  • The outbreak of viral diseases caused by viral haemorrhagic septicaemia virus (VHSV) and red seabream iridovirus (RSIV) have been reported in cultured olive flounder, Paralichthys olivaceus. VHSV has been a serious viral disease that infects the olive flounders in South Korea. Clinical signs of VHSV infection are skin darkening, abdominal distension and haemorrhages. Outbreaks of fish iridoviral disease was first reported from red seabream, Pagrus major farms in Japan. Recently, iridovirus infection have occurred frequently from olive flounder farms in South Korea. In this study, disease surveillance was performed to monitor the prevalence of VHSV and RSIV in olive flounder in 2014. The samples were collected from 60 different olive flounder farms in Jeju from April, May, September, November and December in 2014. RT-PCR (VHSV) or PCR (RSIV) results showed that VHSV were detected in 5 farms, but RSIV has not been detected in any farms. The migration of olive flounder was restricted for the quarantine in 5 farms of VHS outbreak. The nucleocapsid protein (N) gene and glycoprotein (G) gene sequences of the 5 Korean VHSV isolates were successfully amplified and sequenced. Phylogenetic analysis was performed using the VHSV sequences reported here together comparison with the nucleotide sequences available from the GenBank database. Phylogenetic analysis indicated that most of Korea VHSV belong to the genotype IVa and closely related to the strains from Japan and China.

  • The maturation and spawning of Black seabream Acanthopagrus schlegeli were investigated using samples randomly collected in the Southern Sea of Korea from March 2007 to February 2008. The FL at 50% sex-changers maturity was estimated to be 26.7 cm. We estimated monthly changes in maturity stages and gonadosomatic index (GSI). The spawning period ranged from March to July, and the peak spawning occurred in May and June. The fecundity (F) was 839,600 eggs at 32.2 cm fork length (FL) to 2,894,913 eggs at 42.0 cm FL, and the relationship between FL and F was expressed as $F=1.264FL^{3.869}$ ($R^2=0.736$). The FL at 50% group maturity was estimated to be 25.8 cm for females and 19.0 cm for males. The sex ratio was 19.9% for female, 20.9% for early sex-changers, 28.7% late sex-changers, 30.5% for male.

  • On the southern coast of South Korea, dark-banded rockfish, sea bass, and red seabream are caught by single-line fishing. In particular, red seabream and sea bass are caught in the Jeollanam-do region using gear with attached fishing hooks, such as longline and single-line gear, with live shrimp as bait. The objective of this study was to compare the catch efficiency of two types of live shrimp (naturally grown Shiba shrimp [Metapenaeus joyneri] and cultured whiteleg shrimp [Litopenaeus vannamei]) used as bait. The investigation included interviews, on-board surveys, and water tank experiments. Interviews were conducted with relevant parties to determine the preference for live shrimp as bait, and the results showed a greater preference for cultured whiteleg shrimp. Further, an on-board survey was conducted to compare catch efficiency between these two types of live shrimp bait for single-line fishing. The on-board investigations were conducted once or twice a month between June and October. In total, the amounts of fish caught using naturally grown Shiba shrimp and cultured whiteleg shrimp were 56 and 52, respectively. Of these, the numbers of sea bass, the primary target fish species, caught using naturally grown Shiba shrimp and cultured whiteleg shrimp were 43 and 40, respectively. Thus, the results showed that there was almost no difference in the number of fish caught based on the bait used. However, according to a water tank experiment, cultured whiteleg shrimp survived longer than naturally grown Shiba shrimp.

  • CD96 is a membrane-bound receptor discovered in humans in 1992 that is mainly present in natural killer cells and T cells derived from haematopoietic cells and performs immune functions. Based on the sequence of CD96 obtained from red seabream (Pagrus major), phylogenetic analysis with other species, infections of normal fish, Streptococcus iniae and red sea bream iridovirus (RSIV), and expression analysis was conducted using real-time polymerase chain reaction. Phylogenetic analysis showed the highest homology with Sparus aurata, and multiple sequence analysis confirmed the conservation of major domains between different fish species. Normal fish high expression results were confirmed in the head kidney, and spleen, which are the haematopoietic organs of the fish. High expression levels were confirmed in the gills, liver, spleen, and kidney on day three after RSIV infection. After S. iniae infection, high expression was confirmed in the gills and liver on day one, and high expression was confirmed in the spleen from 12 hours. These results show that PmCD96 functions as an immune gene in P. major and is considered a basic research case for CD96 in fish's hematopoietic organ immune system.

  • FK506BP (FK506 binding protein 12) is a small peptide with a single FK506BP domain. It is involved in suppression of immune response, oxidative stress and inflammation. The purpose of this study was to investigate the gene expression of FK506BP in red seabream (Pagrus major) exposure to low water temperature (8℃, 33 psu) and low salinity (20℃, 10 psu). Results showed that, the expression of FK506BP was significantly increased in the experiment groups, such as low water temperature (8℃, 33 psu), and low salinity (20℃, 10 psu). These results suggest that FK506BP was played roles in biomarker gene on the environmental stress such as water temperature and salinity.

  • Interleukin-1 (IL-1) is one of the proinflammatory cytokines, after IL-1 binds to IL-1RI, IL-1RacP (interleukin-1 receptor accessory protein) joins with IL-1/IL-1RI to form a complex, and leading to cell activation. IL-1RAcP is involved in immune response, stress and apoptosis. The purpose of this study was to investigate the gene expression of IL-1RAcP in red seabream (Pagrus major) exposure to low water temperature (8℃, 33 psu) and low salinity (20℃, 10 psu). Results showed that, the expression of IL-1RAcP was significantly increased in the experiment groups, such as low water temperature (8℃, 33 psu), and low salinity (20℃, 10 psu). • Comparison of blood properties in juvenile black seabream (Acanthopans schlegeli) between transfer from seawater to freshwater (sFW) and transfer from freshwater to seawater (fSW) were investigated for 60 days. Plasma cortisol levels in sFW were significantly increased from 34.2 $\pm$ 28.6 ng/mL at the beginning to 365.6 $\pm$ 136.0 ng/mL at 3 hours, and decreased to the beginning level at 24 hours. However, in fSW, no significant differences in the cortisol levels were recognized throughout experimental period. No significant difference was found in the glucose levels between sFW and fSW. The transfer from seawater to freshwater of juvenile black seabream resulted in reduced $Na^{+}\;and\; Cl^{-}$ concentrations for 24 hours (decreasing from 165.3 $\pm$ 2.5, 146.0$\pm$ 2.2 to 118.3 $\pm$ 12.3, 78.0$\pm$ 7.0 mEq/L, respectively), but these were completely recovered the beginning levels at 30 days. Total protein and AST showed no significant differences between the two rearing conditions, while ALT was markedly elevated at 3 hour in sFW and at 24 hours in fSW. Ht in sFW was increased from 18.5 $\pm$ $0.6\%$ at the beginning to 25.3 $\pm$ $4.0\%$ at 12 hours, and was decreased to the beginning level at 24 hours. Ht, RBC and Hb in fSW were also significantly higher at 12 hours, but recovered to their initial levels at 24 hours. All fish were dead until 50 days in sFW while survival rate in fSW was $85\%$ at the end of experiment.

  • This study aims to analyze the market interactions among the main farmed fish species in Korea, using both multivariate and bivariate cointegration analysis. For the analysis of market interactions among farmed fish species, major four farmed fish species, olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), red seabream (Pagrus major), and grey mullet (Mugil cephalus) were selected as the analytical target species. And their real price data by month from January 2000 to December 2011 were used in the analysis. The results of the multivariate cointegration test for four farmed fish showed that there would be no long-term equilibrium relationships among farmed fish species, and consequently they do not share the same market. The results of bivariate cointegration test indicated that there was little evidence to suggest that all farmed fish species were cointegrated each other. However, it was only analyzed that olive flounder and grey mullet might have a long run equilibrium relationship.

  • Two parasitic copepods, poecilostomatoid Colobomatus mylionus Fukui, 1965 and siphonostomatoid Clavellisa chinensis ($Y{\ddot{u}}$, 1933), were reported for the first time from Korea and redescribed based on the females collected from the marine fish of Korean waters. The C. mylionus was collected from the blackhead seabream, Acanthopagrus schelegelii(Bleeker). Colobomatus mylionus had been frequently reorded from marine teleost fishes from of Eden, Newcastle, Brisbane, Yelloon, and Point Sampson. The second record was C. chinensis redescribed based on the speciemens collected from the gills of a Japanese grenadier anchovy, Coilia nasus Temminck and Schlegel. Some differences were observed between the orginal description of known specimens examined in this study, these differences do not warrant a change in the taxonomic staus of the two species. The attachment site of all two copepods reported from Korea were the cephalic sensory canals and gills.

  • In order to improve the present running seawater system for seedling production of marine fishes, rearing experiments with the juveniles of black seabream (Acanthopagrus schlegeli) and oblong rockfish (Sebastes oblongus) were conducted. The recirculating seawater system (F) equipped with the rotating biological contactors and the running seawater system (R) were used. Environmental factors, growth, survival rate and rearing density of the fish were compared between two systems during the experimental period. In the rearing experiment of black seabream, water temperature in F was fluctuated with surrounding air temperature and was higher than that in R. Specific gravity of the rearing seawater in F was relatively stable in the range of 1.0252 to 1.0266, while that in R was greatly fluctuated. pH in F turned out to be 7.51, but pH in R was similar to that in the natural seawater. Dissolved nitrite and nitrate in F were higher than those in R. While no significant differences in the growth of black seabream juveniles were recognized between two systems, survival rates of fishes in F were higher than those of fishes in R. In the rearing experiment of oblong rockfish, water temperature in F was higher than that in R. Specific gravity in F was slightly higher than that in R which showed relatively less fluctuation in the range of 1.0253 to 1.0270. pH in F turned out to be 7.96, but pH in R was similar to that in the natural seawater. Dissolved nitrite and nitrate in F were higher than those in R, but ammonia was lower in F. The juveniles of oblong rockfish reared in F grew significantly faster in their total length and body weight than those in R (P<0.05). However, survival rates of fishes between two systems showed no significant differences.
