-4, interleukin-5, total IgE and OVA-specific IgE were checked. Results : IFN-${\gamma}$ was increased 36% more in the TQT group than that in the control group. IL-4, IL-5, the total IgE and OVA-specific IgE were decreased in the TQT group as compared with the control group and these results were statistically significant. Conclusions : Considering the above experimental results, this study showed that TQT could reduce the allergic reaction in allergic rhinitis. Advanced studies are required to investigate the further mechanisms of TQT.
Objectives This study investigated the effects of Cheongyeonsan for allergic rhinitis. Methods First, the GS/MS was used to analyze the effects of Cheongyeonsan by measuring inflammatory markers. Second, 3 groups of 10 6-week-old BALB/c mice were divided into Ctrl (no treatment), ARE (allergic rhinitis-induced without treatment), and CRT (allergic rhinitis-induced after Cheongyeonsan treatment) groups. Ovalbumin (OVA) was used as an antigen to induce allergic rhinitis and sensitization was performed by intraperitoneal injection of 0.1% OVA solution 21, 14, and 7 days before the onset of allergic rhinitis. Allergic rhinitis was induced by dropping OVA solution on the nasal cavity of each mouse for 5 days after the last sensitization. Seven days after the first induction, second induction was introduced by the same method. After making the section, MMP-9, substance P, $TNF-{\alpha}$, $NF-{\kappa}B$ p65, COX-2, iNOS and Nrf, apoptotic cells were observed by Masson trichrome staining, immunohistochemical staining, TUNEL of nasal mucosal tissues of each group. Results GC/MS results showed undecanoic acid. Masson trichrome results showed that the CRT group had less respiratory epithelial damage. Immunohistochemical staining showed CRT group had 58% decrease in MMP-9, 61% decrease in substance P, 55% decrease in $TNF-{\alpha}$, 38% decrease in $NF-{\kappa}B$ p65, 53% decrease in COX-2, 54% decrease in iNOS, 87% increase in Nrf compared to those of the ARE group. TUNEL showed a positive reaction of 84% increase in apoptotic cells greater than that of the ARE group. Conclusions Cheongyeonsan alleviates nasal mucosal damage and reduces inflammatory mediators from allergic rhinitis-induced mice.
To evaluate the micro leakage of compomer, 4 materials were divided into 4 groups of 15 cavities each.(Group 1: Z-100, Group 2: Dyarct AP, Group 3: Fuji II LC, Group 4: Compoglass) After the class II cavities were prepared using carbide bur No. 553, all specimen were restored by incremental filling technique. In group 3, Z-100 was filled with a base of a light curing glass-ionomer. After 7 days, all specimens were thermocycled between $5^{\circ}C$ and $55^{\circ}C$ for 500 cycles, followed by placement in 50% silver nitrate dye for 2 hours at $37^{\circ}C$. After rinsed in distilled water, these teeth were immersed in photodeveloping solution and exposed to fluorescent light for 6 hours. Teeth were then washed in distilled water to remove the photodeveloping solution, sectioned mesio-distally and evaluated. The results were as follows : 1. In the cervical portion, there was significant difference between Fuji II LC and other groups(Z-100, Dyract AP, Compoglass), Fuji II LC had the least value.(p<0.05) 2. In the cervical portion, there was not significant difference among Dyract AP, Z-100 and Compoglass. 3. In the occlusal portion, there was not significant difference among Dyract AP, Z-100 and Compoglass. From the results above, In enamel, microleakage of compomer such as Dyract AP and Compoglass resemble to that of composite resin. It is thought that it is due to characteristics of composite resin portion of compomer. But in dentin, microleakage of compomer is higher than that of resin modified glass ionomer cement, it is thought that in compomer, acid-base reaction is not developed with dentin.
Objectives: This study aimed to investigate the efficacy of ionic and non-ionic-based contrast media (in vitro study) and the combinatorial effect of chitosan-based endo-radiopaque solution (CERS) (in vivo study) for visualization of the root canal anatomy. Materials and Methods: In vitro study (120 teeth): The root canal of maxillary premolars and molars (in vitro group 1 and 2 respectively, n = 60 each) were analyzed using 4 different contrast media (subgroups: Omnipaque 350, Iopamidol, Xenetix 350, and Urografin 76; n = 15 each) in combination with 5.25% sodium hypochlorite (NaOCl). Based on the results of the in vitro study, in vivo study (80 teeth) was done to compare Xenetix 350 + 5.25% NaOCl with CERS (in vivo group 1 and 2 respectively, n = 40 each) on maxillary and mandibular premolars and molars. Two endodontists used radiovisiography to assess the depth of ingress and identify the aberrant root anatomy after access cavity preparation, and after initial cleaning and shaping of canals. Kruskal-Wallis test was used for in vitro comparison (p < 0.05), and Wilcoxon signed-rank test and Mann-Whitney U test for in vivo analysis (p < 0.01). Results: In vitro study, Xenetix 350 + 5.25% NaOCl facilitated a significant higher visualization (p < 0.05). For in vivo study, CERS had a statistically significant depth of ingress (p < 0.01), and was efficient in identifying the aberrant root canal anatomy of premolars and molars. Conclusions: CERS facilitates better visualization of the root canal anatomy of human premolars and molars.
The aim of this in vitro study was to analyze the composition of human tooth enamel in terms of three components, Ca, P, and F after demineralization and remineralization in acid buffer solution. A total of 8 human premolars without any defects and cracks were selected and buccal and lingual sides of the teeth were cleaned with an ultrasonic device and pumice without fluoride 5$\times$5mm windows were opened, and other areas were completely covered with 3-coats of nail varnish to prevent from being in contact with demineralized and remineralized solutions. After demineralization process, each tooth was sectioned into two slices, highly polished one of them with$\gamma$-alumina, and then analyzed the composition of the demineralized tooth with EPMA(electron probe micro-analyzer). The other slices were put into the remineralized solution for 10 days, polished, and analyzed in the same manner. These data were statistically analyzed with one sample t-test(p<0.05). The results were as follows. 1. Normal tooth enamel consists of 49.76% Ca, 39.80% P, and 0.28% F. 2. After demineralization, percentage of Ca and P ratio were decreased by about 5.57 and 5.07% respectively. Percentage of F ratio was also decreased by about 0.01%, which was not statistically significant. 3. After remineralization, percentage of Ca, P increased about by 4.47 and 4.35% respectively Percentage of F decreased by about 0.01%, which was not statistically significant. In conclusion, remineralized solution used in our study has the potential to induce the uptake the Ca and P into the pore sites of the demineralized enamel. But, in the oral cavity. there were rapid temperature change, organic matrix that inhibits the movement of the ions, and limitation of continuous contact with this remineralized solution. Therefore, further in vivo study is necessary.
This study was designed in order to investigate the effect of copper sulfate to the ultrastructural changes of the hepatocytes in Sprague Dawley rats. The animals were administered with copper sulfate (10mg/kg B.W.), which was dissolved in normal saline. The solution was injected into abdominal cavity every day. The animals were sacrificed at the 6th, 12th, and 24th day from the beginning of administration. The specimens obtained from the liver were observed with electron microscope and significant changes were as follows. 1. A prominent dilatation and disruption of the cisternae of rough endoplasmic reticulum were recognized. Also, the detachment of membrane bound ribosomes was shown. 2. The proliferation of smooth endoplasmic reticulum and the depletion of glycogen particles were noted. 3. The increase of primary lysosomes and autophagic vacuoles was obserbed. 4. The dilatation of mitochondrial cristae was obserbed. And it was irregulary scattered in the stroma of mitochondria. 5. The atrophy of microvilli in the bile canaliculi and space of Disse was prominent. 6. Membrane of hepatocytes was damaged and significant hydrophic degeneration was obserbed in the perisinusoidal regions. 7. The damage of Fat-storing cells was more significant than that of hepatocytes.
