  • Liquefaction potential on the specific site of nuclear power plant is analyzed and reviewed. The layered site fur this study consists of silt and sand. Based on the limited available soil data, maximum shear strength at critical locations using Seed & Idriss method and computer program SHAKE is calculated, and liquefaction potential is reviewed. Seismic input motion used fur the assessment of liquefaction is the artificial time history compatible with the US NRC regulatory Guider .60. Assessment results of the liquefaction are validated by analyzing to the other typical soil fecundations which can show the effects of foundation depth and soil data.

  • An assessment of liquefaction potential is made in principle by comparing earthquake induced shear stress to the liquefaction strength of the soil. In this research a modified method based on Seed and Idriss theory is developed for evaluating liquefaction strength of Jumunjon sand(Korean standard sand). Also the factors affecting liquefaction strenght such as cyclic shear stress amplitude and relative density are investigated and verified by using cyclic triaxial test. From the result the new relationships between cyclic shear stress ratio and number of load cycles are proposed for evaluating liquefaction strength under moderated magnitude(M=6.5) of earthquake.

홍화씨추출물의 피부 주름개선 효과(II) (Anti-wrinkle Effect of Safflower (Carthamus tinctorius L.) Seed Extract (II))

  • 최근 홍화씨의 세로토닌유도체, 리그난 및 플라보노이드와 같은 화합물은 에스트로겐${\cdot}$항에스트로겐 활성을 갖는 식물 유래의 diphenol 화합물인 phytoestrogen 물질로 작용할 수 있음이 밝혀지면서 이를 함유하고 있는 홍화씨추출물[safflower (Carthamus tinctorius L.) seed extract]의 피부주름 개선효과가 기대되고 있다. 본 연구진은 이전에 홍화씨추출물은 피부세포의 산화를 막아주는 항산화작용을 지니고 있으며, 아울러 피부의 matrix protein을 만들어내는 섬유아세포 증식을 촉진할 뿐만 아니라 피부의 구조를 이루고 있는 엘라스틴 단백질을 분해하는 elastase의 활성 저해효과를 보고한 바 있다. 이러한 연구 배경에서 본 연구에서는 홍화씨추출물의 피부주름 개선효과를 알아보기 위해 콜라겐 발현효과 및 MMP-1 발현 억제효과를 각각 측정하였다. 그 결과 홍화씨추출물의 콜라겐 발현효과는 레티놀보다 우수하였으며, 아울러 MMP-1 발현 저해효과는 올레산과 유사하게 농도 의존적으로 저해되었다. 또한 $0.1\%$ 홍화씨추출물 함유 크림은 대조군(홍화씨추출물 미함유 크림)에 비하여 피부주름 개선 효과가 크게 나타났다. 이러한 결과를 볼 때 홍화씨추출물은 주름개선용 기능성화장품 신소재로의 사용이 기대된다.

저지대 이차림지역의 매토종자 발아특성 -예산군의 침엽수림과 활엽수림- (Germination of Buried Seeds in Secondary Forest of Basla Zone - Coniferous and Broadleaved Forest of Low Slope, Yesan-gun, Korea -)

  • 본 연구는 이차림 잠재식생 변화의 기초자료로 제공하기 위하여 예산군의 침엽수림과 활엽수림 지상부 (현존식물)와 지하부 (매토종자 발아식물) 식생을 파악하고 비교하였다. 매토종자 발아의 관속식물은 침엽수림이 20과 27속에 딸린 28종 1변종의 29분류군이었고, 활엽수림은 18과 32속에 딸린 34종 2변종의 36분류군이었다. 국화과는 침엽수림과 활엽수림 모두 가장 많이 발아한 매토종자의 식물 집단이었고, 조사구 출현빈도는 침엽수림의 방동사니와 활엽수림의 고들빼기가 높게 나타났다. 토심별 분류군은 침엽수림이 토심 0~10cm, 활엽수림이 토심 0~5cm에서 가장 많았고, 개체수는 침엽수림과 활엽수림 모두 토심 0~2cm에서 가장 많이 나타났으며 토심이 깊을수록 감소하였다. 매토종자의 발아 개체수가 가장 많은 식물은 침엽수림과 활엽수림 모두 고들빼기이었다. 지상부와 지하부의 유사도지수는 평균 0.22로서 낮았으며, 침엽수림과 활엽수림은 유사도지수가 평균 0.40으로서 지상부와 지하부 비교보다 유사성이 높게 나타났다.

돼지감자 종자의 휴면성과 발아유도를 위한 종자처리 (Seed Dormancy of Jerusalem Artichoke (Helianthus tuberosus L.) and Seed Treatment for Germination Induction)

  • 국내외에서 수집한 5종(JA 2, JA6, Mammoth French White, D-19, K-8) 돼지감자 화기에서 종자결실을 조사하였고 수집 종자들의 발아특성을 밝히고 환경조건을 달리하여 발아유도를 시도하여 다음의 결과를 얻었다. 1. 임실율은 품종에 따라 차이가 있어 JA 2의 2.4%에서 Mammoth French White의 14.7% 범위에 있었으며 1주로부터 채취 가능한 종자입수는 JA2의 88개에서 JA6의 1,058 개 범위이었다. 2. 수확 후 3개월 실내 저장된 종자는 약 0%의 발아율을 나타냈으며 온도, 광 및 GA3에도 발아가 유도되지 않았다. 그러나 실내에서 27개월 저장된 종자는 D-19 품종에서 47.5%까지 발아하였다. 3. 종피제거된 종자는 96.8%의 발아율을 나타냈으며 종피에 바늘로 구멍을 뚫었을 때에도 82.3% 발아하였다. 따라서 종피제거 방법에 의해 완전한 발아유도가 가능하였으며 돼지감자 휴면종자는 종피에 의해 발아가 억제되고 있음을 알 수 있었다. 4. 휴면종자를 약 70일 저온습층처리한 경우 85% 이상의 발아율을 나타냈으며 이 경우 종피존재여부에 관계없이 어느 경우에나 발아하였다.

오크라 씨 기름의 기능성 성분 함량 및 생리활성 평가 (Evaluation of Bioactive Compounds Contents and Biological Activities of Okra Seeds Oils)

  • This study examined the contents of bioactive compounds and the biological activity of okra seed oil. Okra seed oil consisted mainly of linoleic acid (44.2%). The content of total phytosterols was 2.180 mg/g oil, with β-sitosterol being the highest (1.756 mg/g oil). The vitamin E content was 1.278 mg/g oil; the content of α-tocopherol was higher than γ-tocopherol. The total polyphenol and flavonoid contents were 2.463 mg gallic acid equivalent/g and 1.602 mg cathechin equivalent/g, respectively. The 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid and α-α-diphenyl-β-picrylhydrazyl free radical scavenging activities were 15.297% and 22.265%, respectively, and the reducing power was 4.524 mg gallic acid equivalent/g. The okra seed oil inhibited 77.692% of the α-glucosidase activity. The present study showed that okra seed oil had a considerable amount of phytochemicals and exhibited biological activity. These results suggest that okra seed oil is a potential natural therapeutic for the management of metabolic syndromes.

비름(Amaranth)과 명아주(Quinoa) 재배종의 기능성 물질과 변이 (Functional Ingredient and Their Some Variance in Amaranth and Quinoa)

  • Amaranth(Amaranthus spp. L.) and quinoa (Chenpodium quinoa Willd.) are old crops from South, Central America and Central Asia and their grains have been identified as very promising food crops because of their exceptional nutritive value. Squalene is an important ingredient in skin cosmetics and computer disc lubricants as well as bioactive materials such as inhibition of fungal and mammalian sterol biosynthesis, antitumor, anticancer, and immunomodulation. Amaranth has a component called squalene (2,6,10,15,19,23-hexamethyl-2,6,10,14,22-tetraco-sahexaene) about 1/300 of the seed and $5\~8\%$ of its seed oil. Oil and squalene content in amaranth seed were different for the species investigated. Squalene content in seed oil also increased by $15.5\%$ due to puffing and from 6.96 to $8.01\%$ by refining and bleaching. Saponin concentrations in quinoa seed ranged 0.01 to $5.6\%$. Saponins are located in the outer layers of quinoa grain. These layers include the perianth, pericarp, a seed coat layer, and a cuticle like structure. Oleanane-type triterpenes saponins are of great interest because of their diverse pharmacological properties, for instance, anti-inflammatory, antibiotic, contraceptive, and cholesterol-lowering effects. It is known that quinoa contains a number of structurally diverse saponins including the aglycones, oleanolic acid, hederagenin, and phytolaccagenic acid, which are new potential in gredient for pharmacological properties. It is likely that these saponin levels will be considerably affected by genetic, agronomic and environmental factors as well as by processing. With the current enhanced public interest in health and nutrition amaranth and quinoa will most likely remain in the immediate future within the realm of exotic health foods until such time as agricultural production meets the quantities and qualify required by industrial food manufacturers.

Effect of ultrasonication, light and liquid smoke treatment on germination of lettuce seeds

  • Seed priming leading to improved emergences, germination speed and uniformity under unfavorable conditions without loss of longevity are the best level of techniques. We studied the possibility to obtain primed seed with reduced mean germination time (MGT) and uniformity but with simple, fast and effective methods. The experiments were carried out at $15^{\circ}C$ for 16 hours with priming treatment. To compare the effects of each treatment, we used 200mM $CaCl_2$ priming or hydro priming or no imbibition after ultrasonication or red light treatment. The light treatment was performed by irradiating 2000 lux of red light for 15, 30, 60, and 120 minutes. Addition, in order to investigate the effect of the priming solution, treated at 200mM $CaCl_2$ for 8 hours. Ultrasonication treatment was performed for 5, 10, and 20 minutes at exposures of 13.0, 17.3, and 21.6 kHz during priming. For liquid smoke (LS) treatment, seed soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted water and 200mM $CaCl_2$ solution. After each treatment, the seeds were dried to moisture content ranged 5-8% at $25^{\circ}C$ for 24 hours. The effect of seed treatment was evaluated with germination percentage (GP), MGT, germination index(GI), germination rate(GR), Germination Uniformity(GU) and heath seed percentage(HS). For several factors tested, we found that the desired germination improvement could be obtained by treating the seed with ultrasonication at 17.3 kHz for 5 minutes in water or red light exposure at 2000 lux for 120 min in water that resulted very similar to those used to 200mM $CaCl_2$ priming for 16 hrs. However, LS treatment showed no improvement in all diluted solution. Therefore, the methods applied ultrasonication and red light treatment showed high potential for fast and easy treatment avoiding pollution of salt solution waste.

Sodium Hypochlorite 처리가 몇가지 작물의 종자소독에 미치는 효과 (Effect of Sodium Hypochlorite Treatment on Incidence of Seed-borne Fungi in Several Crop Seeds)

  • 일반소독제로 광범위하게 사용되는 NaOCl을 종자소독제로써 이용할 수 있는 방안을 확립하고자 종자전염에 의한 병해 발생이 많은 볍씨와 채소종자를 대상으로 소독 효과를 조사하였던 바 다음과 같은 결과를 얻었다. 1. NaOCl의 참깨 종자 소독 효과를 조사하였던 바 NaOCl의 농도가 높을 수록 그리고 침지시간이 길수록 종자 소독 효과가 뚜렷 하였으나 종자 발아 장해를 일으켰다. 종자발아율을 높이며 소독효과가 우수한 종자 처리 방법은 NaOCl 1-2%에서 10분간 침지하는 것이었고, 벼 종자소독 효과는 NaOCl 농도 1-2%에서 침지시간을 30분 이내로 하는 것이 좋았다. 2. 무, 배추 종자 소독 효과를 조사하였던 바 NaOCl 농도 1%에서 10분간 침지할 경우 각종 Alternaria균의 검출율을 현저히 저하시켰고, 고추종자에서는 NaOCl 농도 1%에서 10분간 침지할 경우 각종 탄저병균의 검출율을 현저히 저하시켰다. 3. 종자전염성 진균에 대한 NaOCl의 소독 효과는 종자소독제인 Benlate T에 비하여 참깨종자에서는 우수하게 나타났고, 볍씨에서는 비슷하였다. 그러나 NaOCl은 처리농도가 높고 시간이 긴 경우에는 발아장해를 초래하였다.

Use of SSR Markers to Complement Tests of Distinctiveness, Uniformity, and Stability (DUS) of Pepper (Capsicum annuum L.) Varieties

  • This study was carried out to assess the potential of SSR markers for variety identification by comparing SSR markers and morphological traits in tests of distinctiveness, uniformity, and stability (DUS) of pepper (Capsicum annuum L.) varieties. Twenty-seven SSR markers were polymorphic in 66 pepper varieties, revealing a total of 89 alleles. Average polymorphism information content (PIC) value was 0.529, ranging from 0.03 to 0.877. Cluster analysis of the band patterns separated the varieties into three groups corresponding to varietal types. Morphological trait-based clustering showed some degree of similarity to dendrogram topologies based on the SSR index. However, no significance correlation was found between the SSR and morphological data. SSR markers could be used to complement a DUS test of a candidate variety and to select complimentary varieties by pre-screening existing varieties in the context of protecting new varieties of pepper.
