Approximately 200 individual skeletal elements, which differ in shape and size, are the building blocks of the vertebrate skeleton. Various features of the individual skeletal elements, such as their location, shape, growth and differentiation rate, are being determined during embryonic development. A few skeletal elements, such as the lateral halves of the clavicle and parts of the skull are formed by a process called intramembranous ossification, whereby mesenchymal cells differentiate directly into osteoblasts, while the majority of skeletal elements are formed via endochondral ossification. The latter process starts with the formation of a cartilaginous template, which eventually is being replaced by bone. This requires co-regulation of differentiation of the cell-types specific for cartilage and bone, chondrocytes and osteoblasts, respectively. In recent years it has been demonstrated that Wnt family members and their respective intracellular pathways, such as non-canonical and the canonical $Wnt/{\beta}$-catenin pathway, play important and diverse roles during different steps of vertebrate skeletal development. Based on the recent discoveries modulation of the canonical Wnt-signaling pathway could be an interesting approach to direct stem cells into certain skeletal lineages.
Morphological and skeletal development in the cultured threeline grunt Parapristipoma trilineatum was studied from 1.96 to 61.39 mm notochord length (NL) or standard length (SL) specimens (n=360). After two days post-hatching (dph, 2.13 mm NL), the mouth opened, and melanophores formed in the eyes and egg yolk were absorbed. Oli globules were absorbed at 3 dph (2.71 mm NL). The total number of fin rays reached that of adults at 28 dph (10.58 mm SL). During skeletal development, the premaxillaray, dentary, opercle, preopercle, and cleithrum first began to ossify at 6 dph (3.51 mm NL). Ossification of all skeletal elements was completed at 28 dph (14.57 mm SL). The vertebrae began to ossify at 17 dph (5.63 mm SL) and were completed at 28 dph (8.48 mm SL), progressing from cranium to caudal bone. The centrum ossified from the dorsal to ventral direction. Vertebral fusion and separation were found to be common skeletal abnormalities.
Objective: The aim of this study was to establish the normative data of dentofacial transverse dimensions according to the skeletal maturation stage in Korean adolescents with good occlusion, assess gender differences and determine correlations between transverse variables. Methods: A total of 577 Korean subjects between ages 7 to 19 years and exhibiting skeletal Class I occlusion were categorized by skeletal maturation index (SMI) of Fishman using hand-wrist radiographs. Dentofacial transverse dimensions were assessed using posteroanterior cephalograms. Independent two-sample t-tests were used to analyze differences between genders. Pearson correlation coefficient was used to determine the correlation between transverse measurements. Results: Dentofacial transverse norms relevant to skeletal maturation stages were established. The average maxillomandibular width difference and ratio at growth completion was 22.16 mm and 77.01% for males; 23.70 mm and 74.06% for females, respectively. Males had greater facial, maxillary and mandibular widths compared to females at every SMI stage. The maxillary and mandibular intermolar widths showed the strongest correlation for both sexes (r = 0.826 for males, r = 0.725 for females). Conclusions: Dentofacial transverse norms of Korean adolescents were established according to developmental stage. All dentofacial widths were greater in males at growth completion. Maxillary and mandibular intermolar widths were strongly correlated. This study may serve as a guideline for the assessment of dentofacial transverse growth according to skeletal maturation stage in Korean adolescents with good occlusion.

Insulin resistance is commonly observed in patients prior to the development of type 2 diabetes and may predict the onset of the disease. We tested the hypothesis that impairment in insulin stimulated glucose-disposal in insulin resistant patients would be reflected in the gene expression profile of skeletal muscle. We performed gene expression profiling on skeletal muscle of insulin resistant and insulin sensitive subjects using microarrays. Microarray analysis of 19,000 genes in skeletal muscle did not display a significant difference between insulin resistant and insulin sensitive muscle. This was confirmed with real-time PCR. Our results suggest that insulin resistance is not reflected by changes in the gene expression profile in skeletal muscle.
Skeletal class III had been classified by the position of the maxilla, the mandible, the maxillary alveolus, the mandibular alveolus and vertical development. This morphologic approach is simple and useful for clinical use, but it is insufficient to permit understanding of the pathophysiology of dysmorphoses. The author hypothesizes that there are different patterns of mutual relation of the skeletal components which have contributed pathologic equilibrium of skeletal class III. The purpose of this study are threefold: 1) to classify skeletal class III in subgroups, which can show the architectural characteristics of the deformity, 2) to analyse the craniofacial architecture of each subgroup on etio-pathogenic basis, and 3) to characterize and visualize the pattern as a prototype. Materials used in this study were lateral cephalograms of 106 skeletal class III adults, which were analysed with modified Delaire's architectural and structural analysis. Linear and angular measurements of the individual subject were obtained and cluster analysis was used for the subgrouping. Data were evaluated for verification of the statistical significances. The following results were obtained. 1. By the modified Delaire's architectural and structural analysis and cluster analysis, skeletal class III adults were classified into 7 clusters and presented as prototypes, which could show the pathophysiology of the skeletal architecture 2. There was significant relationship in measurement variables of each cluster, which could reflect characteristics of the skeletal pattern of growth. 3. The flexure of cranial base had a close relationship to the anterior rotational growth of the maxilla and contributes to understand the etio-pathology of skeletal class III. 4. The proportion of craniospinal area in cranial depth, craniocervical angle and vertical position of point Om had a close relationship to rotational growth of the mandible and direction of condylar growth. They contribute to understand the etio-pathology of skeletal class III. In summary, the cranium and the craniocervical area must be considered in diagnosis and treatment planning of dentofacial deformity. And the occlusal plane can be considered as a representative which shows the mutual relationships of the skeletal components.

Dexamethasone, a synthetic glucocorticoid (GC), is clinically administered to woman at risk for premature labor to induce fetal lung maturation. However, exposure to repeated or excess GCs leads to intrauterine growth restriction (IUGR) and subsequently increases risk of psychiatric and cardio-metabolic diseases in later life through fetal programming mechanisms. GCs are key mediators of stress responses, therefore, maternal nutrient restriction or psychological stress during pregnancy also causes negative impacts on birth and neurodevelopment outcome of fetuses, and other congenital defects, such as craniofacial and skeletal abnormalities. In this study, to examine the effect of prenatal stress on fetal skeletal development, dexamethasone (1 mg/kg [DEX1] or 10 mg/kg [DEX10] maternal body weight per day) was administered intraperitoneally at gestational day 7.5~9.5 and the skeletons were prepared from embryos at day 18.5. Seven out of eighteen (39%) embryos treated with DEX10 showed axial skeletal abnormalities in either the T13 or L1 vertebrae. In addition, examination of the sternum revealed that xiphoid process, the protrusive triangular part of the lower end of the sternum, was bent more outward or inward in DEX group embryos. In conclusion, our findings suggest a possible link to the understanding of the effect of uterine environment to the fetal skeletal features.
The purpose of this study is to investigate the effects of supplementation of mulberry powder, mulberry extract and silkworm powder during the 8 weeks of resistance exercise on muscle increase of rats. Fifty males, Sprague-Dawley rat, were randomly divided into 5 groups: CON(control group, n = 10), REG(resistance exercise group, n = 10), MP REG(mulberry powder intake and resistance exercise group, n = 10), ME REG(mulberry extract intake and resistance exercise group, n = 10) and SP REG(silkworm powder intake and resistance exercise group, n = 10). After climbing the ladder without weights during the 1 week of adaptation period, the rats in the resistance exercise group were trained to climb a 0.98-m vertical(80 degree incline) ladder with weights in their tail during 7 weeks(10 times each day, 2 days per week). After exercise, the skeletal muscle was extracted from the flexor hallucis longus. The absolute weight of skeletal muscle was measured by the electronic balance and the relative weight of skeletal muscle about the weight was calculated. The strength and density of legs muscle were analyzed by the computed tomography scan. After 8 weeks of resistance exercise, the absolute weight of skeletal muscle wasn't significant, but it increased in MP REG 8%, ME REG 8% and SP REG 10% compared to REG. The relative weight of skeletal muscle increased significantly in SP REG compared to REG (p < 0.05). Additionally, there was a significant increase of density and strength of the forelegs in SP REG compared to CON (p < 0.05). However, there was no significant increase of density and strength of the hind legs. In conclusion, regular resistance exercise made rats muscle increase and there was more muscle increase effected by providing silkworm.
The loss of skeletal muscle, called sarcopenia, is an inevitable event during the aging process, and significantly impacts quality of life. Autophagy is known to reduce muscle atrophy caused by dysfunctional organelles, even though the molecular mechanism remains unclear. Here, we have discuss the current understanding of exercise-induced autophagy activation in skeletal muscle regeneration and remodeling, leading to sarcopenia intervention. With aging, dysregulation of autophagy flux inhibits lysosomal storage processes involved in muscle biogenesis. AMPK-ULK1 and the $FoxO/PGC-1{\alpha}$ signaling pathways play a critical role in the induction of autophagy machinery in skeletal muscle, thus these pathways could be targets for therapeutics development. Autophagy has been also shown to be a critical regulator of stem cell fate, which determines satellite cell differentiation into muscle fiber, thereby increasing muscle mass. This review aims to provide a comprehensive understanding of the physiological role of autophagy in skeletal muscle aging and sarcopenia.
The maintenance of skeletal muscle mass is very important for the prevention of life style-related diseases and the improvement of quality of life. It is well-known that resistance exercise and nutrition (especially amino acids) are the most effective interventions for maintaining skeletal muscle mass. It has been reported that many molecules are involved in the regulation of protein synthesis in response to resistance exercise and nutrition. Understanding the molecular mechanisms regulating muscle protein synthesis is crucial for the development of appropriate interventions. The role of intracellular signaling pathways through the mammalian target of rapamycin (mTOR), a serine/threonine protein kinase in the regulation of muscle protein synthesis, has been extensively investigated for these years. Control of protein synthesis by mTOR is mediated through phosphorylation of downstream targets that modulate translation initiation and elongation step. In contrast, upstream mediators regulating mTOR and protein synthesis in response to resistance exercise and amino acid still needed to be determined. In this brief review, we discuss the current progress of intracellular mechanisms for exercise- and amino acid-induced activation of mTOR pathways and protein synthesis in skeletal muscle.
삼세기(Hemitripterus villosus)는 새로운 양식 어종으로 대두되고 있으나, 자원량이 감소하고 있으며, 양식 어종에서 지속적인 문제로 나타나는 골격 기형에 대해 삼세기의 정상적인 골격발달 기준을 제시하고자 한다. 이 연구에 사용된 시료는 어획된 어미로부터 채란된 수정란을 부화시킨 자치어를 포르말린액에 고정한 뒤 Kawamura and Hosoya (1991) 염색법에 따라 염색하여 골격발달을 관찰하였다. 부화 직후 평균전장 13.65±0.71 mm (n=5) 일 때 부설골과 주상악골, 전상악골, 치골이 골화되기 시작하였고, 부화 후 65일째 평균전장 21.24±0.50 mm (n=5) 일 때 사골과 안상골이 골화하면서 두부가 완성되었다. 견대부는 부화 7일째 평균전장 14.61±0.52 mm (n=5) 일 때 쇄골, 주새개골, 관절골이 골화하기 시작하였으며, 부화 44일째 평균전장 18.15±0.61 mm (n=5) 일 때, 4개의 사출골이 골화되어 견대부가 완성되었다. 부화 10일째 평균 전장 14.80±0.65 mm (n=5) 일 때 척추골의 골화가 시작되었으며, 부화 후 54일째 평균전장 18.67±0.54 mm (n=5) 일 때 척추골 추체 39개가 골화하여, 부화 60일째 평균전장 20.25±0.45 mm (n=5) 일 때 척추골의 골격이 완성되었다. 이 연구는 삼세기 자치어의 골격발달에 대한 것으로 삼세기의 자치어 골격 기형의 기준과 기초자료 연구에 사용될 것이다.
