  • Background: Peste des petits ruminants (PPR) is an infectious disease caused by the peste des petits ruminants virus (PPRV) that mainly produces respiratory symptoms in affected animals, resulting in great losses in the world's agriculture industry every year. Single-domain variable heavy chain (VHH) antibody fragments, also referred to as nanobodies, have high expression yields and other advantages including ease of purification and high solubility. Objectives: The purpose of this study is to obtain a single-domain antibody with good reactivity and high specificity against PPRV. Methods: A VHH cDNA library was established by immunizing camels with PPRV vaccine, and the capacity and diversity of the library were examined. Four PPRV VHHs were selected, and the biological activity and antigen-binding capacity of the four VHHs were identified by western blot, indirect immunofluorescence, and enzyme-linked immunosorbent assay (ELISA) analyses. ELISA was used to identify whether the four VHHs were specific for PPRV, and VHH neutralization tests were carried out. ELISA and western blot analyses were used to identify which PPRV protein was targeted by VHH2. Results: The PPRV cDNA library was constructed successfully. The library capacity was greater than 2.0 × 106 cfu/mL, and the inserted fragment size was approximately 400 bp to 2000 bp. The average length of the cDNA library fragment was about 1000 bp, and the recombination rate was approximately 100%. Four single-domain antibody sequences were selected, and proteins expressed in the supernatant were obtained. The four VHHs were shown to have biological activity, close affinity to PPRV, and no cross-reaction with common sheep diseases. All four VHHs had neutralization activity, and VHH2 was specific to the PPRV M protein. Conclusions: The results of this preliminary research of PPRV VHHs showed that four screened VHH antibodies could be useful in future applications. This study provided new materials for inclusion in PPRV research.

  • The conspectus, based on a set number of subject descriptors derived essentially from LC subject classifications, provides a consistent framework within which libraries could report subject intensity. While library collection development has been benefitted by using this conspectus, its effectiveness in this digital era is under controversy. The purpose of this study is to survey the concepts and the various types of conspectus in library collection development and to test the applicability of a revised conspectus model to electronic resource management in libraries. For this, the focus was especially put on investigating the characteristics of electronic resources as considered the collection depth usually identified in collection development policies. Previous research has been also surveyed to consolidate their outcomes in order to design a matrix model of conspectus, which put the characteristics, persistence, and collection level in consideration in oder for the design of the conspectus model.

  • This paper intend to analyze, review and point out the actual conditions of "Reserve Book System" of university libraries in Korea, then to present several proposals. I took six national universities and six private universities as researching objects and the materials were collected by questionnaires. 1) There are so many libraries which have no seperate reserve books room, while every colleges, and their departments are putting into forces the reserve book system. This makes many unresonable problems. 2) Many volumes were not adequate to the courses of each depertment and so many problems arose to set up reserve books owing to Lhe lack of library budget. 3) Insincerities of professors and librarians make no good conditions for the reserve book system. 4) For insufficiency of understanding about the reserve book system cf professors and students with university authorities, we must persuade and guide them about this system. 5) We imust evaluate about the problems of the reserve book system. I show the methods which is necessary to solve the problems of this system in the following. 1) As to methods of improving the field of menagement for running about the reserve book system, a) Establishment of the independent reserve books room. b) Gradual expansion of the reserve book system. c) Evaluation of the actual using of reserve books. 2) As to contents to improve the reserve book system, a) Improving the selection method of reserve books. b) Increasing the library budget enable to purchase more reserve books. c) Making more close connections between the curricula ard reserve books. 3) Settlment of coordinating system for carrying out reserve book system, a) We must have mutual understanding and co-operation among university authorities, faculties and libraries. b) We must have co-operation between professors and librariars.ibrariars.

  • The purpose of this study is to explore the establishment and administrative strategies for shared repositories for academic libraries in Korea, using the US ReCAP (Research Collections and Preservation Consortium) Project as a case study. The US ReCAP Project was a collaboration between the New York Public Library and the university libraries of Columbia University, Harvard University, and Princeton University. In-depth interviews were conducted with librarians and administrators in charge of the ReCAP Project to understand the original vision of the project, as well as how the strategy and approach evolved over the years. The analysis results revealed that shared repositories for libraries have evolved into shared collections enabling shared use. But shared repositories require a central management system to be successful in the long-term. Additionally, findings showed that the shared repository filled more quickly than anticipated. This in turn required collaborative planning on shared collection development and selection policies to determine which materials to send to the shared repository.
