Effects of Natural and Chemical Disinfectants to Epithelium and Endothelium of Rabbit's Cornea - Scanning Electron Microscopy (천연보존제와 화학보존제가 가토안의 각막 상피, 내피에 미치는 영향- 주사전자현미경적 관찰)

  • The cytotoxicity that chemical disinfectants of K products and natural disinfectants of naringin & chitosan well-known preservative, have on endothelium and epithelium of rabbit's cornea were obseved by scanning electron microscope. The main component of naringin is grapefruit seed extract, which is one of the flavonoid widely recognized as natural antioxidants and used as preserved food and cosmetics. The chitosan also wildly distributed epithelium of crustacea, epidermis of insects, mould, fungi and so on. It is no harm of cytotoxicity of human body and recognized of antibacterial for various of bacteria. This study was performed to examine the cytotoxicity of natural and chemical preservative.

Shape-dependent Adhesion and Friction on Au Nanoparticles Probed with Atomic Force Microscopy

  • Shape control of metal nanocrystals has broad applications, including catalysis, plasmonics, and sensing. It was found that controlling the atomic arrangement on metal nanocrystal surfaces affects many properties, including the electronic dipole or work function. Tuning the surface structure of exposed facets of metal nanocrystals was enabled by shape control. We investigated the effect of shape on nanomechanical properties, including friction and adhesion forces. Two nanoparticles systems, high-index {321} and low-index {100}, were used as model nanoparticle surfaces. Scanning force microscopy was used to probe nanoscale friction and adhesion. Because of the abundant presence of high-density atomic steps and kinks, high-index faceted nanoparticles have a higher surface energy than low-index faceted cubic nanoparticles. Due to this high surface energy, high-index faceted particles have shown stronger adhesion and higher friction than low-index nanoparticles. We discuss the results in light of the differences in surface energy as well as the effect of capping layers in the measurement.

  • Filtration by fibrous filter is one of the Principle methods used for removing pollutant particles in the liquid. Because of the increasing need to protect both human health and valuable devices from exposure to fine particles, filtration has become more important. Filters have been developed with modified surface charge characteristics to capture and adsorb particles by electrokinetic interaction between the filter surface and particles contained in water. The main purposes of this study are to develop and evaluate the performance evaluation of the apparatus for making a positively charged porous filter media and to analyze the surface characteristics of the filter media for capturing negavitely charged contaminants mainly bacteria and virus from water. The experimental apparatus consists of a mixing tank, a vacuum pumping system, a injection nozzle, a roller press and a controller. The filter media is composed of glass fiber(50-750 nm), cellulose($10-20{\mu}m$) and colloidal charge modifier. The characteristics of filter media is analyzed by SEM(Scanning Electron Microscopy), AFM(Atomic Force Microscopy) and quantified by measuring the zeta potential values.

Differentiation of the Fetal Rat Pulmonary Epithelial Cells in Organotypic Culture (기관형 배양에서 흰쥐 태자 폐상피세포의 분화)

  • In order to study the differentiation of the epithelial cells during the development of fetal rat lung tissue, histological changeB in organotypic culture and in vivo were examined. Light microscopy and scanning electron microscopy were used to analvre the histological change in rat lung from the 15th nary of gestation to the 111th nary after birth. In organotypic culture system, the pulmonary epithelial cell differentiation was studied by scanning electron microscopy. The results obtained from this study were as follows. 1. During deveiopment of lung, the glandular stage lasted from the Isth day to the lsth naut of gestation; the canalicular stage from the 17th nay to the 19th naut of gestation; the saccuiar stage from 20th nary to the birth. Alveolar stage was observed at the 3rd nary of postnatal rat lung. 2. In organotvpic culture of fetal rat lung cells organized alveolar-like structures resembling those of in uiuo state were observed on the gelatin matrix. In contrast with in vivo state, fetal lung cells formed group of type ll pneumocytes predominently along the contours of the matrix. These cells have large apical surface, short microvilli and secreted materials which may be sunactant. These results suggested that an orsanotypic culture retaining epithelial- -mesenchvmal relationships is appropriate culture model to study the pulmonary epithelial cell (especially type ll pneumocvte) differentation.

Analysis of dislocation density in strain-hardened alloy 690 using scanning transmission electron microscopy and its effect on the PWSCC growth behavior

  • The dislocation density in strain-hardened Alloy 690 was analyzed using scanning transmission electron microscopy (STEM) to study the relationship between the local plastic strain and susceptibility to primary water stress corrosion cracking (PWSCC) in nuclear power plants. The test material was cold-rolled at various thickness reduction ratios from 10% to 40% to simulate the strain-hardening condition of plant components. The dislocation densities were measured at grain boundaries (GB) and in grain interiors of strain-hardened specimens from STEM images. The dislocation density in the grain interior monotonically increased as the strain-hardening proceeded, while the dislocation density at the GB increased with strain-hardening up to 20% but slightly decreases upon further deformation to 40%. The decreased dislocation density at the GB was attributed to the formation of deformation twins. After the PWSCC growth test of strain-hardened Alloy 690, the fraction of intergranular (IG) fracture was obtained from fractography. In contrast to the change in the dislocation density with strain-hardening, the fraction of IG fracture increased remarkably when strain-hardened over 20%. From the results, it was suggested that the PWSCC growth behavior of strain-hardened Alloy 690 not only depends on the dislocation density, but also on the microstructural defects at the GB.

Synthesis of ZnO/Zn(OH)2 Nanosheets Using Ionic Layer Epitaxy (이온층 에피택시법을 이용한 ZnO/Zn(OH)2 나노시트의 합성)

  • ZnO nanosheets have been used for many devices and antibacterial materials with wide bandgap and high crystallinity. Among the many methods for synthesizing ZnO nanostructures, we report the synthesis of ZnO/Zn(OH)2 nanosheets using the ionic layer epitaxy method, which is a newly-developed bottom-up technique that allows the shape and thickness of ZnO/Zn(OH)2 nanosheets to be controlled by temperature and time of synthesis. Results were analyzed by scanning electron microscopy and atomic force microscopy. The physical and chemical information and structural characteristics of ZnO/Zn(OH)2 nanosheets were compared by X-ray photoelectron spectroscopy and X-ray diffraction patterns after various post-treatment processes. The crystallinity of the ZnO/Zn(OH)2 nanosheets was confirmed using scanning transmission electron microscopy. This study presents details of the control of the size and thickness of synthesized ZnO/Zn(OH)2 nanosheets with atomic layers.

Morphological and molecular investigation of Melosira octogona Schmidt (Melosirales, Bacillariophyta) isolated from the Jeju Island

  • Melosira octogona Schmidt has been documented primarily through illustrations and light microscopy photographs, resulting in incomplete morphological descriptions. In this study, the morphological characteristics of M. octogona were investigated using scanning electron microscopy, and the small subunit rDNA of the strain isolated from the coast of Jeju Island, South Korea, was analyzed for the first time. The cells were organized in short chains, with the frustules taking on an octagonal shape in the girdle view because of the straight, obliquely oriented valve margin. The cells were joined together in specific pairs by a cingulum. Imaging analysis via SEM revealed the presence of a hemispherical valve with a well-developed corona on the valve face, along with stellate thickening and rimoportulae. The epicingulum was composed of six ligulate bands punctuated by rows of circular to elongated pores. Genetic analysis of the 18S rDNA sequence suggested a close relationship between M. octogona and Melosira sp. (KJ961704), demonstrating a high degree of similarity. Meanwhile, it exhibited comparatively low similarity (99.0 %) to Melosira cf. octogona (AY485518). The results of our analysis can enhance the precise recognition of M. octogona and enrich our understanding of its phylogenetic position within the order Melosirales.

  • Next generation lithography technologies, such as EBL(Electron Beam Lithography), X-ray lithography, SPL(Scanning Probe Lithography), have been studied widely for getting over line width limitation of photolithography. Among the next generation lithography technologies, SPL has been highlighted because of its high resolution advantage. But is also has problem which are slow processing time and sample size limitation. The purpose of this study is complement of present SPL system. Brand new SPL system was made. SPL test was performed with the system in ultra thin PMMA(polymethlymethacrylate) film.

  • In a scanning electron microscopy, image distortion is a critical issue and it is needed to be eliminated by some kinds of schemes. In this work, scanning frequency and scanning wave form are adjusted to have an improved image. The relationship between scan coil and its driver is investigated and appropriate frequency and wave form are suggested. It is proved that the selected frequency and wave form showed an enhanced image with less distortion, which were done by experiments. In addition, a noise elimination is addressed, providing improved image with a GROUND signal integration with the amplifier and the scan driver.

  • We attempt to investigate morphology of self-assembled dipyridinium dithioacetate on Au(111) substrate by Scanning Tunneling Microscopy(STM). Also, we measured electrical properties using Scanning Tunneling Spectroscopy(STS). Sample that use this experiment acquires thiol function beside quantity by dipyridinium dithioacetate, is structure that can be self-assembled easily to Au(111) substrate. The same self-assembly procedure was used for two different concentrations, 0.5mmol/ml and 1mmol/ml. Dilute density of sample by 0.5mmol/ml, 1mmol/ml and observed dipyridinium dithioacetate's image by STM after self-assembled on Au(111) substrate. The structure of Tip/SAMs/Au(111) has been used measurement for electrical properties(i-v) using STM. The current-voltage measurement result, observed negative differential resistance(NDR) properties.
