Phytotoxic Effect of Xanthium occidentale Leaf Extract on Seed Germination and Early Seedling Growth of Alfalfa and Barnyard Grass

  • Compositae plants are known to contain biologically active substances that are allelopathic to agricultural crops as well as weed species. Aqueous extracts from leaves of Xanthium occidentale were assayed against alfalfa (Medicago sativa) to determine their allelopathic effects, and the result showed that the extracts applied onto filter paper significantly inhibited seed germination as well as root growth of alfalfa. Untreated seeds germinated in 60h, but extract concentrations greater than 30g $\textrm{L}^{-1}$ delayed seed germination. The extracts significantly inhibited seed germination of alfalfa, and $\beta$-amylase activity of alfalfa and barley seeds during 24-36 hours after treatment. Aqueous extracts of 40 g $\textrm{L}^{-1}$ from X. occidentale were completely inhibited the hypocotyl and root growth of alfalfa. Aqueous leaf extracts showed the highest inhibitory effect and followed by root and stem extracts. Early seedling growth of both alfalfa and barnyard grass (Echinochloa crus-galli) was significantly reduced by methanol extracts. By means of high-performance liquid chromatography, chlorogenic acid and trans-cinnamic acid were quantified as the highest amounts from water and EtOAc fractions, respectively. BuOH and EtOAc fractions of X. occidentale reduced alfalfa root growth more than did hexane and water fractions. The findings of the bioassays for aqueous or methanol extracts reflected that the inhibitory effect of extract was closely related to the level of responsible allelochemicals found in plant extracts.

Bioassessment and Comparison of Toxicity of Arsenics based on the Results of Various Bioassays (다양한 생물 검정법에 근거한 비소의 위해성 평가 비교)

  • The acute toxicity of arsenic compounds was assessed and compared using following four bioassays; bioluminescence activity of the recombinant strain RB1436, germination of four different seeds, ${\alpha}$-glucosidase activity produced by Bacillus lichemiformis, acute genetic revertant mutation using mutant strain Salmonella typhimurium. Different sensitivities were observed among tested bioassays, but generally the toxicity by arsenite was greater than that of arsenate. Among tested four seeds, sensitivities of Lactucus and Raphanus were greater than others, and these two seed types were appeared as proper type for bioassay. High revertant mutation ratio (5.1) was observed with 1 mg/L arsenite, indicating high mutagenicity. The sensitivity of ${\alpha}$-glucosidase activity on arsenic compounds was much lower than other methods. The evaluation of interactive toxic effects using various bioassays may comprise a useful tool for the bioassessment of environmental pollutants.

Isolation of a New Herbicidal Compound Angelicin from Curly Dock (Rumex crispus L.) (소리쟁이(Rumex crispus L.)로부터 신규 살초활성물질 Angelicin의 분리)

  • The objective of this study was to isolate a herbicidally active compound from curly dock (Rumex crispus), a native weed in Korea and to identify its' chemistry. The $GR_{50}$ value of methanol extracts which is determined by a seed bioassay using rapeseed (Brassica napus) seedlings was $935\;{\mu}g\;g^{-1}$. Activity-directed isolation of ethylacetate extract led to the isolation of ECDA fraction with $GR_{50}$ value $53\;{\mu}g\;g^{-1}$. Based on data of GC-MS, $^1H$-NMR and $^{13}C$-NMR, the chemical structure of ECDA was determined as 2H-furo[2,3-H]-[1]-benzopyran-2-one which is known as angelicin. The $GR_{50}$ values of angelicin to barnyardgrass (Echinochloa crus-galli), southern crabgrass (Digitaria ciliaris), and indian jointvetch (Aeschynomene indica) were 426, 66 and $216\;{\mu}g\;g^{-1}$, respectively. Our results suggest that angelicin could be used as a lead compound for the development of new herbicides.

Purification and Identification of Antioxidant Compounds from Dolichos lablab L. Seeds (백편두의 항산화 물질 분리 및 동정)

  • Background: This study aimed to identify antioxidant compounds from the seeds of Dolichos lablab L. by bioassay-guided isolation and recrystallization. Methods and Results: The water layer of D. lablab L. seed extract inhibits intracellular reactive oxygen species (ROS) expressing the 2',7'-dichlorofluorescein diacetate (DCF-DA), Cu/Zn superoxide dismutase (SOD) and catalase genes, as determined by quantitative real-time PCR (qRT-PCR). Two compounds were purified from the water layer of the seeds of D. lablab L. using column chromatography and prep-high performance liquid chromatography (HPLC). Using nuclear magnetic resonance (NMR) and electrospray Ionization mass spectrometry (ESI-MS), their chemical structures were identified as 5-[(2-acetyl-2,3-dihydro-1H-indazol-1-yl)carbonyl]-4,5-dihydro-3H-furan-2-one (C14H14N2O4) and stachyose. Conclusions: Two active antioxidant compounds were purified from the seed extract of D. lablab L. seed extract and the structures of these compounds were identified as C14H14O4N2 and stachyose.

  • This study was carried out to investigate the sea sonal changes of the contents of inhibitors in leaves and fruits of Ginseng plant during ripening. Three kinds of inhibitors in leaves and all parts of fruit, i.e., seed, sarcocarp and endocarp were recognized at the Rf 0.1, 0.4-0.6 and 0.8-1.0 zones by the bioassay of lettuce seed germination. Among them, the level of the inhibitor at the Rf 0.4-0.6 zone in leaf and seed increased most significantly in accordance with fruit ripening. The activities of three inhibitors found in endocarp gradually decreased during ripening.

Insecticidal Activity of Essential Oils against Larvae of Culex pipiens pallens (빨간집모기(Culex pipiens pallens) 유충에 대한 식물오일의 살충효과)

  • Larvicidal activity of 34 plant essential oils were tested against the 4th instar larvae of Culex pipiens pallens. Among them, six oils (basil, fennel, caraway seed, lime, thyme red, and thyme white) showed more than 80% mortality at 100 mg $litre^{-1}$ concentration. GC and GC-MS analyses of the six essential oils and bioassay of their components revealed that anethole (a major component of fennel and basil oil) and (+)-limonene (a major component of caraway seed and lime oil) showed higher larvicidal activities than others. In addition to the above two components, ${\alpha}$-phellandrene, p-cymene, $\gamma$-terpinene, ${\beta}$-pinene, and thymol showed higher larvicidal activity than Bt product.

  • To analyze the relationship between CGMMV occurrence and seed contamination by cucumber cultivars, cucumber cultivars of CGMMV infected field and diseased degree were investigated in cucumber main cultivation regions of Jeonnam province from 1999 to 2002. While the diseased degree was different by years and cultivars, CGMMV occurred for 4 years in 'Janghyeongnakhap', for 3 years in 'Jangjukcheongjang', and for 2 years in 'Gyeoulsali' and 'Gyeoulnagi'. CGMMV was detected in seeds of 'Janghyeongnakhap' and 'Jangjukcheongjang' by ELISA test, and seed of 'Jangjukcheongjang' showed positive reaction in bioassay. As a result of resistant test of 34 cultivars to CGMMV, all cultivars showed mosaic symptoms in pot test but only 'Hanboksamcheok' showed mild mosaic symptom in field test.

Autotoxicity of alfalfa flower extract and its allelopathy to Echinochloa crus-galli (알팔파 꽃 추출물의 Autotoxicity와 돌피에 대한 Allelopathy)

  • The aim of this study was to separate or purify some bioactive compounds from flowers of alfalfa(Medicago sativa L.) and to test of the isolated compounds on alfalfa for their autotoxicity and on Echinochloa crus-galli for their allelopathy for seed germination and seedling weight. Using thin layer chromatography(TLC) of $CHCl_3$ extracts, the most inhibitory band to alfalfa seed germination was determined. Germination inhibition of this extract suggested a complex chemical interaction. Separation and purification of compounds with CHCl$_3$ extract of fresh alfalfa flowers were conducted by a silica gel TLC, and microcrystalline cellulose TLC(MCTLC), followed by droplet countercurrent chromatography(DCCC) bioassay. Preliminary identification was done by high perfomance liquid chromatography(HPLC) on the most inhibitory fractions in DCCC. Ferulic acid, caffeic acid, vanillic acid, rutin, narringin were identified in fraction 5 and ferulic acid, caffeic acid, vanillic acid, rutin, coumarin in fraction 6. The phytotoxicity of their individual compound was tested on alfalfa and Echinochloa crus-galli seed germination and seedling weight. Coumarin and ferulic acid showed the most inhibitory effect on alfalfa seed germination and Echinochloa crus-galli seedling fresh and dry weight. These compounds may be, at least in part, involved in autotoxicity and allelopathy.

Toxicity Assessment of Nanopariticles Based on Seed Germination and Germination Index (씨앗발아 및 발아지수에 근거한 나노입자 독성평가)

  • Nanomaterials have been widely used in many fields. This study investigates the effects of metal oxide nanoparticles (CuO, NiO, $TiO_2$, $Fe_2O_3$, $Co_3O_4$, ZnO) on germination and germination index (G.I.) of seeds, Lactuca and Raphanus. Under aqueous exposure, CuO on Lactuca shows the most significant impacts on activities compared to others, showing $EC_{50s}$ for germination and G.I. as 0.46 mg/L and 0.37%, respectively. The effects of nanoparticle phytotoxicity on seed Lactuca was much higher than that of Raphanus. In general, the toxicities on seed germination and germination index were as following orders : CuO > ZnO > NiO ${\gg}$ $TiO_2$, $Fe_2O_3$, $Co_3O_4$. No measurable inhibition was observed at 1,000 mg/L (maximum exposure concentration) of $TiO_2$, $Fe_2O_3$, $Co_3O_4$.

Effect of Growth Medium and Extract Sterilization on a Seedling Bioassay of Alfalfa Autotoxicity (생장배지 및 추출액의 멸균이 알팔파 Autotoxicity 의 유묘 생물검정에 미치는 영향)

  • Autotoxic substance(s) from alfalfa(Medicago sativa L.) plants reduces germination and growth of adjacent new alfalfa after alfalfa. The autotoxic chemical(s) in alfalfa are clearly unknown. Our objective was to improve the sensitivity of an alfalfa seedling bioassay for evaluating autotoxic leaf extracts. We determined critical extract concentrations that inhibit seed germination and seedling growth, compared two different culture media, and evaluated the effects of extract sterilization on the sensitivity of the assay, by using streptomycin and autoclaving method. An agar medium in petri plate gave better responses of germination and seedling growth to the extracts than using filter paper in the plate. On agar medium, the concentration of extract required to reach 50% inhibition of root length was 2.7 g $kg^{-1}$, and of germination and hypocotyl length were 3.8 and 9.9 g $kg^{-1}$, respectively. Leaf extracts with 100 ppm streptomycin stimulated germination significantly compared to Leaf extract alone but reduced root length of control by 43%. Root length was more sensitive to the autotoxin(s) than was germination or hypocotyl length. These results suggest that agar medium mixed with extract and sterilization by autoclaving could be improved the consistency and precision of bioassay, and that root length was the best parameter of autotoxic effect of alfalfa leaf extract.
