The Effect of Seed-borne Mycoflora from Sorghum and Foxtail Millet Seeds on Germination and Disease Transmission

  • The seed-borne mycoflora of sorghum and foxtail millet collected from different growing areas in South Korea were isolated and taxonomically identified using dry inspection, standard blotter and the agar plate method. We investigated the in vitro and in vitro germination rates of disinfected and non-disinfected seeds of sorghum and foxtail millet using sterilized and unsterilized soil. The percent recovery of seed-borne mycoflora from the seed components of sorghum and foxtail millet seeds was determined and an infection experiment using the dominant species was evaluated for seedling emergence and mortality. A higher number of seed-borne fungi was observed in sorghum compared to that of foxtail millet. Eighteen fungal genera with 34 fungal species were identified from the seeds of sorghum and 13 genera with 22 species were identified from the seeds of foxtail millet. Five dominant species such as Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme and Phoma sp. were recorded as seed-borne mycoflora in sorghum and 4 dominant species (Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme) were observed in foxtail millet. The in vitro and in vitro germination rates were higher using disinfected seeds and sterilized soil. More seed-borne fungi were recovered from the pericarp compared to the endosperm and seed embryo. The percent recovery of seed-borne fungi ranged from 2.22% to 60.0%, and Alternaria alternata, Curvularia lunata and 4 species of Fusarium were isolated from the endosperm and embryo of sorghum and foxtail millet. Inoculation of the dominant seed-borne fungi showed considerable mortality of seedlings. All the transmitted seed-borne fungi might well be a primary source of infection of sorghum and foxtail millet crops.

  • Disease is the major problem in ginseng cultivation from seed stratification, soil preparation prior to planting, right through to drying of the roots. There are many soil-borne disease pathogen in rhizosphere soil environment, furthermore occurrence of diseases by a diverse group of fungi and related organisms are closely related to various soil condition. Observable symptoms for soil-borne diseases include wilting, leaf death and leaf fall, death of branches and limbs and in severe cases death of the whole plant. The fungus Cylindrocarpon destructans is the cause of root rot characterized by a decay of the true root system in many ginseng production areas in Korea. Some pathogens are generally confined to the juvenile roots whilst others are capable of attacking older parts of the root system. However, the relation between the soil environmental characteristics and ginseng root rot by soil-borne disease pathogen is not clearly identified in ginseng field. In this paper, we reviewed soil-borne plant pathogen causing root rot disease of ginseng with respect to soil environment.

Effects of ectomycorrhizal fungi on soil-borne plant pathogenic fungi in red pine seedlings

  • Disease suppression by ectomycorrhizal(ECM) fungi has been demonstrated on red pine seedlings. Culturing of pathogenic fungi on petri plates containing culture filtrates of ECM fungi showed that culture filtrates of the ECM fungus Hebeloma cylindrosporum may inhibit the mycelial growth of all tested soil-borne plant pathogenic(SBPP) fungi upto 60%, In order to examine the effects of ECM fungi on SBPP fungi and on red pine seedlings, both symbiotic and pathogenic fungi were inoculated into the soil with red pine seedlings by three inoculation methods; pre-inoculation of SBPP fungi 10 days before inoculation of ECM fungi, simultaneous inoculation of both fungi, post-inoculation of SBPP fungi 60 days after inoculation of ECM fungi. Seedling mortality, seedling growth, and ectomycorrhizal formation by the combined treatments were examined and compared. Pine seedlings were dead by the pre-inoculation of pathogenic fungi, except Rhizina undulate which required 9-12 days, within 6 days after inoculation. Among pathogenic fungi tested, Fusarium oxysporum was the most pathogenic with the mortality of 44%. However, no dead seedlings were shown by simultaneous inoculation of both fungi or pre-inoculation of ECM fungi. In addition, pine seedlings treated by simultaneous or post-inoculation of SBPP fungi were relatively higher than those treated by pre-inoculation in diameter at root crown and the number of ectomycorrhizal roots. There were no significant differences among inoculation methods in root length and dry weight of treated seedlings. It means that ECM fungi somehow play a role in protecting primary roots of red pine seedlings against invasion by the SBPP fungi.

Effects of Solar Heating for Control of Pink Root and Other Soil-borne Diseases of Onions

  • These experiments were carried out to examine efficacy of soil solarization for control of pink root disease by means of mulching with transparent polyethylene sheets in the hot season. The effects of soil solarization on incidence of pink-root disease caused by Pyrenochaeta terrestris and on onion growth and on populations of soil fungi were investigated. Solarization was dramatically effective in reducing pink root incidence in onion seedling and harvested onion bulb. A 30-day and 40-day solarization treatment significantly improved seedling survival and increased yield of 'Changnyeong-deago' onion while decreasing incidence of pink root. Populations of soil fungi from fields planted to onion were assayed on selective media. Solarization treatment was effective in reducing populations of P. terrestris, Pythium spp., and Rhizoctonia sp. in soil. Increase of yield of onion bulbs was associated with control of soil-borne pathogenic fungi. Soil solarization had beneficial effects on yield, bulb diameter, or incidence of pink root.

Isolation and Structure Elucidation, Molecular Docking Studies of Screlotiumol from Soil Borne Fungi Screlotium rolfsii and their Reversal of Multidrug Resistance in Mouse Lymphoma Cells

  • A new compound namely (13-(3,3-dihydroxypropyl)-1,6-dihydroxy-3,4-dihydro-1H-isochromen-8(5H)-one (1) was isolated from an ethyl acetate extract of the borne fungi Screlotium rolfsii. Its chemical structure was elucidated by spectroscopic analysis. Screlotiumol 1 were evaluated for their effects on the reversion of multidrug resistant (MDR) mediated by P-glycoprotein (P-gp) of the soil borne fungi. The multidrug resistant P-glycoprotein is a target for chemotherapeutic drugs in cancer cells. In the present study rhodamine-123 exclusion screening test on human mdr1 gene transfected mouse gene transfected L5178 and L5178Y mouse T-cell lymphoma which showed excellent MDR reversing effect in a dose dependent manner against mouse T-lymphoma cell line. Moreover, molecular docking studies of compound-1 also showed better results as compared with the standard. Therefore the preliminary results obtained from this study suggest that screlotiumol 1 could be used as a potential agent for the treatment of cancer.

Effect of Bacterial Wilt on Fungal Community Composition in Rhizosphere Soil of Tobaccos in Tropical Yunnan

  • Bacterial wilt, which is a major soil-borne disease with widespread occurrence, poses a severe danger in the field of tobacco production. However, there is very limited knowledge on bacterial wilt-induced microecological changes in the tobacco root system and on the interaction between Ralstonia solanacearum and fungal communities in the rhizosphere soil. Thus, in this study, changes in fungal communities in the rhizosphere soil of tobaccos with bacterial wilt were studied by 18S rRNA gene sequencing. The community composition of fungi in bacterial wilt-infected soil and healthy soil in two tobacco areas (Gengma and Boshang, Lincang City, Yunnan Province, China) was studied through the paired comparison method in July 2019. The results showed that there were significant differences in fungal community composition between the rhizosphere soil of diseased plants and healthy plants. The changes in the composition and diversity of fungal communities in the rhizosphere soil of tobaccos are vital characteristics of tobaccos with bacterial wilt, and the imbalance in the rhizosphere microecosystem of tobacco plants may further aggravate the disease.

Development of a Selective Medium for the Fungal Pathogen Cylindrocarpon destructans Using Radicicol

  • The soil-borne ascomycete fungus Cylindrocarpon destructans causes ginseng root rot disease and produces various secondary metabolites such as brefeldin A and radicicol. The slow growth of this fungus compared with other plant pathogenic and saprophytic fungi in soil disturbs isolation of this fungus from soil and infected ginseng. In this study, we developed a selective medium for C. destructans using radicicol produced by this fungus. Supplementing 50 mg/L of radicicol to medium inhibited the mycelia growth of other fungi including Botrytis cinerea, Rhizoctonia solani and Alternaria panax, but did not affect the growth of C. destructans. In addition, conidia germination of other fungal species except for C. destructans was inhibited in submerged culture supplemented with radicicol. This medium provides a very efficient tool for isolating C. destructans and also can be used as an enrichment medium for this fungus.

국내에 분포하는 Sordariales목 균에 관한 보고 (New Report on Sordarialean Fungi in Korea)

  • 토양 희석방법을 이용하여 토양에 분포하는 곰팡이에 대한 분포 조사를 실시하였다. 토양 시료는 경상남도 진주시에 분포하는 머스크멜론 시설 재배지에서 채취하였으며 총 7종의 자낭균을 분리, 동정하였다. 이들 중 Sordariales목에 속하는 5종의 균을 국내에 처음 보고하며 이들의 형태적 특징을 기재하였다. 이들 균은 완전세대와 불완전세대를 동시에 가지는 Corynascus sepedonium과 Petriella sordida와 완전세대만을 가지는 Thielavia fragilis, Chaetomium indicum과 Chaetomium brasiliense이다. 본 논문에 기재된 균들은 농업과학기술원 농용미생물 보존센타에 보관하였다.

Characterization of Soil Microorganism from Humus and Indigenous Microorganism Amendments

  • This study was conducted to understand the dynamics of microbial communities of soil microorganisms, and their distribution and abundance in the indigenous microorganisms (IMOs) manipulated from humus collected from the forest near the crop field. The soil microorganisms originated from humus and artificially cultured microbial-based soil amendments were characterized by molecular and biochemical analyses. The bacterial population (2 × 106~13 × 106 CFU/g sample) was approximately 100-fold abundant than the fungal population (2 × 104~8 × 104 CFU/g sample). The 16S rDNA and ITS sequence analyses showed that the bacterial and fungal communities in humus and IMOs were mainly composed of Bacillus and Pseudomonas, and Trichoderma and Aspergillus species, respectively. Some of the bacterial isolates from the humus and IMOs showed strong inhibitory activity against soil-borne pathogenic fungi Fusarium oxysporum and Sclerotinia sclerotiorum. These bacteria also showed the siderophore production activity as well as phosphate solubilizing activity, which are requisite traits for biological control of plant pathogenic fungi. These results suggest that humus and IMOs could be a useful resource for sustainable agriculture.

한국(韓國)에 있어서 태양열(太陽熱)을 이용(利用)한 토양소독(土壤消毒)의 가능성(可能性) (Possibility of Soil Solarization in Korea)

  • 1. 시설(施設)하우스내(內)에서 피해(被害)가 큰 토양전염성병원균(土壤傳染性病原菌) Fusarium oxysporum f. sp. lycopersici, Fusarium oxysporum f. sp. niveum, Rhizoctonia solani, Sclerotinia sclerotiorum, Sclerotium rolfsii, Pythium-debaryanum 6종(種)에 대(對)한 살균(殺菌)에 요(要)하는 처리시간(處理時間)은 $45^{\circ}C$에서 $2{\sim}10$일간(日間), $50^{\circ}C$에서는 4일이내(日以內)이었고 이 중 가장 내성(耐性)이 강한 것은 Fusarium균(菌)과 Pythium균(菌)이었다. 2. 7월(月) 28일(日)${\sim}$8월(月) 18일(日)까지 21일간(日間) house내(內) 일중(日中) 온도변화(溫度變化)는 5cm, 15cm 깊이에서 각각 최고(最高) $57^{\circ}C,\;47^{\circ}C$ 최저(最低) $38^{\circ}C,\;40^{\circ}C$이었고 15cm 깊이에서의 $43^{\circ}C$이상(以上) 지속기간은 12시간정도(時間程度)였다. 3. 턴널은 하우스보다 축열효과(蓄熱效果)가 더 좋았으며 멀칭은 멀칭하지 않은 것보다 $3{\sim}7^{\circ}C$정도(程度)의 승온효과(昇溫效果)가 있었고 단열구(斷熱區)가 무단열구(無斷熱區)보다 $5{\sim}9^{\circ}C$정도(程度) 승온효과(昇溫效果)가 좋았다. 4. 약(約) 60%정도(程度)의 관수(灌水)를 하여준 구(區)에서는 $1{\sim}3^{\circ}C$정도(程度)가 높았다. 5. 매몰병원균(埋沒病原菌)의 살균효과(殺菌效果)는 하우스-턴널-단열구(斷熱區)에서 15cm 깊이까지는 처리(處理) 7일간(日間)에 사멸(死滅)했고 20cm 깊이에서는 Pythium균(菌)과 Fusarium균(菌)을 제외(除外)하고 모두 14일간(日間)에 사멸(死滅)하였다. 6. 최고(最高)에 달(達)했다가 최저(最低)에 이르는 온도(溫度)의 반복적(反復的)인 파동(波動)이 있는 것에서 없는 것보다 단시간(短時間)에 사멸(死滅)하였다. 이상(以上)의 결과(結果)에서 볼 때 하기고온기(夏期高溫期)의 재배(栽培) 휴한기(休閑期)에 PVC film을 사용(使用)하여 하우스를 밀폐(密閉)하거나 턴널을 만들어서 태양열(太陽熱)을 이용(利用)한 토양소독(土壤消毒)의 가능성(可能性)이 충분(充分)히 있는 것으로 생각된다.
