A Study on Shelf-life of Seasoned and Fermented Squid (오징어 조미젓갈의 품질유지기한에 관한 연구)

  • Traditional fermented fish products 'Jeodtgal' have been prepared with addition of about $25\%$ salt to prevent spoilage, but domestic seasoned and fermented products have only about $8\%$ salt, so they have been required careful treatment during distribution and could have a different fermentation pattern from traditional 'Jeodtgal' This study was conducted to establish shelf-life of seasoned and fermented squid at different distribution conditions. To establish shelf-life of seasoned and fermented squid stored at $10^{\circ}C,\;20^{\circ}C$ and $30^{\circ}C$, chemical components and sensory evaluation were investigated. Chemical components such as volatile basic nitrogen(VBN), ammonia nitrogen ($NH_3-N$) and total nitrogen(T. N) rapidly increased during storage at $20^{\circ}C$ and $30^{\circ}C$, but pH value and Hunter color value(${\Delta}E$) slowly decreased during storage without respect of storage temperature. Judging from chemical components analysis and senseory evaluation, seasoned and fermented squid could be storage for 3days at $30^{\circ}C$, 7days at $20^{\circ}C$ and 25days at $10^{\circ}C$, respectively. The ratio of quality deterioration per hour were 0.00166 at $10^{\circ}C$, 0.00595 at $20^{\circ}C$ and 0.01385 at $30^{\circ}C$.

Effect of Sodium Chloride Intake Related to the Composition of the Diet (식이조성(食餌組成)에 따른 식염섭취량(食鹽攝取量)에 관(關)한 연구(硏究))

  • Sodium chloride plays an important role as the main condiment at daily meal. It is well known that humans require sodium chloride as an essential nutrient to keep the homeostasis of electrolytes. The amounts of salt intake may be a reflection of geography, culture and food habit rather than necessity. Lee has reported (1962) that Koreans ingest high amounts of sodium chloride in their meals, with an intake of excess carbohydrate (80-90% of total Calories) and low protein in their diet. This includes large amounts of rice, Kimchi and other fermented soybean products common in the Korean diet. This investigation was designed to study the dietary relations of sodium chloride to other nutrients in the Korean diet. Twenty four albino male rats, weighing from 290-300g, were divided into four dietary groups according to the amounts of carbohydrate, protein and fat in the basal diet. Each diet contained a rice powder as a carbohydrate source. Diet I was a control diet, Diet II, low protein, Diet III, low protein and low fat diet and Diet IV, low fat diet. All rats were provided with 3% sodium chloride solution. Diet and salt solution were given ad libitum. The experiment was carried out for 9 weeks during which time the body weight, the food intake, and 3% sodium chloride solution consumption were determined. At the 9th week, the urine was collected the blood sample from the artery of each rat for the analysis of sodium and potassium and other chemical studies. The rats were sacrificed and the kidney, adrenal, liver and spleen were measured, and observed changes of the pathological tissue in the kidney and adrenal. The results were summarized as follows: 1) The growth rate was higher in Diet I than in the other experimental diets (II, III and IV) after 4 weeks. There was no significant difference found between the experimental Diets II, III and IV. 2) The daily food intake was greater in the experimental diets II, III and IV than in the control diet. However, there was no difference among the high carbohydrate diets Diet II, III and IV. 3) The daily water (3% sodium chloride solution) intake was also greater in the Diets II, III and IV, than in the control diet. However, there was no difference between Diets II, III and IV. 4) The concentration of sodium and potassium in the blood were within the normal range in all diets. 5) The amount of sodium chloride in the urine was significantly greater in Diets II, III and IV than in the control diet. Diets II, III, IV had a larger amount of sodium solution consumption. 6) Observation of pathological tissue in the experimental diets found a cell proliferation in the glomerlulus of the kidney, while such change was not found in the control diet.

Effect of Na Salt on the Formation of MgO Obtained from Mg(OH)2 by Precipitation Method (침전법으로 제조된 Mg(OH)2의 잔류 Na염이 MgO 입자 형성에 미치는 영향)

  • The particle size of MgO was examined as a function of the Na content in $Mg(OH)_2$ powders and the calcination temperature. $Mg(OH)_2$ suspension was obtained by dropwise precipitation of $Mg(NO_3)_2{\cdot}6H_2O$ and NaOH solutions. The suspension was diluted by varying the dilution volume ratio of distilled water to $Mg(OH)_2$ suspension to change the Na salt concentration in the suspension. $Mg(OH)_2$ slurry was filtered and dried at $60^{\circ}C$ under vacuum, and then its $Mg(OH)_2$ powder was calcined to produce MgO with different amount of Na content at $500\sim900^{\circ}C$ under air. Investigation of the physical and chemical properties of the various MgO powders with dilution ratio and calcination temperature variation was done by X-ray diffraction, transmission electron microscopy, BET specific surface area and thermal gravimetric analysis. It was observed that MgO particle size could depend on the condition of calcination temperature and dilution ratio of the $Mg(OH)_2$ suspension. The particle size of the MgO depends on the Na content remaining in the $Mg(OH)_2$ powder, which powder was prepared by changing the dilution ratio of the $Mg(OH)_2$ suspension. This change increased as the calcination temperature increased and decreased as the dilution ratio increased. The growth of MgO particle size according to the increase of temperature was more effective when there was a relatively high content of Na. The increase of Na content lowered the temperature at which decomposition of $Mg(OH)_2$ to MgO took place, thereby promoting the crystal growth of MgO.

Isolation and Characterization of a Formate Dehydrogenase cDNA in Poplar (Populus alba ${\times}$ P. glandulosa) (현사시나무에서 Formate Dehydrogenase cDNA의 분리와 특성 구명)

  • Formate dehydrogenase (FDH), catalyzing the oxidation of the formate ion to carbon dioxide, is known as the stress protein in response to drought, low temperature and pathogen infection. To study the functions of FDH in poplar (Populus alba ${\times}$ P. glandulosa), we isolated a FDH cDNA (PagFDH1) and examined its expressional characteristics. The PagFDH1 is 1,499 base pairs long and encodes a putative 388 amino acid protein with an expected molecular mass of 42.5 kDa. The PagFDH1 protein has N-terminal mitochondria signal peptide and $NAD^+$ binding domain. Southern blot analysis indicated that a single copy of the PagFDH1 is present in the poplar genome. PagFDH1 is expressed highly in the suspension cells (especially in the lag and early exponential phases) and moderately in roots, flowers and leaves. ABA-mediated enhanced expression of PagFDH1 in response to drought and salt stress treatments indicates that the gene product could play an important role in the development of stress resistant trees.

Growth and Yield of Strawberry $(Fragaria\;\times\;ananassa\;Duchesne)$ 'Nyoho' and Salt Accumulation in PE Film House Soil as Affected by Fertilization Program (시비방법이 무기염의 토양집적과 딸기 생육 및 수량에 미치는 영향)

  • This research was conducted to determine the effect of fertilization programs on crop growth, nutrient uptake and yield of strawberry and salt accumulation in PE film house soil. To achieve these, experiments were conducted through two years with various fertilization program such as traditional fertilization(A), recommended fertilization of Rural development administration (B), Tochigi prefectural experiment station (C) and Nonsan strawberry experiment station (D), and control (E). In 1st year, statistical differences in growth characteristics were not observed among treatments except (E). Yield per 10 was the highest in (D) followed by (A), (B), (C) and (E) and there was the statistical difference between (D) and (E). In the 2nd year experiment, growth characteristics and yield showed similar trends to those of 1st year experiment. In the results of soil analysis of 2nd year, the soil pH of all treatments were in the acceptable range, while electrical conductivity of (A) and (D) were 2.36 and 2.19 %$dS{\cdot}m^{-1}$, respectively. After finishing of strawberry cultivation, nitrate concentration in soil solution of (A) and (D) were 74.6 and 65.0%$mg{\cdot}L^{-1}$ and Na in those were 3.71 and 3.53 $cmol+kg^{-1}$, respectively. Above results indicated that (A) and (D) were good fertilization program for strawberry cultivation, but those also resulted in accumulation of $NO_{3-}N$ and Na in PE house soil.

PARAMYOSIN OF THE ABALONE, NOTOHALIOTIS DISCUS (전복 Paramyosin의 분리 및 그 성질)

  • The protein composition of abalone muscle was estimated with the following result: on a series of samples analyzed, water-soluble protein, $19\~22\%$, salt-soluble protein, $27\~39\%$: alkali-soluble protein, $20\~26\%$ : and stroma $20\~28\%$ : respectively. It was demonstrated by ultracentrifugal analysis that approximately $65\%$ of the salt-soluble protein is accounted for by paramyosin, $30\%$ by actomyosin, and $5\%$ by myosin, respectively. The ultracentrifugally homogenous paramyosin was prepared by BAILEY's ethanol-dried method. It showed a $S^{\circ}\;_{20,\;{\omega}$ of 3.14s, and was completely salted in with KCl beyond $0.35{\mu}$. The intrinsic viscosity at $25^{\circ}C$ was estimated at 3.1. The paramyosin is rich in several amino acids such as arginine, aspartic acid, glutamic acid, etc., and lacking of both proline and tryptophane, in rough accord with other paramyosins reported. The abalone paramyosin did not show ATPase activity over a pH range of 5 to 9,5 even in the presence of Ca++ or Mg++. So was the case with the paramyosin specimen prepared by BAILEY's wet-extraction method.

Effects of Health Practices and Oral Health Behaviors on Obesity in University Students (일부 대학생의 건강 실천행위 및 구강건강행위가 비만에 미치는 영향)

  • This study was to identify the obesity(BMI), health practices and oral health behaviors of university students and provide basic data for the development of a health promotion program for healthy campus communities. Total 177 students filled out a self-administered questionnaire. Data were analyzed for descriptive statistics, chi-square test and multivariate logistic regression to evaluate the effects of health practice and oral behaviors on obesity. This study shows that the prevalence of obesity was 27.7%. Univarate analysis demonstrated that gender, age, the preference of salt, sleep in every day, self-perceived general health, the visiting the dentist during the past year were significantly different according to BMI. In multivariable regression, preference of salt, sleep in every day, the visiting the dentist during the past year were independent predictors of BMI, after adjustment of gender and age. The findings of this study emphasize the importance of obesity prevention program in overall, eating habit, mental health and oral health behavior in students.

A Study on the Level of Perception of the Health Condition of Koreans and Americans in Virginia, USA and Their Preference for Salty Taste (미국 버지니아주 거주 한국인과 미국인의 건강상태 인식도 및 짠맛 기호도 비교)

  • This study was conducted as a comparative analysis, using the SPSS 17.0 program for a survey conducted from March to July 2013, on the level of perception and preference factors of Koreans and Americans residing in Virginia, USA(89 male and female adults of 20 years of age or above). For the information on the degree of obesity, a survey was used and the subjects provided with their heights and weights. The researcher used this data to calculated their BMI and distinguished the degrees of obesity. For the BMI, the obesity diagnosis of Korean Society for the Study of Obesity was used for Koreans, and the WHO's obesity diagnosis on adults were employed for Americans. First, regarding the Korean participants, the level of obesity appeared in the order of normal (79.6%), overweight (14.3%), and underweight (6.1%). Also, the level of perceptions of their health conditions appeared in the order of average (51.7%) and good (34.7%). No one perceived their health to be in poor condition. Meanwhile, for the American participants, the order was as follows: overweight at about 45%, normal weight at about 35%, obesity at about 12.5%, and underweight at about 7.5%. In terms of the level of perception of health during ordinary times, about 55% of the American participants answered that they are in good health. Therefore, the Americans were found to perceive their health to be in relatively better condition than the Koreans residing in the USA. Also, concerning the level of interest in health while eating food at ordinary times, the American participants were discovered to be more interested (about 62.5%) than the Korean participants. The research on their preference for salty taste was conducted as a survey, and the subjects were asked to choose an item according to their subjective feeling. Regarding the preference for salty taste, both Koreans and Americans residing in the USA responded as moderate. However, a higher proportion of Americans than Koreans responded to favor salty taste. Concerning the reason for their preference for salty taste, Koreans responded with various reasons whereas Americans mostly responded that it is because they eat salty food frequently. For any changes in dietary behavior related to eating salty food, both Korean and American respondents thought that the amount of their daily intake of salt is appropriate and that they consider taste as most important in selecting a snack. Based on the above data, the results of this research indicate a need for actual improvement in the diet with regard to salty food along with diverse policies for promoting life with a low salt diet in future.

The Identification and Physiological Properties of Lactobacillus plantarum JK-01 Isolated from Kimchi (김치로부터 분리한 Lactobacillus plantarum JK-01의 동정 및 생리적 특성)

  • In order to identify probiotic microorganisms, 25 isolates of Lactobacillus sp. were selected from kimchi based on their growth rates, lactic acid production and salt tolerance. The isolate JK-01 was identified as Lactobacillus plantarum by the API kit and 16S rDNA analysis (99.9% of homology), and named as L. plantarum JK-01. The maximum number of L. plantarum JK-01 was reached at 18 hr fermentation in MRS broth and the pH gradually decreased to 4.5. L. plantarum JK-01 showed high enzyme activities for xylanase, amylase, protease, and phytase on MRS agar plates containing each substrate. L. plantarum JK-01 showed high resistance to acidic pH and bile salts, and grew well even at pH 2.0 and 1.0% bile salt. In particular, L. plantarum JK-01 showed high heat stability as shown by $3.3{\times}10^3$ CFU/mL at $60^{\circ}C$. The isolate showed remarkable antimicrobial activity against E. coli in MRS broth based on its disappearance after 18 hr and clear zone formation using a paper disk assay. These results suggest that L. plantarum JK-01 may be probiotic in nature.

Physical Properties Analysis for Automated Process Design of Solar Salts (천일염 자동화 공정설계를 위한 물리적특성 분석)

  • In this paper, the particle size, bulk density, friction coefficient and angle of repose were measured for the automatic processing of solar salt, which was classified with 5 levels of moisture content (16.5, 13.2, 8.2, 4.9, 3.7%). Average diameter was 1.98~1.60mm according to moisture content, and bulk density increased with increasing moisture content. As a result of measuring the coefficient of dynamic friction, the maximum value was shown in the stainless steel plate according to the material, and ABS plate and acrylic plate were lower in the order. Movement occurred at the moisture content of 8.2% or less, and the empty angle of repose was increased and the filling angle of repose was decreased as the moisture content increased.
