A Cre/loxP-${\delta}$-integration system was developed to allow sequential and simultaneous integration of a multiple gene expression cassette in Saccharomyces cerevisiae. To allow repeated integrations, the reusable Candida glabrata MARKER (CgMARKER) carrying loxP sequences was used, and the integrated CgMARKER was efficiently removed by inducing Cre recombinase. The XYLP and XYLB genes encoding endoxylanase and ${\beta}$-xylosidase, respectively, were used as model genes for xylan metabolism in this system, and the copy number of these genes was increased to 15.8 and 16.9 copies/cell, respectively, by repeated integration. This integration system is a promising approach for the easy construction of yeast strains with enhanced metabolic pathways through multicopy gene expression.
Rapid urbanization, resource depletion, and limited land are further increasing the need for skyscrapers in city centers; therefore, it is imperative to enhance tall building performance efficiency and energy-generative capability. Potential performance improvements can be explored using parametric multi-objective optimization, aided by evaluation tools, such as computational fluid dynamics and energy analysis software, to visualize and explore skyscrapers' multi-resource, multi-system generative potential. An optimization-centered, software-based design platform can potentially enable the simultaneous exploration of multiple strategies for the decreased consumption and large-scale production of multiple resources. Resource Generative Skyscrapers (RGS) are proposed as a possible solution to further explore and optimize the generative potentials of skyscrapers. RGS can be optimized with waste-energy-harvesting capabilities by capitalizing on passive features of integrated renewable systems. This paper describes various resource-generation technologies suitable for a synergetic integration within the RGS typology, and the software tools that can facilitate exploration of their optimal use.
After the US government declared 3D printing technology a next-generation manufacturing technology, there have been many practical studies conducted to expand 3D printing technology to manufacturing technologies, called AMERICA MAKES. In particular, the Keck Center, located at the University of Texas at El Paso, has studied techniques for easily combing the 3D stacking process with space mobility and expanded these techniques to simultaneous staking techniques for multiple materials. Additionally, it developed convergence manufacturing techniques, such as direct inking techniques, in order to produce a module structure that combines electronic circuits and components, such as CUBESET. However, in these studies, it is impossible to develop a unified system using traditional independent through simple sequencing connections. This is because there are many problems in the integration between the stacking modeling of 3D printers and post-machining, such as thermal deformations, the precision accuracy of 3D printers, and independently driven coordinate problems among process systems. Therefore, in this paper, the integration method is suggested, which combines these 3D printers and subsequent machining process systems through an Internet-based cloud. Additionally, the sequential integrated system of a 3D printer, an NC milling machine, machine vision, and direct inking are realized.
A numerical study is carried out on the detonation wave propagation through a T-shaped flame tube, which represents a crucial part of the combustion wave ignition (CWI) system aimed for simultaneous ignition of multiple combustion chambers by delivering detonation waves. The formulation includes the Euler equations and an induction-parameter model. The reaction rate is treated based on a chemical kinetics database obtained from a detailed chemistry mechanism. A second-order implicit time integration and a third-order TVD algorithm are Implemented to solve the theoretical model numerically. A total of more than two-million grid points are used to provide direct insight into the dynamics of the detonation wave. Several important phenomena including detonation wave propagation, degeneration, and re-initiation are carefully examined. Information obtained can be effectively used to facilitate the design and optimization of the flame tubes of CWI systems.
Future microcellular systems will require distributed network control. A packet-switched network is suitable for this requirement. The packet reservation multiple access(PRMA) is a Reservation-ALOHA like protocol for wireless terminals to transmit packet speech to a base station. It allows spatially distributed users in cellular systems to transmit packeted voice and data to a common base station using a shared channel. In the existing PRMA, the problem is that the voice packets may collide with the data packets due to simultaneous channel access. the problem may be a major performance degradation factor to a voice and data mixed system. We propose a new PRMA method that integrates voice and data traffic efficiently by resolving the collision problem between data and voice packets. The proposed PRMA method gives a performance improvement than the existing PRAMA method in terms of voice packet dropping probability and data delay characteristic. From analytic results, we can confirm that the proposed PRMA method show a performance improvement than the existing PRMA protocol.
Distributed energy resources (DERs) are essential for coping with growing multiple energy demands. A microgrid (MG) is a small-scale version of the power system which makes possible the integration of DERs as well as achieving maximum demand-side management utilization. Hence, this study focuses on the analysis of optimal power dispatch considering economic aspects in a multi-carrier microgrid (MCMG) with price-responsive loads. This paper proposes a novel time-based demand-side management in order to reshape the load curve, as well as preventing the excessive use of energy in peak hours. In conventional studies, energy consumption is optimized from the perspective of each infrastructure user without considering the interactions. Here, the interaction of energy system infrastructures is considered in the presence of energy storage systems (ESSs), small-scale energy resources (SSERs), and responsive loads. Simulations are performed using GAMS (General Algebraic modeling system) to model MCMG, which are connected to the electricity, natural gas, and district heat networks for supplying multiple energy demands. Results show that the simultaneous operation of various energy carriers, as well as utilization of price-responsive loads, lead to better MCMG performance and decrease operating costs for smart distribution grids. This model is examined on a typical MCMG, and the effectiveness of the proposed model is proven.
The extent and depth of the event plan determines the scope of pedagogical experience in situations and consequently the quality of immersive learning based on our simulated world. In contrast to planning in conventional narrative-based systems mainly pursuing dramatic interests, planning in virtual world-based pedagogical systems strive to provide realistic experiences in immersed situations. Instead of story plot comprising predetermined situations, our inter-event planning method aims at simulating diverse situations that each involve multiple events coupled via their associated agents' conditions and meaningful associations between events occurring in a background world. The specific techniques to realize our planning method include, two-phase planning based on inter-event search and intra-event decomposition (down to the animated action level); autonomous and independent agents to behave proactively with their own belief and planning capability; full-blown background world to be used as the comprehensive stage for all events to occur in; coupling events via realistic association types including deontic associations as well as conventional causality; separation of agents from event roles; temporal scheduling; and parallel and concurrent event progression mechanism. Combining all these techniques, diverse exogenous events can be derived and seamlessly (i.e., semantically meaningfully) integrated with the original event to form a wide scope of situations providing chances of abundant pedagogical experiences. For effective implementation of plan execution, we devise an execution scheme based on multiple priority queues, particularly to realize concurrent progression of many simultaneous events to simulate its corresponding reality. Specific execution mechanisms include modeling an action in terms of its component motions, adjustability of priority for agent across different events, and concurrent and parallel execution method for multiple actions and its expansion for multiple events.

The purpose of the study was to apply and to expand the six sigma to reduce waiting times for computed tomography (CT) examination which manipulated by the department of radiology. It was preceded by DMAIC (Define, Measure, Analyze, Improve, and Control). In the stage of definition, it wereselected for total 5 critical to quality (CTQ), which were the kindness, the waiting time, the examination explanation, the waiting day and the waiting stand environment, that increased the reserved time of CT examination. In the stage of measurement, the number of examinations and of reservation waiting days performed and resulted in final CTQ(Y) which measured each 1.68 and 1.85 sigma. In the stage of analysis, the examination concentrated on morning time, non-scheduled examination of the day, the delayed time of booking, frequent telephone contacting and equipment malfunction were determined as variable key causes. In the stage of improvement, it were performed with expansion of the examination in the morning time, integration of laboratories that used to in each steps, developing the ability of simultaneous booking schedule for the multiple examinations, developing program of examination request, and the customer management team operations. For the control, the number of examinations and reserved waiting days were measured each 3.14 and 1.13 sigma.
