Breeding of inbred lines 'Wongyo 3115' and 'Wongyo 3116' in Strawberry (딸기 근교계통 '원교3115호'와 '원교 3116호' 육성)

  • Fragaria ${\times}$ ananassa (Duch.) cultivars, 'Wongyo 3 115' a nd 'Wongyo 3 116', we re bre d as a n inte rme diate parent l ine using heterosis of $F_1$ hybrids in octoploid strawberry. These two new cultivars were developed as inbred lines ($S_9$ generation) propagated from both seed and runners/stolons. 'Wongyo 3115', an inbred line derived from the 'Benihoppe' cultivar, has a semi-upright plant shape showing a vigorless growth habit, weaker than the original 'Benihoppe' cultivar; it has early flower bud differentiation and produces 10 flowers per cluster when grown from healthy seed. 'Wongyo 3115' has low yields of conical type fruit with pink colored skin; average fruit weight is approximately 11 g. The important characteristic of 'Wongyo 3115' is its excellent fruit firmness, firmer than the original variety. 'Wongyo 3116', an inbred line derived from the 'Doyonoka' cultivar, has an upright plant shape, vigorless growth habit, weaker than that of the original 'Doyonoka' cultivar; it has early flower bud differentiation and produces 8 flowers per cluster when grown from healthy seed. 'Wongyo 3 116' has oblate type fruits with red skin and an ave rage weight of approximately 12 g. The important characteristics of 'Wongyo 3116' are excellent fruit shape and higher yield than other inbred lines, although it produces small fruits compared to the original cultivar. Both cultivars have excellent taste, high sugar/acid ratio, and good texture. In regards to disease and pest resistance, 'Wongyo 3115' and 'Wongyo 3116' have a tendency to be sensitive to powdery mildew, anthracnose and the two-spotted spider mite.

Antioxidant Activities of Perilla frutescens Britton Seed Extract and Its Inhibitory Effects against Major Characteristics of Cancer Cells (들깨 추출물의 항산화 활성과 암세포 기본 특성에 대한 억제 효과)

  • The aim of the study was to investigate the antioxidant activities of ethanol extract of perilla seed (PSE) and its inhibitory effects against major characteristics of cancer cells, such as unrestricted growth and activated metastasis in vitro. The total polyphenol and flavonoid levels of PSE were 222.6 mg gallic acid equivalent/100 g and 285.7 mg quercetin equivalent/100 g, respectively. The radical scavenging activity and ferric reducing antioxidant power of PSE at concentration of 87.5 to $350{\mu}g/mL$ were 24~45% and 28~62%, respectively. Treatment of HCT116 colorectal carcinoma cells and H1299 non-small cell lung carcinoma cells with PSE dose-dependently inhibited growth by 18~94% (at concentration range of 87.5 to $350{\mu}g/mL$) and completely abolished colony formation (at concentration of $175{\mu}g/mL$). PSE was also effective in inhibiting migration of H1299 cells (by 30~37% at concentration range of 87.5 to $350{\mu}g/mL$) and adhesion of both HCT116 and H1299 cells (by 14~16% at concentration of $350{\mu}g/mL$). These results indicate that PSE exerts antioxidant and anti-cancer activities in vitro. It needs to be determined whether or not similar effects can be reproduced in vivo.

  • (1) Relatively active polyphenol oxidase influence was seen at $0{\circ}C$, and the optimal pH level for the enzyme from the seeds of a small type sweet pepper Zairaisisi is 6.5. (2) The starting stage of the brown coloration with low-temperature injuries showed a strong activity of polyphenol oxidase, and the activity drops to 0 as the entire seed became brownish. (3) The browning effect with enzyme solution of polyphenol extracts suggested that the brown coloration continues in vitro even if polyphenol oxidase activity is nil. (4) Although cytochrome oxidase activity dropped when an abnormality occurrs in electron pathways of respiration at the starting stage of the browning with low temperature injuries, there was no marked influence of it on the total respiration, indicating the fact that polyphenol oxidase can take place of terminal oxidase in the compensatory respiration process.

A Taxonomic Review of Phytolacca insularis (Phytolaccaceae) (섬자리공(Phytolacca insularis, 자리공과)의 분류학적 재검토)

  • The purpose of this study is to review the taxonomical position of Phytolacca insularis Nakai based on morphological characters and ITS sequences. Phytolacca insularis was similar to P. acinosa in the stem shape, inflorescence, apocarpous, eight pistils, eight stamens, pink anther, and eight fruits. But the unique difference between P. insularis and P. acinosa was leaf size and pistil color. Phytolaccri insularis and P. acinosa have a similar sculpturing patterns as an foveolate and microscabrate in pollen, but they differed from P. americana in having a bigger foveolae size in sculpture pattern. Phytolacca insularis and P. acinosa were similar in the seed size but different from P. americana by having a small size of seed. The seed surface of P. insularis and P. acinosa was similar with a irregularly undulate shape, but its feature of P. americana differed in having the reticulate shape. Phytolacc a insularis had identical ITS sequences with P. acinosa. Phytolacca insularis and P. acinosa have a close relationship in the morphological characters and ITS sequences. The examinations of morphology and ITS sequences suggest that P. insularis be a form of P. acinosa although it has been formerly treated as a distinct species.

Addition Effect of Seed-associated or Free Linseed Oil on the Formation of cis-9, trans-11 Conjugated Linoleic Acid and Octadecenoic Acid by Ruminal Bacteria In Vitro

  • The effects of seed-associated or free linseed oil on fermentation characteristics and long-chain unsaturated fatty acids composition, especially the formation of conjugated linoleic acid (CLA) and octadecenoic acid (trans-11 $C_{18:1}$, $t-C_{18:1}$) by mixed ruminal bacteria were examined in vitro. Concentrate (1% of culture solution, w/v, as-fed basis) with ground linseed (0.6% of culture solution, w/v, DM basis) or linseed oil as absorbed onto ground alfalfa hay was added to 600 ml mixed solution consisting of strained rumen fluid and artificial saliva at the ratio of 1:1 in a glass culture jar. The culture jar was covered with a glass lid with stirrer, and placed into a water-bath ($39^{\circ}C$) and incubated anaerobically up to 24 h. Seed-associated or free linseed oil did not significantly affect the pH and ammonia concentration in the culture solution. Molar percent of acetate tended to increase while that of propionate decreased with the addition of free oil treatment throughout the incubation. Differences in bacterial number were relatively small, regardless of the form of supplements. Decreasing trends in the compositions of linoleic acid ($C_{18:2}$) and linolenic acid ($C_{18:3}$) but increasing trends of stearic acid ($C_{18:0}$), $t-C_{18:1}$ and CLA compositions were found from culture contents up to 12h incubation when incubated with both ground linseed and linseed oil. The compositions of $C_{18:0}$, $C_{18:2}$ and $C_{18:3}$ were greater but those of oleic acid ($C_{18:1}$), $t-C_{18:1}$ and CLA were smaller in a culture solution containing ground linseed than those containing linseed oil. The ratio of $t-C_{18:1}$ to CLA was lower in the culture solutions containing linseed oil up to 12h incubations as compared to those containing ground linseed.

Effect of Dietary Supplementation of Ground Grape Seed on Growth Performance and Antioxidant Status in the Intestine and Liver in Broiler Chickens (천연 항산화제로서 포도씨 분말 첨가가 육용계의 성장 및 항산화 작용에 미치는 영향)

  • A total of twenty, 2-wk-old male broiler chickens were allotted into control diet(CON) or a diet supplemented with 1% ground grape seed(GGS). They had free access to feed and water for 3 wk. Growth performance and antioxidant markers in plasma, intestine and liver were determined. Dietary addition of 1% GGS did not affect weight gain, feed intake, feed conversion and organ weight in 35 day-old broiler chickens significantly. There was no difference in plasma levels of glucose, triglyceride, cholesterol, AST, ALT and LDH activity. However, total antioxidant status(TAS) in blood increased(P<0.05) in chickens fed the diet supplemented with 1% GGS compared to those fed the control diet. In addition, the specific activity of intestinal superoxide dismutase(SOD) increased(P<0.05) in birds fed the diet supplemented with GGS. However, the activities of intestinal gluthathione peroxidase(GSHPx) and gluthathione -S- transferase(GST) and hepatic SOD, GSHPx and GST were not affected by the dietary GGS. The levels of reduced glutathione and lipid peroxidation in the small intestine and liver were not different between the two groups. In conclusion, dietary supplementation of 1% GGS did not result in a negative effect on growth performance. In addition, some antioxidant indicators including blood TAS and intestinal SOD were markedly elevated in response to dietary GGS. Therefore, dietary addition of 1% GGS may be beneficial to improve antioxidant capacity in broiler chicken.

Floral Characteristics and Cross Compatibility of Collected Paeonia lactiflora PALL (작약(芍藥) 수집종(蒐集種)의 화기특성(花器特性)과 교잡친화성(交雜親和性))

  • In order to obtain the basic information on cross breeding of peony, the growth and floral characteristics were investigasted for local collected strains in main cultvation area in Korea. Local peony strains were classified by early emergence lines and late emergence lines according to the emergence date. Flower color of single-petaling flower was varied with red, pink and white, on the other hand, red+red, pink+white, pink+red and pink+pink in double-petaling flower. Number of pistil of the most local peony strains was 3-4/flower, number of stamen was varied from 0 to more than 200 ea/flower, classifing by 4 groups according to the diameter of flower. Most of the cross combinations showed the cross comptibility, and number of follicle was 3-5 ea/flower, average number of seed was 15.6 ea/follicle, seed size was grouped : by small grains with less than 10g, medium grains with 10-15g and large grains with more than 15g of 100 seed weight.

Preparation and Pervaporative Alcohol Dehydration of Crystallographically b/c-axis Oriented Mordenite Zeolite Membranes (결정학적으로 b/c-축 방향으로 배향된 모데나이트 제올라이트 분리막의 제조 및 투과증발 알코올 탈수 거동)

  • In the present study, crystallographically b- and c-axis oriented mordenite zeolite membranes were prepared and their pervaporative ethanol dehydration was investigated. The seed layer with a high coverage grew to be c-axis oriented dense layer, while the seed layer with a low coverage grew to be b-axis oriented layer. This phenomenon could be explained by the evolutionary selection growth mechanism. The b-axis grown membrane with 8-membered rings showed a high separation factor of above 1000 and a considerable total flux of around $0.2kg/m^2h$. The c-axis grown, columnar structured membrane with 8- and 12-membered rings showed a low separation factor of less than 200 and a relatively high total flux of around $0.25kg/m^2h$. The high performance of b-axis grown membrane was due to the relatively small opening of 8-membered rings. Water molecules can freely permeate through the openings, but ethanol molecules, difficultly. Therefore, in the present study, we introduced a new method to control crystallographic orientation of mordenite membrane by changing seeding amount of needle-like crystals, and elucidated that b-axis oriented mordenite membrane showed better performance than c-axis grown mordenite membrane.

The Effect of Polyphenols from Safflower Seed on HMG-CoA Reductase (HMGR) Activity, LDL Oxidation and Apo A1 Secretion (홍화씨 폴리페놀이 HMG-CoA reductase, LDL 산화 및 Apo A1 분비에 미치는 영향)

  • This study was conducted to examine the effect of polyphenols from safflower seed on HMG-CoA reductase (HMGR) activity, LDL oxidation and Apo A1 secretion from Hep3B cell. The safflower seed polyphenols were matairesinol (Iignan), enterolactone (lignan metabolite), acacetin (flavone) and serotonin derivative. In addition to safflower polyphenols, mevastatin, ${\alpha}-estradiol,\;{\alpha}-tocopherol$ and soy genistein were tested as reference compounds depending on the type of the test. HMGR source was liver microsome obtained from rat fed 2% cholestyramine for 10 days. Inhibition of HMGR activity was greater with mevastatin (53%) than safflower serotonin derivatives (45%), followed by genistein (35%), but was very small with matairesinol, enterolactone and acacetin. LDL oxidation induced by $CuSO_4$ was suppressed by all the test material used in the present study and in the order of safflower serotonin derivatives> matairesinol > ${\beta}-estradiol$ > genistein > acacetin > enterolactone. Apo A1 secretion from Hep3B cell was significantly stimulated by mevastatin, but moderately (p<0.1) by ${\beta}-estradiol$ and genistein as well as enterolactone. These results suggest that the safflower polyphenols improve body lipid status via inhibition of cholesterol synthesis and suppression of LDL oxidation.

Effect of Chemical Treatments and Electric and Magnetic Field Treatments on Germination of Onion Seeds (화학적 처리와 전장 및 자기장 처리가 양파 종자의 발아에 미치는 영향)

  • This study was conducted to investigate the effects of chemical treatments and electric and magnetic field treatments on germination and seedling growth of onion (Allium cepa L.). Germination ratios of the onion seed were not much different among the tested species. However, germination ratio was much higher in species of small and medium size seed than that of large one. Moisture content of the seeds was shown to be more suitable in the range of 10-15% for seed germination compare to that of 20%. The germination ratio of onion seeds was found to be increased by the chemical treatments in the order of GA<$NaNO_3$<$KNO_3$. In particular, the treatments with 0.1 and 0.2% $KNO_3$ increased the germination ratio by 13 and 15% compared to that of the non-treatment. The seeds treated with electric or magnetic field resulted in much higher germination ratio and better initial growth. Germination ratios of 86 and 88% were found in the 10 kV and 4 Gauss treatments, respectively. The length and weight of the seedlings grown for 40 days after transplanting were increased by 23-45% with 8 kV and 12 Gauss treatments compared to those of non-treatment.
