정수장 원생동물 분석방법의 국내 실험실간 교차시험 연구 (Interlaboratory Study for Cryptosporidium and Giardia Test Methods in Water)

  • This interlaboratory study was designed to evaluate protozoan test methods in water and to predict the major causes of deviation of the methods. Each of four laboratories with previous experience of protozoa analysis in water participated, and met the initial performance criteria of EPA 1623 method provided. The protozoan analysis procedure consists of filtrations, concentration, immunomagnetic separation, dyeing (staining) and counting with observation. We tested three different filtration equipments: capsule filter for 10 L of surface water, and high volume (HV) capsule filter and membrane filter for 100 L of finished water. When the recovery of each step of the procedure was evaluated with EasySeed, the commercial stock of each 100 Cryptosporidium and Giardia, immunomagnetic separation and filtration step were the most crucial steps affecting the stability of the recovery, especially for Cryptosporidium. There was no significant difference of recovery among the filtration methods. Recovery of protozoa from source water were evaluated with spiked EasySeed as matrix tests. The recoveries of Giardia increased significantly in the matrix tests compared those in the deionized water. We also applied red stained mixture stocks of Cryptosporidium and Giardia called ColorSeed as internal standards of water sample tests. The recoveries of both EasySeed and ColorSeed in samples tested were within the range of the criteria, however, the Giardia recoveries using ColorSeed decreased significantly. Further optimization study with ColorSeed will be necessary, considering the convenience of using the internal standard without additional sample analysis. The significant factors of the recovery variation were identified as the differences of laboratories as well as water quality and type of the stock for spiking. The results of this study emphasize the importance of the quality assurance program for protozoan analysis lab in water.

Disc Electrophoresis에 의한 면화의 Genome과 종간관계 분석 (Analysis of Genome and Species Relationships in Cotton by Disc Electrophoresis)

  • Protein spectra from 6 species of Gossypium were obtained by disc electraphoresis of seed extracts. Protein extracts were made by soaking 0.5g. of seed in 15ml of Tris-glycine buffer for 24 hours. Gels 24 hours. Gels were stained in 0.5% Amido Black solution for 1 hour, and destained in 7% acetic acid for 72 hours. Nine to 15 bands were visible in each gel. Homologies of Protein bands among the species were determined by migration velocity. Evidences obtained from electrophoretic separation of seed Protein were consistent with those from genetic, cytological, morphological and Phenogenetic methods regarding the origin of New World cultivated cottons. Possibility, however, does not exist to exclude Gossypium herbaceum from one of the Progenitors of New World cultivated cottons from electrophoretic evidences alone.

Quantitative Determination of Sesaminol Glucosides in Sesame Seed

  • The sesaminol glucosides in 80% EtOH extract from sesame seeds were separated by high performance liquid chromatography (HPLC). A HPLC system using a Develosil ODS-5 column and gradient elution system from 30% to 80% methanol was selected for separation and quantitative determination of sesaminol triglucoside, sesaminol diglucoside, and sesaminol monoglucoside. Quantitative analyses for these sesaminol glucosides, sesaminol triglucoside, sesaminol diglucoside, and sesaminol monoglucoside were determined on the basis of standard curve of sesaminol glucosides. Sesaminol triglucoside, sesaminol diglucoside and sesaminol monoglucoside contents of the seed of one Korean sesame cultivar, Danbaekggae, were 56.4 mg/100g, 9.6 mg/100g, and 7.5 mg/100g, respectively. Core-Shell Polymerization with Hydrophilic Polymer Cores

  • Two-stage emulsion polymerizations of hydrophobic monomers on hydrophilic seed polymer particles were carried out to make core-shell composite particles. It was found that the loci of polymerization in the second stage were the surface layer of the hydrophilic seed latex particles, and that it has resulted in the formation of either eccentric core-shell particles with the core exposed to the aqueous phase or aggregated nonspherical composite particles with the shell attached on the seed surface as many small separated particles. The driving force of these phenomena is related to the gain in free energy of the system in going from the hydrophobic polymer-water interface to hydrophilic polymer-water interface. Thermodynamic analysis of the present polymerization system, which was based on spreading coefficients, supported the likely occurrence of such nonspherical particles due to the combined effects of interfacial free energies and phase separation between the two polymer phases. A hypothetical pathway was proposed to prepare hydrophilic core-hydrophobic shell composite latex particles, which is based on the concept of opposing driving and resistance forces for the phase migration. It was found that the viscosity of the monomer-swollen polymer phase played important role in the formation of particle morphology.

Cross-linked Leucaena Seed Gum Matrix: An Affinity Chromatography Tool for Galactose-specific Lectins

  • A cross-linked leucaena (Leucaena leucocephala) seed gum (CLLSG) matrix was prepared for the isolation of galactose-specific lectins by affinity chromatography. The matrix was evaluated for affinity with a known galactose-specific lectin from the seeds of snake gourd (Trichosanthes anguina). The matrix preparation was simple and inexpensive when compared to commercial galactose-specific matrices (i.e. about 1.5 US$/100 ml of matrix). The current method is also useful for the demonstration of the affinity chromatography technique in laboratories. Since leucaena seeds are abundant and inexpensive, and the matrix preparation is easy, CLLSG appears to be a promising tool for the separation of galactose-specific lectins.

레미콘 슬러지를 이용한 불소제거 (Fluoride Removal Using Ready-Mixed Concrete Sludge)

  • 본 연구의 목적은 칼슘염을 함유한 레미콘 슬러지를 이용하여 불소함유폐수를 물에 난용성인 $CaF_2$ 화합물 형태로 침전시켜 처리하는데 있어 pH, 레미콘슬러지 주입량, Seed물질 주입량, 교반속도, 교반시간의 최적 조건을 도출하고자 하는데 있다. 실험결과 레미콘 슬러지를 이용한 대상폐수의 $CaF_2$ 침전 반응에서 함수율 및 불소의 제거효율 등을 고려할 경우 최적의 pH 6, 레미콘 슬러지 주입량은 10 g/L, Seed 물질 주입량은 2 g/L, 교반속도은 100 rpm, 교반시간은 60 min으로 관찰되었다. 이 때 Seed 물질의 주입은 불소제거효율에는 영향이 없는 것으로 나타났지만 침전물의 형성이 되는 반응을 촉진시키고 플럭형성을 원활하게 하여 고액 분리가 잘 되어 함수율이 낮아지는 결과가 나타났다.

종실유(seeds oil)의 위치별 지방산 및 트리아실글리세롤의 조성 연구 (Study on the Positional Distribution of Fatty Acids, and Triacylglycerol Separation, of Seed Oils)

  • 본 연구에서는 6종류의 종실에서 기름을 착유하여 조지방 함량(crude fat content) 측정 및 총 지방산 조성과 위치별 지방산 조성을 비교 분석 하였으며, triacylglycerol(TAG)의 조성과 tocopherol 함량을 분석하였다. Folch법을 이용하여 분석된 조지방 함량은 들깨 21.64%, 홍화씨 13.85%, 고추씨 9.60%, 석류씨 8.85%, 녹두 2.25%, 결명자 2.00%로 나타났다. 추출된 종실유의 지방산 분석결과 불포화 지방산인 linoleic acid(C18:2)가 81.57 wt%~46.17 wt%로 가장 높은 비중을 차지하였으며, sn-2 위치의 지방산 조성에서도 linoleic acid의 함량이 88.30 wt%~15.99 wt%로 높은 함량을 나타내었다. Triacylglycerol(TAG)의 조성은 reversed-phase HPLC를 이용하여 분석하였으며 partition number(PN)=36~48의 분포를 보였다. Total tocopherol의 함량은 석류씨에서 가장 높은 377.74 mg /100 g으로 나타났으며 이 외에 각각 녹두(141.16 mg/100 g), 결명자(107.23 mg/100 g), 고추씨(33.88 mg/100 g), 들깨(30.05 mg/100 g), 홍화씨(29.80 mg/100 g) 순으로 측정되었다.

퇴화처리에 따른 참깨, 파, 상추 종자의 당, 아미노산, 단백질 누출 (Leakage of Sugars, Amino Acids and Protein from Differently - Aged Seeds of Sesame, Welsh Onion and Lettuce)

  • 비파괴적인 방법으로 활력이 높은 종자를 선별하는 방법을 개발하기 위하여, 참깨, 파, 상추 종자를 상대습도 90%, 온도 45$^{\circ}C$에서 인위적으로 퇴화시켜 활력이 다른 종자를 물에 침지하였을 때 종자가 분비하는 당, 아미노산, 단백질의 양과 속도 등을 조사한 결과를 요약하면 다음과 같다. 1. 참깨 종자는 활력과 침지시간에 관계없이 당, 아미노산, 단백질을 거의 분비하지 않았다. 2. 파의 죽은 종자는 건전종자나 활력이 중인 종자보다 아미노산을 더 많이 분비하였으나 전당과 단백질 분비량은 크지 않았다. 3. 상추의 죽은 종자는 당, 아미노산, 단백질을 모두 많이 분비하였다. 건전종자와 활력이 중인 종자는 당은 거의 분비하지 않았으나 아미노산과 단백질은 다소 많이 분비하였다.

Pumpkin Seed Oil as a Partial Animal Fat Replacer in Bologna-type Sausages

  • Beef fat was replaced with cold press pumpkin seed oil (PSO; 0%, 5%, 15%, and 20%) in the production of bologna-type sausages. A value of pH, water-holding capacity (WHC), jelly-fat separation, emulsion stability and viscosity values were determined in meat batters. Thiobarbituric acid reactive substances (TBARS), color, and textural characteristics (TPA, shear test, penetration test) were determined in end-product at 1, 7, 14, 21, and 28 days of storage at 4℃. The pH values were varied between 6.06 and 6.08. With the increase in the level of PSO in meat batters, there was a significant increase in WHC, jelly-fat separation and viscosity values (p<0.05) while a significant decrease in emulsion stability (p<0.05). TBARS values of sausages were found to be significantly higher than in the control group (p<0.05), and this trend continued during storage. Increasing of PSO level were caused a significant increase in L* and b* values while a decrease in a* value (p<0.05). Hardness, adhesiveness and chewiness values were significantly reduced whereas cohesiveness and resilience values increased (p<0.05). Maximum shear force and work of shear was significantly decreased as the level of PSO increased (p<0.05). Hardness, work of penetration and the resistance during the withdrawal of the probe values (penetration tests) increased significantly with the increase in the level of PSO (p<0.05). These results indicate that PSO has potential to be use as a replacement of animal-based fats in the production of bologna-type sausages.

  • The seed, or grain, of modern cultivars of Lupinus angustifolius, commonly known as Australian sweet lupins (ASL), is an established feed resource for the intensive animal industries of Australia, Japan, Korea and several other countries in Asia and Europe. Since the introduction of ASL to the world marketplace about 25 years ago, researchers in many countries have found them to be a valuable component of the diet of beef and dairy cattle, sheep, pigs, poultry, finfish and crustaceans. The seed of ASL contains ~32% crude protein (CP) (~35% DM basis) and 5% oil. The main storage carbohydrates in the seed are the ${\beta}$-galactans that comprise most of the cell-wall material of the kernel and the cellulose and hemicellulose of the thick seed coats. ASL seeds contain about 40% non-starch polysaccharides (NSP) and a negligible amount of starch. This makes them an excellent ingredient for ruminant diets, as the risk of acidosis is very low. The seed of modern cultivars of domesticated Lupinus species contain negligible amounts of lectins and trypsin inhibitors so they do not require preheating before being used as an ingredient in feeds for monogastric species. They have a high digestibility coefficient for protein, >90% for most species, but a low energy digestibility, ~60%, which is mostly due to the high content of NSP. The low content of methionine (0.22%) and of lysine (1.46%) is typical of the legumes. The lysine availability for pigs is >70%. Lupin kernels contain ~39% CP (~42% DM basis), 6% oil and 30% NSP. They have a higher digestible energy for pigs and finfish and a higher metabolisable energy for poultry than whole seed. Commercial operations rarely achieve complete separation of kernel from hull and it is more likely that the kernel fraction, called splits or meats, will contain ~36% CP. The replacement of soybean meal or peas with ASL in cereal-based diets for most intensively reared animals, birds and fish is possible provided lysine, methionine and digestible energy levels are kept constant. This makes ASL economically competitive in many, but not all, circumstances.
