Studies on Flacherie and Ina-flacherie Viruses of the Silkworm, Bambyx mori II. Some Properties of Polypeptide of Flacherie Virus (가잠의 연화병 바이러스에 관한 연구 II. 연화병 바이러스 Polypeptide의 성상)

  • Purified preparations of flacherie virus capsid protein were fractionated by SDS-polyacrylamide gel electrophoresis, and amino acid composition was determined by amino acid analyzer. Three polypeptide components, FP I, FP II and FP III were detected, and the molecular weights of these components were 37,500, 30,500 and 26,500 respectively. The FP III was major poly-peptide comprised about 68.4% of the total virus capsid protein. Seventeen amino acids were detected by an amino acid analyzer from hydrolyzate of the virus capsid protein and the pattern of amino acid composition was similar to those of several other insect viruses.

Heritability and Repeatability Estimates for Reproductive Traits of Japanese Black Cows

  • Reproductive data collected from more than 20,000 Japanese Black cows of Hyogo and Shimane Prefectures were analyzed. Averages of age at first calving, gestation length, days open and calving interval were 25.1 mo, 289 d, 112 d and 401 d, respectively. Variance components were obtained by REML procedure and the heritability estimate of age at first calving was 0.22. In gestation length the heritability estimate was 0.40 and no permanent environmental effect was estimated. Estimated variance components of calving interval were similar to those of days open and the heritability and repeatability of calving interval were 0.05 and 0.09, respectively. Random farm effects accounted for approximately 10% of phenotypic variations in all traits. Genetic and farm correlations between age at first calving and calving interval were 0.27 and 0.39, respectively. It was found that temporary environment was an important source of variation for calving intervals of Japanese Black.

Longevity regulation by NMD-mediated mRNA quality control

  • Proper maintenance of biological components is crucial for longevity and healthy aging. Although the role of homeostatic maintenance systems for DNA and protein in longevity is established, it remains largely unknown for RNA. In our recent work, we show that nonsense-mediated mRNA decay (NMD) promotes longevity in the roundworm C. elegans by enhancing RNA quality control. We find that the activity of NMD decreases during aging, raising the possibility that RNA quality declines in old animals. We then show that key components of NMD complex are required for prolonged lifespan in C. elegans. In addition, animals with reduced insulin/insulin-like growth factor-1 (IGF-1) signaling (IIS), a representative longevity model, display increased NMD activity. Thus, up-regulation of NMD appears to play crucial roles in longevity conferred by reduced IIS via enhancing mRNA quality control. As both IIS and NMD pathways are evolutionarily conserved, mammals including humans may be equipped with similar RNA quality control systems to achieve longevity.

  • In this study to cope with the decreasing product's life-cycle a virtual simulator to realize the simulation environment similar to a real manufacturing line is developed. The developed simulator plays a role in reducing the product conversion time by alternating manufacturing components and work plans on the simulation as manufacturing lines change and actuating a virtual manufacturing lines change and actuating a virtual manufacturing line before a real production. The developed simulator realized a virtual manufacturing line on the simulation using various manipulators and work cells as manufacturing components. Also It can be shown that the simulator can cope with rapid change of a manufacturing line by developing a interface that a separate process is managed for each manufacturing module and a manipulator component and a work cell are changed for a user to become convenient to teach tasks of each work module. using Microsoft Visual C++ 6.0 and OpenGL of Silicon Graphics for libraries to realize 3-dimensional graphic and constructing a database system, a hybrid type of hierachical and relational model to develop a progra that has efficiency and standardization.

  • In this paper, transparent dynamic photoelastic experimental hybrid method for propagating cracks in orthotropic material was developed. Using transparent dynamic photoelastic experimental hybrid method, we can obtain stress intensity factor and separate the stress components from only isochromatic fringe patterns without using isoclinics. When crack is propagated with constant velocity, the contours of stress components in the vicinity of crack tip in orthotropic material are similar to those of isotropic material or orthotropic material with stationary crack under the static load. Dynamic stress intensity factors are decreased as crack growths. It was certified that the dynamic photoelastic experimental hybrid method was very useful for the analysis of the dynamic fracture mechanics.

Angle of View Polarization Characterization of Liquid Crystal Displays and Their Components

  • LCD performance is generally evaluated in terms of luminance and color versus viewing angle. In the present paper, we show that this type of display can be favorably characterized in terms of polarization. We show that ELDIM EZContrast instrument which is routinely used for viewing angle measurements can be upgrade for measuring the polarization state of the light at each incidence and azimuth angle. More precisely, the degree of polarization of light, its ellipticity and polarization direction can be measured at each incidence angle between 0 and $88^{\circ}$ and for all the azimuth angles (from 0 to $360^{\circ}$). Important differences between the displays can be detected and related to their internal structures when luminance and color profiles are quite similar. The same setup can also be used to characterize optical components of the LCDs.

Fixture Planning Using Case-Based Reasoning (사례기반 추론방법을 이용한 치공구의 선정)

  • The aim of this research is the development of an automated fixture planning system for prismatic parts using the case-based reasoning (CBR). CBR is the problem solving paradigm that uses the similarity between a new problem and old cases to solve the new problem. This research uses CBR for the fixture planning. A case is composed with the information of the part, the components of fixture and the method of fixing for the part. The basic procedure is the retrieval and adaptation for the case, and this research presents the method of retrieval that selects most similar case to the new situation. The retrieval-step is divided into an index matching and an aggregated matching. The adaptation is accomplished by the modification, which transforms the selected case to the solution of the situation of the input part by the specified CBR algorithm. The components of fixture and the method of fixing are determined for a new part by the procedure.

  • In order to investigate the effect of strain states attributed to the lubrication during shape rolling, shape rolling for V-sectioned sheets was carried out with and without lubrication. The evolution of strain states during shape rolling was studied by three-dimensional finite element method (FEM). Shape rolling with and without lubrication produces shape-rolled samples in fairly similar outer shapes, since the distribution of normal strain components is nearly independent of the lubrication condition. In contrast, the distribution of shear strain components strongly depends on the lubrication condition.

Production and Characterization of Crystalline Cellulose-Degrading Cellulase Components from a Thermophilic and Moderately Alkalophilic Bacterium

  • A moderately thermophilic, alkalophlic and powerful crystalline cellulose-digesting bacterium, Bacillus K-12, was isolated from filter paper wastes and found to be similar to Bacillus circulans or Bacillus pumilis, except for its ability to grow at a moderately high pH and temperature. The isolate grew at a pH ranging from 6 to 10 and at a temperature ranging from 35 to $65^{\circ}C$ and produced a large amount of cellulase components containing avicelase, xylanase, CMCase, and FPase when grown in avicel medium for 5 to 7 days at $50^{\circ}C$. The crude enzyme preparation from the culture broth hydrolyzed xylan, raw starch, pullulan and ${\beta}-1,3$ glucan such as laminarin. Furthermore, the enzyme hydrolyzed crystalline cellulose to cellobiose and glucose and had a broad pH activity curve (pH 6~9). The enzyme was stable up to $70^{\circ}C$.

  • In this paper, we present an nonlinear PID controller with network based compensator which consists of a conventional PID controller that controls the linear components and neuro-compensator that controls the output errors and nonlinear components. This controller is based on the Harris's concept where he explained that the adaptive controller consists of the PID control term and the disturbance compensating term. The resulting controller's architecture is also found to be very similar to that of Wang's controller. This controller adds a self-tuning ability to the existing PID controller without replacing it by compensating the output errors through the neuro-compensator. Various simulations and comparative studies have proven that the proposed nonlinear PID controller produces superior results to other existing PID controllers. When applied to an actual magnetic levitation system which is known to be very nonlinear, it has also produced an excellent results.
