The present study was carried out to investigate the preventive effect of Epimedii Herba combined Samgijiwhang-Tang(SJTE) on streptozotocin(STZ)-induced diabetic nephropathy. SJTE was given to rats with oral administration. The experimental animals were divided into normal group of rats, control group of STZ-induced diabetic rats, and sample group with SJTE administration. Experimental diabetic nephropathy was induced by the injection of STZ(60mg/kg) to the rat via the peritoneum. The effect of SJTE on STZ-induced diabetic nephropathy was observed by measuring the serum level of insulin, glucose, creatinine and BUN. Urine secretion of albumin for 24 hours and urine level of glucose measures too. Anti-oxidative stress of STZ administration in living body was estimated by measuring lipid peroxide in cortex of kidneys. STZ induced increase of serum glucose. creatinine, urine albumin secretion and renal cortical lipid peroxidation were lowered by SJTE administration. In conclusion, the SJTE treatment showed protective effect on rat diabolic nephropathy model, and action mechanism of the effect was thought to be concerned with internal glucose metabolism.
This study was conducted to examine the hypoglycemic effects of complexcity extract(DB 55) from five oriental medicinal plants on the intake of food, body weight, blood glucose level, insulin content, serum GOP, GPT, BUN and hematocrit value in streptozotocin(STZ)-induced diabetic rats. General nutritional composition was also assessed. Thirty- five male Sprague-Dawley rats were assigned to five groups : normal control group(NC), STZ-diabetic control group (STZ-control), STZ-diabetic DB-1 group(DB 55-1), STZ-diabetic DB-2 group(DB 55-2) and STZ-diabetic DB-3 group(DB 55-3). The animals were fed an experimental diet for 12 weeks. Body weight gain of the STZ-diabetic group was significantly lower but diet intake was significantly higher than the NC group. Blood glucose levels of the DB-1, DB-2 and DB-3 rats were significantly lower than the STZ-control animals. Insulin level of STZ-control rats was lower than the NC serum GOT, GPT and BUN levels were significantly higher in the NC group, but hematocrit value was not significant as compare to the STZ-diabetic group. Blood glucose level was lower observed for the DB 55-1, DB 55-2 and DB 55-3 group. The results will be useful in oriental diet therapy and in the developing functional food resources.
Objectives : This study was performed to investigate the anti-diabetic effect of the fruit extract of Lycium chinense Mill(Lycii Fructus, LF) on multiple low-dose streptozotocin-induced diabetic rats.Methods : Male Sprague-Dawley rats were divided into three groups; normal, STZ-control, Lycii Fructus extractorally administrated 300 ㎎/㎏ group (STZ-LF). Diabetes was induced in rats by consecutive injection of streptozotocin (STZ) at doses of 30 ㎎/㎏ for 5 days. After 4 weeks, all rats were sacrificed, Fasting blood glucose, total cholesterol (CHO), triglyceride (TG) and HDL-Cholesterol were measured in sera of rats. Histopathological changes of pancreas, kidney, liver and lung tissues were observed by microscope after H&E, Periodic acid-Schiff (PAS) and Masson's trichrome staining. The changes of body weight, blood glucose, and food and water intake were also measured.Results : There were no differences in body, food intake and water intake in LF-administrated groups compared with STZ control group. However, LF extract significantly decreased the levels of serum glucose, CHO, TG and HDL-Cholesterol in diabetic rats. In histopathological analysis of kidney, liver and lung, LF-administrated groups showed the inhibition of morphological damage.Conclusions : These results suggest that LF have a biological action on multi low-dose STZ-induced diabetes in rats via decreasing the serum glucose, TG and TG levels and may protect the morphological changes of kidney, liver and lung.
This study was performed to explore the effects of the fermented functional extracts (FE) on blood glucose and lipid levels in diabetes. FE were created by mixing 9 kinds of plants with sea water and then allowing the mixture to ferment for 1 year. FE were supplemented in the feed of streptozotocin (STZ)-induced diabetic rats at 1%, 3% and 5%. The 1% feeding group showed the lowest weight loss of the three experimental groups. The blood glucose and glycosylated hemoglobin level were significantly decreased in the FE fed rats compared to the diabetic control (DMC) group. The lipid levels in serum were decreased in 1% and 3% FE fed rats in comparison to the DMC group, and there was no significant difference in triglyceride levels due to the FE concentration. The HDL-C level was significantly increased in rats with FE supplemented diets, compared to the DMC group. The levels of lipid peroxides in liver tissue were significantly decreased in FE fed diabetic rats, and the hepatic glycogen content was increased in rats receiving supplements. As a result of these studies, we believe 1% FE may be the optimum level for controlling blood glucose and alleviating hyperlipidemia in STZ-induced diabetic rats.
Hypertension is associated with cardiovascular disease. The environmental and genetic factors can cause the development of hypertension. In this study, the relationship between hypertension and family history of hypertension in Koreans was analyzed in consideration of serum fasting blood glucose levels and age. The study subjects were 2,484 subjects who had a medical examination at a university hospital. The main statistical analysis method was multiple logistic regression analysis. Hypertension prevalence was 16.4% of all subjects, and subjects with a family history of hypertension were 23.5%. The risk of hypertension was 2.36 times higher in subjects with a family history of hypertension than subjects without a family history of hypertension. In addition, in the subjects with fasting blood glucose levels more than 120 mg/dL, the risk of hypertension was 4.44 times higher in subjects with a family history of hypertension compared with subjects without a family history of hypertension. The relationship between family history and hypertension was slightly higher in the older group than in the younger group. To assess the association between hypertension and family history, further cohort study is necessary in the future.
The effects of Geumguesingitang and Dohongsamultang on rats with nephrosis induced by a single tail-intravenous injection of puromycin aminonucleoside(PAN), 2.5mg/l00g of body weight were evaluated in the present study. The effects of Geumguesingitang and Dohongsamultang on PAN nephrosis were evaluated by measuring (1)the concentrations of albumin, total protein, total lipid, cholesterol, triglyceride, creatinine, blood urea nitrogen(BUN) and uric acid in the serum, (2)the concentrations of protein, creatinine, glucose, occult blood and volume of the 24 hours urine and (3)the volume of intake water. The results are summerized as follows; 1. In the Control group as compared to the Normal. the amount of protein of 24 hours urine was significantly increased, the concentrations of albumin and total protein were significantly decreased. Total lipid, cholesterol and triglyceride in the serum were significantly increased. The concentrations of creatinine, BUN, uric acid in the serum, the amount of glucose and occult blood of 24 hours urine were also increased significantly. 2. In the Geumguesingitang group as compared to the Control, the increase in the amount of urinary protein during 24 hours induced by PAN was significantly suppressed, and the concentrations of total protein and albumin in the serum were significantly increased. The concentrations of total lipid, cholesterol and triglyceride in the serum were significantly inhibited. The decrease of the concentrations of creatinine and uric acid in the serum were also observed significantly. 3. In the Dohongsamultang group as compared to the Control, the increase of the amount of protein and glucose of the 24 hours urine induced by PAN were significantly inhibited, and the concentrations of total protein and albumin in the serum were increased significantly. The concentrations of total lipid, cholesterol and triglyceride in the serum were decreased significantly. The decrease of the concentrations of creatinine and uric acid in the serum were observed significantly. It can be inferred that Geumguesingitang has effects on improving proteinuria, hypoproteinemia effectively. It has an effect on hyperlipidemia significantly relieved. And relieving azotemia when nephrotic syndrome is accompanied by the acute renal failure. It can be inferred that Dohongsamultang improves hyperlipidemia effectively. It has effects on proteinuria, hypoproteinemia in nephrotic syndrome significantly relieved. And relieving azotemia when nephrotic syndrome is accompanied by the acute renal failure.
Objective : This study was undertaken to investigate how Bombycis corpus (BC) effects the development and progress of complications occurring in Diabetes Mellitus (DM). Methods : Laboratory rats were seperated into three groups; normal, rats with DM and treated with BC, and rats with DM and not treated. In this study DM was experimentally induced through injection of streptozotocin. The BC treated group was given BC extract p.o. for 15 days. Then, The activities of glucose phosphatic enzymes and polyol pathway channels were observed. Results : The blood glucose level greatly increased in the DM groups after injection of streptoztocin, but it significantly decreased in the BC treated group. Significantly enhanced levels of serum insulin levels were seen in the BC treated group, while supressed levels were seen in the untreated DM group. Weight was recovered by the BC treated group, matching the normal group. Decreased enzyme activity of aldose reductase, sorbitol dehydrogenase and glucose-6-phosphatase were seen in BC treated diabetic rats. Increased enzyme activity, of the glucokinase and hexokinase were seen in BC treated diabetic rats. Conclusions : This study suggests that BC normalized the blood glucose and serum insulin levels destablized by DM. Because increased activity of glucose phosphatic enzymes, glucokinase and hexokinase, and decreased glucose-6-phosphatase activity, and suppression of polyol pathway enzymes, aldose reductase and sorbitol dehydrogenase, were all seen, these observations suggest that BC suppresses blood glucose levels and prevents complications due to DM.
The purpose of this study was to investigate the effect of soybean on blood glucose and lipid concentrations, and antioxidant enzyme activity in type 2 diabetes mellitus(DM) patients. We divided patients into two groups and fed them, respectively, a basal diet(control group) and a basal diet with 69 g/d of soybean(soybean group) for 4 weeks. Pills with roasted soybean powder were provided to the soybean supplementation group three times a day. Macronutrients intake except dietary fiber was similar between the two groups. No significant differences were observed in dietary intakes or body weight before and after the supplementation. Energy composition ratio of C:F:P was 65:19:16 in the control group, 64:20:16 in the soybean group. The blood parameters of subjects before supplementation, such as fasting blood glucose, postprandial glucose level, total cholesterol, triglyceride, LDL-cholesterol and $HbA_1C$ were not different between the two groups. After supplementation, fasting blood glucose(p<0.001), postprandial glucose level(p<0.001) and serum triglyceride level(p<0.05) were significantly reduced in the soybean group in comparison with the control group. The total cholesterol level was not significantly different between the control and the supplemented group after 4 weeks of treatment. TBARS levels of the soybean group were not significantly different from those of the control group. The activities of catalase(p<0.01) and glutathione peroxidase(p<0.05) were significantly higher in the soybean group compared to the control group. The results of this study suggest that soybean supplementation would be helpful to control blood glucose and serum lipid in diabetic patients. Also, soybean showed an antioxidant activity that may contribute to enhance the effect of antioxidant defense. This activity contributes to protection against oxidative damage in type 2 DM patients. Soybean may have potential use in the disease management of patients with DM.

Effects of raw soy flour(RSF) and RSF with magnesium supplementation(RSF+Mg) feeding on lipid metabolism and serum glucose concentration of diabetic and nondiabetic rats were studied. Male rats(Sprague-Dawley, body weight $332.8\pm19.3g)$ were assigned to four dietary groups and fed for 4 weeks. For each experimental diet some rats were injected with streptozotocin in intraperitonium (i.p.) to induce diabetes, and other rats were injected with buffer i.p. as a nondiabetic group. The diabetic rats fed diets of control, RSP and RSF+Mg showed significant body weight decrease(-67~-79g) compared with nondiabetic group. Pancreas weights in diabetic RSF and RSF + Mg groups were heavier than those of the control and nondiabetic groups. , Fasting serum glucose level of diabetic RSF and RSF+Mg groups significantly decreased by 73 and 44% compared with the diabetic control group, but they significantly increased by 77 and 111% compared with the nondiabetic group. Serum triglyceride, phospholipid and total cholesterol concentration of diabetic RSF and RSF + Mg groups significantly decreased compared with the diabetic control group. Serum HDL-cholesterol, HDL-cholesterolitoal cholesterol ratio and liver total cholesterol were not affected by RSF and RSP+Mg feeding, Liver total lipid and triglyceride concentration were significantly lower in diabetic groups than those in the nondiabetic group. The results indicated that RSF and RSF+Mg feeding did not produce hyperlipemia and somewhat improved the glucose levels in diabetic rats.
Objectives : We conducted this study to examine the association between serum GGT levels within the normal range and the risk factors of cardiovascular diseases Methods : We examined the cross-sectional association between serum GGT and the systolic and diastolic blood pressure, fasting blood glucose, total cholesterol, triglyceride, high density lipoprotein cholesterol (HDLcholesterol), and uric acid among 975 subjects that participated in the health examination of a university hospital located in Daegu city. All the patients' GGT levels were within the normal range. Results : After adjustment were made for age, body mass index (BMI), smoking status, drinking frequency, exercise frequency and coffee intake, the serum GGT level was positively associated with fasting blood glucose (p<0.01), total cholesterol (p<0.01), and triglyceride (p<0.01) in men, and it was positively associated with fasting blood glucose (p<0.01), total cholesterol (p<0.05), triglyceride (p<0.01), and uric acid (p<0.01) in women. The associations were not significantly different depending on the status of alcohol drinking or obesity, except for the associations of serum GGT with diastolic blood pressure (P for interaction=0.04) and uric acid (P for interaction=0.04) between the lean and obese subjects. Conclusions : Serum GGT levels within the normal range were positively associated with fasting blood glucose, triglyceride and uric acid in most subgroups irrespective of the drinking or obesity status. These results suggest that GGT has important clinical implications as being more than just a marker of alcohol consumption and hepatobiliary disease.
