This study was conducted to establish the efficient protocols of the germination and cryopreservation of dehisced Korean ginseng seeds for long-term germplasm conservation. $GA_3$ and BA treated on seed for 24 hr facilitated germination at 5 and $10^{\circ}C$. Germination percentage of desiccated seeds was decreased under moisture content (MC) of below $7.2\%$. Dehisced ginseng seeds were dried under airflow of laminar floor cabinet and seed drying room. The high levels (more than $90\%$) of germination after cryogenic exposure were obtained after drying under vertical airflow of laminar floor for 12-30 hours (MC $10.6{\~}7.2\%$). Decrease in germination percentage of ginseng seeds due to desiccation damage and freezing injury was observed at MC of below $7.2\%$ and of above $12.1\%$, respectively. Therefore, MC of ginseng seeds need to be controlled with a range of $8{\~}10\%$ to avoid damages from both desiccation and freezing.

Direct-seeded rice can have comparable yield with transplanted rice if its inherent problems can be solved. It is a labor-saving technology and can significantly reduce production cost because seedling nursery , pulling , and transplanting are omitted. Turnaround time between cropping is reduced hence the possibility of a third annual crop. But direct-seeded rice is very vulnerable to pest attack (by birds, rats, and golden snails), desiccation, weed infestation, and prone to lodging resulting to unstable crop establishment and inconsistent yield. These problems can be solved by anaerobic seeding (sowing pre-germinated seeds under the soil). It requires precise seed placement into the soil to optimize its benefits. We developed a four-row anaerobic direct seeder (US $ 200 commercial price) for this purpose . It consist of a structural framework mounted with a drum -hopper metering device, flotation type drivewheels, spring-loaded and adjustable furrow closers, and furrow open rs, and a plastic rainguard. It can sow in line pre-germinated seeds into the soil thus permitting the use of mechanical weeders for a chemical-free weed control. Its performance was comparable with the Japanese two-row anaerobic seeder (costing US$400) in terms of seed placement and crop establishment. It was tested with five cultivars. Seeding rate varied from 38 kg/ha to 80kg/ha. Crop establishment ranged from 64 to 99 percent while grain yield varied from 3.0 t/ha to 5.4t/ha. A six-row anaerobic seeder was also developed and adapted to a powertiller for increased capacity , field efficiency , and easier operation. The anaerobic seeder is useful to farmers shifting to direct seeding to reduce rice production cost and to researchers conducting agronomic studies in direct-seeded rice. A cDNA encoding desiccation-related protein was isolated from a flower bud cDNA library of Chinese cabbage (C-DH) and its nucleotide sequence was characterized. It contains 679 bp nucleotides with 501 bp open reading frame. The amino acid sequence of the putative protein showed the highest amino acid sequence homology (79 % identity) to dehydrin protein in Gossypium hirsutum. Also, the C-DH shares 48-52% amino acid sequence identity with the other typical dehydrin proteins in plant cells. When the amino acid sequence of their proteins were aligned, several peptide motifs were well conserved, of which function has to be solved. Particularly the C-DH contains 15 additional amino acids at its N-terminus. Genomic Southern blot analysis using the coding region of C-DH showed that the C-DH consists of a single copy gene in Chinese cabbage genome. The C-DH mRNA, whose transcript size is 0.7 kb, was expressed with a tissue-specific manner. It was highly expressed in seed, flower buds and low expression as detected in root, stem or leaf tissues of Chinese cabbage. And the transcript level of C-DH was significantly induced by the treatment of plant hormone, abscisic acid and water-deficit conditions.
BACKGROUND: Seed of perilla (Perilla frutescens var. japonica Hara) is short-lived in conventional storage conditions. For long-term conservation of plant species, cryopreservation is the method currently available. This study was performed to find out reliable methods for a long-term storage of seeds of perilla as a genetic resource. METHODS AND RESULTS: Using seeds of 9 perilla cultivars, the effects of desiccation, aging, and cryopreservation on seed germinability and ascorbate peroxidase activity in the seeds were investigated. Initial germinability of the seeds was various, and dry seeds of all cultivars survived cryopreservation without loss of viability. The highest germination was achieved at 4-5% moisture content, and stimulatory effect of cryogenic temperature on the seed germination was observed in some cultivars. Accelerated aging of perilla seeds led to reduction in germination and ascorbate peroxidase activity, and the susceptibility of seeds to aging was different among the tested cultivars. No significant difference in germination was observed for the aged seeds of control and liquid nitrogen exposed. CONCLUSION: The results of this study suggest that cryopreservation at 4-5% moisture content would be a suitable method for long-term conservation of perilla seeds without detrimental effects on germination.
The disaccharide trehalose ($\alpha$-D-glucopyranosyl-$\alpha$-D-glucopyranoside) is found in variety of organ-isms that are able to withstand almost complete desiccation. In order to identify the function of trehalose in plants, we isolated Arabidopsis trehalase (AtTRE) gene that encodes the enzyme able to hydrolyze trehalose to glucose, and trehalose-6-phosphate synthase isolog, TPS3 gene by RT-PCR. The AtTRE had the substrate specificity to hydrolyze only trehalose, and a broad pH range of enzyme activity. The AtTRE promoter/GUS reporter gene was expressed in cotyledons, mature leaf tissues including guard cells, and developing siliques. The GUS expression driven by AtTPS3 promoter was significant in root tissues, and the level of GUS activity was much higher than that of the pBll 21 control seedlings. The knockout of AtTPS3 gene in Arabidopsis resulted in the retarded root development, whereas the overexpression of AtTPS3 increased the root elongation in the presence of sucrose in MS medium. Possible functions of AtTRE and AtTPS3 in plant will be discussed. In addition, ectopic expression of yeast TPS1 driven by the inducible promoters in tobacco and potato conferred the plants on the drought and freezing tolerances.
This experiment was conducted on the fowl pox embryo vaccine for the production immunity, and stability, using an attenuated fowl pox virus (Nakano strin). Burnet's window method was applied, that is, 0.1 ml. of seed virus was inoculated on the chorioallantoic membrane of 12-day old chicken embryos, and incubated for 5 to 6 day, and then the result were read. Four kinds of suspensions of different embyo tissue were prepared and tested for the infectivity in chickens. Finally the suspension of chorioallantoic membrane was used as the vaccine throughout the experiment. Results obtained in this experiment are summarized as follows: (1) Of embryo tissue infected with the vaccine virus, chorioallantoic membrane had the highest virus titer of $10^{-5.4}$$EID_{50}$, and albumen the lowest titer of $10^{-0.7}$$EID_{50}$. (2) Suspensions of infected whole embryo with or without saline, and de-embryonated whole egg had about the same virus titer of $10^{-4.4}$$EID_{50}$, whereas the chorioallantoic membrane had $10^{-5.7}$ EID 50 or higher. The virus titer droped one log from $EID_{50}$ when inoculated into chickens. Takes were observed 35.6% of 500 chickens by stick method and 89% of 500 chickens by brush method. (3) The chorioallantoic membrane conferred almost perfect immunity for chickens by 10 days after vaccination. (4) Satisfactory immunity was observed in the chickens when eruption in a single follicle. (5) Eight of 10 vaccinated chickens revealed durable immunity for 307 days following vaccination. (6) The vacuum-dried vaccine maintained its infectiviy for 899 days at $5^{\circ}C$ or below and maintained the vius titer of $10^{-3.6}$$EID_{50}$. On the other hand, non-desiccated wet vaccine maintained the titer of $10^{-3.0}$$EID_{50}$ for 50 days of preservation period at $5^{\circ}$. However, in 50% glycerin-saline the infectivtiy of the same wet vaccine dropped to $10^{-1.5}$$EID_{50}$ (7) The vartation of virus titer of the vaccine before and after desiccation was $10^{-0.5}$$EID_{50}$ on the average. (8) As suspending media, 0.85 per cent saline and distilled water showed nearly the same effect on the infectivity of the vaccine by retaining the titer $10^{-3.0}$$EID_{50}$ after 50 days of preservation both at $5^{\circ}C$ and $20^{\circ}C$, while 50 percent cent glycerine-saline dropped the titer to $10^{-2.5}$ EID and $10^{-1.5}$$EID_{50}$ respectively at $5^{\circ}C$ and $2^{\circ}C$ after the same period.

Metagenomics is a powerful tool to isolate novel biocatalyst and biomolecules directly from the environmental DNA libraries. Since the metagenomics approach bypasses cultivation of microorganisms, un-cultured microorganisms that are majority of exists can be the richest reservoir for natural products discovery. To discover novel herbicidal substances from soil metagenome, we established three easy, simple and effective high throughput screening methods such as cucumber cotyledon leaf disc assay, microalgae assay and seed germination assay. Employing the methods, we isolated two active single clones (9-G1 and 9-G12) expressing herbicidal activity which whitened leaf discs, inhibited growth of microalgae and inhibited root growth of germinated Arabidopsis seeds. Spraying butanol fraction of the isolated active clones' culture broth led to growth retardation or desiccation of Digitalia sanguinalis (L) Scop. in vivo. These results represent that the screening methods established in this study are useful to screen herbicidal substances from metagenome libraries. Further identifying molecular structure of the herbicidal active substances and analyzing gene clusters encoding synthesis systems for the active substances are in progress.

Based on the differential growth response to exogenous gibberellic acid ($GA_{3}$) between semi-dwarf wheat(Triticum aestivum) and wild oat(Avena fatua), we examined the possibility of improving the selective performance of several herbicides by $GA_{3}$ application and the physiological background of $GA_{3}$-induced increase in herbicidal activity. Growth of wild oat was 4 to 5 times higher than that of wheat by $GA_{3}$ treatment. Pretreatment of wild oat seed with 300 ppm $GA_{3}$ increased the herbicidal activities of trifluralin and isoproturon by soil-surface application, but not of alachor and metsulfuron-methyl. $GA_{3}$ applied simultaneously with post-emergence herbicides resulted in a significant or moderate improvement of the efficacy of such herbicides as tralkoxydim, fenoxaprop-ethyl, metsulfuron-methyl, metribuzine and isoproturon, but not in the mixtures of oxyfluorfen or paraquat with $GA_{3}$. In the sequencial treatment of tralkoxydim and $GA_{3}$ at interval of one-day, $GA_{3}$ applied prior to tralkoxydim significantly increased a chlorosis and desiccation of leaf without affecting the growth inhibition by tralkoxydim. Tralkoxydim followed by $GA_{3}$ application had lower herbicidal activity than that of $GA_{3}$ followed by tralkoxydim treatment. Electrolyte leakage response of $GA_{3}$-pretreated or $GA_{3}$-untreated wild oat leaf against several compounds inducing membrane. peroxidation was compared. Differencial responses were observed in oxyfluorfen and isoproturon treatments with an increased electrolyte leakage in $GA_{3}$-pretreated tissue, but not in paraquat and rose bengal treatments. These results suggest that $GA_{3}$-induced increase in herbicidal activity is likely to be dependent on a herbicide type and may be due to activation of a metabolic ability related with herbicidal reponse as well as an increase in the herbicide absorbtion and translocation, rather than due to membrane and cell wall extention induced by $GA_{3}$, which in turn makes the herbicides easily enter.
